The effects of the methanol subfraction from Pueraria Radix on hydrogen peroxide-induced lipid peroxidation and cadmium-induced cytotoxicity were investigated in NIH 3T3 fibroblast cells. After the methanol subfraction treatment, the content of MDA induced by 600 ${\mu}g$$H_2O_2$ significantly decreased in proportion to the subfraction concentrations as well as 50 ${\mu}M$$CdCl_2$-induced cytotoxicity. Especially, 200 ${\mu}g/mL$ concentration of methanol subfraction was strongly shown inhibition of lipid peroxidation and detoxification of cadmium. These results suggest that the methanol subfraction from Pueraria Radix retains a potential antioxidant and protective effect against cadmium.
Inositol polyphosphate multikinase (IPMK) is required for the biosynthesis of inositol phosphates (IPs) through the phosphorylation of multiple IP metabolites such as IP3 and IP4. The biological significance of IPMK's catalytic actions to regulate cellular signaling events such as growth and metabolism has been studied extensively. However, pharmacological reagents that inhibit IPMK have not yet been identified. We employed a structure-based virtual screening of publicly available U.S. Food and Drug Administration-approved drugs and chemicals that identified the antidepressant, vilazodone, as an IPMK inhibitor. Docking simulations and pharmacophore analyses showed that vilazodone has a higher affinity for the ATP-binding catalytic region of IPMK than ATP and we validated that vilazodone inhibits IPMK's IP kinase activities in vitro. The incubation of vilazodone with NIH3T3-L1 fibroblasts reduced cellular levels of IP5 and other highly phosphorylated IPs without influencing IP4 levels. We further found decreased Akt phosphorylation in vilazodone-treated HCT116 cancer cells. These data clearly indicate selective cellular actions of vilazodone against IPMK-dependent catalytic steps in IP metabolism and Akt activation. Collectively, our data demonstrate vilazodone as a method to inhibit cellular IPMK, providing a valuable pharmacological agent to study and target the biological and pathological processes governed by IPMK.
장미무당버섯은 담자균문, 주름버섯목, 무당버섯과에 속하며 맛이 좋은 식용 버섯으로 예로부터 암의 치료에 효과가 있다고 알려져 있다. 본 연구에서는 장미무당버섯의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하고 ICR mice에 주사하여 항암 및 면역증강 효과를 조사하였다. Sarcoma 180, HepG2, HT-29, NIH3T3 등의 암세포에 대한 독성을 조사한 각각의 암세포는 $10{\sim}2000\;{\mu}g/ml$의 조다당류 농도에서 세포독성을 나타내지 않았다. 각각의 조다당류가 투여된 실험군은 대조군에 비해 수명이 각각 $21.4{\sim}45%$ 연장되었다. 열수로 추출한 조다당류는 B 임파구의 alkaline phosphatase 활성을 $500\;{\mu}g/ml$의 농도에서 대조군에 비해 약 6.8배의 증가시켰다. 중성염추출 조다당류를 50 mg/kg body weight의 농도로 투여한 생쥐의 총 복강 세포 수와 백혈구의 수는 대조군에 비하여 각각 6배 와 2.6배로 증가하였다. 또한 면역에 관련된 장기인 간, 비장 및 흉선의 체중이 대조군에 비하여 소폭 증가된 것으로 조사되었다. 따라서 장미무당버섯의 자실체에서 추출한 조다당류가 Sarcoma 180에 대한 항암작용을 나타내는 것은 이들 조다당류가 Sarcoma 180에 대한 세포독성효과가 아니고 면역을 증강시킨 효과에 의한 것으로 사료된다.
송이는 담자균문, 주름버섯목, 송이과에 속하며 맛이 좋은 식용 버섯으로 예로부터 사람의 위궤양이나 위암의 치료효과는 물론 생쥐의 Sarcoma 180과 Ehrlich Sarcoma에도 효과가 있다고 알려져 있다. 본 연구에서는 송이의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하고 ICR mice에 주사하여 항암 및 면역증강 효과를 조사하였다. NIH3T3, Sarcoma 180, HepG2, HT-29 등의 암세포에 대한 독성을 조사한 결과 각각의 암세포는 2 mg/ml의 조다당류 농도에서는 세포독성을 나타내지 않았다. Sarcoma 180이 접종된 ICR mouse에 자실체에서 추출한 각각의 조다당류를 투여한 실험군은 대조군에 비해 평균수명이 각각 23.4$\sim$37.2% 연장되었다. 메탄올과 열수로 추출한 조다당류를 0.2$\sim$0.5 mg/ml의 농도로 투여한 실험군 생쥐의 B 임파구 alkaline phosphatase 활성은 대조군에 비해 각각 2.17$\sim$11.9배의 증가하였다. 중성염추출 조다당류를 50 mg/kg body weight의 농도로 투여한 생쥐의 총 복강 세포 수와 백혈구의 수는 대조군에 비하여 각각 6배와 1.5배 증가하였다. 따라서 송이의 자실체에서 추출한 조다당류는 생쥐의 Sarcoma 180에 대해 항암작용 및 면역증강 작용을 나타내는 것으로 확인되었다.
본 연구에서는 검은콩의 육모 촉진 효과를 평가하기 위하여 검은콩 추출물을 이용하여 모유두 세포의 성장 촉진과 모세혈관 확장에 미치는 효과를 평가하였으며, C57BL/6 마우스 모델을 이용하여 검은콩 추출물의 경구 섭취가 모발 성장에 미치는 효과를 2% minoxidil과 비교하여 평가하였다. 그 결과 검은콩 추출물이 모유두 세포의 증식을 촉진하고 모세혈관을 확장 시키는 효과가 있으며, 우수한 성장기 모낭 유도 촉진 효과를 통해 모발의 성장을 촉진하는 것을 확인하였다. 또한, 혈액의 항산화능을 증가시키고 혈중 중성 지질을 감소하는 효과를 가져 혈액을 맑게 하고 혈액 순환에 도움을 주어 육모 및 모발 건강 증진에 기여할 수 있음을 확인하였다. 검은콩 추출물의 우수한 육모 효과를 확신하기 위해서는 사람을 대상으로 6개월 이상의 인체 효능 평가와 검은콩 추출물 내의 유효 성분을 규명하는 연구가 후속으로 진행되어야 할 것이다. 그러나 본 연구는 민간에서 널리 알려진 검은콩의 육모 촉진 효과를 체계적으로 평가하였다는 것에 큰 의의가 있으며, 이를 통하여 검은콩 추출물이 혈액 순환 개선 및 모발 성장 주기 조절을 통해 모발 건강 개선 및 육모 촉진제로써 이용될 수 있다고 기대할 수 있다.

EP was obtained through 20% ethanol extraction of Pueraria lobata root, and the fermented form of EP, FEP, was prepared from the EP after incubating with Lactobacillus rhamnosus vitaP1. There was no significant toxicity by EP and FEP up to $1000{\mu}g/ml$ in NIH-3T3, HaCaT, and B16F10 cells. In addition to antioxidant potentials of EP and FEP determined by DPPH and ABST assays, we confirmed increase of procollagen type I and elastin synthesis by supplementation of the EP and FEP at the concentration of $50{\mu}g/ml$ using ELISA kits. The protein expression levels of matrix metalloprotease (MMP)-1, -3, and -9, those are involved in the degradation of collagen or other skin matrix proteins, were remarkably suppressed while their inhibitory protein metallopeptidase inhibitor 1 (TIMP-1) was greatly up-regulated by supplementation of the EP and FEP at a concentration of $50{\mu}g/ml$. Taken together, both EP and FEP supplementation could be involved in the suppression of the skin wrinkle formation through inhibiting degradation of collagen and stimulating the synthesis of collagen and elastin. The results showed that the anti-wrinkle potential of the EP and FEP will be a promising candidate for developing cosmeceutical compounds or products.
Nerve growth factor(NGF) has a critical role in peripheral nerve regeneration. The aim of this study is to construct a well-functioning hNGF-$\beta$ recombinat adenovirus for the ultimate development of improved method to promote peripheral nerve regeneration with adenovirus mediated hNGF-$\beta$ gene transfection into Schwann cells. First PCR associated cloning of GFP-tagged hNGF-$\beta$ which was ligated into E1/E3 deleted adenoviral vector was performed and tranfected into E. coli to construct hNGF-$\beta$ recombinant adenovirus. After production of recombinat adenovirus in a large scale, its transfection efficiency, expression, and function were evaluated using cell lines or primarily cultured cells of HEK293 cells, Schwann cells, fibroblast(NIH3T3) and myocyte(CRH cells). GFP expression was observed in 90% of infected cells compared to uninfected cells. Total mRNA isolated from hNGF-$\beta$ recombinat adenoviru infected cells showed strong RT-PCR band, however, LacZ recombinant adenovirus infected or uninfected cells did not. NGF quantification by ELISA showed a maximal release of 18.865 +/- 0.31ng/mL at 4th day. PC-12 cells exposed to media with hNGF-$\beta$ recombinant adenovirus infected Schwann cell demonstrated higher levels of differentiation compared with controls. We generated hNGF-$\beta$ recombinant adenovirus and induced over expression of NGF successfully in nonneuronal and neuronal cells. Following these result, it is expected to develop an improved treatment strategy peripheral nerve regeneration using the hNGF-$\beta$ gene transfected cells.
점액질이 풍부한 꼼치 조직에서 NIH 3T3 세포주를 이용하여 subtracted cDNA 라이브러리를 얻어 200례의 클론을 제작하였다. 이 클른 중에서 비반복성 유전자를 선택하고, RNA in situ hybridization을 실행하여 꼼치 조직에서 특이하게 발현되는 곰신 클론(C90-171)을 선택하였다. 이 클론은 사람의 타액선 조직에서도 특이하게 발현되는 유전자로서 이를 확인하기 위하여 C90-171(곰신) 항체를 제작하였다. 꼼치의 cDNA 라이브러리에서 곰신의 항체를 통하여 스크리닝한 결과 PRP(proline-rich protein)와 가장 많이 교차반응하며, 면역조직화학적 염색으로 PRP와 유사한 양성반응으로 나타나 PRP와 유사한 기능을 하는 단백질로 사료된다. 또한 타액 내에서의 꼼치 단백질의 분해에 대한 실험결과 거의 분해가 일어나지 않는 것으로 보아, 곰신은 꼼치의 몸통을 보호하는 유전물질일 뿐만 아니라, PRP와 유사하게 조직을 보호하는 안정된 새로운 기능성 단백질로 사료된다.
PURPOSE. This study was to evaluate the effects of bacterial cellulose (BC) membranes as a barrier membrane on guided bone regeneration (GBR) in comparison with those of the resorbable collagen membranes. MATERIALS AND METHODS. BC membranes were fabricated using biomimetic technology. Surface properties were analyzed, Mechanical properties were measured, in vitro cell proliferation test were performed with NIH3T3 cells and in vivo study were performed with rat calvarial defect and histomorphometric analysis was done. The Mann-Whitney U test and the Wilcoxon signed rank test was used (${\alpha}<.05$). RESULTS. BC membrane showed significantly higher mechanical properties such as wet tensile strength than collagen membrane and represented a three-dimensional multilayered structure cross-linked by nano-fibers with 60 % porosity. In vitro study, cell adhesion and proliferation were observed on BC membrane. However, morphology of the cells was found to be less differentiated, and the cell proliferation rate was lower than those of the cells on collagen membrane. In vivo study, the grafted BC membrane did not induce inflammatory response, and maintained adequate space for bone regeneration. An amount of new bone formation in defect region loaded with BC membrane was significantly similar to that of collagen membrane application. CONCLUSION. BC membrane has potential to be used as a barrier membrane, and efficacy of the membrane on GBR is comparable to that of collagen membrane.
세포의 이동은 많은 생리적 반응뿐만 아니라 암 세포 침윤과 전이에 중요한 역할을 수행한다. 본 연구에서는 마늘이 암세포의 이동에 미치는 영향을 확인하기 위해, 표준 마늘과 흑마늘을 준비하고 이들을 각각 물을 이용하여 추출하거나 건조하여 추출한 추출물 4 종류를 이용하여 항침윤성과 항전이성에 대해 조사하였다. 실험결과, 암세포의 이동은 건조 후 헥산으로 추출한 분획에 암세포의 이동 억제 활성이 관찰되었다. 이 분획을 박막 크로마토그래피를 이용하여 분리정제하였으며, 이를 inhibitor of cancer metastasis from garlic #27 (ICMG-27)이라 명명하였다. ICMG-27 (6 ${\mu}g/ml$)을 세포에 처리하였을 때, IGF-1에 의한 OVCAR-3와 NIH-3T3 세포의 이동을 억제함을 확인하였다. 그러나 ICMG-27은 mouse embryonic fibroblast (MEF) 세포에서 IGF-1에 의한 이동에는 영향을 주지 않았다. 이러한 ICMG-27은 OVCA-3, SKOV-3와 MDA-MB-231 세포와 같은 암세포에서 모두 IGF-1에 의한 이동을 억제함을 관찰하였다. 마지막으로 세포이동을 일으키는 인자에 따른 ICMG-27의 영향을 확인한 것으로, IGF-1, lysophosphatidic acid (LPA), sphingosine-1-phosphate (S1P), leukotriene B4 (LTB4) 그리고, angiotensinII (AngII)에 의한 OVCAR-3 세포의 이동을 모두 억제하였다. 이러한 결과를 바탕으로, ICMG-27은 암세포의 이동을 유도하는 많은 인자들에 의한 필수적인 단계를 차단함으로써, 암세포의 이동을 억제하는 것을 확인 할 수 있었으며, ICMG-27에 의한 암세포의 항 침윤 메커니즘의 규명은 암환자의 치료에 기초적인 발판을 제공할 것입니다.
