Studies on the Synthesis of Etodolac Derivatives as Potential Anti-inflammatory Agents (항염증제인 Etodolac 유도체의 합성에 관한 연구)

  • For the synthesis of new anti-inflammatory agents as indol derivatives, we have synthesized ${\alpha}$-benzoyl-1-ethyl-1,3,4,9-tetrahydro-8-ethyl-9-(N-benzoyl)pyrano[3,4-b]indole-1-acetic acid methyl ester. It was a new method for ${\alpha}$-substituted etodolac carboxylic acid. The synthetic process was composed of four steps, and 7-ethylindole and oxalyl chloride were used as starting materials. The third step, cyclization was carried out by addition of borontrifluoride diethyl etherate in 66% yield. The step of reduction and cyclization were simplified successfully. The final product, ${\alpha}$-benzoyl-1-ethyl-1,3,4,9-tetrahydro-8-ethyl-9-(N-benzoyl)pyrano[3,4-b]indole-1-acetic acid methyl ester was obtained in 66% yield by the reaction of methyl 1,8-dimethyl-1,3,4,9-tetrahydropyrano[3,4-b]indole-1-acetate (etodollic acid methyl ester) and benzoyl chloride.

Another Evidence for Nitric Oxide as Mediator of Relaxation of Isolated Rabbit and Human Corpus Cavernosum

  • To prove the hypothesis that NO- and N $O_2$-carrying molecules potentiate photorelaxation by generating NO, investigation was carried out using isolated rabbit and human corpus cavernosum. Corporal smooth muscle, in the presence or absence of endothelium, relaxed only slightly upon ultraviolet light (366 nm) irradiation. But, NO-and/or N $O_2$-containing compounds such as streptozotocin and $N^{G}$-nitro-L-arginine methyl ester significantly (p<0.01) enhanced photorelaxation in this tissue. In addition, $N^{G}$-nitro-D-arginine methyl ester, known to lack inhibitory action on NO synthase, showed concentration-dependent potentiation of the photorelaxation. Oxygen radical generating system via copper+ascorbic acid and guanylate cyclase inhibitor, methylene blue, significantly (p<0.05) inhibited the streptozotocin-potentiated photorelaxation. Nitrite was accumulated by photolysis of streptozotocin, $N^{G}$-nitro-L-arginine methyl ester and $N^{G}$-nitro-D-arginine methyl ester, in a concentration and exposure time dependent manner. These observations indicate that NO is a potent relaxant of rabbit and human corpus cavernosum and further support the hypothesis that NO is released by photolysis from NO- and N $O_2$-carrying molecules.lecules.

  • Green mustard leves were found to effectively prevent lipid peroxidation of bovine-brain tissue by ascor-bate/Fe system, The 50% methanol extracts mustard leaves were separated into four solvent faction using n-hexane,. EtOAc, n-BiOH and water. Then n-BiOH fraction exclusively exhibited the antioxidative activities at concentration above 100 $\mu\textrm{g}$/mL/ The n-BuOH fraction was further isolated to a single compound using TLC analysis and silica gel chromatography. The active antiodidative compounds were identified as sinapic acid methyl ester and ferulic acid methyl ester by $^{1}$H-NMR and $^{13}$ C-NMR, The sinapic acid methyl ester and ferulic acid methyl ester were prepared by methylating of sinapic acid and ferulic acid with diazomethane. The results strongly suggested that sinapic acid and ferulic acid could be emplyed as a potential antioxiative agents for preventing the bovine brain lipid peroxidation. lipid peroxidation.

Antimutagenic Effects of Methyl Alcohol Extracts from Auricularia auricula and Gyrophora esculenta (목이 및 석이 메틸 알콜 추출물의 항돌연변이원성)

  • This study was conducted to investigate the antimutagenic effects of methyl alcohol extracts from Auricularia auricula and Gyrophora esculenta on the SOS response induced by 4-nitroquinoline-1-oxide (4NQO), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), mitomycin C (MMC) and 3-amino-1,4- dimethyl-5H-pyrido-[4,3-b]indol (Trp-P-1) in E. coli PQ37/plasmid pKM101. In the mutagenic test on test strain, both methyl alcohol extracts did not show mutagenic activity. In the antimutagenic test, each sample strongly inhibited the mutagenecity induced by 4NQO, MNNG, MMC and Trp-P-1. Methyl alcohol extracts from Auricularia auricula and Gyrophora esculenta showed inhibitory effects of 52% and 59% against 4NQO, 49% and 58% against MNNG, 53% and 64% against MMC, and 61% and 64% against Trp-P-1, respectively. Gyrophora esculenta extracts on the antimutagenicity showed relatively higher inhibitory effects than that of Auricularia auricula.

  • The absorption rate of carbon dioxide with 2-amino-2-methyl-1-propanol(AMP) was measured in such non-aqueous solvents as methanol, ethanol, n-propanol, n-butanol, ethylene glycol, propylene glycol, and propylene carbonate, and in water at 298 K and 101.3 kPa using a semi-batch stirred tank with a plane gas-liquid interface. The overall reaction rate constant, obtained under the condition of fast reaction regime, from the measured rate of absorption was used to get the elementary reaction rate constants in complicated reactions represented by reaction mechanism of carbamate formation and the order of overall reaction of $CO_2$ with amine. The correlation between the elementary reaction rate constant and the solubility parameter of the solvent was also presented.

Cd(II) and Zn(II) Complexes Containing N,N'-Bidentate N-(Pyridin-2-ylmethylene)cyclopentanamine: Synthesis, Characterisation and Methyl Methacrylate Polymerisation

  • The reaction between $[CdBr_2{\cdot}4H_2O]$ and anhydrous $[ZnCl_2]$ with N,N'-bidentate N-(pyridin-2-ylmethylene)-cyclopentanamine (impy) in ethanol yields dimeric $[(impy)Cd({\mu}-Br)Br]_2$ and monomeric $[(impy)ZnCl_2]$ complexes, respectively. The X-ray crystal structure of Cd(II) and Zn(II) complexes revealed that the cadmium atom in $[(impy)Cd({\mu}-Br)Br]_2$ and zinc in $[(impy)ZnCl_2]$ formed a distorted trigonal-bipyramidal and tetrahedral geometry, respectively. Both complexes showed moderate catalytic activity for the polymerisation of methyl methacrylate (MMA) in the presence of modified methylaluminoxane (MMAO), with polymethylmethacrylate (PMMA) syndiotacticity of about 0.70.

  • The kinetic studies on the reactions of O-methyl N,N-diisopropylamino phosphonochloridothioate with X-anilines and X-pyridines have been carried out in acetonitrile. The free energy relationship with X in the anilines exhibits biphasic concave upwards with a break region between X = (H and 4-F), giving unusual negative ${\beta}_X$ and positive ${\rho}_X$ values with weakly basic anilines. The unusual phenomenon is rationalized by isokinetic relationship. A stepwise mechanism with a rate-limiting leaving group departure from the intermediate is proposed based on the selectivity parameter and variation trend of the deuterium kinetic isotope effects with X. The free energy relationship with X in the pyridines exhibits biphasic concave upwards with a break point at X = 3-MeO. A concerted mechanism is proposed based on relatively small ${\beta}_X$ value, and frontside and backside nucleophilic attack are proposed with strongly and weakly basic pyridines, respectively.

  • Reaction of thianthrene cation radical perchlorate (1) with 1-methyl-4-benzenesulfonylaminobenzene (10) afforded thianthrene (5), N-(4-tolyl)-N-thianthrenylbenzenesulfonamide (14), 1-methyl-3-[N-(4-tolyl)-N-benzenesulfonyl-amino- 4-benzene-sulfonylaminobenzene (16), cis-thianthrene-5,10-dioxide (17), 5-(3'-methyl-6-benzenesulfonylaminobenzene)thian threnium perchlorate (18), and benzenesulfonate. In the meantime, reaction of 1 with 1-ethyl-4-benzenesulfonylaminobenzene (12) afforded 5, 1-ethyl-3-[N-(4-ethylphenyl)-N-benzenesulfonyl]a mino-4-benzenesulfonylaminobenzene (19), 1-benzenesulfonyl-amino-4-[1-(2-benzenesulfonyla mino-5-ethylphenyl)ethyl]benzene (20), and 1-(1-acetamidoethyl)-4-benzenesulfonylaminobenz ene (21). The formations of these products except for 18 and benzenesulfonate could be rationalized by assuming a sulfonamidyl radical as an intermediate.

Effects of Various Nootropic Candidates on the Impaired Acquisition of Ethanol-treated Rats in Step-through Test (에탄올 급성 투여로 유발된 학습획득 손상에 미치는 수종 뇌기능개선 후보 물질의 작용)

  • Effects of single and repeated administration of various nootropic candidates were examined on impaired acquisition by single oral administration of 3 g/kg ethanol (EtOH) in step through test. The inhibitory effect of EtOH on acquisition was significantly reduced by single picrotoxin, but not affected by diazepam, acetyl-L-carnitine and apomorphine. Single or repeated red ginseng total saponin and deprenyl, single piracetam, repeated N-methyl-D-glucamine, but not single or repeated protopanaxadiol, protopanaxatriol and centrophenoxine significantly ameliorated the impairment of acquisition by EtOH. On the other hand, the inhibitory effect of repeated red ginseng total saponin but not that of repeated N-methyl-D-Glucamine, was significantly blocked by pretreatment of $\alpha$-methyl-$\rho$-tyrosine, a inhibitor of catecholamine synthesis. Whereas, the inhibitory effect of repeated deprenyl on EtOH amnesia was exaggerated by $\alpha$-methyl-$\rho$-tyrosine. These results suggest that the amelioration processes of drugs on ethanol amnesia involve complex mechanism between the central GABAergic and dopaminergic neuronal activity in memory and learning, although the effects of repeated drugs administration are not yet clear.
