Changes of Ginsenoside Content by Mushroom Mycelial Fermentation in Red Ginseng Extract

  • To obtain microorganisms for the microbial conversion of ginsenosides in red ginseng extract (RGE), mushroom mycelia were used for the fermentation of RGE. After fermentation, total sugar contents and polyohenol contents of the RGEs fermented with various mushrooms were not a significant increase between RGE and the ferments. But uronic acid content was relatively higher in the fermented RGEs cultured with Lentus edodes (2155.6 ${\mu}g/mL$), Phelllinus linteus (1690.9 ${\mu}g/mL$) and Inonotus obliquus 26137 and 26147 (1549.5 and 1670.7 ${\mu}g/mL$) compared to the RGE (1307.1 ${\mu}g/mL$). The RGEs fermented by Ph. linteus, Cordyceps militaris, and Grifola frondosa showed particularly high levels of total ginsenosides (20018.1, 17501.6, and 16267.0 ${\mu}g/mL$, respectively). The ferments with C. militaris (6974.2 ${\mu}g/mL$), Ph. linteus (9109.2 ${\mu}g/mL$), and G. frondosa (7023.0 ${\mu}g/mL$) also showed high levels of metabolites (sum of compound K, $Rh_1$, $Rg_5$, $Rk_1$, $Rg_3$, and $Rg_2$) compared to RGE (3615.9 ${\mu}g/mL$). Among four different RGE concentrations examined, a 20 brix concentration of RGE was favorable for the fermentation of Ph. linteus. Maximum biotransformation of ginsneoside metabolites (9395.5 ${\mu}g/mL$) was obtained after 5 days fermentation with Ph. linteus. Maximum mycelial growth of 2.6 mg/mL was achieved at 9 days, in which growth was not significantly different during 5 to 9 days fermentation. During fermentation of RGE by Ph. linteus in a 7 L fermenter, $Rg_3$, $Rg_5$, and $Rk_1$ contents showed maximum concentrations after 5 days similar to flask fermentation. These results confirm that fermentation with Ph. linteus is very useful for preparing minor ginsenoside metabolites while being safe for foods.

불로초버섯 노랑썩음병의 발생과 병원균의 분류 동정 (Occurrence and Identification of the Fungus Causing Yellow Rot on Ganoderma lucidum)

  • 1990년을 전후하여 불로초버섯 재배원목에 노랑 얼룩무늬를 형성하며 버섯균을 심하게 썩히는 토양 전염성 곰팡이에 의한 병이 발생되었다. 불로초버섯 재배기간이 10년 이상인 철원과 강화 지역의 발병율은 각각 61%와 94%로 매우 높았으며 재배 년수가 비교적 짧은 문경과 홍성에서는 아직 병발생이 없었다. 피해원목을 절개하여보면 감염 초기에는 원목내부에 연한노랑 얼룩무늬를 형성하면서 영지버섯균사와 대치된 부분에서 갈색의 띠를 형성하였는데, 병이 많이 진전된 경우에는 원목의 내부 전체가 약간 푸르스름한 색깔을 띄면서 병원균의 자낭각이 다량으로 형성되어 있었다. 심하게 이병된 원목에서는 불로초버섯의 자실체가 전혀 생산되지 못하였으며 자실체가 형성된 경우라도 매우 불량하고 거의 죽어있었다. 단포자로 분리에 의해 배양된 병원균은 배양기상에서나 재배원목에서 불로초버섯 균사체를 심하게 용해시키는 강한 병원성을 나타내었으며, 병원균이 분비한 항균물질에 의하여 불로초버섯 균사체는 전혀 생장하지 못하였다. 본 병원균은 Arthrographis cuboidea로 동정되었는데 이들은 일반 배양기에서 비교적 빠르게 자라며 노란 색소를 형성하고 무성세대인 분절포자를 형성하였으며 포자낭병은 분지되지 않고 연속적으로 포자를 형성하였다. 무성포자의 폭이 길이보다 넓은 특징을 가지고 있었다. A. cuboidea에 의한 불로초버섯 노랑썩음병에 대한 병원성 및 분류 동정한 결과를 보고하고자 합니다.

황기에 표고버섯 균사체를 배양한 추출물이 항암효과 및 알레르기 억제효과에 미치는 영향 (Effect of Mycelia Extracts from Lentinus edodes Mushroom-Cultured Astragalus membranaceus Bunge on Anti-cancer and Anti-allergy Activities)

  • 황기를 이용하여 황기균사체를 접종, 배양하여 얻어진 황기균사체 추출액의 간암세포, 유방암세포, 자궁경부암세포 그리고 고형암의 증식에 미치는 영향을 조사하였다. 3가지 암세포의 형태변화 및 증식억제에 미치는 영향은 황기균사체 추출물이 황기추출물보다 효과적인 것으로 나타났으며, 황기균사체를 간암세포에 3 mg/mL로 처리했을 때 65.23%, 유방암세포에 5 mg/mL로 처리했을 때 69.42%의 높은 암세포 증식 억제 효과가 있었다. 자궁경부암세포에서는 황기균사체를 3 mg/mL로 처리했을 때 황기를 처리했을 때보다 암세포 증식억제효과가 23.82%더 높았다. 특히 간암세포와 유방암세포에서 황기균사체 추출물을 처리하였을 때 강력한 암세포 증식억제효과가 있는 것으로 나타났다. 고형암 억제효과에서도 대조군에 비해 황기균사체 추출물이 47%의 고형암 억제효과가 있었고, 황기추출물보다 고형암 억제 효과 10% 더 높았다. 히스타민 유리 억제효과를 측정한 결과 compound 48/80 처리군에 비해 황기추출물은 7.6%, 황기균사체 추출물은 44.6%의 히스타민 분비 억제효과가 있는 것으로 나타났다. 이상의 결과로 볼 때 한방기능성 소재에 균사체 배양기법의 접목으로 효능의 상승과 복합기능을 기대할 수 있겠다.

Mating Relationship between the parent and the mutant strains in Pleurotus ostreatus

  • Pleurotus ostreatus 'Miso' is a mutant strain showing white color in pileus from the known parent strain 'Wonhyeong 1'. Shape and several other characters also vary with culture conditions. Mating experiments were performed to understand interstrain mating relationship using monokaryons of the parent and the mutant strains. All monokaryons were grown from single spores isolated from freshly collected fruit bodies. Pairings were performed in 90 mm petri dishes on PDA. They were allowed to grow at 25 until two fronts of the advancing mycelia met and developed a conspicuous contact zone. The contact zone and the outer edges of paired colonies on each plate were examined for clamp connections. The parent and the mutant resulted in tetrapolar incompatibility in intrastrain crosses. In interstrain crosses, each monokaryotic tester strain of the parent strain was out-crossed to monokaryotic tester strains of the mutant. As a result of these crosses it was found that both strains share the same A and B incompatibility factors yielding 25% compatibility.

A comprehensive review of the therapeutic effects of Hericium erinaceus in neurodegenerative disease

  • Mushrooms are considered not only as food but also for source of physiologically beneficial medicines. The culinary-medicinal mushrooms may important role in the prevention of age-associated neurological dysfunctions, including Alzheimer's and Parkinson's diseases. Hericium erinaceus (H. erinaceus), is edible mushrooms, is a parasitic fungus that grows hanging off of logs and trees and well established candidate for brain and nerve health. H. erinaceus contains high amounts of antioxidants, beta-glucan, polysaccharides and a potent catalyst for brain tissue regeneration and helps to improve memory and cognitive functions. Its fruiting bodies and the fungal mycelia exhibit various pharmacological activities, including the enhancement of the immune system, antitumor, hypoglycemic and anti-aging properties. H. erinaceus stimulates the synthesis of Nerve Growth Factor (NGF) which is the primary protein nutrient responsible for enhancing and repairing neurological disorders. Especially hericenones and erinacines isolated from its fruitin body stimulate NGF, synthesis. This fungus is also utilized to regulate blood levels of glucose, triglycerides and cholesterol. H. erinaceus can be considered as useful therapeutic agents in the management and/or treatment of neurodegeneration diseases. However, this review focuses on in vitro, in vivo and clinical trials for neurodegerative disease.

  • 한국산 느타리버섯 균주의 균사, Primordia, 갓, 줄기의 전피영동 isozyme 패턴을 비교한 결과 자실체중 Primordia, 갓, 줄기의 패턴은 모두 아주 유사하였으나 균사의 패턴은 자실체의 것과 많은 차이가 있었다. 자생지역이 다른 한국산 Pleurotus ostreatus의 전기영동 isozyme 패턴중 Malate Dehydrogenase, Acid phosphatase는 유사하였으나 Esterase, Peroxidase, Leucine amino peptidase, Superoxide Dismutase의 패턴은 차이가 있었다. Pleurotus osteatus, P. cornucopiac, P. florida의 종간 Esterase 밴드 패턴은 많은 차이가 있어 전기영동에 의한 느타리버섯 종의 가능하였다.

  • 한국에 자생하는 느타리버섯 균주중 지역별로 기주가 다른 균주들을 수집하여 Esterase 동위효소와 Leucine amino peptidase 동위효소를 등전점 전기영동으로 균사와 자실체 부위별로 비교하였다. Pleurotus ostreatus의 Esterase 밴드 패턴은 균사와 자실체가 많은 차이가 있었으나 자실체중 Primordia, 갓, 줄기 등의 isozyme 패턴은 유사하였다. P. ostreatus, P. cornucopiae, P. florida의 Esterase 밴드 패턴에 많은 차이가 있어 종간 균주구별이 가능하였다. Leucine Amino Peptidase 밴드는 P. ostreatus, P. cornucopiae, P. florida 간에 뚜렷이 구별지을 수 없었으며, 균사와 자실체 간에는 약간의 밴드 패턴의 차이가 있었다.

Cd처리가 느타리버섯 균사체 생장에 미치는 영향 (Effect of Cd treatment on the mycelial growth of Pleurotus ostreatus)

  • In this study, we determined the effect of different concentrations of Cd on the growth of Pleurotus ostreatus mycelia, which was confirmed using PDA, PDB, and a column test. The efficacy of treatment was evaluated using Cd at two concentrations, 10 ppm and 100 ppm. The extent of mycelial growth on PDA and PDB was similar to that observed in the untreated groups and those treated with 10 ppm Cd. However, it was found that the mycelial growth was suppressed in a system treated with Cd at concentrations of 100 ppm. Moreover, the extent of mycelial growth observed upon conducting a column test was similar to that obtained using PDA and PDB. When the composition of free amino acids in PDB was determined, their levels in the group treated with 100 ppm Cd were found to be similar to those of the control. However, the amounts of 15 amino acids in the group treated with 10 ppm of Cd had increased compared to those detected in the control.

Favorable Culture Conditions for Mycelial Growth of Korean Wild Strains in Ganoderma lucidum

  • Ganoderma lucidum (Fr.) Karst (Polyporaceae), belonging to basidiomycota, is one of the most famous medicinal mushrooms. This study was carried out to investigate favorable mycelial growth conditions, such as pH, temperature, growth media, carbon sources and nitrogen sources of Korean strains in G. lucidum. The most suitable temperature for the mycelial growth was obtained at $30^{\circ}C$. In general, optimal temperature range for the mycelial growth was found at $25{\sim}30^{\circ}C$. This Mushroom has a broad pH range ($5{\sim}9$) for its mycelial growth and mostly favorable growth was found at pH 5. Generally, Hamada, Glucose peptone, YM, Mushroom complete and Lilly media were the most suitable for the mycelial growth of G. lucidum. Among 10 different carbon sources, dextrin, galactose and fructose were best but the rest of other carbon sources also facilitated the growth of mycelia. The most suitable nitrogen sources were ammonium acetate, glycine, arginine and calcium nitrate, but to a certain extent, all of the supplemented nitrogen sources also stimulated the mycelial growth.

Utilization of Robinia pseudoacacia as Sawdust Medium for Cultivation of Edible and Medicinal Mushrooms

  • This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.
