Lignosus rhinocerus is a macrofungus that belongs to Polyporaceae and is native to tropical regions. This highly priced mushroom has been used as folk medicine to treat diseases by indigenous people. As a preliminary study to develop a culture method for edible mushrooms, the cultural characteristics of L. rhinocerus were investigated in a range of culture media under different environmental conditions. Mycelial growth of this mushroom was compared on culture media composed of various carbon and nitrogen sources in addition to C/N ratios. The optimal conditions for mycelial growth were $30^{\circ}C$ at pH 6 and 7. Rapid mycelial growth of L. rhinocerus was observed on glucose-peptone and yeast extract peptone dextrose media. Carbon and nitrogen sources promoting mycelial growth of L. rhinocerus were glucose and potassium nitrate, respectively. The optimum C/N ratio was approximately 10 : 1 using 2% glucose supplemented as a carbon source in the basal media.

Mycelial growth of L. edodes by pretreatments of softwood was studied on a sawdust medium. The sawdust used was from the following softwood species : Larix leptolepis, Pinus densiflora and Pinus koraiensis. The pretreatment consisted of cold-water (48 h), hot-water (3 h) and steam extractions (3 h) at a ratio of 500 g : 3,000 mL (sawdust : distilled water). The sawdust medium was a mixture of 76% sawdust, 20% rice bran, 3% glucose, 0.4% potassium nitrate and 0.6% calcium carbonate. Following sawdust pretreatments proved most suitable : L. leptolepis (steam extraction), P. densiflora (hot-water extraction) and P. koraiensis (hot-water extraction). Mycelial growth on P. koraiensis sawdust increased in proportion to an increase in hot-water extraction time. Mycelial growth was optimum on the sawdust extracted for 12 hours, hot-water extraction beyond this period proved unsuitable. With the exception of P. densiflora at 100 ㎍/mL, antifungal activity occurred in every sample. Maximum inhibition of mycelial growth was obtained from following concentration of hot-water extractives : P. densiflora (104 ㎍/mL) and P. koraiensis (104 ㎍/mL). This study has provided useful preliminary information for the cultivation of L. edodes.
Cultural characteristics of Cordyceps militaris preserved in EFCC, Kangwon National University were investigated for the mass production. The higher mycelial density of C. militaris was observed in Sabouraud's yeast and Yeast Malt agars, but the higher mycelial growth in Mushroom Minimal agar than other agars. The mycelium of C. militaris was observed to grow well at $25^{\circ}C$ and pH 6.0 respectively. The dextrose was found the best suitable energy source among the carbohydrates used for its mycelial growth, while the fructose or lactose observed to be well for mycelial growth. Hemoglobin was observed to be the best among the protein sources used for mycelial growth, while tryptone found to be the best in the spore formation. Similarly, the mycelial growth was best in mineral salts of $KH_2PO_4$ or $K_2HPO_4$ and the optimum C/N ratio was 100 : 1.

The influences of inoculum size, pH, and medium composition on mycelial growth and exopolysaccharides (EPS) production were investigated in shake flasks and in a bioreactor. The optimum inoculum size for both mycelial growth and EPS production was identified to be 10% (v/v) in shake flask cultures. The optimal initial pH for mycelial growth and EPS production in shake flask cultures were found to be 5.0 and 7.0, respectively. However, the optimal pH was 5.0 for both mycelial growth and EPS production in bioreactor cultures where the pH was regulated. The optimal mass ratio of the two major carbon sources, glucose to dextrin, was 1:4. The optimal mass ratio of the two major nitrogen sources, yeast extract to soy tone peptone, was 2:1. When 500 mg $1^{-1}$ of $MnSO_4-5H_2O$ was added to the bioreactor culture, both mycelial growth and EPS production were enhanced by approximately 10%. Under the optimized conditions, a mycelial biomass of 9.85 g $1^{-1}$ and an EPS concentration of 4.92 g $1^{-1}$ were obtained in 4 days.
For the practical using of liquid spawn we carried out selection test of nutrient sources, cultural methods and cultural apparatus for liquid spawn production of oyster mushroom(ASTI 2001, ASTI 2018, ASTI 2072, ASTI 2016, ASTI 2070, ASTI 2180, ASTI 2183, ASTI 2042). The optimal temperature and pH range for mycelial growth of Pleurotus species were $25^{\circ}C\;to\;30^{\circ}C$ and 5.5 to 6.5, respectively. The effect of carbon sources, nitrogen sources and mineral salts on the mycelial growth was studied using petridish culture. Generally, the disaccharides and polysaccharides showed good effect for mycelial growth of Pleurotus species, and the polysaccharides were superior to the other classes of carbon sources for mycelial growth. For the mycelial growth of the 8 oyster mushroom stains, soybean flour was superior to the other kinds of nitrogen sources. On the other hand, addition of mineral salts did not affect, and even poor under certain mineral salts, the mycelial growth of the 8 oyster mushroom stains. The brown sugar selected out the carbon source of the agricultural medium. Also the soybean flour selected out the nitrogen source of agricultural medium. In the medium selection, we selected out agricultural optimum medium composed of brown sugar 3%, soybean flour 0.3%, potassium phosphate 0.05%, magnesium sulfate 0.05%. Under the 250 ml erlenmeyer culture, the effects of such factors as the inoculum rate, the working volume, cultural method and flask shapes on the mycelial growth were examined. The optimal inoculum rate and working volume on the mycelial growth of oyster mushroom was 2 mycelial disk (diameter 6 mm) and 100 ml, respectively. The shake flask culture was enhanced the mycelial growth than at the erlenmeyer flask. Pulp form growth of mycelium in the erlenmeyer flask culture was obtained in the culture with glass rod of length 50 mm, diameter 10 mm.
This study was carried out to investigate effect of adding tobacco waste to promote oyster mushroom, Pleurotus ostreatus, mycelial growth and control of Trichoderma disease. Tobacco waste was treated on the surface of rice straw inoculated with oyster mushroom strains. The mycelial growth treated with 0.1g tobacco waste was somewhat faster than non-treatment. In the case of mixed media with 1.0g tobacco waste, mycelial growth was increased compare to control, but adding 2.0 g tobacco waste decreased the mycelial growth. We also treated the tobacco waste on Trichoderma disease. All of the tobacco waste treatment force down the growth of Trichoderma disease. We treated tobacco waste on the surface of rice straw substrates. ASI 2072 mycelial growth was decreased regardless of amount of tobacco waster and method of treatment. Mycelial growth of ASI 2180 was somewhat decreased with treatment of 0.1g and 0.2g In the case of media mixed, mycelial growth of ASI 2072 was decreased in the overall treatment. Mycelial growth of other strains were decreased as only 2.0g treatment. Field test of mycelial growth was conducted using rice straw and waste cotton with substrates. Rice straw culture with treated 3% tobacco waste and cotton waste culture with treated 6% tobacco waste showed the highest yield. There was no effect to Trichoderma disease.

Batch cultures were carried out to optimize the exo-polysaccharide production by liquid cultures of Pleurotus ferulae. Among the various carbon sources, when $5\%$ of glucose was used, the maximum mycelial growth and exo-polysaccharide concentration reached were 8.78 g/l and 3.59 g/l, respectively. Yeast extract and polypeptone were identified as the most suitable nitrogen sources. In particular, when a mixture of $1\%$ of polypeptone and $0.8\%$ of yeast extract was used, 9.52 g/l of mycelial growth and 4.09 g/l of exo-polysaccharide were obtained. In the case of mineral sources, K$_2$HPO$_4$ and MgSO$_4$$\codt$7H$_2$0 were found to be the best mineral sources for mycelial growth and exo-polysaccharide production. Under the optimized culture conditions, the agitation speed and aeration were investigated for mycelial growth and exopolysaccharide production in a jar fermentor. The maximum mycelial growth and exo-polysaccharide concentration at 1.5 vvm and 200 rpm obtained were 13.2 g/l and 4.95 g/l, respectively, after 10 days of culture, which were $76\%$ and $79\%$ higher than those of the basal medium. The specific growth rate was decreased with the increase of mycelial growth. However, the specific production rate of the exo-polysaccharide was proportionally increased with the specific growth rate. The proposed model profiles showed good agreement with the experimental results for the mycelial growth and exo-polysaccharide production. The specific production rate using the optimized medium was higher than that of basal medium.

The objective of this study was to determine effects of corn germ meal(CGM) and condensed molasses fermentation solubles(CMS) as the replacement of rice bran on mycelial growth and density in various mushrooms. The results of this study showed that CGM and CMS might be utilized more efficiently than rice bran in culturing mushrooms. The results are as follows; 1. Mycelial growth of Pleurotus ostreatus was remarkably higher than that of control when the concentration of CGM and CMS was 10%, respectively. However, the mycelial density was lower than that of control. Thus, Considering the mycelial growth as well as density, it was found that 20% supplements of CGM and CMS resulted in the best with 78mm/10d and 71mm/10d, respectively. 2. Mycelial growth of Pleurotus eryngii in the treatment group of 10% CMS and 20% CGM showed 74mm/10d and 67mm/10d, respectively. This result tended to be equal or somewhat lower compared with that of control(74mm/10d). 3. Mycelial growth of Flamulina velutipes was 87mm/10d in the treatment of 10% CMS, 79mm/10d in the treatment of 20% CGM, which showed significantly higher than that of control(56mm/10d). When the contents of CGM and CMS were mixed with the amount of 10-30%, Thus, it was assumed that the quantity of mushrooms may increase with the supplements of CEM and CMS. 4. Mycelial growth of Lentinus edodes in the treatment group of 10-40% CMS and 10-30% CGM showed 58-52mm/10d and 56-49mm/10d, respectively. This result was significantly higher that control(47mm/10d). 5. When CMS and CGM were supplememted with the level of 10-30%, mycelial growth rate of Ganoderma lucidum showed 76-72mm/10d and 74-69mm/10d, respectively, which are higher than those of control.
Some factors affecting mycelial growth of Pleurotus sajor-caju and P. ostreatus. were investigated. The optimal temperature and pH of the medium for mycelial growth of both the species were $25^{\circ}C$ to $30^{\circ}C$, and 5 to 6, respectively. The effect of carbon and nitrogen sources on mycelial growth of the species was studied using modified Waksman's special medium as the basal medium. Generally, the disaccharides and polysaccharides showed good effect for mycelial growth of P. sajor-caju, and the polysaccharides were superior to the other classes of carbon sources for mycelial growth of P. ostreatus. Generally the organic nitrogen compounds showed good mycelial growth of both the species. The mycelial growth of the species were favored by the ammonium nitrogen more than by the nitrate nitrogen source. Asparagine was superior to the other kinds of amino acid. Six percentage of glucose and 0.6% ammonium tartarate was optimum for mycelial growth of P. sajor-caju whereas the optimum concentration for the P. ostreatut was 4% glucose and 0.4% ammonium tatarate.

In order to develop the culture material for breeding of Lentinula edodes, dikaryotic and monokaryotic mycelia were cultured in agar and sawdust medium. The cultivars were used Sanjo No. 701, Sanjo No. 705, Chamaram and Chuje No. 2 in this study. The mycelial growth of the cultivars were highest in PDA except for Sanjo No. 705, but Sanjo No. 705 showed the best mycelial growth in MCM. Regardless of the cultivars, the mycelial growth showed the most stable in MEA. There was no significant difference in mycelial growth among the cultivars in sawdust medium, but it was about 10% faster than that of Sanjo No. 701 and 705. Monokaryotic strains of Sanjo No. 701 and Chamaram showed less mycelial growth than dikaryotic mycelium. Dikaryotic mycelium of Chamaram showed better mycelial growth than that of Sanjo No. 701, but monokaryotic mycelium of Chamaram showed lower mycelial growth than Sanjo No. 701. The selected monokaryotic mycelium has a wide varience of mycelial growth, and the morphologies of the colonies are very diverse, so those are presumed that wide variences of monokaryotic are selected in genetically, and these monokaryotic mycelium are expected to be a good breeding materials.
