Hohenbuehelia petaloides의 배양학적 특성에 관한 연구 (Studies on the Cultural Characteristics of Hohenbuehelia petaloides)

  • This study was carried out to obtain the basic data on artificial culture of Hohenbuehelia petaloides. The optimum medium are glucose peptone medium (GP), Hennerberg medium, Phellinus igniarius medium (PIM), Lentinus edodes medium (LEM), Czapek dox medium. The optimum condition for the mycelial growth was $30^{\circ}C$ and pH 6.0. The carbon sources such as dextrine, fructose and lactose were favorable to mycelial growth. The optimal concentrations of carbon sources are 10% dextrin and fructose. As nitrogen sources, tryptone, casamino acid and histidine appeared to be favorable. The optimal concentrations of nitrogen sources are 1% soy tone and 0.3% ammonium nitrate. The optimal concentration of yeast extract is 0.4%. The mineral nutrients of $KH_2PO_4$, $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were effective and the optimal concentrations were 0.046, 0.1 and 0.05%, respectively.

양식 가물치(Channa argus)에 대한 Saprolegnia sp.의 병리학적 특성과 물곰팡이의 생장을 제어하는 정유의 영향 (Pathological Characteristics of Saprolegnia sp. to Snakehead and Effect of Essential Oils to the Growth of the Water Mold)

  • 양식 가물치로부터 Saprolegnia sp.를 분리하고 그들의 일부 생리학적 특성과 제어를 위한 식물 추출물(精油)을 탐색하였다. 분리된 Saprolegnia sp. 영양체 생장 최적 온도는 $25^{\circ}C$이었고, 최적 pH는 6.0이었으며, 최적 NaCl 농도는 0%이었다. 또한 phosphate 10 mM 농도와 casamino acid 10mg/L농도에서 균사의 생장은 잘되었다. Saprolegnia sp.의 제어 정도를 조사하기 위해 쑥, 편백, 측백나무로부터 정유를 분리하여 10, 100, 500, 1,000, 1,500, 2,000ppm에서 처리한 결과 쑥 정유는 처리농도 10 ppm 이상 농도에서부터 억제 양상이 나타나다가 농도가 높아질수록 더 많은 억제양상을 나타냈다. 측백과 편백나무 정유를 처리한 것에서는 10ppm이상 처리 농도에서부터 억제되기 시작하여 그 이상의 처리 농도에서는 상당한 균사 생장 제어 효과를 나타내다가 500 ppm 이상 처리 농도에서는 완전한 제어 효과를 나타냈다. 또한 Saprolegnia sp.에 감염된 가물치의 조직 병리학적인 조사를 실시한 결과 균사가 감염된 근육조직 부분에는 깊은 곳까지 균사가 침투하였고, 아가미의 세포는 곤봉형태를 나타냈으며, 간조직 세포에서는 일부가 괴사되었고 부분적으로 ceroid 물질이 침착되어 있었다. 또한 비장조직 세포에서는 일부 괴사된 세포를 관찰하였고, ceroid 물질의 침착을 볼 수 있었다. 신장조직은 세뇨관의 상피세포에서 일부 괴사를 관찰하였고, 조혈 조직에는 ceroid 물질의 침착상태가 나타났다.

고추 갈색점무늬병원균 (Cercospora capsici)의 배양특성과 다량 포자형성법 (Sporulation of Cercospora capsici causing Cercospora leaf spot of Pepper)

  • 고추 갈색점무늬병(Cercospor capsici)병원균의 균사특성과 포자형성 방법 조사하였다. C. capsici 병원균의 균사생장 적온은 $25^{\circ}C$이였고 $5^{\circ}C$ 이하와 $35^{\circ}C$ 이상에서는 균사생장이 일어나지 않았다. pH 4.0~pH 8.0의 넓은 범위에서 균사생장에 차이가 없었고 일장도 균사생장에는 영향을 주지 않는 것으로 나타났다. 분생포자 다량 형성 시키기는 배지로는 0.5% 고추잎배지가 가장 좋았으며 배지 상에서 포자를 다량으로 형성시키는 방법은 고추잎배지에 균사현탁액을 도말 한 후 2,3일간 $20^{\circ}C$에서 배양한 후 1ml/접시 살균수를 첨가하여 배지표면을 긁어 자극을 준 후 2, 3일정도 배양을 하면 포자형성이 양호하였다.

Reutilization of Enokitake Cultural Waste as Lentinus edodes Cultivation Substrate

  • The availability of enokitake cultural waste for Lentinus edodes cultivation was investigated, although hardwood sawdust has traditionally been used as a substrate for this fungus. Firstly, physiochemical characteristics of cultural waste were analysed. Secondly, mycelial growth characteristics and fruiting yields of L. edodes on waste treated in some methods were determined. Physiochemical characteristics of enokitake cultural waste showed that the millwaste complex was a little degraded by enokitake fungus and suggested the probability that most component lost by enokitake could be rice bran. Mycelia of L. edodes grew and fruited well on waste supplemented by fresh rice bran and Quercus sawdust although didn't on waste only. Mycelial growths of these fungi on waste were accelerated when supplemented by rice bran to the percent of 40(w/w) but decreased or suppressed at above ratios(30, 40%, w/w). Supplementations of oak sawdust at above 40%(w/w) of the waste and rice bran at 20%(w/w) of the sawdust allowed such a good mycelial growth as to be selected as a pertinent mixing ratio for fruiting medium. A fruiting yield on enokitake cultural waste supplemented by oak sawdust (at 40% of the waste, w/w) and rice bran (at 20% of the sawdust, w/w) was not inferior to that on oak sawdust supplemented by rice bran only (at 20% of the sawdust, w/w). These results indicated strongly the potentiality of enokitake cultural waste as raw materials for shiitake cultivating substrates.

Physiological Characteristics of Symbiotic Fungi Associated with the Seed Germination of Gastrodia elata

  • This study was carried out to investigate the possibility for seeds germination of Gastrodia elata using symbiotic fungi. Since seeds of G. elata are very small and lack an endosperm and other nutrients, their germination is difficult without requirement for external nutrients. Out of twenty six isolates collected from protocorms of G. elata and roots of native orchids inhabited in wild, two strains(H-2 and H-21) were observed to stimulate the seed germination of G. elata. The seed germination of G. elates was excellent on oak tree leaves medium. The optimal conditions for mycelial growth of symbiotic fungi were $25^{\circ}C$ and pH 6.0, respectively. The mycelial growth of H-2 strain was excellent on YMA medium, while H-21 was poor on PDA medium. In case of carbon sources, the mycelial growth of H-2 and H-21 was good on media supplemented with glucose and dextrin, respectively. Calcium nitrate was good for mycelial growth of H-2 strain as a nitrogen sources, whereas urea was effective to H-21 strain.

플라스크배양에서 버들송이버섯 균사체 배양에 관한 연구 (A Study on the Mycelial Growth of Agrocybe aegerita in Flask Culture.)

  • 버들송이버섯의 균사체 대량 생산의 기초자료를 얻을 일환으로 균사의 영양생장에 필요한 적합한 조건을 연구하고자 균사생장을 위한 최적 배양 조건 및 영양원을 조사한 결과는 다음과 같다 5종의 공시배지 중 ME 배지에서 버들송이 버섯의 균사생장 및 밀도가 가장 양호하였으며, 균사생장최적 온도는 $25^{\circ}C$이고, 최적 pH는 5.5이며 최적 배양일수는 12일이었다. 버들송이 버섯의 균사생장을 위한 최적 배지조성은 탄소원에서는 다당류인 dextrin이었으며, 최적 탄소원인 dextrin의 적정농도는 3% (w/v)였다. 질소원에서는 유기태 질소원인 yeast extract이었고, yeast extract 최적농도는 2% (w/v)였으며, 최적 무기염류는 MgSO$_4$와 KH$_2$PO$_4$이고, 최적 농도는 MgSO$_4$0.05% (w/v), KH$_2$PO$_4$0.15 (w/v)를 혼합하였을 때 가장 많은 생산량을 보였다. 기본배지인 ME 배지와 본 연구에서 얻어진 최적배지로 액체 배양하여 균체량을 비교한 결과 기본배지는 배양 12일째 최대 건조 균체량 7.59 g/l을 얻었으며, 최적배지는 배양 12일째 17.6 g/l의 최대 건조 균체량을 얻어 기본배지 보다 최적배지의 균체량이 훨씬 높은 균사체 생산수율을 보였다.

Dichlofluanid 저항성 및 감수성 잿빛곰팡이병균(Botryits cinerea)의 약제 반응과 생태 적응력과 관련된 특성 (Chemical Responses and Fitness-Related Characteristics of Dichlofluanid-Resistant and -Sensitive Botrytis cinerea Isolates)

  • Nine hundred and ninety-two isolates of Botrytis cinerea were obtained from infected strawberries, tomatoes and cucumbers in Taejon, Gongju, Puyo, Nonsan and Kimhae in Korea. Six hundred forty-two (64.7%) isolates were benomyl resistant (BR), 245 (24.7%) were procymidone resistant (PR), and 105 (10.6%) were dichlofluanid resistant (DR). In the minimum inhibitory concentration (MIC) test, DR isolates showed mycelial growth on the PDA incorporated with 100 or 500 $\mu\textrm{g}$/ml of dichlofluanid, while dichlofluanid sensitive (DS) isolates did not grow on the PDA incorporated even with 10 $\mu\textrm{g}$/ml of dichlofluanid. Chemical concentrations for inhibition of spore germination were much lower than those for inhibition of mycelial growth. IC50 values, the effective concentrations for 50% inhibition of spore germination, for DR were 0.11~0.29 $\mu\textrm{g}$/ml, whereas they were 0.04~0.09 $\mu\textrm{g}$/ml for DS isolates. In comparison of fitness-related characteristics such as virulence, sclerotial formation, and sporulation, DR isolates were inferior to DS isolates. However, mycelial growth was little different between DR isolates and DS isolates.

Comparison of Cenangium Dieback Fungus Isolated from Three Different Species of Pine

  • Dieback of pine branches or twigs with brown needles occurs most commonly on Pinus species after severe winter in Korea. In this study, Cenangium ferruginosum was isolated from infected stems, branches, and twigs of Pinus koraiensis (C1), P. densiflora (C2), and P. thunbergii (C3). Morphological and cultural characteristics of the isolates were than compared. There were no significant differences in the morphological characteristics of conidia and ascospores produced by the three isolates. However, cultural differences were observed among the isolates. Optimum temperatures for mycelial growth of C1, C2, and C3 were 15, 20, and $20^{\circ}$, respectively. C1 produced a few conidia and no ascospores, while C2 and C3 produced abundant ascospores and conidia. While optimum temperatures for mycelial growth ranged from 15 to $20^{\circ}$, mycelial growth was also relatively good at lower temperatures of 5-$10^{\circ}$. Conidiomata and conidia were produced on MSA (malt extract soya peptone agar) after 25-30 days of incubation in the dark at $15^{\circ}$. Apothecia were produced by altering culture condition from 15 to $20^{\circ}$, and incubating for 35-60 more days. Optimum temperature for ascospore and conidium germination was $20^{\circ}$. RAPD analysis revealed that there was high similarity of 0.78 between C2 and C3, and low similarity of 0.31 between C2 or C3 and C1.

Cultural Characteristics of A Medicinal Mushroom, Phellinus linteus

  • For comparison of mycelial colonization of Phellinus linteus on logs, several techniques of inoculation were tested; sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation. The mycelial colonization of P. linteus on logs was good in the treatment of sterilized short log inoculation, but poor in the traditional methods such as drilling inoculation and log-end sandwich inoculation. The initial mycelial growth and the full mycelial colonization of P. linteus in logs were the best in case of 20 cm logs under the condition of 42% moisture content. Also, the initial mycelial growth of P. linteus was accelerated over 12 hours of sterilization. Burying method of logs after $5{\sim}6$ months of incubation was the best for formation of basidiocarps of P. linteus. The formation of fruiting body of P. linteus was quite good in the cultivation house at $31{\sim}35^{\circ}C$ and over 96% of relative humidity.

단감나무 둥근갈색무뉘병원균 Pestalotiopsis theae의 배양적 특성 (Studies on Cultural Characteristics Pestalotiopsis theae causing Leaf Blight on Oriental Persimmon Tree)

  • 단감나무 둥근 갈색무늬병을 일으키는 P. theae, SP2, SP3 및 P. longiseta의 균사 생장과 포자 형성에 미치는 여러 요인을 조사하였다. 균사 생육 및 포자 형성 적온은 $25{\sim}30^{\circ}C$으로 나타났으며, $35^{\circ}C$$0{\sim}8^{\circ}C$에서는 균사 생장 및 포자 형성이 불량하거나 정지되었다. 균사 생육 및 포자 형성 최적 pH은 4.5~5.0과 6.0~6.0으로 나타났다. 모든 공시 균은 Leonian, PSA 그리고 오트밀 배지에서 양호한 균사 생육을 보였으며, 포자 형성은 오트밀과 PSA 비재에서 많은 량의 포자를 형성하였으나 대체적으로 PSA 배지에서 많은 량의 포자를 형성하는 것으로 나타났다. 질소원 typtone과 glycine, 탄소원 starch, dextrose, galactose 그리고 lactose 등에서 균사 생장이 가장 좋았다. 또한 질소원 glutamic acid, peptone과 tryptone, 탄소원으로 starch, sucrose와 galactose은 포자형성에 영향을 주었다. 비타민은 균사 생육 및 포자 형성에 거의 영향을 미치지 않았다. 배양 기간에 따른 배지의 pH 변화에서 배양전 pH 7인 배지는 배양 10일 후에는 SP2, SP3균은 각각 pH 4.5~4.7로 P. theae와 P. longiseta은 각각 5.0~5.4로 낮아졌지만, 배양전 pH가 5.0인 배지는 배양 10일 후에 모든 균이 pH 4.5~4.7로 유지되었다.
