Objectives : It is demonstrated that oxygen free radicals have cytotoxic effect on NIH3T3 fibroblast cells. Recently, many of herb extracts have an effect of antioxidant in oxygen free radical-induced cytotoxicity. But, the toxic mechanism of oxygen free radical is left unknown. The purpose of this study was to examine the cytotoxicity of hydrogen peroxide ($H_2O_2$) and antioxidant effect of Citri reticulatae pericarpium (CRP) on NIH3T3 fibroblasts. Methods : The cytotoxicy was measured by cell viability by XTT assay in NIH3T3 fibroblasts. XTT assay is regarded as a very sensitive screening method for the determination of the cell viability on various chemicals. Results : In this study, H2O2 decreased cell viability according to the dose- and time dependent manners after NIH3T3 fibroblasts were treated with various concentrations of H2O2 for 4 hours. And also, CRP showed the effect of antioxidant on $H_2O_2-induced $ cytotoxicity in cultured NIH3T3 fibroblasts. Conclusion : These results suggest that $H_2O_2$ has highly cytotoxic effect on cultured NIH3T3 fibroblasts by the decrease of cell viavility, and the herb extract such as CRP was showed the effect of antioxidant on $H_2O_2-induced$ cytotoxicity in these cultures.
To clerify the protective effect of omega-3 of polyunsaturated fatty acid docosahexaenoic acid (DHA) on the cytotoxicity induced by organic mercury in cultured NIH3T3 fibroblasts. The measurement of cell viability on ogranic mercury wad done by XTT assay after NIH3T3 fibroblasts were cultured with various concentrations of methyl mercuric chloride (MMC). And also, the effect of DHA on the MMC-mediated cytotoxicity was examined by cell viability, and antioxidant effect of DHA was also assessed by superoxide dismutase (SOD)-like activity and the lipid peroxidation activity in cultured NIH3T3 fibroblasts. In this study, MMC decreased cell viability and $XTT_{50}$ value was determined at $50{\mu}M$ of MMC in these culture. In the effect of DHA against the cytotoxicity induced by MMC, DHA significantly increased the cell viability damaged by MMC in cultured NIH3T3 fibroblasts. And also, DHA showed the antioxidant effect by showing the increase of SOD-like activity and the decrease of lipid peroxidation activity. From these results, it is suggested that organic mercury such as MMC has highly toxic effect on cultured NIH3T3 fibroblasts, and also, omega-3 of polyunsaturated fatty acid, DHA showed the protection on MMC-induced cytotoxicity and antioxidant effect.
This study was carried out to investigate cytotoxicity of several herbicides (Bentazone, Butachlor. Paraquat and Ethalfluralin) in cultured mouse NIH 3T3 fibroblasts. Tetrazolium (MTT), neutral red (NR) and sulforhodamine protein B (SRB) of the colorimetric assays were performed to evaluate the cytotoxicity on cell organelles. 2 x 10$^4$cell/$m\ell$ of NIH 3T3 fibroblast in each well of 24 multidish were cultured. After 24 hours, the cells were treated with solution (1, 25, 50 or 100 $\mu$M) of each herbicide. After the NIH 3T3 fibroblasts of all groups were cultured in the same condition for 48 hours, MTT, NR and SRB assays were performed to evaluate the cytotoxicity. The light microscopic study was carried out to examine morphological changes of cultured NIH 3T3 fibroblasts. The MTT$_{50}$ of Bentazone, Butachlor, Paraquat and Ethalfluralin were 1560.97 $\mu$M, 56.15 $\mu$M, 3138.81 $\mu$M and 1301.82 $\mu$M, respectively. The NR$_{50}$ of Bentazone, Butachlor. Paraquat and Ethalfluralin were 1763.93 $\mu$M, 45.98 $\mu$M, 1030.85 $\mu$M and 1808.29 $\mu$M, respectively. The SRB$_{50}$ of Bentazone, Butachlor. Paraquat and Ethalfluralin were 1913.38 $\mu$M, 65.30 $\mu$M, 1860.73 $\mu$M and 1086.93 $\mu$M, respectively. The morphological changes of NIH 3T3 fibroblasts showed severe degeneration in Butachlor 50 $\mu$M and 100 $\mu$M concentrations. These results indicate that Butachlor has high cytotoxicity, Bentazone, Paraquat and Ethalfluralin very weak cytotoxicity against NIH 3T3 fibroblasts.lasts.
Objectives : It is well known that hexavalent chromium has toxic effect on normal cells. Recently, toxic effect of hexavalent chromium is diminished by the some extracts derived from herbs or plants. But, the toxic or protective mechanism of hexavalent chromium is well unknown. This study was performed to examine the protective effect of poncirin against $Na_2Cr_2O_7$-induced cytotoxicity on NIH3T3 fibroblasts. Methods : The protective effect of the cytotoxicity induced by $Na_2Cr_2O_7$ was measured by the cell viability after NIH3T3 fibroblasts were cultured with or without $Na_2Cr_2O_7$ for 48 hours. Antitoxic effects of poncirin on the cytotoxicity induced by $Na_2Cr_2O_7$ were examined by colorimetric assays such as MTT or XTT assay. Results : $Na_2Cr_2O_7$ decreased cell viability by the decreased absorbance in MTT or XTT assay, but, the poncirin increased cell viability which was decreased by $Na_2Cr_2O_7$-induced cytotoxicity on NIH3T3 fibroblasts. Conclusion : These results suggest that $Na_2Cr_2O_7$ showed cytotoxicity effect on NIH3T3 fibroblasts by the decrease of cell viavility, and poncirin was effective in the protection of $Na_2Cr_2O_7$-induced cytotoxicity in these cultures.

This study was carried out to evaluate the cytotoxic effect of phenolic compound on normal NIH 3T3 fibrolasts. The colorimetric assay for phenol compound, syringic acid was performed by MTT assay or XTT assay. MTT or XTT assays are known as a very sensitive method in measuring the cytotoxic effect of chemical agents in vitro. In the present study, syringic acid on normal Nlli 3T3 fibroblasts did not show any cytotoxicity for MTT assay or XTT assay compared with control after cells were treated with various concentrations of syringic acid for 48 hours. MTT/sub 50/ and XTT/sub 50/ were 3,340.9 μM and 2,462.4 μM of syringic acid, respectively. From the above the results, it is suggested that phenolic compound of syringic acid did not have any cytotoxicity on normal NIH 3T3 fibroblasts.

The cytotoxic activity of Cratalariae sessiliflorae on cultured NIH 3T3 fibroblasts and human oral epithelioid carcinoma cells (KB) were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) colorimetric method These fractions of methanol extract of Cratalariae sessiliflorae showed inhibitory effect in vitro in the milligram range against KB cell lines. In general, the antitumor activities of these fractions were does-dependent over the milligram range. The comparison of IC50 values of these fractions in tumor cell lines showed that their susceptibility to these fractions decrease in the following order: Fr. 4> Fr. 6> Fr. 10> Fr. 2> Fr. 11> Fr. 3> Fr. 8> Fr. 7> Fr. 9> Fr. 1> Fr. 5 by the MTT assay. These fractions were tested for their cytotoxic effects on NIH 3T3 fibroblasts using MTT assay. They exhibited potent cytotoxic activities in vitro in the milligram range against NIH 3T3 fibroblasts. In general, the cytotoxic activities of these fractions were does-dependent over the milligram range. The comparison of CD50 values of these fractions in NIH 313 fibroblasts shows that their susceptibility to these fractions in decrease the following order: Fr. 10> Fr. 9> Fr. 2 = Fr. 4> Fr. 8> Fr. 11> Fr. 1 = Fr. 7> Fr. 3> Fr. 5 = Fr. 6 by the MTT assay. These results suggests that fraction 5 has the most growth - inhibitory activity against KB cell lines.
Despite extensive investigation, the mechanisms of immune responses to Candida albicans infection remain poorly understood. Using RT-PCR and Northern blot analysis, this study demonstrates the pattern of IL-6 mRNA expression in thioglycollate-elicited mouse peritoneal macrophages and NIH3T3 fibroblasts (NIH3T3) in response to C. albicans. The expression of IL-6 mRNA was detectable in both cell types. However, IL-10 mRNA was only expressed in the macrophages, and IL-4 mRNA was not expressed in neither of the two cell types. Although the phagocytic function of the macrophages was inhibited by Cytochalasin D, these macrophages could still induce the expression of IL-6mRNA. These findings indicated that the phagocytosis of C. albicans is not pivotal in the induction of IL-6 mRNA expression. A Northern blot analysis was used to investigate the dose effects of C. albicans and time-course kinetics of IL-6 mRNA expression at various time points. IL-6 mRNA was expressed in a dose-independent manner, and was detectable as early as 30min after C. albicans stimulation. It was evenly sustained up to 4h. These results can contribute to understanding the mechanism of IL-6 mRNA expression in macrophages and NIH3T3 cells in response to C, albicans.
Cytotoxicity of cadmium on NIH 3T3 fibroblasts was utilized in order to discover antitoxic compound in Galgeuntang extract in this study. Treatment groups were chosen as follows; control (medium only), $MTT_{50}$ group and five experimental groups. MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide} method was performed to evaluate the cytotoxicity of cell organelles and $IC_{50}$ was also measured. Accordingly we have examined the detoxification effects of Galgeuntang extract on cadmium-treated NIH 3T3 fibroblasts to observe morphological changes by the light microscopy. Galgeuntang extract showed cytoprotective effects on cadmium-induced cytotoxicity. Furthermore, Galgeuntang showed a dose-dependency in detoxication. The phenolic content of Galgeuntang ethanol extract was higher than that of water content. These results suggest that Galgeuntang extract may be used as a cytoprotective agent against cadmium (II)-mediated cytotoxicity.
It is demonstrated that phenolic compound has cytotoxic effect on cancer cells. Recently, ferulic acid is involved in anticancer activity by showing the decrease of cell viability in cancer cells. But, the anticancer mechanism of ferulic acid is left unknown. The purpose of this study was to examine the anticancer activity of ferulic acid on NIH3T3 fibroblasts and human skin melanoma cells (SK-MEL-3). The anticancer activity was measured by determining the cytotoxicy of ferulic acid on these cells. The cytotoxicity was measured by cell viability via XTT assay in these cells. In this study, ferulic acid decreased cell viability according to the dose-dependent manners after human skin melanoma cells were treated with various concentrations of ferulic acid for 48 hours. especially, ferulic acid remarkably decreased cell viability at a concentration of $120{\mu}M$ compared with control in human skin melanoma cells. While, ferulic acid did not show the significant decrease of cell viability at concentrations of $30{\sim}120{\mu}M$ in NIH3T3 fibroblasts. These results suggest that ferulic acid showed anticancer activity in cancer cells such as human skin melanoma cells by the decrease of cell viability significantly.
The anticancer and antioxidant effect of different lengths of saturated fatty acids was tested on NIH3T3 fibroblasts and human skin melanoma cellsn in this study. The cell existence rate and antioxidizing capacity and optic reservation of cells were observed. This saturated fatty acid was concentration-dependent. IC50 Concentrations in NIH3T3 fibroblasts, human skin melanoma cells and DPPH radical scavenging activity of fatty acid was increasing the order of carbochain length ; caprylic acid < lauric acid < palmitic acid < stearic acid. The reduction in cell number and morphological change in human skin melanoma cells was increasing the order of carbochain length ; caprylic acid < lauric acid < palmitic acid < stearic acid. These results suggest that carbochain length of fatty acid can be used as structure-activity relationships for anticancer and antioxidant.
