국내산 약용식물 추출물에 대한 항균성 검색과 농도별 및 분획별 항균특성 (Antimicrobial Effect of Ethanol Extracts from Some Medicinal Herbs and Their Fractionates against Food-Born Bacteria)

  • 국내에서 생산되는 한약재 56종을 75% 에탄올로 추출하여 10종의 균주, Listeria monocytogenes ATCC 15313, 19111, 19112, 19113, 19114, Bacillus cereus YUFE 2004, Staphylococus aureus KFCC 11764, P. fluorescens 1645, 2344 and Leuconostoc mesenteroides KFCC 12031에 대하여 항균성을 비교한 결과 귤피, 연교, 우방자, 진교, 호황련, 희첨등은 몇 균주에 대하여 항균 효과를 보이는 반면 가자육, 금앵자, 소목 그리고 육두구는 각 균주에 대해 뚜렷한 증식억제효과를 보였다. 그중 소목 조추출물을 10 ppm 첨가수준에서 모든 시험 균주에 대해 뚜렷한 증식 저해 효과를 나타냈고 가자육은 L. monocytogenes ATCC 19114, B. cereus YUFE 2004, S. aureus KFCC 111764 그리고 P. fluorescens KCTC 1645에 대해 농도에 비례하여 상당한 항균효과를 보였다. 또한 뚜렷한 항균효과가 있는 추출물을 클로로포름, 에틸아세테이트, 부탄올로 분획하여 검색한 결과 대부분의 균주가 가자육, 금앵자, 소목 추출물은 에틸아세테이트 분획물에서 육두구는 클로로포름 분획물에서 더 높은 항균성을 보였다.

  • The methanol extract from seed of Myristica fragrans (Myristicaceae) demonstrated a potent inhibition on the pro-liferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro. By the continuous effort to purify the active components responsible far the anti-proliferative effect on tumor cell lines, we have isolated eleven kinds of lignan components, i. e., safrole (1), machilin A (2), licarin B (3), macelignan (4), mere-dihydroguaiaretic acid (5), mγnstargenol A (6), methoxyeugenol (7), machilin F (8), licarin A (9), nectandrin B (10), and 2-(4-allyl-2,6-dimethoxyphenoxy)-1-(4-hydroxy-3-methoxyphenyl)propan-1-ol (11) together with a novel furan fatty acid, (E)-3-(3-methyl-5-pentylfuran-2-yl) acrylic acid (12) from seed extract of M. fragrans. Chemical structures of the isolated components (1-12) were established bγ the aid of NMR spectroscopic analyses, i. e., COSY HMQC and HMBC. Each of the Isolates demonstrated a potent inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OY-3 (ovary), SK-MEL-2 (melanoma) and HCT-15 (colon) in vitro.

  • In this study, we investigated the antimicrobial activity of herbs related with treatments of intestinal diseases against intestinal pathogens under anaerobic broth system. The water extract of Saussurea lappa Clarke and Myristica fragrans Houtt. showed no growth inhibition against tested pathogens(Eubacterium limonsum ATCC 10825, Escherichia coli ATCC 25922, Bacteroides fragilis KCTC 5013, Clostridium perfringens STCC 3627, Staphylococcus aureus KFCC 11764 및 Salmonella typhimurium ATCC 14028). All tested pathogens were not inhibited in broth containing 100$\mu\textrm{g}$/$m\ell$ of Areca catachu L. Water extract but its extract strongly inhibited the growth of Eubacterium limonsum STCC 10825, Bacteroides fragilis KCTC 5013, Clostridium perfringens ATCC 3627 and Salmonella typhimurium ATCC 14028 at 1,000 to 2,000$\mu\textrm{g}$/$m\ell$ of concentration. Escherichia coli ATCC 25922 and Staphylococcus aureus KFCC 11764 hardly grew in broth containing 2,000$\mu\textrm{g}$/$m\ell$ of Terminalia chebula Retz. water extract.

The Crystal Structure of Licarin-B $(C_{20}H_{20}O_4)$, A Component of the Seeds of Myristica fragrans

  • The crystal structure of licarin-B, a component of Myristicae Semen was determined by single crystal X-ray diffraction analysis. Crystal of the compound, which was recrystallized from the mixture of hexane and ether, is monoclinic with a=12.740(1), b=7.219(1), c=9.284(1) ${\AA}$, ${\beta}=94.75(1)^{\circ}$, $D_x=1.26$, $D_m=1.27\;g/cm^3$, space group P21, and Z=2. The structure was solved by direct method and refined by least-squares procedure to the final R value of 0.040 for 1532 independent reflections ${F{\ge}3{\sigma}(F)}$. The compound is a dimeric phenylpropanoid, and belongs to the neolignan analogues. The molecules are arranged along with the screw axis. The intermolecular contacts appear to be the normal van der Waals' forces.

The Effect of the Water Extracts of Digestive Medicinal Plants on the Shelf-life of Pork Patties

  • This study was performed to examine the possibility of water extracts of several digestive medicinal plants (DMPEs), such as Amomum tasoko, Alpinia oxyphylla, Citrus unshiu, and Myristica fragrans, as a natural antioxidant. Total phenol contents of each extract were expressed as gallic acid equibalents (GAE) and those were significantly different among A. tasoko ($39.87{\pm}5.77$ mg GAE/g), A. oxyphylla ($30.28{\pm}3.36$ mg GAE/g), C. unshiu ($28.13{\pm}5.01$ mg GAE/g) and M. fragrans ($6.36{\pm}0.30$ mg GAE/g) (p<0.05), and extract of A. tasoko showed significantly higher antioxidative effect than butylated hydroxyanisole (BHA) on linoleic acid peroxidation at 72 h after incubation (p<0.05). Addition of extracts in pork patties did not affect the pH value and total microbes during cold storage. However, thiobarbituric acid reative substances (TBARS) of treated patties were lower in dose dependant manner than that of control as storage period increased (except patties treated with C. unshiu extract), and patties treated with 0.5% A. tasoko extract showed no significant difference with patties treated with 0.5% BHA at day 7.

The Crystal Structure of a $\beta$-Allyl Type Phenylpropanoid 2-(4-allyl-2, 6-dimethoxyphenoxy)-1-(4-hydroxy-3-methoxyphenyl) propan-1-ol, from the Seeds of Myristica fragrans

  • The structure of a $\beta$-allyl type phenylpropanoid was determined by single crystal X-ray diffraction analysis. The compound was recrystallized from a mixture of n-hexane and benzene in monoclinic crystal system with a = 24.782 (2), b = 10.537 (1), c = 7.871 (1) ${\AA}, \beta=95.74$ (1)$^\circ, $D_x$=1.216, $D_m$=1.22g/$cm^3$, space group $P2_1/a$, and Z=4. The structure was solved by direct method and refined by least-squares procedure to the final R value of 0.054 for 2824 observed reflections {$F{\geq}3\sigma(F)$}. The molecular geometry shows a most stable trans-form with respect to the bulky phenyls, and this conformation is settled by an intramolecular hydrogen bond. In the crystal, the molecules are arranged along with the screw axis, and stabilized by the $O{\cdot}H{\cdots}O$ type intermolecular hydrogen bonds. The other intermolecular contacts appear to be the normal van der Waals' interactions. The compound is a dimeric phenylpropanoid, and belongs to the neolignan analogues.

Inhibitory Activity of Edible Plant Extracts on Proliferation of Human Umbilical Vein Endothelial Cells (HUVECs)

  • Thirteen edible plants previously reported to show inhibitory activities on farnesyl protein transferase (FPTase) and phosphatase of the regenerating liver-3 (PRL-3) were evaluated for inhibitory activity on the proliferation of human umbilical vein endothelial cells (HUVECs). Four plant extracts, Oenothera erythrosepala, Perilla frutescens, Panicum miliaceum, and Quercus acutissima, significantly inhibited the proliferation of HUVECs induced by the basic fibroblast growth factor (bFGF) without cytotoxicity at 100 ${\mu}g/mL$. Myristica fragrans, Rosmarinus officinalis, and Syringa patula also showed inhibitory activity on the proliferation with only mild cytotoxicity.

  • The screening of anti-cancer activity for the MeOH extracts of 163 natural sources, which were registered as edible plants by Korea Food & Drug Administration, exhibited 9 extracts to have significant inhibitory effects on farnesyl-protein transferase (FPTase) and phosphatase of regenerating liver-3 (PRL-3). In order to confirm the inhibitory activity of these active extracts, the activity assay was repeated for some fractions obtained from the active extracts using Medium Pressure Liquid Chromatography (MPLC). Some fractions of Carya illinoensis, Chlorella vulgaris, Panicum miliaceum, Perilla frutescens, Rosmarinus officinalis showed over 50% inhibitory activity on FPTase as well as those of Capsella bursa-pastoris, C. illinoensis, C. vulgaris, Coix lacrymajobi, Myristica fragrans, P. miliaceum, R. officinalis did over 50% inhibitory activity on PRL-3.

Rosemary, Sage, Clove 및 Nutmeg의 휘발성 및 비휘발성 성분의 항산화성 (Antioxidative Effects of Volatile Oil and Oleoresin Extracted from Rosemary, Sage, Clove and Nutmeg)

  • 천연 향신료의 항산화력을 탐색하기 위하여 항산화력이 있다고 보고된 rosemary, sage, clove, nutmeg 등 4종의 휘발성 정유성분 및 비휘발성 추출물을 수증기 증류법 및 용매 추출법을 써서 추출하였다. 항산화성은 lard를 기질로 하여 랜시매트법과 POV를 측정하여 비교한 바, 다음과 같은 결과를 얻었다. Rosemary, sage, clove 및 nutmeg 등 4종의 향신료를 랜시매트법에 의하여 휘발성 정유성분의 유도기간을 측정한 결과, 농도를 0.01, 0.05, 0.1%로 증가시켰으나, 4종 공히 유의적인 차이가 없어 항산화력이 없는 것으로 평가되었다. 상기 4종 향신료의 비휘발성 정유성분을 랜시매트법에 의하여 유도기간을 측정한 결과, 농도를 0.01, 0.05, 0.1%로 증가시킴에 따라, 항산화성이 증가하였고, 특히 rosemary, nutmeg가 항산화효과가 컸다. 4종 향신료의 항산화력을 POV를 측정하여 비교한 결과, 랜시매트법으로 측정한 결과와 동일함을 알 수 있었다. 따라서, lard사용 제품류에 상기 4종 향신료의 비휘발성 정유성분을 0.1% 첨가시키면 현저한 항산화효과를 얻을 수 있을 것이다.

Staphylococcus aureus와 Salmonella gallinarum에 대한 한약재의 항균활성 (Antimicrobial Activity of Medicinal Herbs against Staphylococcus aureus and Salmonella gallinarum.)

  • 40여종의 한약재추출물을 제조하여 가금질병에 관련이 있는 세균에 대한 항균활성을 조사하였다. 오미자, 천련자, 소목 및 오배자 추출물은 Salmonella gallinarum에 대해 높은 항균활성을 보였으며 향유, 육두구, 초두구, 지실, 하고초, 소목 및 오배자 추출물은 Staphylococcus aureus에 대해 높은 항균활성을 보였다. 천련자, 소목 및 오배자 추출물의 S. gallinarum에 대한 최소저해농도는 1.2 ㎎/ml이며 오배자 추출물의 S. aureus에 대한 최소저해농도는 0.6 mg/ml로 가장 낮은 농도에서 항균활성을 보였다. 열에 의한 항균활성 변화에 있어서 오미자와 오배자 추출물은 S. gallinarum에 대한 항균럭이 감소하였고 초두구와 소목 추출물은 S. aureus 에 대해 항균력이 안정하였다. pH에 의한 항균활성의 변화에서 S. gallinarum에서 오미자가, S. aureus에서는 향유, 지실 및 오배자는 산성역에서는 비교적 안정하였으나 알칼리역에서 불안정하였으며 소목은 산성과 알칼리역에서 안정하였다. 항균력이 가장 우수한 오배자와 소목 추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 0, 100, 300 및 500 ppm의 추출물을 첨가하여 균주의 증식 억제효과를 조사하였다. 배양후 24시간에 무첨가구에 비해 100 ppm이상의 오배자 추출물 첨가시 시험 균의 증식을 현저히 억제하였고, 소목 추출물에서도 무첨가구보다 300 ppm 이상의 추출물 첨가시 시험 균의 증식을 현저히 억제하였다. 이런 결과를 종합하면 가금 질병을 유발하는 S. gallinarum과 S. aureus를 예방 할 수 있는 사료첨가제 개발 가능성을 확인하였다.
