In Vitro Biological Characterization of DCUN1D5 in DNA Damage Response

  • Background: Novel prognostic biomarkers or therapeutic molecular targets for laryngeal squamous cell carcinoma (LSCC) are an urgent priority. We here sought to identify multiple novel LSCC-associated genes. Methods: Using high-density microarray expression profiling, we identified multiple genes that were significantly altered between human LSCCs and paired normal tissues. Potential oncogenic functions of one such gene, DCUN1D5, were further characterized in vitro. Results: Our results demonstrated that DCUN1D5 was highly expressed in LSCCs. Overexpression of DCUN1D5 in vitro resulted in 2.7-fold increased cellular migration, 67.5% increased invasive capacity, and 2.6-fold increased proliferation. Endogenous DCUN1D5 expression was decreased in a time-dependent manner after genotoxic stress, and silencing of DCUN1D5 by siRNA decreased the number of cells in the S phase by 10.2% and increased apoptosis by 11.7%. Conclusion: Our data suggest that DCUN1D5 in vitro might have vital roles in DNA damage response, but further studies are warranted to assess its significance in vivo.

A Study on the Automatic Detection and Extraction of Narrowband Multiple Frequency Lines (협대역 다중 주파수선의 자동 탐지 및 추출 기법 연구)

  • Passive sonar system is designed to classify the underwater targets by analyzing and comparing the various acoustic characteristics such as signal strength, bandwidth, number of tonals and relationship of tonals from the extracted tonals and frequency lines. First of all the precise detection and extraction of signal frequency lines is of particular importance for enhancing the reliability of target classification. But, the narrowband frequency lines which are the line formed in spectrogram by a tonal of constant frequency in each frame can be detected weakly or discontinuously because of the variation of signal strength and transmission loss in the sea. Also, it is very difficult to detect and extract precisely the signal frequency lines by the complexity of impulsive ambient noise and signal components. In this paper, the automatic detection and extraction method that can detect and extract the signal components of frequency tines precisely are proposed. The proposed method can be applied under the bad conditions with weak signal strength and high ambient noise. It is confirmed by the simulation using real underwater target data.

Autofocus Phase Compensation of Velocity Disturbed UUV by DPC Processing with Multiple-Receiver (다중 수신기 DPC 처리에 의한 속도 교란 수중 무인체의 자동초점 위상 보상)

  • In the case of a small UUV operating an active synthetic aperture sonar, various velocity disturbances may occur on the path due to the influence of external underwater environment, and this causes phase errors in coherent synthetic aperture processing, which has a large influence on the detected image. In this paper, when a periodic sinusoidal velocity disturbance is generated in the traveling direction, the phase generated by the round trip slope range at each position is estimated the cross correlation coefficient for multiple received signals and compensated the position variation in the overlapped DPC by the average value within the maximum allowable width. Through simulations, it has been confirmed that the images degraded by the velocity disturbance amplitude and fluctuating frequency of the UUV are removed from the false targets and the performance of azimuth resolution is improved by the proposed phase compensation method.

  • In this study, we have described a method for the fabrication of a protein chip on silicon substrate using hydrophobic thin film and microfluidic channels, for the simultaneous detection of multiple targets in samples. The use of hydrophobic thin film provides for a physical, chemical, and biological barrier for protein patterning. The microfluidic channels create four protein patterned strips on the silicon surfaces with a high signal-to-noise ratio. The feasibility of the protein chips was determined in order to discriminate between each protein interaction in a mixture sample that included biotin, ovalbumin, hepatitis B antigen, and hepatitis C antigen. In the fabrication of the multiplexed assay system, the utilization of the hydrophobic thin film and the microfluidic networks constitutes a more convenient method for the development of biosensors or biochips. This technique may be applicable to the simultaneous evaluation of multiple protein-protein interactions.

  • In the Cloud, for the scheduling of parallel jobs, there are many tasks in a job and those tasks are executed concurrently on different VMs (Visual machines), where each task of the job will be executed synchronously. The goal of scheduling is to reduce the execution time and to keep the fairness between jobs to prevent some jobs from waiting more time than others. We propose a Cloud model which has multiple Clouds, and under this model, jobs are in different lists according to the waiting time of the jobs and every job has different parallelism. At the same time, a new method-ZOMT (the scheduling parallel tasks based on ZERO-ONE scheduling with multiple targets) is proposed to solve the problem of scheduling parallel jobs in the Cloud. Simulations of ZOMT, AFCFS (Adapted First Come First Served), LJFS (Largest Job First Served) and Fair are executed to test the performance of those methods. Metrics about the waiting time, and response time are used to test the performance of ZOMT. The simulation results have shown that ZOMT not only reduces waiting time and response time, but also provides fairness to jobs.

The Effect of Multicultural Education Experiences on the Multicultural Acceptability of Military Officers (군 간부의 다문화교육경험이 다문화수용성에 미치는 영향)

  • This research seeks to examine the relationship between multicultural education experiences and the multicultural acceptability of military officers. The sample included 552 Republic of Korea Army officers and non-commissioned officers(NCOs) who were located in Gangwon-Do and Gyeonggi-Do. For the statistical analysis, SPSS 18.0 was used to perform descriptive statistical analysis, frequency analysis, differential verification, and multiple regression analysis. The major analysis results are as follows: First, The multicultural acceptability of the military officers and NCOs, including sub-factors, was above the mean level; in terms of the ranking per sub-factor, the scores were in the order of diversity, relationship, and universality. The experience frequency of the targets and the evaluation was lowest in the educational experience within the military, and the evaluations were in the order of public media, school, and military. Second, The differential verification, depending on the characteristics of the sociology of the population and the existence of the multicultural educational experience, showed that the duration of education(personal factor), marital status(family factor), existence of a foreign relative(family factor), and existence of experience of managing multicultural personnel as soldiers under command(military-related factor) showed a significant difference in multicultural acceptability. Additionally, the existence of multicultural education through schools and public media lead to a significant difference in multicultural acceptability, but the existence of education within the military did not lead to a significant difference. Third, The analysis of the influencing relationship through multiple regression analysis showed that the higher the evaluation of multicultural education through the military, school, and public media, the higher the multicultural acceptability.Therefore, this research proposes multicultural educational plans that may improve the multicultural acceptability of military officers and NCOs as a way to establish a multicultural composition in the army.

Multiple vertical depression-based HMS active target detection using GSFM pulse (GSFM 펄스를 이용한 다중 수직지향각 기반 선체고정소나 능동 표적 탐지)

  • In decades, active sonar, which transmits signals and detects incident signals reflected by underwater targets, has been significantly studied since passive sonar in Anti-Submarine Warfare (ASW) detection performance becomes lowered, as underwater threats become their radiated noise reduced. In general, active sonar using Hull-Mounted Sonar (HMS) adjusts vertical tilt (depression) and sequentially transmits multiple Linear Frequency Modulation (LFM) subpulses which have non-overlapped bands, i. e. 1 kHz ~ 2 kHz, 2 kHz ~ 3 kHz, in order to reduce shadow zones. Recently, however, Generalized SFM (GSFM), which is generalized form of SFM, is proposed, and it is confirmed that subpulses of GSFM have orthogonality among each other depending on setting of GSFM parameters. Hence, in this paper, we applied GSFM to active target detection using HMS to improve the performance by the signal processing gain obtained from enlarged bandwidths of GSFM subpulses compared to those of LFM subpulses. Through simulation, we verified that when the number of subpulses is three, the matched filter gain of GSFM is approximately 5 dB higher than that of LFM.

An Efficient Multiple Event Detection in Sensor Networks (센서 네트워크에서 효율적인 다중 이벤트 탐지)

  • Wireless sensor networks have a lot of application areas such as industrial process control, machine and resource management, environment and habitat monitoring. One of the main objects of using wireless sensor networks in these areas is the event detection. To detect events at a user's request, we need a join processing between sensor data and the predicates of the events. If there are too many predicates of events compared with a node's capacity, it is impossible to store them in a node and to do an in-network join with the generated sensor data This paper proposes a predicate-merge based in-network join approach to efficiently detect multiple events, considering the limited capacity of a sensor node and many predicates of events. It reduces the number of the original predicates of events by substituting some pairs of original predicates with some merged predicates. We create an estimation model of a message transmission cost and apply it to the selection algorithm of targets for merged predicates. The experiments validate the cost estimation model and show the superior performance of the proposed approach compared with the existing approaches.

  • Chromatin remodelers that include histone methyl transferases (HMTases) are becoming a focal point in cancer drug development. The NSD family of three HMTases, NSD1, NSD2/MMSET/WHSC1, and NSD3/WHSC1L are bona fide oncogenes found aberrantly expressed in several cancers, suggesting their potential role for novel therapeutic strategies. Several histone modifiers including HMTase have clear roles in human carcinogenesis but the extent of their functions and regulations are not well understood, especially in pathological conditions. The extents of the NSDs biological roles in normal and pathological conditions remain unclear. In particular, the substrate specificity of the NSDs remains unsettled and discrepant data has been reported. NSD2/MMSET is a focal point for therapeutic interventions against multiple myeloma and especially for t(4;14) myeloma, which is associated with a significantly worse prognosis than other biological subgroups. Multiple myeloma is the second most common hematological malignancy in the United States, after non-Hodgkin lymphoma. Herein, as a first step before entering a pipeline for protein x-ray crystallography, we cloned, recombinantly expressed and purified the catalytic SET domain of NSD2. Next, we demonstrated the catalytic activities, in vitro, of the recombinantly expressed NSD2-SET on H3K36 and H4K20, its biological targets at the chromatin.

Administration of red ginseng regulates microRNA expression in a mouse model of endometriosis

  • Objective: Red ginseng (RG) exerts anti-inflammatory, anti-proliferative, and immunomodulatory effects on endometriosis through the regulation of microRNA (miRNA) expression. It may also ameliorate endometriosis by affecting the expression of multiple miRNAs simultaneously, rather than acting on a single miRNA at a given time. Since studies on the overall effects of RG on endometriosis via the regulation of miRNA expression are lacking, the current study aimed to explore the global effect of RG on miRNA expression in a mouse model of endometriosis. Methods: To establish the mouse model, the uterine horn of donor mice was implanted into the lateral side of the recipients' peritoneum, followed by vehicle or RG treatment for 8 weeks. Results: To confirm the effects of RG on the established mouse model, the size of the implanted uterus was measured; it was found to be lower in mice from the RG group than in mice from the control group. miRNA expression profiles in the implanted uterus of the mouse model of endometriosis after vehicle or RG administration were analyzed using microarray technology. Thereafter, seven candidate miRNAs and 125 candidate genes (miRNA targets) were identified through a bioinformatics analysis. Conclusion: The present findings suggest that RG regulates the expression of multiple miRNAs and mRNAs, thereby alleviating endometriosis in a mouse model of the disease.
