Effect of Glutathione on Lead Induced Modulation of NO Synthesis in RAW 264.7 Cell (RAW 264.7 Cell에서 납에 의한 NO 생성의 조절에 미치는 Glutathione의 효과)

  • Objectives : To evaluate the elect of glutathione(GSH) on lead induced modulation of nitric oxide(NO) synthesis, and to examine how lead modulates NO production in macrophages. Methods : This study was observed in a culture of RAW 264.7 cells, which originated from a tumor in a Balb/c mouse that was induced by the Abelson murine leukemia virus. The compounds investigated were lead chloride, N-acetyl-cystein(NAC), and Buthionine Sulfoximine( BSO). Results : ATP synthesis in RAW 264.7 cells was unchanged by each lead concentration exposure in a dose dependent manner. The NO synthesis was decreased when exposed to lead($PbCl_2$) concentration $0.5{\mu}M$. The presence of $300{\mu}M$ NAC, used as a pretreatment in the culture medium, caused the recovery of the lead induced decrease in NO synthesis, but in the presence of $300{\mu}M$ BSO as a pretreatment, there was no recoverey. Pretreatment with NAC and BSO had no affect on ATP synthesis at any of the lead concentrations used. Conclusions : These results indicated that GSH has a protective effect toward lead toxicity, and suggested that the inhibition of NO production in macrophage due to lead toxicity may be related to cofactors of iNOS (inducible nitric oxide synthase)

Inhibitory effect of Petalonia binghamiae on neuroinflammation in LPS-stimulated microglial cells (LPS에 의해 활성화된 미세아교세포에서 미역쇠 추출물의 신경염증 보호 효과)

  • Purpose: Neuroinflammation is mediated by activation of microglia implicated in the pathogenesis of neurodegenerative disorders such as Alzheimer's disease and Parkinson's disease. Inhibition of neuroinflammation may be an effective solution to treat these brain disorders. Petalonia binghamiae is known as a traditional food, based on multiple biological activities such as anti-oxidant and anti-obesity. In present study, the anti-neuroinflammatory potential of Petalonia binghamiae was investigated in LPS-stimulated BV2 microglial cells. Methods: Cell viability was measured by MTT assay. Production of nitric oxide (NO) was examined using Griess reagent. Expression of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) was detected by Western blot analysis. Activation of nuclear factor ${\kappa}B$ ($NF-{\kappa}B$) signaling was examined by nuclear translocation of $NF-{\kappa}B$ p65 subunit and phosphorylation of $I{\kappa}B$. Results: Extract of Petalonia binghamiae significantly inhibited LPS-stimulated NO production and iNOS/COX-2 protein expression in a dose-dependent manner without cytotoxicity. Pretreatment with Petalonia binghamiae suppressed LPS-induced $NF-{\kappa}B$ p65 nuclear translocation and phosphorylation of $I{\kappa}B$. Co-treatment with Petalonia binghamiae and pyrrolidine duthiocarbamate (PDTC), an $NF-{\kappa}B$ inhibitor, reduced LPS-stimulated NO release compared to that in PB-treated or PDTC-treated cells. Conclusion: The present results indicate that extract of Petalonia binghamiae exerts anti-neuroinflammation activities, partly through inhibition of $NF-{\kappa}B$ signaling. These findings suggest that Petalonia binghamiae might have therapeutic potential in relation to neuroinflammation and neurodegenerative diseases.

A Study on Characteristics of NOx and Ozone by Plasma Reaction (Plasma반응에 의한 NOx와 Ozone의 특성에 관한 연구)

  • In this experiment, we made the plasma reactor which adhere needle electrode in order to treat safely an NOx which was included in the gas. Also we experimently investigated characteristics of equipment and inspected efficiency. As a reaction gas, by using mixture gas of $NO/N_2$ and $N_2/O_2$, we setted up initial NO concentration and gas flow rate was set at 2 ${\iota}$/min. As a reaction characteristics of NOx, when discharge input power was high, NO concentration decreased and when the oxygen concentration increased, the NO decomposition was easy and decomposition energy efficiency was high. Also in case that NO concentration increased, NO decomposition energy efficiency was high but decomposition rate was low. The characteristics of ozone, when discharge input power was high, ozone increased and when $NO/N_2$ concentration increased, the ozone decreased.

Roles of Key Elementary Reaction for NO Formation in Premixed Flame and Counterflow Diffusion Flame (예혼합 및 대향류확산 화염에서 NO의 생성에 미치는 소반응의 역할)

  • In this paper it is investigated the roles of key elementary reactions for NO formation in methane-air one-dimensional premixed flame and counterflow diffusion flame, which were studied numerically by using NO kinetics and $C_{2}$ -chemistry complied by Miller and Bowman. The spatial distributions of the reaction rates of 9 main elementary reactions directly related to NO formation and destruction were calculated. Integration of the rates of all reactions in the NO formation across the flame yields the quantitative reaction path diagram, which shows clearly relative importance of each reaction path in NO formation and how it changes with the type and parameters of the flame. The results show that the thermal and Fenimore mechanisms are dominant respectively for learn and rich premixed flames, and the latter is dominant for diffusion flames. In addition, it was found that the HCN recycle route is important for diffusion flame, and that the routes of mutual transformation between NO and NO$^{2}$, and between NO and HNO do not contribute to the net NO formation.

  • 外治에 응용되는 蛇床子의 抗炎症效果를 살펴보고자 NO의 生成濃度, 細胞 生存度, iNOS의 합성 및 $TNF-{\alpha}$의 生成濃度를 實驗하였다. 그 結果 NO의 濃度는 顯著하게 減少하였고, 細胞 生存度는 蛇床子에 의하여 影響을 받지 않았으며, iNOS의 합성에 있어서 iNOS의 濃度가 顯著하게 낮아졌고, $TNF-{\alpha}$의 濃度는 顯著하게 減少하였다. 以上을 살펴볼 때 蛇床子가 iNOS 및 $TNF-{\alpha}$의 생성자체의 억제를 통하여 NO의 合性을 조절함으로써 蛇床子가 충분한 抗炎症效果가 있음을 알 수 있었다. 향후 蛇床子의 성분분석등 다양한 연구가 진행되어야 할 것으로 생각되며, 이를 통하여 앞으로 抗炎症效果가 우수한 약제의 개발이 가능할 것으로 思料된다.

Production of NO in Interacting Laminar Diffusion Flames (상호작용하의 층류 확산 화염에서 NO의 생성)

  • In order to find the effect of flame interaction on NO production, dual laminar diffusion flames issuing from two rectangular nozzles were investigated theoretically. Chemical equilibrium model and Zeldovich mechanism were used in numerical model. The effect of four major parameters on NO production were inspected. These parameters are nozzle spacing, Raynolds number, aspect ratio of nozle cross section and velocity of secondary flow. It is found that interaction of flames enhances production of n. It is also found that multiflames with large spacing, small aspect ratio and strong secondary flow product less n.

  • 목적 : 본 연구는 염증유발 내독소인 LPS 자극을 통한 면역 스트레스에 대한 약침의 치료효과와 기전을 알아보기 위해 설계되었다. LPS를 통한 염증반응은 다양한 기전을 통해 이루어지는 데 특히, 산화질소(NO)를 생성하는 효소인 iNOS의 활성은 시상하부에서 NO의 생성을 촉진함으로써 염증반응을 발현시키는 중요한 역할을 하는데 약침이 LPS로 인해 활성화된 iNOS의 발현에 어떠한 영향을 미치는지 알아보고자 하였다. 실험방법 : 흰쥐에 LPS를 투여하고 2시간 경과한 후 약침 (소염방, 대한약침학회)을 경혈 (합곡, 족삼리)에 좌,우 각각 0.2cc 씩 피하 투여하고 RT-PCR 방법을 통하여 면역활성에 중요한 역할을 하는 시상하부에서의 iNOS의 mRNA 발현을 알아 보았다. 실험결과 : LPS 투여로 인하여 증가하였던 iNOS mRNA가 약침치료를 통해 현저하게 감소하였다. 그러나 비경혈점 (미추부위 임의혈)에 약침을 투여한 군 경혈점에 생리식염수를 투여한 군에서는 iNOS mRNA 가 감소하지 않았다. 이는 약침이 LPS를 투여한 흰쥐의 시상하부에서 iNOS의 mRNA 발현에 조절작용을 가지고 있는 것을 보여주는 결과로 약침이 면역활성에 중요한 역할을 하는 NO의 생성을 억제함으로써 면역 스트레스에 치료효과를 나타내는 것으로 보여진다. 결론 : 본 연구는 약침치료가 면역반응의 중추기관인 시상하부에서 면역반응의 중요한 조절인자인 NO의 생성을 조절하는 기전을 통해 그 치료효과를 나타내는 것을 밝혔고 더 나아가 약침에 대한 현대과학적 연구에 중요한 발판을 마련했다는 점에서 그 의의를 가지고 있다고 사료된다.

Anti-Inflammatory Effect of Erigeron annuus L. Flower Extract through Heme Oxygenase-1 Induction in RAW264.7 Macrophages (RAW264.7 대식세포에서 Heme Oxygenase-1의 유도에 의한 개망초 (Erigeron annuus L.) 꽃 Methanol 추출물의 항염증 효과)

  • This study investigated the anti-inflammatory effect of Erigeron annuus L. flower (EAF) methanol extract. We examined the involvement of heme oxygenase-1 (HO-1) in the inhibitory activities of EAF methanol extract on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. Cell viability and NO assays were performed. In addition, inducible nitric oxide synthase (iNOS) and HO-1 expressions were detected by Western blotting and blocking HO-1 activity on NO production. EAF methanol extract (25, 50, 100, 200 ${\mu}g$/mL) significantly inhibited LPS-stimulated NO production (p<0.05; 12.82, 9.61, 6.83, 2.52 ${\mu}m$) in a concentration-dependent manner. EAF methanol extract also reduced the expression of iNOS protein. The EAF methanol extract induced the expression of HO-1 in a dose-dependent manner. Blockage of HO-1 activity by zinc protoporphyrin suppressed EAF methanol extract-induced reductions in the production of NO. The present results suggest that EAF methanol extract has a potent anti-inflammatory effect in RAW264.7 macrophages through the induction of HO-1.

Anti-Inflammatory Effects of Vegetable Soup in Murine Macrophage RAW 264.7 Cells (야채스프의 RAW 264.7 세포에서 항염증 효과)

  • The aim of the present study is to investigate the anti-inflammatory effect of a vegetable soup (VS). The present study was designed to determine the effect of the vegetable soup on pro-inflammatory factors such as NO, iNOS and TNF-$\alpha$ in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The cell toxicity was determined by MTS assay. To evaluate the anti-inflammatory effect of vegetable soup, amount of NO was measured using the NO detection kit and the iNOS expression was measured by reverse transcriptase polymerase chain reaction (RT-PCR). Also, proinflammatory cytokines were measured by ELISA kit. The results showed that the vegetable soup reduced NO, iNOS and TNF-$\alpha$ production without cytotoxicity. Our results suggest that the vegetable soup may have an anti-inflammatory property through suppressing inflammatory mediator productions and appears to be useful to develop the functional food realted to anti-inflammation.

Comparisons of Ginsenosides and Anti-inflammatory Effects of White Ginseng and Puffed Red Ginseng (인삼과 팽화홍삼의 Ginsenoside 함량 및 항염효과 비교)

  • In this study, the ginsenoside contents and anti-inflammatory effects of white ginseng (WG) and puffed red ginseng (PRG) were compared. The contents of Rb1, Rg5 and Rk1 were significantly higher in PRG than in WG, whereas the contents of Rg1 and Rb2 were decreased in PRG. The levels of NO production and iNOS expression were suppressed in LPS-stimulated cells by treatment with WG and PRG. Further, the production of cytokines (TNF-$\alpha$ and INF-$\gamma$) and inflammatory proteins (NF-${\kappa}B$ and COX-2) was decreased in cells upon treatment with any of the ginsenosides. The high NO inhibitory activity and cytokine production of PRG is caused by differences in the composition of ginsenosides produced.
