Optimization of Media Composition and Culture Conditions for the Mycelial Growth of Coriolus versicolor and Lentinus edodes (Coriolus versicolor와 Lentinus erodes의 영양배지 조성 및 배양조건의 최적화)

  • The media compositions and culture conditions were optimized for mycelial growth of Coriolus versicolor and Lentinus edodes. Media composition for optimal growth of Coriolus versicolor was 2.0% glucose 0.4% peptone and 0.6% yeast extract. Media composition for optimal growth of Lenttnus edodes was 2.0% glucose 2.0% starch 0.4% bacto-soytone and 0.6% yeast extract. The media supplemented with KH2PO4, 0.046% KH2PO4 0.1% and MgSO4, .7H2O 0.05% supported better mycelial growth than the media without mineral salts. The optimum temperature for mycelial growth ranged from $25^{\circ}C$-28$^{\circ}C$. The optimum pH range for mycelial growth of Coriolus versicolor was 5.2~5.6 while that of Lentinus edodes appeared to be 5.75.

Suitable Conditions for Mycelial Growth of Phellinus spp.

  • The fungus Phellinus is a mushroom that is widely used medicinally. The optimal conditions for mycelial growth of 13 strains of the fungus were investigated. Mycelial growth was optimal at 25$^{\circ}C$ and was uniformly minimal at 15$^{\circ}C$ and 35$^{\circ}C$. Growth was optimal at pH 6$\sim$7. The mycelial phenotype was best promoted by growth using Potato Dextrose agar, Hamada, Glucose peptone, and Yeast-Malt media, whereas Czapek Dox, Hennerberg, and Lilly media were the most unfavorable for the mycelial growth of Phellinus spp. Glucose, sucrose, fructose, and dextrin were the most suitable carbon sources for mycelial growth, while lactose, maltose, and galactose were unsuitable. Among tested nitrogen sources, ammonium phosphate, potassium nitrate, and arginine best promoted mycelial growth, while alanine, urea, and histidine least promoted mycelial growth.

Favorable Culture Conditions for Mycelial Growth of Korean Wild Strains in Ganoderma lucidum

  • Ganoderma lucidum (Fr.) Karst (Polyporaceae), belonging to basidiomycota, is one of the most famous medicinal mushrooms. This study was carried out to investigate favorable mycelial growth conditions, such as pH, temperature, growth media, carbon sources and nitrogen sources of Korean strains in G. lucidum. The most suitable temperature for the mycelial growth was obtained at $30^{\circ}C$. In general, optimal temperature range for the mycelial growth was found at $25{\sim}30^{\circ}C$. This Mushroom has a broad pH range ($5{\sim}9$) for its mycelial growth and mostly favorable growth was found at pH 5. Generally, Hamada, Glucose peptone, YM, Mushroom complete and Lilly media were the most suitable for the mycelial growth of G. lucidum. Among 10 different carbon sources, dextrin, galactose and fructose were best but the rest of other carbon sources also facilitated the growth of mycelia. The most suitable nitrogen sources were ammonium acetate, glycine, arginine and calcium nitrate, but to a certain extent, all of the supplemented nitrogen sources also stimulated the mycelial growth.

  • Lignosus rhinocerus is a macrofungus that belongs to Polyporaceae and is native to tropical regions. This highly priced mushroom has been used as folk medicine to treat diseases by indigenous people. As a preliminary study to develop a culture method for edible mushrooms, the cultural characteristics of L. rhinocerus were investigated in a range of culture media under different environmental conditions. Mycelial growth of this mushroom was compared on culture media composed of various carbon and nitrogen sources in addition to C/N ratios. The optimal conditions for mycelial growth were $30^{\circ}C$ at pH 6 and 7. Rapid mycelial growth of L. rhinocerus was observed on glucose-peptone and yeast extract peptone dextrose media. Carbon and nitrogen sources promoting mycelial growth of L. rhinocerus were glucose and potassium nitrate, respectively. The optimum C/N ratio was approximately 10 : 1 using 2% glucose supplemented as a carbon source in the basal media.

  • Characteristic growth patterns of Cordyceps militaris isolates on various media, under varying light conditions and at varying incubation periods were examined. Light was found to be the most critical single factor in determining the density, texture, and pigmentation of the mycelial culture of the fungus. However, under the light condition, the degree of pigmentation and mycelial density were found to be affected by the incubation period and type of medium. Irrespective of the variations in medium type or incubation period, there was no pigmentation of the mycelium under dark condition. Radial growth of the mycelium was faster under dark incubation rather than under light incubation. Abundant mycelial density and darkest pigmentation of C. militaris isolates were produced in nutritionally rich media like SDAY, SMAY and CZYA, suggesting that these media may fulfill all the requirements for vegetative growth of the fungus. Growth characteristics of C. militaris isolates could be easily observed by the simple agar culture method, which would be useful to characterize the phenotypic characteristics of large number of pure cultures of the fungus under given conditions of growth factors such as medium, light and temperature.

Characteristics of mycelial growth and fruit body of Sparassis latifolia strains and selection of suitable incubation conditions in liquid spawn

  • Sparassis latifolia is called "Cauliflower Mushroom" and is known as an edible mushroom that has high content of ${\beta}$-glucan. Recently, artificial cultivation of S. latifolia has been done by bottle, plastic bag and wood cultivation in Korea. However it is not widely used because there are low incubation ratio and yield. For the high efficiency of production, we aim to find the superior strains and media for better mycelial and fruit body growth. First, we analyzed the genetic relationship among 31 strains and divided five groups with three kinds of URP primers. And then ten strains were selected from five groups based on the experiment of mycelial growth. The suitability of media for mycelial growth was different according to media type. The suitable solid and liquid media for mycelial growth of S. latifolia isolates were PDA and M2, respectively. In addition, with regard to C/N ratio, the mycelial growth increased even until C/N 160. Second, we investigated the production of fruitbody of the strains by plastic bag cultivation. The substrate was mixed with larch sawdust, corn flour, and wheat flour (8:1:1, v/v). Moisture content of substrate was controlled by about 60% with 10% molasses solution. Out of 31 strains, 19 strains formed primordia. The eight strains produced more than 140g/1kg in fresh weight. Third, molasses culture media was selected for the mycelial growth. And molasses suitable sugar content and input aeration were around 8Brix% and 0.3~0.6vvm, respectively. The longer the incubation period is, the more dried weight of mycelia increased, but medium volume decreased. Therefore, the best incubation period was 9 to 11 days depending on strains. In the future, research project entitled development of culture system and new variety for stable production of S. latifolia will be considered as a new item.

  • To elucidate the optimal conditions of mycelial growth of Pholiota adiposa, the effect of a kinds of media, incubation temperature and shaking culture were investigated. Mushroom complete medium was selected as superior media for mycelial growth in this fungus. The optimal temperature for mycelial growth of this fungus shows at 25℃. Shaking culture could be obtain the mycelia in two or three times than the stationary culture.

Physicochemical Requirement for the Vegetative Growth of Schizophyllum commune Collected from Different Ecological Origins

  • Schizophyllum commune is an edible and medicinal mushroom widely distributed in the world. The optimal growth conditions for the mycelia of 10 strains of the fungus were investigated. The temperature suitable for the mycelial growth and density was obtained at $30{\sim}35^{\circ}C$. Among the tested conditions, the minimum mycelial growth was found at $15^{\circ}C$. In case of pH, the most favorable growth was found at pH 5. The results indicated that this mushroom well adapted to high temperature and low pH for its mycelial growth. Considering growth phenotype of mycelia, Hamada, Hennerberg, PDA and YM were the most suitable and Lilly, Glucose triptone, Glucose peptone and Hoppkins were the most unfavorable among tested media for the mycelial growth of S. commune. Out of tested carbon sources, dextrin and fructose were the most suitable and lactose, mannose and sorbitol were the unsuitable for the fungus. Compact mycelial density was obtained from most of the carbon sources. Among used nitrogen sources, calcium nitrate, potassium nitrate and alanine were the most appropriate and the most incompatible were ammonium phosphate, histidine, urea and arginine for mycelial growth of S. commune on the culture media. Calcium nitrate, histidine and potassium nitrate showed moderately thin or thin, and rest of nitrogen sources showed compact or moderately compact mycelial density.

  • The mycelial growth of Pythium graminicola, P. ultimum, P. rostratum was best at -3 osmotic potential. P. monospermum was best grown at -8 bars osmotic potential. Mycelial growth by Pythium spp. was prevented at -42 bars for P. graminicola, P. ultimum and P. monospermum. Sporangial formation by P. graminicola and P. ultimum was influenced by water potential in much the same way as mycelial growth: -3 bars was maximum and -42 bars or below was minimal. Pythium spp. tested was grown faster on PDA than other media including oat meal agar. P. ultimum was best grown on any media among Pythium spp. tested.

Sawdust Media Affecting the Mycelial Growth and the Fruiting Body Formation of Sparassis crispa

  • Six strains of Sparassis crispa such as S. crispa DUM-01, DUM-02, DUM-03, DUM-04, DUM-05, and DUM-06 were tested for their mycelial growth on 6 different kinds of sawdust media and primordial formation on 10 different compositions of larch sawdust media. The highest mycelial growth was recorded on the larch sawdust. Of the 6 strains of S. crispa, S. crispa DUM-04 recorded the favorable formation of primordia. The primordial formation of S. crispa DUM-04 was more favorable on L-3 medium than 9 kinds of larch sawdust media. When S. crispa DUM-04 was cultured on the media of larch sawdust+pine sawdust, the formation of its fruiting bodies was more outstanding on the media of larch sawdust+pine sawdust than those of larch sawdust.
