Acute Toxicity Studies of Octachlorostyrenes for Environment Organisms (Octachlorostyrenes의 환경생물에 대한 급성독성 연구)

  • Octachlorostyrene (OCS) is a persistant and bioaccumulative toxic subtance (PBTs). In this study, acute toxicity tests on algae, daphnia and fish for octachlorostyrene and its isomers were done to determine effective concentration ($EC_{50}$), Lethal concentration ($LC_{50}$), no observed effect concentration (NOEC) or lowest observed effect concentration (LOEC). As a result, NOEC on algae growth inhibition test for octacholorostyrene and 2-, 3-chlorostyrene was determined as $0.50\;mg\;L^{-1}$, and NOEC for 4-chlorostyrene was determined as $0.13\;mg\;L^{-1}$. NOEC on daphnia, acute immobilisation test for octachlorostyrene and 2-, 3-chlorostyrene was determined as $5.00\;mg\;L^{-1}$ and $EC_{50}$ for 4-chlorostyrene was determined as $2.128\;mg\;L^{-1}$. NOEC on Oryzias Latipes, acute toxicity test for octachlorostyrene was determined as $80.0\;mg\;L^{-1}$ and NOEC for 2-, 3-chlorostyrene was determined as $60.0\;mg\;L^{-1}$. $LC_{50}$ for 4-chlorostyrene was determined as $39.0\;mg\;L^{-1}$ (48h) and $22.6\;mg\;L^{-1}$ (96h).

  • Objectives : This study was aimed to examine the acute toxicity assessment of two new algicides, thiazolidinediones derivatives (TD53 and TD49), which were synthesized to selectively control red tide, to the marine ecosystem. Methods : The assessment employed by a new method using Ulva pertusa Kjellman which has been recently accepted as a standard method of ISO. The toxicity was assessed by calculating the $EC_{50}$ (Effective Concentration of 50%), NOEC (No Observed Effect Concentration) and PNEC (Predicted No Effect Concentration) using acute toxicity data obtained from exposure experiments. $EC_{50}$ value of TD49 and TD53 was examined by 96-hrs exposure together with Solutol as a TD49 dispersing agent and DMSO as a TD53 solvent. Results : $EC_{50}$ value of TD53 was $1.65\;{\mu}M$. From the results, values of NOEC and PNEC were calculated to be $0.63\;{\mu}M$ and 1.65 nM, respectively. DMSO under the range of $0{\sim}10\;{\mu}M$, which is same solvent concentration used in examining TD53, showed no toxic effect. $EC_{50}$ value of TD49 was $0.18\;{\mu}M$ and that of Solutol was $1.70\;{\mu}M$. NOEC and PNEC of TD49 were $0.08\;{\mu}M$ and 0.18 nM, respectively and those for Solutol were $1.25\;{\mu}M$ and 1.25 nM, respectively. Conclusions : From the values of NOEC, PNEC of TD53 and TD49, TD49 showed 9 times stronger toxicity than TD53. On the other hand, DMSO showed no toxicity on the Ulva pertusa Kjellman, but Solutol was found to be a considerable toxicity by itself.

The Acute and Chronic Toxicity Studies of Herbicide, Molinate to Waterfleas (Molinate의 물벼룩에 대한 급성 및 만성독성 연구)

  • To assess the impact of molinate on freshwater aquatic organisms, acute and chronic toxicity studies for waterfleas were conducted. In acute toxicity studies for Daphnia magna, and Moina macrocopa, the 48-h $EC_{50}$ values were 11.4 and 8.3 mg/L respectively. And in reproduction toxicity studies for the same species, the NOEC's were 2.5 and 2.0 mg/L respectively. These results suggest that waterfleas have simillar sensitivity to molinate. On the other hand, the NOEC for 3-generation toxicity of moina macrocopa 0.16 mg/L, was much lower than those of acute values. This studies concludes that molinate has minimal risk to waterfleas in river.

Characteristics of Bacteriophage Isolates and Expression of Shiga Toxin Genes Transferred to Non Shiga Toxin-Producing E. coli by Transduction

  • A risk analysis of Shiga toxin (Stx)-encoding bacteriophage was carried out by confirming the transduction phage to non-Stx-producing Escherichia coli (STEC) and subsequent expression of the Shiga toxin genes. The virulence factor stx1 was identified in five phages, and both stx1 and stx2 were found in four phages from a total of 19 phage isolates with seven non-O157 STEC strains. The four phages, designated as ϕNOEC41, ϕNOEC46, ϕNOEC47, and ϕNOEC49, belonged morphologically to the Myoviridae family. The stabilities of these phages to temperature, pH, ethanol, and NaClO were high with some variabilities among the phages. The infection of five non-STEC strains by nine Stx-encoding phages occurred at a rate of approximately 40%. Non-STEC strains were transduced by Stx-encoding phage to become lysogenic strains, and seven convertant strains had stx1 and/or stx2 genes. Only the stx1 gene was transferred to the receptor strains without any deletion. Gene expression of a convertant having both stx1 and stx2 genes was confirmed to be up to 32 times higher for Stx1 in 6% NaCl osmotic media and twice for Stx2 in 4% NaCl media, compared with expression in low-salt environments. Therefore, a new risk might arise from the transfer of pathogenic genes from Stx-encoding phages to otherwise harmless hosts. Without adequate sterilization of food exposed to various environments, there is a possibility that the toxicity of the phages might increase.

Toxic Effects of Heavy Metal (Pb, Cr, As) on the Hatching Rates of Fertilized Eggs in the Olive Flounder (Paralichthys olivaceus) (중금속(Pb, Cr, As)이 넙치(Paralichthys olivaceus) 수정란 부화율에 미치는 독성 영향)

  • Toxic effects of heavy metal (Pb, Cr, As) were examined by the hatching rates of fertilized eggs in the oliver flounder, Paralichthys olivaceus. Eggs were exposed to Pb, Cr, As (0, 10, 100, 500, 1,000, 2,500, 5,000 ppb) and then normal hatching rates were investigated after 48 h. The normal hatching rates in the control condition (not including heavy metal) were greater than 80%, but suddenly decreased with increasing of heavy metal concentrations. Pb, Cr and As reduced the normal hatching rates in concentration-dependent way and a significant reduction occurred at concentration grater than 100, 100, 500 ppb, respectively. The ranking of heavy metal toxicity was Cr>As>Pb, with $EC_{50}$ values of 415, 518 and 1,029 ppb, respectively. The no-observed-effect-concentration (NOEC) and lowest-observed-effect-concentration (LOEC) show each 100 bbp and 500 ppb of normal hatching rates in exposed to Pb and As. The NOEC and LOEC of normal hatching rates in Cr were 10 ppb and 100 ppb, respectively. From these results, the normal hatching rates of P. olivaceus have toxic effect at greater than the 100 ppb concentrations in Pb, As and the 10 ppb concentrations in Cr in natural ecosystems. These results suggest that biological assay using the normal hatching rates of P. olivaceus are very useful test method for the toxicity assessment of a toxic substance as heavy metal in marine ecosystems.

  • In this study, TI(Toxic Index) of Daphnia toximeter corresponded to ecological toxicity standard 1 TU(Toxic Unit) was set up using Daphnia toximeter and when operating NOEC(water quality standards for drinking water) and $EC_{50}$ Daphnia toximeter alarm was issued appropriately, which enables real time ecological toxicity evaluation. I studied to get a good shot and the research was conducted by investigating domestic and international related data and conducting a preliminary study. 6 of 59 hazardous substances (As, Hg, Cr, Diazinon, Dioxane, and Phenol) recommended by the water quality monitoring items for artificial river water were selected and static, dynamic and quality management test, TI was shown to be good in other materials except Diazinon, and as a result of $EC_{50}$ spiking test, TI was matched to TU by distinguishing between 1 TU and 1 TU. in suggesting the complementary point of ecological toxicity management system and the future of research on water Daphnia toximeter.

Biological Study on the Increment of Survival Rate during Early Life Cycle in th Rockfish, Sebastes schlegeli (Telostei: Scorpaenidae) - I.   • The effect of ammonia on survival and growth of the larval rockfish, Sebastes schlegeli was examined by a static renewal bioassay method. The tolerance of larvae to ammonia toxicity was more sensitive at the early larvae, but increased with the development of larvae. In 14 day-old-larvae after parturition, the mortality with treatment of each concentration of ammonia was 5% at control group and 0.0112 $NH_3$mg/l, whereas it was increased up to 27.5% at exposure group of 0.1230 $NH_3$mg/l with higher concentration. Regression equation between ammonium concentration(X) and mortality(Y) was followed; Y=0.516+3.482 X($r^2=0.4737$, P<0.01). The NOEC(no-observable-effect concentration) and LOEC (lowest-observable-effect concentration) to mortality compared to control group were 0.100 $NH_3mg/l$ and 0.1230 $NH_3$mg/l, respectively and chronic value(ChV) which is the geometric mean of the NOEC and LOEC was 0.1110 $NH_3$mg/l. Body length after 7-days exposure in control group, 0.0112 $NH_3$mg/l and 0.1230 $NH_3$mg/l were 7.8325mm, 7.700mm and 7.05mm, respectively. The NOEC, LOEC and chronic value(ChV) were 0.0335 $NH_3$mg/l, 0.0558 $NH_3$mg/l and 0.0432 $NH_3$mg/l, respectively.

Toxicity Assessment of Heavy Metals (As, Cr and Pb) Using the Rates of Survival and Population Growth in Marine Rotifer, Brachionus plicatilis (해산로티퍼 (Brachionus plicatilis)의 생존 및 개체군 성장률을 이용한 중금속 (As, Cr, Pb) 독성평가)

  • Toxicity assessment of heavy metals (As, Cr and Pb) has been investigated by using the rate of survival and population growth(r) of marine rotifer, Brachionus plicatilis. The survival rate was determined after 24 hours of exposure to As, Cr and Pb. As and Cr reduced survival rate in dose-dependent manner and a significant reduction were occurred at concentration of greater than 30 and $150mg\;L^{-1}$, but Pb had no effect on survival rate. The r was determined after 72 hours of exposure to As, Cr and Pb. As, Cr and Pb reduced r in dose-dependent manner and a significant reduction were occurred at concentration of greater than 5, 25 and $50mg\;L^{-1}$. The toxicity of heavy metals were ranked As>Cr>Pb, with $EC_{50}$ values of 12.98, 82.34 and $110.14mg\;L^{-1}$, respectively. The no-observed-effect-concentration (NOEC) of r in As, Cr and Pb exposure were 1, 12.5 and $50mg\;L^{-1}$, respectively. The lowest-observed-effect-concentration (LOEC) of r in As, Cr and Pb exposure were 5, 25, and $50mg\;L^{-1}$, respectively. From the results, the concentration of As, Cr and Pb (greater than 5, 25 and $50mg\;L^{-1}$, respectively) have toxic effect on the r of B. plicatilis in natural ecosystems. These results (including NOEC and $EC_{50}$) might be useful for the mixing toxicity assessment and toxic guide line of heavy metals in marine ecosystems.

Effects of Ammina on Survival and Growth of the Flounder Larva, Paralichthys olivaceus (넙치, Paralichthys olivaceus 자어의 생존과 성장에 미치는 암모니아의 영향)

  • The effects of ammonia on survival and growth of the flounder larva, Paralichthys olivaceus, were examined by a static renewal bioassay method. The $96\;hr-LC_{50}$ with the developmental stages during the period from 1day to 23 day-old larvae ranged 0.273 to 1.023 mg $NH_3/\ell$. Tolerance of the larvae to ammonia toxicity was much sensitive at the early larval stage, and increased with the growth of the larvae. Threshold $96hr-LC_{50}$ in 1, 3 and S day-old larvae after hatching were 0.293, 0.248 and 0.379 mg $NH_3/\ell$, respectively. Survival rate and growth in body weight and body weight of the larva were reduced with increase of ammonia concentration in the range of 0.055 and 0.341 mg $NH_3/\ell$. The no-observable-effect concentration (NOEC) and lowest-observable- effect concentration (LOEC) of the flounder larve were 0.102 and 0.174 mg $NH_3/\ell$ for body length, and 0.151 and 0.198 mg $NH_3/\ell$ for body weight, respectively. Chronic value (ChV), which is the geometric mean of the NOEC and $NH_3/\ell$ to body length of the larvae were 0.124 mg $NH_3/\ell$. The coefficient of variation (CV) for body length was higher at high concentration than at low concentration.
