고리형 알콕시기를 함유한 크리신 유도체의 합성 및 일산화질소 생성 억제작용 (Synthesis of 8-Cycloalkoxychrysin Analogs and their Inhibitory Activity Against NO Production)

  • Several 8-cycloalkoxychrysin analogs were synthesized from 6-benzyloxy-5,8-dihydroxyflavone and cycloalkanols in 2 steps and we evaluated their inhibitory activities against NO production from LPS-induced RAW 264.7 cells. Among tested analogs, two analogs with cyclopentyl substructure (796 and 798) showed strong inhibitory activity against NO production.

  • Objectives : The objective of this study is to study the effects of Euonymi Lignum Suberatatum pharmacopuncture solution on NO and IL-6 production in macrophage. Methods : At first, the RAW 264.7 macrophage was subclutured. In order to evaluate cytotoxicity, MTT assay performed. Then, the cell was induced by LPS, INF-$\gamma$ and Experimental groups were divided into five(Normal, Control, Euonymi Lignum Suberatatum 100, 200, $300{\mu}g/m{\ell}$). Then Euonymi Lignum Suberatatum pharmacopuncture solution was put into cell. We measured IL-6, iNOS, NO. Results : The cytotoxic effect of Euonymi Lignum Suberatatum pharmacopuncture solution in RAW 264.7 macrophage was not appeared. $300{\mu}g/m\ell$ Euonymi Lignum Suberatatum pharmacopuncture solution inhibited IL-6 production in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages significantly. Euonymi Lignum Suberatatum pharmacopuncture solution inhibited iNOS revelation in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages. All group of Euonymi Lignum Suberatatum pharmacopuncture solution inhibited NO production in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages significantly. Conclusions : Our study demonstrated that Euonymi Lignum Suberatatum pharmacopuncture solution had an inhibition effect on NO production, iNOS revelation, IL-6 production. So Euonymi Lignum Suberatatum pharmaco puncture solution may have an Anti-inflammation effect.

Augmentation of Macrophage Antitumor Activities and Nitric Oxide Production by Oregonin

  • Oregonin, a diarylheptanoid derivative from Alnus hirsuta Turcz, Betulaceae, was evaluated for its antitumor activity. Oregonin, known to have an antitumor function, and is a novel immunomodulator, which may augment macrophage activity. MTT assays and NO production tests were performed in order to investigate the cytotoxicity of oregonin in tumor cells and to examine its influence on macrophage in detail. In this study, the tumoricidal activity was also evaluated by a MTT assay. The cytotoxicity measurements in the oregon in-treated group both in vitro and in vivo showed a significant difference from that of the control group. In vivo, oregonin significantly increased NO production in a dose-dependent manner, and in vitro, the thioglycolate-induced inflammatory macrophages increased NO production in a dose-dependent manner after incubation. These results suggest that oregonin reacts with both the inflammatory and non-inflammatory macrophages in a similar way.

다양한 촉매들을 통한 모델 바이오매스-초임계수 촉매 가스화에서 수소 생산 성능에 대한 연구 (The Study on of Hydrogen Production Performance by Model Biomass-supercritical Water Gasification with Various Catalysts)

  • In this study, the model biomass was used for hydrogen production by supercritical water gasification (SCWG). Model biomasses were glycerol, glycine, lignin and cellulose. The feed concentration was set to 1 wt%. Experiments were conducted in a reactor at $440^{\circ}C$ and above 26.3 MPa for 30 min. The effects of catalysts such as alkali metal salt ($K_2CO_3$ and $Na_2CO_3$) and transition metal salts ($Ni(NO_3)_2$, $Fe(NO_3)_3$ and $Mn(NO_3)_2$) on the gasification were systematically investigated. No tar or coke was observed in all experiments. The results showed that the gasification efficiency increased with various catalysts. For the cellulose and glycerol, all catalysts were effective for the promoted $H_2$ production compared with no catalyst. The significant decrease of $H_2$ production compared with no catalyst was observed with $Na_2CO_3$ and $Fe(NO_3)_3$ for glycine and lignin. respectively. The highest H2 production, 1.24 mmol was obtained for glycerol-SCWG with $Mn(NO_3)_2$. Conclusively, the addition of $Mn(NO_3)_2$ enhanced all model biomass gasification efficiency and increased the hydrogen production promoting the supercritical water reaction.

당귀약침액이 대식세포에서 산화질소(NO) 및 프로스타글란딘 (Prostaglandin) 생성에 미치는 영향 (Effects of Angelicae Gigantis Radix pharmacopuncture on Nitric Oxide and Prostaglandin E2 Production in Macrophage)

  • Objectives : Angelicae Gigantis Radix has been known traditional medicine with antimicrobial activities and it has been widely used for treatment of blood and inflammatory diseases. In the present study, some studies examined anti-inflammation effects of Angelicae Gigantis Radix but they usually were performed by ethanol extracted Angelicae Gigantis Radix pharmacopuncture. So We investigated the inhibitory effects of Angelicae Gigantis Radix pharmacopuncture by hot water and ethanol extract on Nitric oxide(NO) and Prostaglandin $E_2$($PGE_2$) production in lipopolysaccharide(LPS) induced macrophage cell. Methods : Angelicae Gigantis Radix was extracted by ethanol and hot water. Cell viability was determined by MTT assay. To evaluate anti-inflammation effects of Angelicae Gigantis Radix pharmacopuncture, we examined NO and $PGE_2$ production in LPS induced macrophages. The concentrations of NO and $PGE_2$ were measured by Griess assay and Enzyme Immuno-Assay. Results : 1) The MTT assay demonstrated that cytotoxic effect of Angelicae Gigantis Radix pharmacopuncture by hot water extract and ethanol extract in RAW 264.7 macrophage cells were not appeared. 2) Angelicae Gigantis Radix pharmacopuncture by ethanol extract and hot water extract inhibited NO production in LPS induced macrophages significantly. 3) Angelicae Gigantis Radix pharmacopuncture by ethanol extract tended to inhibiting $PGE_2$ production in LPS induced macrophages. And Angelicae Gigantis Radix pharmacopuncture by hot water extract inhibited LPS induced production of $PGE_2$ in RAW 264.7 macrophage cells significantly. Conclusions : This study suggests that Angelicae Gigantis Radix pharmacopuncture may have an anti-inflammatory property through the inhibition of NO and $PGE_2$ production in LPS induced macrophages. It may have a therapeutic potential for the treatment of various inflammatory diseases.

Emission of NO2 Gas Causing Damage to Plants in an Acid Soil under Conditions Favorable for Denitrification

  • Nitrogen dioxide ($NO_2$) gas damage on vegetable crops commonly occurs in plastic film houses where relatively large amounts of $NO_3{^-}$ are applied in acid soils. In acid soils, $HNO_2$ can be formed from the $NO_2{^-}$ accumulated during denitrification, and $NO_2$ can be evolved from the chemical self-decomposition of $HNO_2$. In this study, $NO_2$ gas production and its detrimental effects on plants were investigated in soils of various conditions to elucidate the mechanisms involved in the gas production. A silty loam soil was amended with $NO_3{^-}$ (500 mg N $kg^{-1}$) and glucose, and pH and moisture of the soil were adjusted respectively to 5.0 and 34.6% water holding capacity (WHC) with 0.01 M phosphate buffer. The soil was placed in a 0.5-L glass jar with strawberry leaf or $NO_2$ gas absorption badge in air space of the jar, and the jar was incubated at $30^{\circ}C$. After 4-5 days of incubation, dark burning was observed along the outside edge of strawberry leaf and $NO_2$ production was confirmed in the air space of jar. However, when the soil was sterilized, $NO_2$ emission was minimal and any visible damage was not found in strawberry leaf. In the soil where water or $NO_3{^-}$ content was reduced to 17.3% WHC or 250 mg N $kg^{-1}$, $NO_2$ production was greatly reduced and toxicity symptom was not found in strawberry leaf. Also in the soil where glucose was not amended, $NO_2$ production was significantly reduced. In soil with pH of 6.5, $NO_2$ was evolved to the level causing damage to strawberry leaf when the soil conditions were favorable for denitrification. However, compared to the soil of pH 5.0, the $NO_2$ production and its damage to plants were much less serious in pH 6.5. Therefore, the production of $NO_2$ damaging plants might be occurred in acid soils when the conditions are favorable for denitrification.

High molecular weight water-soluble chitosan acts as an accelerator of macrophages activation by recombinant interferon ${\gamma}$ via a process involving $_L$-arginine -dependent nitric oxide production

  • High molecular weight water-insoluble chitosan alone has been previously shown to exhibit in vitro stimulatory effect on macrophages nitric oxide (NO) production. However, high molecular weight water-soluble chitosan (WSC) had no effect on NO production by itself. When WSC was used in combination with recombinant $interferon-{\gamma}\;(Rifn-{\gamma})$, there was a marked cooperative induction of NO synthesis in a dose-dependent manner. The optimal effect of WSC on NO synthesis was shown at 24 h after treatment with $rIFN-{\gamma}$. The increased production of NO from $rIFN-{\gamma}$ plus WSC-stimulated RAW 264.7 macrophages was decreased by the treatment with $N^G$ $monomethyl-_L-arginine$. The increase in NO synthesis was reflected, as an increased amounts of inducible NO synthase (iNOS) protein. Synergy between $rIFN-{\gamma}$ and WSC was mainly dependent on WSC-induced nuclear $factor-_KB$ activation. The present results indicate that WSC may provide various activities such as anti-microbial, anti-tumoral, and anti-viral. In addition, since NO has emerged as an important intracellular and intercellular regulatory molecule having functions as diverse as vasodilation, neural communication, cell growth regulation and host defense, it is tempting to hypothesize that this WSC is involved in the local control of the various fundamental processes such as cardiagra, cardiac infarction, impotence etc.

보음면역단의 면역 증강 효과 (Immune Enhancing Effect of Boummyunyuck-dan)

  • Objective : To investigate immune enhancing effects of Boummyunyuck-dan (BMD) Methods : In this study I investigated the effect of BMD on cell proliferation and viability. In addition, I investigated production of cytokines (IL-2, IL-4 and $IFN-{\gamma}$), NO, and $TNF-{\alpha}$ in human T-cell leukemia, MOLT-4 cells. The cells were cultured for 24h in the presence or absence of BMD. Result : BMD increased the cell viability by 15% (P<0.05) and enhanced IL-2, IL-4 and $IFN-{\gamma}$ production compared with media control in a dose-dependent manner (P<0.01) at 24h. BMD also increased mRNA and protein expression levels of $IFN-{\gamma}$ in MOLT-4 cells. In addition, I also assessed the effects of BMD on production of NO and $TNF-{\alpha}$ from the peritoneal macrophages because NO and $TNF-{\alpha}$ as a potent macrophage-derived immune reaction regulatory molecule has received increasing attention. However, BMD had no effect on NO and $TNF-{\alpha}$ production in the cells. Conclusion : These data indicate that BMD has some immune-enhancing effect, and that its action may be due to the proliferation and cytokine production of T cells.

Activation of ATM/Akt/CREB/eNOS Signaling Axis by Aphidicolin Increases NO Production and Vessel Relaxation in Endothelial Cells and Rat Aortas

  • Although DNA damage responses (DDRs) are reported to be involved in nitric oxide (NO) production in response to genotoxic stresses, the precise mechanism of DDR-mediated NO production has not been fully understood. Using a genotoxic agent aphidicolin, we investigated how DDRs regulate NO production in bovine aortic endothelial cells. Prolonged (over 24 h) treatment with aphidicolin increased NO production and endothelial NO synthase (eNOS) protein expression, which was accompanied by increased eNOS dimer/monomer ratio, tetrahydrobiopterin levels, and eNOS mRNA expression. A promoter assay using 5'-serially deleted eNOS promoters revealed that Tax-responsive element site, located at -962 to -873 of the eNOS promoter, was responsible for aphidicolin-stimulated eNOS gene expression. Aphidicolin increased CREB activity and ectopic expression of dominant-negative inhibitor of CREB, A-CREB, repressed the stimulatory effects of aphidicolin on eNOS gene expression and its promoter activity. Co-treatment with LY294002 decreased the aphidicolin-stimulated increase in p-CREB-Ser133 level, eNOS expression, and NO production. Furthermore, ectopic expression of dominant-negative Akt construct attenuated aphidicolin-stimulated NO production. Aphidicolin increased p-ATM-Ser1981 and the knockdown of ATM using siRNA attenuated all stimulatory effects of aphidicolin on p-Akt-Ser473, p-CREB-Ser133, eNOS expression, and NO production. Additionally, these stimulatory effects of aphidicolin were similarly observed in human umbilical vein endothelial cells. Lastly, aphidicolin increased acetylcholine-induced vessel relaxation in rat aortas, which was accompanied by increased p-ATM-Ser1981, p-Akt-Ser473, p-CREB-Ser133, and eNOS expression. In conclusion, our results demonstrate that in response to aphidicolin, activation of ATM/Akt/CREB/eNOS signaling cascade mediates increase of NO production and vessel relaxation in endothelial cells and rat aortas.

Thieny/Furanyl-hydroxyphenylpropenones as Inhibitors of LPS-induced ROS and NO Production in RAW 264.7 Macrophages, and Their Structure-Activity Relationship Study

  • Twelve thienyl/furanyl-hydroxyphenylpropenones were systematically designed and synthesized, and evaluated for their inhibitory effect on LPS-induced ROS and NO production in RAW 264.7 macrophages. Compound 11 displayed the most significant inhibitory activity of LPS-induced ROS and NO production in RAW 264.7 macrophages. Structure-activity relationship study indicated that para-hydroxyphenyl moiety plays an important role for inhibitory activities on both LPS-induced ROS and NO production as well as 3-thienyl moiety on molecule.
