Effect of Tree Species, Inoculation Amount and Inoculation Methods on Mycelium Growth and Sclerotia Formatino of Poria cocos Wolf (樹種, 接種量 및 接種方法이 茯笭 菌絲生長과 結笭에 미치는 영향)

  • When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.

Mycelial growth and wood decaying enzymatic activity analysis by various addition rates of oak powder in the liquid spawn of Lentinula edodes (참나무분 첨가에 따른 표고 액체종균의 균체생산 및 효소 활성)

  • This study was carried out to establish a suitable method for liquid spawn production from Lentinula edodes. The optimum production of liquid spawn (OLS) was achieved using soybean meal medium (SMM) with 0.3% of 850 um oak powder and 10-day incubation period and 0.6 vvm aeration volume. OLS showed activities of laccase on ABTS agar plate and carboxymethyl cellulase (CM-cellulase) on CMC agar plate. In case of liquid spawn, fruiting-body development period was delayed approximately 1 day compared to that of sawdust spawn, however, the yield of 153 g per 1.2 kg polypropylene bag was similar to that of sawdust spawn.

Thermophile mushroom cultivation in Cambodia: Spawn production and development of a new substrate, acacia tree sawdust

  • To minimize cultivation costs, prevent insect-pest infestation, and improve the production efficiency of thermophilic mushrooms, plant substrates obtained from local areas in Cambodia were used for production of both spawn and mushrooms. In this experiment, different sawdusts different organic wastes and grain ingredients and analyzed for improvement of spawn-production efficiency. Four thermophilic mushroom species, Pleurotus sajor-caju (oyster mushroom, Sambok), Ganoderma lucidum (deer horn shaped), Auricularia auricula (ear mushroom), and Lentinula edodes (shiitake), were used to identify efficient new substrates for spawn and mushroom production. Although the mycelia in the rubber tree sawdust medium showed a slightly slower growth rate (10.9 cm/15 days) than mycelia grown in grains (11.2 cm/15 days in rice seeds), rubber tree sawdust appeared to be an adequate replacement for grain spawn substrates. Th findings indicate that rubber tree sawdust, sugarcane bagasse, and acacia tree sawdust supplemented with rice bran and calcium carbonate could be new alternative the substrates for. Although sugarcane bagasse and rubber tree sawdust showed similarly high biological efficiencies (BE) of 60% and 60.8%, respectively, acacia tree sawdust exhibited relatively a low biological efficiency of 22.4%. However, it is expected that acacia sawdust has potential for the mushroom cultivation when supplemented with currently used sawdust substrates in Cambodia, because of its relatively low price. The price of the sawdust (20 kg sawdust= 6500 Riel or 1.6 USD) currently used was 6.5 times higher than the price of acacia sawdust (201000 Riel or 0.25 USD). Therefore, utilization for acacia sawdust for mushroom cultivation could become feasible as it would reduce by producing costs of mushrooms in rural areas of Cambodia.

Development of Simple Colorimetric Method for Detecting Contamination of Liquid Spawn of Oyster Mushroom by pH Indicator (pH지시약을 이용한 느타리버섯 액체종균 오염 간이진단법 개발)

  • For the detection of contaminated liquid spawn, we selected suitable medium, indicator and developed method of diagnosis. The growth of pathogenic bacteria, Pseudomonas sp., and fungi, Trichoderma sp., in YPL media was better than in PDA and NA. In addition, the changes of color and absorbance of media were obviously showed when contaminated liquid spawn by pathogenic bacteria and fungi was incubated on YPL including phenol red for 48 hour at $25^{\circ}C$. The color of YPLP after incubating of infected liquid spawn by Pseudomonas sp. and Trichoderma sp. were changed from orange to red and to scarlet, respectively. Whereas, the color of YPLP after incubation of only Pleurotus ostreatus indicated yellow at liquid spawn. Therefore, it is possible to easily distinguish contaminated liquid spawn by color of change in YPLP.

The effect of different culture conditions of liquid spawn on the quality characteristics of shiitake mushroom (Lentinula edodes) (표고의 액체종균 배양 조건 및 품질 특성)

  • To improve the productivity of shiitake mushroom (Lentinula edodes), seven different types of media for liquid spawn (denoted as "A" to "G") were prepared with 0.3% soybean meal and varying sugar and glucose concentrations. During 14 days of incubation, the pH of the liquid culture gradually acidified with increasing incubation period. Additionally, there was a significant, but not prominent, difference in the degree of acidification depending on the sugar to glucose ratio. Liquid spawn culture "G," which had the highest sugar content was the most acidic on the last day of incubation. Mycelium dry weight increased significantly with increasing incubation period, and there was no significant difference in mycelium dry weight irrespective of the sugar to glucose ratio even after 14 days of culture. The inoculation of liquid spawn in sawdust medium with an inoculation volume ${\geq}45mL$ and incubation period of 15 to 18 days were the optimal culture conditions. Productivity of fruit bodies in sawdust medium and mushrooms treated with liquid spawn was significantly higher compared to solid spawn treatment. The mushrooms treated with liquid spawn had better chewiness, and the free amino acid content, which is associated with savory taste, was higher in these mushrooms compared to those treated with solid spawn.

Development on Artificial Cultivation method of Hatakeshimeji (Lyophyllum decastes) using fermented sawdust substrate (잿빛만가닥버섯(Lyophyllum decastes)의 발효톱밥에 의한 인공재배 특성에 관한 연구)

  • These experiments were carried out to determine the optimal culture conditions for nine strains of collected Hatakeshimeji, Lyophyllum decastes (Fr.:Fr.) Sing. SPA 202 and SPA 205 strains were selected because mycelium grew fast and showed fine density. All strains showed fast mycelial growth and mycelial density on BC(Burke compost) media for 20 days of incubation. The optimal sawdust species for the mycelial growth were the fermented sawdusts of Quercus aliena and Populus deltoides. Spawn running period on the fermented sawdust substrate required 50 days at 20 to $25^{\circ}C$ and additional 7 days after soil casing. Cultivation period and temperature forprimordia formation and fruitbody development appeared from 10 to 11 days and from 7 to 8 days at 17 to $18^{\circ}C$ respectively. The length of pilei and stipes of SPA 202 harvested in optimal stage showed 60mm and 67mm, respectively. Yield of SPA 202 strain grown on fermented sawdust substrate was 130g per 1,100ml in bottle cultivation. The length of pilei and stipes of SPA 205harvested in optimal stage showed 51mm and 81mm, respectively. Yield of SPA 205 strain grown on fermented sawdust substrate was 129g per 1,100 ml in bottle cultivation. SPA 202 strain and SPA 205 strain in artificial bottle cultivation of Lyophyllum decastes used in fermented sawdust substrate were selected as themost appropriate strain in yield.

Comparison in the productivity of Pleurotus ostreatus sawdust spawn based on the medium composition (느타리 톱밥종균 배지조성에 따른 생산성 비교)

  • This study was performed to find a medium material that can replace Douglas fir sawdust and rice bran in spawn media for growing spawn directly in the oyster mushroom farm. The pH range, total nitrogen source, and total carbon source of the mixed spawn medium were 5.3~5.9, 0.65~1.11%, and 47.0~49.1%, respectively. The C/N ratio was high in the mixed medium of poplar sawdust, with a low total nitrogen content. The protein content was high in the medium containing fermented Douglas fir sawdust. The mycelium growth rate was higher in the medium containing wheat bran than that in the medium containing rice bran. The highest yield per bottle was observed with poplar sawdust, oak sawdust, and rice bran mixed at a ratio of 40:40:20 (v/v/v); however, there was no significant difference observed in terms of productivity with the other treatments. Thus, when growing sawdust spawns in farms, it is efficient to use poplar sawdust, oak sawdust, and wheat bran, which are also easily available, instead of Douglas fir sawdust and rice bran.

Comparison of productivity among various spawn shapes of middle-temperature type strain for sawdust cultivation of Shiitake (표고 톱밥재배시 중온성 품종의 종균 형태에 따른 생산성 비교)

  • Studies were processed to confirm the difference of the shiitake productivity according to different spawn shapes(sawdust, plug-shaped and liquid spawns) on middle-temperature type strain. A tendency of fruiting was similar among three treatments, and treatments inoculated with plug-shaped spawn and liquid spawn produced over 50 % of total yield until 2nd flushing period and 80 % of total yield until 4th flushing period. In investigation of deformed fruit-bodies, all of three treatments occurred until 2nd flushing period. However, in rate of deformed fruit-bodies, treatment inoculated with liquid spawn was lower than others. In investigation of yield, the amount produced on treatment inoculated with liquid spawn was 411 g per medium and it was highest among treatments. And the amount of fruit-bodies over 10 g was higher than others. Also, the number of fruit-bodies between sawdust and plug-shaped spawn was different, but each weight of fruit-bodies among three treatments was not different.

  • Currently, the spawn of the mushroom Agarcus bisporus is produced by a method developed in the 1980s, and anew spawning method needs to be developed to improve the quality of the spawn. In this study, the condition for a maximum mycelium weight(5.92±0.52 g/L) was shaking culture (24 hours/day) at 24℃ and 120 rpm in CDB (compost dextrose broth). Based on this, the ventilated liquid culture method (2.5 L/min) was cultured for 10 days. This method was appropriate, andwhen the inoculum was cultured at 50 g/mL for about 10 days, it was cultured well without agglomeration and shaking of seed.

  • In this study, whether Giemsa staining solution can accurately determine bacterial contamination of liquid spawn for Flammulina velutipes in a short period of time was investigated. Giemsa solution staining cells of blood, bone marrow, lymph node, malaria parasites, rickettsia et al. was prepared by dissolving basic methylene azul and methylene blue, and acidic eosine in methyl alcohol-glycerine. Supernatant samples of Flammulina velutipes liquid spawn cultured under explosive aeration were placed on a slide, mixed with Gimesa solution and examined with optical microscope after staining. In 40 to 60 seconds bacterial cells were distinguishable from soybean meal residual and hyphal cell fragments. Thus we conclude that microscopy using Gimesa staining solution is a quick, simple and accurate method for the mushroom growers to effectively use to detect bacterial contamination of the liquid spawn.
