Chemical Modification of Tryptophan Residue in Bovine Brain succinic Semlaldehyde Reductase

  • Incubation of an NADPH-dependent succinic semialdehyde reductase from bovine brain with N-bromosuccinimide (NBS) resulted in a time-dependent loss of enzyme activity. The inactivation followed pseudo-first-order kinetics with the second-order rate constant of $6.8\times{10}^3$ $M^-1$ $min^{-1}$. The inactivation was prevented by preincubation of the enzyme with substrate succinic semialdehyde, but not with coenzyme NADPH. There was a linear relation-ship between oxindole formation and the loss of enzyme activity. Spectro-photometric studies indicated that about one oxindole group per molecule of the enzyme was formed following complete loss of enzymatic activity. It is suggested that the catalytic function of succinic semialdehyde reductase is modulated by binding of NBS to a specific tryptophan residue at or near the substrate binding site of the enzyme.

HS-1200 Overcomes the Resistance Conferred by Bcl-2 in Human Leukemic U937 Cells

  • Bcl-2 protects tumor cells from the apoptotic effects of various anti-neoplastic agents. Increased expression of Bcl-2 has been associated with a poor response to chemotherapy in various malignancies, including leukemia. Hence, bypassing the resistance conferred by anti-apoptotic factors such as Bcl-2 represents an attractive therapeutic strategy against cancer cells, including leukemic cells. This study was undertaken to examine whether the anticancer drug, cisplatin and the synthetic chenodeoxycholic acid (CDCA) derivative, HS-1200 show anti-tumor activity in U937 and U937/Bcl-2 cells. Viability assays revealed that HS-1200 overcomes the resistance conferred by Bcl-2 in human leukemic U937 cells. Various apoptosis assessment assays further demonstrated that HS-1200 overcomes the resistance conferred by Bcl-2 in human leukemic U937 cells by inducing apoptosis. In addition HS-1200, but not cisplatin, overcomes the anti-apoptotic effects of Bcl-2 in Bcl-2 over-expressing human leukemic cells (U937/Bcl-2 cells). Notably, we observed that the HS-1200-induced formation of mature promyelocytic leukemia (PML) nuclear bodies (NBs) correlates with a suppression of the anti-apoptotic effects of Bcl-2 in human leukemic cells over-expressing this protein (U937/Bcl-2 cells). Furthermore, HS-1200 was found to induce the association between PML and SUMO-1, Daxx, Sp100, p53 or CBP in the aggregated PML-NBs of U937/Bcl-2 cells. Thus, PML protein and the formation of mature PML-NBs could be considered as therapeutic targets that may help to bypass the resistance to apoptosis conferred by Bcl-2. Elucidating the exact mechanism by which PML regulates Bcl-2 will require further work.

  • $\alpha$-Functionalized benzylamine(Nu=OAc, OMe, $N_3$)은 유기합성의 출발물질 또는 synthon으로서 매우 유용할 것이다. 본 연구에서는 benzylphthalimide를 137$^{\circ}C$, chlorobenzene 용매 하에서 NBS/NaOAc/HOAc조건을 사용하여, 높은 수율로 phthalimide-benzyl acetate을 얻는 방법을 개발하였다. 마지막으로, phthalimide-benzyl acetate을 $NH_2NH_2$/HOAc 조건 하에서, amino(phenyl)methyl acetate (1)을 합성하였다.

Evaluation of urban pollutant washoff characteristics and treatment efficiency of a small constructed wetland

  • Nature-based solutions (NBS) offer a wide variety of techniques that promote cost-efficient stormwater management practices. In particular, low impact development facilities utilize NBS principles to restore the ecosystem services in a highly-urbanized area. Despite the advancements in these technologies, several considerations should still be addressed to ensure optimum functionality and attainment of desired pollutant removal efficiency a LID facility. This study evaluated the mass flushing characteristics of pollutants in an urban catchment and the efficiency of a small constructed wetland (SCW) in treating urban stormwater runoff. 21 rainfall events from 2010 to 2018 were monitored to determine and quantify stormwater pollutants. The highest pollutant washoff was observed on rainfall depths ranging from 0.1mm to 10mm, whereas events with greater rainfall depths exhibited lower pollutant concentrations due to dilution effect. However, the SCW manifested lower pollutant-removal performance on rainfall depths exceeding 10mm due to the exceedance of the facility's design rainfall. This study is beneficial in assessing the dynamics of pollutant washoff and efficiency of LID facilities subjected under various external factors.

안정화 CdS-CdSe계 채료에 관한 연구 제3보 $ZrSiO_4-Cd(SxSe_{1-x})$채료의 응용연구 (A Study on Stabilized CdS-CdSe Red Stain part III, Application of $ZrSiO_4-Cd(SxSe_{1-x})$ Stain to Ceramic Body)

  • 본 연구는 제2보에서 합성한 $ZrSiO_4-Cd(SxSe_{1-x})$ 채료의 실용성을 조사하기 위하여 행하였다. 기존의 유명회사 제품의 채료와 본 합성채료를 비교 검사하였다. 우선 각각의 채료에 Frit를 적당한 비로 혼합하고 일정조성의 Tile에 시유하여 각각다른 온도(850-120$0^{\circ}C$)에서 소성한 다음 colorimeter를 사용하여 XYZ 주파장에 따른 색도도를 측정하였다. 또한 Munsell 계의 ISCC-NBS 색명법을 적용하여 색을 정량화 시킴으로서 채료의 적응성을 연구하였다. 그 결과 $ZrSiO_4-Cd(SxSe_{1-x})$ 채료는 V나 P 계통의 여타 채료와 같이 Frit 대 Stain의 비가 9:1의 최소비로 사용이 가능하며 100$0^{\circ}C$의 높은 온도에서도 밝은 적색으로 재현 될 뿐만 아니라 내산성이 강하고 고온에서 적응성이 있음을 확인하였다.

Genome-wide analysis of heterosis-related genes in non-heading Chinese cabbage

  • Heterosis or hybrid vigor describes a phenomenon that superior phenotypes compared to the two parents are observed in the heterozygous $F_1$-hybrid plants. Identification and characterization of heterosis-related genes (HRGs) will facilitate hybrid breeding in crops. To identify HRGs in Brassica rapa, we analyzed transcriptome profiling using a Br300K microarray in non-heading Chinese cabbage at three developmental stages. A large number of genes were differentially expressed in $F_1$ hybrids and non-additive expression was prominent. Genes that are expressed specifically for $F_1$ hybrid at all three stages were Brassica-specific uncharacterized genes and several defense-related genes. Expression of several photosynthesis- and stress-related genes were also $F_1$ hybrid-specific. Thirteen NBS-LRR class genes showed high and specific expression in $F_1$ hybrid Shulu: some of them were characterized as defense genes in Arabidopsis, but most have not been. Further characterization of these defense-related genes in Brassica species and its application will be helpful for understanding the role of defense responses in heterosis. In addition, results obtained in this study will be valuable to develop molecular markers for heterosis and disease resistance in B. rapa.

Synthesis of Au Nanoparticles Functionalized 1D α-MoO3 Nanobelts and Their Gas Sensing Properties

  • A novel sensor material of Au nanoparticles (NPs) functionalized 1D ${\alpha}-MoO_3$ nanobelts (NBs) was fabricated by a facile lysine-assisted approach. The obtained $Au/{\alpha}-MoO_3$ product was characterized by means of X-ray diffraction (XRD), scanning electron microscope (SEM), transmission electron microscope (TEM) and energy dispersive X-ray (EDX), and X-ray photoelectron spectra (XPS). Then, in order to investigate the gas sensing performances of our samples, a comparative gas sensing study was carried out on both the ${\alpha}-MoO_3$ NBs before and after Au NPs decoration by using ethanol vapor as the molecular probe. The results turned out that, after the functionalization of Au NPs, the sensor exhibited improved gas-sensing characteristics than the pure ${\alpha}-MoO_3$, such as response and recovery time, optimal operating temperature (OT) and excellent selectivity. Take for example 200 ppm of ethanol, the response/recovery times were 34 s/43 s and 5.7 s/10.5 s, respectively, while the optimal operating temperature (OT) was lower to $200^{\circ}C$ rather than $250^{\circ}C$. Besides, the functionalized sensor showed a higher response to ethanol at $200^{\circ}C$, and response was 1.6 times higher than the pure $MoO_3$. The mechanism of such improved sensing properties was interpreted, which might be attributed to the spillover effect of Au NPs and the electronic metal-support interaction.

Contemporary Chinese Households' Food Away From Home Expenditure and Becker's Household Production Theory

  • This study examines factors determining contemporary Chinese households' food away from home (FAFH) expenditures using Becker's household production theory. Data came from the 2000 urban household survey in Guangdong Province, collected by National Bureau of Statistics (NBS) of China. It was revealed that the contemporary urban Chinese wives also substitute their household work by time-saving product, FAFH, as Becker's household production theory postulated. This suggests the important role of time-value (opportunity cost) in determining household FAFH expenditure across the cultures.

NBS-or NCS-induced lactonization of N,N-dimethyl-2-(phenylsulfonyl)-4-pentenamide in DME-$H-2O(2:1 vol)$ gave 2-halo-4-(halomethyl)-2-(phenylsulfonyl)-${\gamma}$-butyrolactone.

  • Novel halactolactonizations using NBS (2), NIS(3), and NSP(4) in dry DMF were found to be the most reasonable and efficient procedures. The participation of X as the cylization initiator could be clarly rationalized by experimental results and the regioselectivities of formed halolactones were reasonably elucidated by weakly bridged carbonium ion intermediate.
