결장암에 대한 활성 자연살해세포의 항암효능 (Anticancer Effect of Activated Natural Killer Cells on Human Colorectal Tumor)

  • Colorectal cancer is one of the most common alimentary malignancies. In this study, the antitumor activity of activated human natural killer (NK) cells against human colorectal cancer was evaluated in vivo. Human NK cells are the key contributors of innate immune response and the effective functions of these cells are enhanced by cytokines. Human peripheral blood mononuclear cells (PBMC) were cultured with interleukin-2 (IL-2)-containing medium for 14 days and resulted in enriched NK cell population. The resulting populations of the cells comprised 7% $CD3^+CD4^+$ cells, 25% $CD3^+CD8^+$ cells, 13% $CD3^-CD8^+$ cells, 4% $CD3^+$CD16/$CD56^+$ cells, 39% $CD3^+$CD16/$CD56^-$ cells, and 52% $CD3^-$CD16/$CD56^+$ cells. Tumor necrosis factor alpha (TNF-$\alpha$), interferon gamma (IFN-$\gamma$), IL-2, IL-4, and IL-5 transcripts of the activated NK cells were confirmed by RT-PCR. In addition, activated NK cells at doses of 2.5, 5 and 10 million cells per mouse inhibited 10%, 34% and 47% of SW620-induced tumor growth in nude mouse xenograft assays, respectively. This study suggests that NK cell-based immunotherapy may be used as an adoptive immunotherapy for colorectal cancer patients.

Tissue-resident natural killer cells exacerbate tubulointerstitial fibrosis by activating transglutaminase 2 and syndecan-4 in a model of aristolochic acid-induced nephropathy

  • Despite reports suggesting that tissue-resident natural killer (trNK) cells cause ischemic kidney injury, their contribution to the development of tubulointerstitial fibrosis has not been determined. This study hypothesized that the depletion of trNK cells may ameliorate renal fibrosis by affecting transglutaminase 2/syndecan-4 interactions. Aristolochic acid nephropathy (AAN) was induced in C57BL/6 mice as an experimental model of kidney fibrosis. The mice were treated with anti-asialo GM1 (ASGM1) or anti-NK1.1 antibodies to deplete NK cells. Although both ASGM1 and NK1.1 antibodies suppressed renal $NKp46^+DX5^+$ NK cells, renal $NKp46^+DX5^-$ cells were resistant to suppression by ASGM1 or NK1.1 antibodies during the development of tubulointerstitial fibrosis in the AAN-induced mouse model. Western blot analysis showed that both antibodies increased the expression of fibronectin, transglutaminase 2, and syndecan-4. These findings indicate that trNK cells played an exacerbating role in tubulointerstitial fibrosis by activating transglutaminase 2 and syndecan-4 in the AAN-induced mouse model.

  • 본 논문은 홍삼 산성 다당류 (RGAP)의 생체외 쥣과 NK 세포 활성 향상에 대한 내용으로 주요 내용은 다음과 같다. RGAP는 Yac-1 종양 세포에서 NK 세포 활성을 향상시켰다. 비장 세포에 16시간 동안 RGAP 10 및 $100{\mu}g$/ml을 처리한 결과, NK 활성이 239와 250% 증가 하였다. 인터페론 (IFN)-$\gamma$ 생성은 17-125% 증가하였고, 종양 괴사 인자 (TNF)-${\alpha}$ 생성도 15-100% 증가 하였다. NK 세포의 세포 독성 증가는 NK 세포가 생성하는 이 두 시토카인 증가가 원인이다. 따라서 RGAP는 NK 세포에 미치는 면역증진 효과가 있다는 내용이다.

k-ras와 c-myc, wnt 억제에 의한 NKG2D 리간드의 발현변화 (Inhibition of Oncogenes Affects the Expression of NKG2D Ligands in Cancer Cells)

  • 자연살상세포(NK cells)은 림프구계의 세포로서 외부 침임 병원균을 막고 체내 형질변환세포를 제거하는데 참여하고 있다. 이러한 자연살상세포의 활성은 특정한 항원이 필요 없고 활성화 신호와 억제성 신호의 균형에 의해 조절되고 있다. 자연살상세포의 중요한 활성화 신호 중의 하나는 NKG2D 수용체를 통한 것인데, 이 NKG2D 수용체를 통해 자연살상세포는 암세포에 있는 NKG2D 리간드를 인식할 수 있다. 지금까지 인간에서는 여덟개의 NKG2D 리간드가 밝혀져 있고 이러한 리간드의 발현은 다양한 기전을 엄격하게 조절되고 있다. 암세포는 암유전자(oncogenes)에 의해 세포내 다양한 유전자의 발현이 정상세포와 확연히 달라지는데, 이러한 암유전자에 의해서 NKG2D 리간드의 발현이 영향을 받을 것으로 생각되어 진다. 이 연구는 인간의 암세포에서 가장 자주 발현되는 세가지 암유전자 k-ras와 c-myc, wnt의 억제를 통해 NKG2D 리간드의 발현이 어떻게 변화되는 지를 알아보았다. k-ras와 c-myc의 억제는 NKG2D 리간드의 발현을 효과적을 증가시켰고 암세포가 자연살상세포에 더욱 잘 죽게 변화되었다. 그러나 wnt 억제는 MICA와 ULBP1의 전사를 감소시켰다. wnt 억제에 의한 NKG2D 리간드의 전사억제에도 불구하고 세포막의 단백질 발현은 변하지 않아서 암세포의 자연살상세포에 대한 감수성은 별다른 변화를 보이지 않았다. 따라서 k-ras와 c-myc, wnt 억제는 각각 다른 반응을 보였으며 최종적인 자연살상세포에 대한 감수성은 NKG2D 리간드의 세포표면단백질 발현정도에 의해 결정됨을 알 수 있었다.

Anticancer and Anti-Inflammatory Activity of Probiotic Lactococcus lactis NK34

  • The anticancer and anti-inflammatory activities of probiotic Lactococcus lactis NK34 were demonstrated. Treatment of cancer cells such as SK-MES-1, DLD-1, HT-29, LoVo, AGS, and MCF-7 cells with 106 CFU/well of L. lactis NK34 resulted in strong inhibition of proliferation (>77% cytotoxicity, p < 0.05). The anti-inflammatory activity of L. lactis NK34 was also demonstrated in lipopolysaccharide-induced RAW 264.7 cells, where the production of nitric oxide and proinflammatory cytokines (tumor necrosis factor-α, interleukin-18, and cyclooxygenase-2) was reduced. These results suggest that L. lactis NK34 could be used as a probiotic microorganism to inhibit the proliferation of cancer cells and production of proinflammatory cytokines.

Cytotoxic activity and subset populations of peripheral blood natural killer cells in patients with chronic pain

  • Background: Chronic pain reportedly exerts complex effects on immune function. Natural killer (NK) cells are lymphocytes that play a critical role in cellular and innate immunity. This study examined changes in the subset populations and cytotoxic activity of peripheral blood NK cells in patients with chronic pain. Methods: Thirty patients with chronic moderate-to-severe pain (group P) and age-matched pain-free subjects (group NoP) were enrolled. Peripheral whole blood was analyzed for the percentage and expression of NK cell surface markers (CD56 and CD16) by flow cytometry. Cytotoxic activity was assayed by evaluating CD69 expression on $CD3^-/CD56^+NK$ cells. Results: The percentage of NK cells among total lymphocytes was not significantly different between groups P and NoP ($16.3{\pm}9.3$ vs. $20.2{\pm}10.5%$). Likewise, the percentages of two major NK cell subsets, $CD56^{bright}$ and $CD56^{dim}$, were also not significantly different between the two groups. However, the percentage of $CD56^{bright}/CD16^+$ subset, was slightly but significantly increased in group P ($1.0{\pm}0.9%$; P< 0.01) compared with group NoP ($0.5{\pm}0.6%$). The cytotoxicity of NK cells was not different between the two groups, showing similar CD69 expression (P vs. $NoP=29.2{\pm}15.2$ vs. $32.0{\pm}15.0%$). These findings were not influenced by pain intensity, opioid use, or disease causing pain in group P. Conclusions: NK cell cytotoxic activity and major subset populations, with the exception of an increased percentage of the $CD56^{bright}/CD16^+$ subset, are not significantly altered in patients with chronic severe pain.

H2-M3의 세포 표면 발현이 NK 세포의 활성에 미치는 영향 분석 (The Cell Surface Expression of H2-M3 Does Not Directly Effect on the Killing Activity of NK Cell)

  • H2-M3 (M3) is a unique antigen presenting molecule which provides N-formylated peptide to certain type of T cells. Previous observation indicated that NK cell activity is significantly diminished during listerial infection in $H2-M3^{-/-}$ mice. To explore the possibility that M3 expression directly effect on NK cell activity, we measured NK cell activity with or without stimulation of N-formylated peptide on antigen presenting cells. Results indicated that the expression of M3 is not directly influence on NK cell activity. Further study will be focused on the indirect effect of M3 on regulating NK cell activity.

Enhancement of NK Cytotoxicity, Antimetastasis and Elongation Effect of Survival Time in B16-F10 Melanoma Cells by Oregonin

  • We investigated the antitumor activity of oregonin, a diarylheptanoid derivative purified from Alnus hirsuta Turcz, Betulaceae. Oregonin is a potential novel immunomodulator, which augments the activation of natural killer (NK) cells, and thereby leads to a powerful antitumor activity. To evaluate the cytotoxicity of oregonin against tumor cells, we examined the effectiveness of NK cells and determined that oregonin could increase NK cell cytotoxicity. This was confirmed by MTT assay. In addition, the survival time of C57BL/6 mice were measured by inoculating 816-F10 melanoma cells to mice via intra muscular (i.m.) injection. Oregonin treatment after 10 hours of inoculation at 10 mg/kg dosage showed a significant extension of survival time by up to 51.32%, when compared to the control group. Moreover, oregonin significantly reduced the incidence of pulmonary metastasis, which may be developed from 816-F10 melanoma cells. These findings suggest that oregon in may be classified as a new and novel immunomodulator due to its potential antitumor activity.

CCR5-mediated Recruitment of NK Cells to the Kidney Is a Critical Step for Host Defense to Systemic Candida albicans Infection

  • C-C chemokine receptor type 5 (CCR5) regulates the trafficking of various immune cells to sites of infection. In this study, we showed that expression of CCR5 and its ligands was rapidly increased in the kidney after systemic Candida albicans infection, and infected CCR5-/- mice exhibited increased mortality and morbidity, indicating that CCR5 contributes to an effective defense mechanism against systemic C. albicans infection. The susceptibility of CCR5-/- mice to C. albicans infection was due to impaired fungal clearance, which in turn resulted in exacerbated renal inflammation and damage. CCR5-mediated recruitment of NK cells to the kidney in response to C. albicans infection was necessary for the anti-microbial activity of neutrophils, the main fungicidal effector cells. Mechanistically, C. albicans induced expression of IL-23 by CD11c+ dendritic cells (DCs). IL-23 in turn augmented the fungicidal activity of neutrophils through GM-CSF production by NK cells. As GM-CSF potentiated production of IL-23 in response to C. albicans, a positive feedback loop formed between NK cells and DCs seemed to function as an amplification point for host defense. Taken together, our results suggest that CCR5-mediated recruitment of NK cells to the site of fungal infection is an important step that underlies innate resistance to systemic C. albicans infection.

Effects of Armillaria mellea Extract on Macrophage and NK Cell Activity

  • Objective : The purpose of this study was to investigate the effects of Armillaria mellea extract (AME) on immune modulation focused on anti-cancer activity. Methods : To prove the effects of AME, we performed NO assay, NK cytotoxicity assay and RT-PCR of cytokine related with macrophage and NK cell activity. Results : AME increased NO production produced by macrophages in part. AME also enhanced the NK cell activities in destroying target cells (YAC-1 cells). AME up-regulated gene expression of IL-l, iNOS, TNF-a in RAW 264.7 cells and IL-l, IL-2, IFN-(equation omitted), TNF-a in splenocytes, respectively. Conclusion : From the above results, we assumed that AME is a potential drug for anti-cancer by activation of the macrophages and NK cells.
