Mar (Multiple-Antibiotic-Resistance) Operon 돌연변이 대장균의 그람음성 세균들간 전파 가능성에 대한 근거 (Evidences that Suggest the Spread of Multiple-Antibiotic-Resistance (mar) Operon of Escherichia coli Mutants among Gram-Negative Bacilli)

  • Mar (multiple-antibiotic-resistance) 대장균의 marRAB 돌연변이가 그람음성 세균들 사이로 전파 될수 있는지를 알아보기 위해서, 염색체 위의 marRAB 돌연변이를 $\lambda$placMu9 (Km$^{r}$)를 이용하여 대장균의 다른 균주내의 pUC19 (Lac$^{+}$, Ap$^{r}$)의 클로닝 사이트, S. typhimurium 및 P. aeruginosa의 염색체 위로 각각 전달시킨 후, Mar phenotype, Km$^{r}$, Lac$^{-}$, 혹은 Ap$^{r}$ 균주를 선택한 다음, salicylate (SAL)의 유도 (induction) 유무에 따른 항생제 내성검사를 실시하였다. pUC19의 보유 유무에 관계없이 대조군인 야생형 대장균 JM109, NM522에 비해서, pUC19::marRAB 돌연변이를 가진 JM109 혹은 NM522의 대부분 균주들은 항생제 내성 수준들이 훨씬 더 높았으며, Mar표현형의 유도도 SAL에 의해서 훨씬 더 높게 나타났다. 하지만 일부 소수의 항생제에 대해서는 예외적으로 내성 수준이 더 낮게 나타났는데, 야생형 JM109 균주에서는 chloramphenicol (Cm)과 tetracycline (Tc)이 , pUC19::marRAB 돌연변이를 가진 NM522 균주 에서는 Tc와 ciprofloxacin (Cp)에 대한 내성 수준이 각각 더 낮게 나타났다. S. typhimurium 및 P. aeruginosa에 대한 같은 실험의 결과도 야생형 대장균의 경우와 거의 대부분 일치하였다. 형질전환 (transformation)과 형질도입 (transduction)의 방법을 주로 사용한 이 실험의 결과는, marRAB 돌연변이가 그람음성 세균들 사이로 전파될 수 있음을 간접적으로 암시한다.

남해안 어류양식장에서 분리된 Vibrio parahaemolyticus와 Vibrio aiginolyticus의 항균제 감수성 (Antimicrobial Susceptibility of Vibrio parahaemolyticus and Vibrio alginolyticus from Fish Farms on the Southern Coast of Korea)

  • The antimicrobial resistance patterns to the 10 antimicrobial agents of potential pathogenic vibrios isolated from seawater and a variety of farmed fishes, including oliver flounder (Payalichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagyus major) and sea bass (Lateolabyax japonicus), were investigated from May to October, 2004. A total of 314 strains of the genus vibrios were isolated from 126 collected samples, and the number of isolated strains of Vibrio payahaemolyticus and Vibrio alginolyticus were 194 and 120, respectively. Apparently $98.5\%$ of V. parahaemolyticus and $100\%$ of V. alginolyticus isolates demonstrated antimicrobial resistance against at least one antimicrobial agent. The resistance of V. parahaernolyticus isolates to ampicillin ($97.9\%$) was highest, followed by oxolinic acid ($26.8\%$), amikacin ($19.1\%$) and tetracycline and amoxicillin/clavulanic acid ($6.7\%$). V. alginolyticus isolates were resistant to ampicillin ($100\%$), solfamethoxazoleit,imethopenem ($25\%$), amikacln ($21.7\%$), amoxicillin/clavulanic acid ($15.8\%$), ciprofloxacin ($13.3\%$), and tetracycline and doxycycline ($11.7\%$). The rate of multiple antimicrobial resistance to at least four antimicrobials was higher in the V. alginolyticus isolates ($20.8\%$) than in the V. parahaemolyticus ($6.7\%$).

한산거제만 및 자란만사량도 해역 육상오염원 배출수에서 분리한 대장균의 항균제 내성 패턴 (Antimicrobial Resistance Patterns of Escherichia coli Isolated from Discharged Water from Inland Pollution Sources in the Hansan-Geojeman and Jaranman-Saryangdo Areas of Korea)

  • We investigated patterns of antimicrobial resistance in Escherichia coli isolated from the water discharged from inland pollution sources in the Hansan-Geojeman and Jaranman-Saryangdo areas of Korea. A total of 217 strains of E. coli were isolated from 23 point-sources. These strains were tested for their susceptibility to 16 antimicrobial agents used in Korea for medical or veterinary therapy. The highest level of antibiotic resistance among the isolated strains was to tetracycline 10.6%, followed by ampicillin (3.2%), nalidixic acid (2.8%), rifampin (1.8%), trimethoprim (1.8%), trimethoprim/sulfamethoxazole (1.8%), chloramphenicol (1.4%), streptomycin (1.4%), cephalothin (0.5%) and gentamicin (0.5%). Resistance to at least one antimicrobial agent was present in 17.1% of the E. coli isolates. Only four of the isolated strains of E. coli showed multiple antibiotic resistance, which is defined as resistance to more than four antibiotics.

젖소의 유방염 원인균 분리 및 약제 감수성 검사 (Identification and Antimicrobial Susceptibility of Microorganisms Isolated from Bovine Mastitic Milk)

  • Microorganisms were isolated and identified from bovine 296 quarters which showed positive reaction by California Mastitis Test (CMT) in 40 farms of Jeju from September 1999 to June 2000. The organisms associated with the mastitis of bovine were 11 different bacterial species in this study. Which of them, Staphylococcus aureus was the most predominant species as 152 (51.4%) isolates. Other identified species included 49 (16.5%) coliform, 47 (15.8%) Streptococcus dysgalactiae, 15 (5.1%) Bacillus spp., 8 (2.7%) Staphylococcus epidermidis, 6 (2.1%) Streptococcus agalactiae, 5 (1.7%) Enterococcus faecalis, 5 (1.7%) Corynebacterium spp., 3 (1.0%) Streptococcus uberis, 1 (0.3%) Pseudomonas aeruginosa and 1 (0.3%) Pasteurella haemolytica. Almost of all the islolated beacterial species showed high sensitivity against kanamycin (98.6%), cephalothin (98.0%), streptomycin (94.9%), gentamicin (94.6%), ampicillin (92.2%) and polymyxin B (90.2%). On the contrary, they showed resistance against penicillin (47.0%), tetracycline (37.2%), cefazolin (26.0%), bacitracin (22.6%) and erythromycin (19.9%). Eighty-one isolates were not resistant to any antibiotics and 215 drug resistant isolates showed 89 different drug resistance patterns from single to nine multiple antibiotics resistance patterns.

Profiles of Enterotoxin Genes and Antimicrobial Resistance in Staphylococcus pseudintermedius Strains Isolated from Livestock and Companion Animals

  • Staphylococcus pseudintermedius는 개에서 기회감염을 유발하는 병원체이며, 공중보건학적으로도 주요한 인수공통 병원체이다. 개에서 분리된 S. pseudintermedius 균주들은 주로 항생제 내성 및 개에서 피부 감염을 유발하는 주요 원인균으로 연구되어 왔지만, 가축에서 분리된 S. pseudintermedius 균주들의 항생제 내성 및 장내 독소 생성에 대한 정보는 매우 제한적이다. 본 연구에서는 개, 돼지, 육우에서 분리된 S. pseudintermedius 균주들에서 18가지의 장내 독소 (staphylococcal enterotoxin; SE) 유전자와 toxic shock syndrome toxin 유전자(tst-1)의 분포양상을 조사하였다. 또한, S. pseudintermedius 균주들의 항생제 내성 양상과 더불어 mecA 유전자 및 SCCmec type 또한 확인하였다. 육우에서 분리한 하나의 균주를 제외한 모든 개와 돼지 분리주 들이 4개 이상의 항생제에 내성을 보였으며, 개에서 분리된 6개의 균주 중 4개의 S. pseudintermedius 균주들이 메티실린 내성과 더불어 SCCmec V를 가진 것으로 확인 되었다. 총 11개의 SE 유전자들 (seb, sec, see, seg, sei, sej, sel, seo, sep, seq, seu) 및 tst-1가 개, 돼지 및 육우로부터 분리된 S. pseudintermedius 균주들에서 확인 되었으며, 대부분의 분리주들 (83%)에서 2개 이상의 SE 유전자들이 확인 되었고, 그 중 sel (42%) 및 sep (42%)가 가장 빈번하게 검출 되었다. 본 연구를 통하여 반려견에서 뿐만 아니라 주요 가축에서 존재하는 S. pseudintermedius 균주들에서 높은 항생제 내성 양상을 확인 하였으며, 항생제 내성과 더불어 여러 staphylococcal enterotoxin 및 tst-1 유전자들을 전파할 가능성을 확인 하였다.

Simplex PCR Assay for Detection of blaTEM and gyrA Genes, Antimicrobial Susceptibility Pattern and Plasmid Profile of Salmonella spp. Isolated from Stool and Raw Meat Samples in Niger State, Nigeria

  • The global evolution of antibiotic resistance has threatened the efficacy of available treatment options with ravaging impacts observed in developing countries. As a result, investigations into the prevalence of antibiotic resistance and the role of plasmids are crucial. In this study, we investigated the presence and distribution of blaTEM and gyrA genes, plasmid profiles, and the antimicrobial susceptibility pattern of Salmonella strains isolated from raw meat and stool sources across Niger State, Nigeria. Ninety-eight samples, comprising 72 raw meat and 26 stool samples, were screened for Salmonella spp. The antimicrobial susceptibility of Salmonella isolates to 10 commonly used antimicrobial agents was determined using the KirbyBauer disc diffusion method. Isolates were further analyzed for plasmids, in addition to PCR amplification of beta-lactamase (blaTEM) and gyrA genes. A total of 31 Salmonella spp. were isolated, with 22 from raw meat (70.97%) and 9 from stool (29.03%). Salmonella spp. with multiple resistance patterns to ceftazidime, cefuroxime, ceftriaxone, erythromycin, ampicillin, cloxacillin, and gentamicin were detected. Ofloxacin and ciprofloxacin were found to be the most effective among the antibiotics tested, with 67.7% and 93.5% susceptible isolates, respectively. Nine (29.03%) isolates harbored plasmids with molecular sizes ranging between 6557 bp and 23137 bp. PCR amplification of gyrA was detected in 1 (3.23%) of the 31 isolates while 28 isolates (90.32%) were positive for blaTEM. This study shows the incidence of antibiotic resistance in Salmonella isolates and the possible role of plasmids; it also highlights the prevalence of ampicillin resistance in this local population.

소아청소년 영역에서의 새로운 항미생물제 (New Antimicrobial Agents for Children)

  • There are relatively few novel antimicrobial agents despite the dramatic increase in antimicrobial resistance and multiple drug resistance of clinical isolates worldwide. Vancomycin is still the most widely used antibiotic for treating resistant Gram-positive coccal infections in children, especially for methicillin-resistant Staphylococcus aureus. For children with Gram-positive coccal infections where vancomycin is not effective or older therapeutic agents cannot be tolerated, linezolid, quinupristin-dalfopristin or daptomycin may be useful in the appropriate clinical setting. For Gram-negative bacterial infections, new carbapenems await clinical application. Tebipenem pivoxil is a novel oral carbapenem undergoing clinical trials for acute otitis media in pediatric patients. Antiviral drug development is now progressing at the pace of antibiotic development 30 years ago. Newer antiviral agents used for the treatment of herpes viruses and hepatitis C virus infections in children are included in this review.

자란만사량도 해역의 굴(Crassostrea gigas) 및 육상오염원에서 분리한 대장균(Escherichia coli)의 항생제 내성 (Antimicrobial Resistance of Escherichia coli Isolated from Oysters Crassostrea gigas and Major Inland Pollution Sources in the Jaranman-Saryangdo Area in Korea)

  • This study evaluated the abundance and antimicrobial resistance of Escherichia coli in oysters Crassostrea gigas and major inland pollution sources sampled in 2014-2015 from Jaranman-Saryangdo on the southern coast of Korea. The ranges of the geometric mean of E. coli concentrations in oysters and major inland pollution sources were <20-29.8 MPN/100 g and 7.5-137.2 MPN/100 g, respectively. We isolated 247 strains of E. coli (28 from oysters and 247 from major inland pollution sources) and examined the antimicrobial resistance patterns of all isolates. Isolates from both sources were highly resistant to rifampin (99.5-100%) and cephalothin (70.8-78.6%). The resistance rate was higher in E. coli isolated from oysters those from inland pollution sources. Multiple resistance against at least four antimicrobials was observed in 85.7% and 21.0% of the oyster and major inland pollution sources isolates, respectively.

  • Salmonella spp. is of increasing public health concern as causative pathogens of food poisoning. The aim of this study was to investigate the serotypes and antimicrobial resistance pattern of Salmonella spp. isolated from duck farms in Daegu-Gyeongbuk province. Also, S. Enteritidis and S. Typhimurium isolates were further examined for plasmid analysis, phage typing and pulsed-field gel electrophoresis (PFGE). A total of 34 Salmonella spp. (16.4%) were isolated from duck farms and ten serotypes were identified. The predominant serotypes were S. Typhimurium (23.5%) S. Fyris (17.6%) and S. Haardt (11.8%), S. Agona and S. Enteritidis (respectively 8.8%). Of 34 Salmonella isolates, 15 (44.1%) isolates were resistant to at least one antimicrobial agent and multiple resistance (resistance to more than 4 drugs) was observed in 9 strains (26.5%). The high resistance was found to streptomycin (32.4%), tetracycline (29.4%), ampicillin, kanamycin and nalidixic acid (respectively, 26.5%), all Salmonella isolates were susceptible to cefoxitin, cefotaxime, gentamicin, amikacin and ciprofloxacin. All S. Enteritidis and S. Typhimurium isolates were found to contain only one plasmid (ca. 54 or 55kb, respectively). Among the S. Enteritidis isolates, two phage types were found, PT32a and PT1c, respectively, one isolates did not react with any of the phages used. Whereas, all S. Typhimurium isolates were RDNC (reacts but does not conform). PFGE showed to be a useful typing method better than plasmid analysis and phage typing for discrimination of isolates especially, S. Typhimurium isolates. Our results indicated that the serotypes of Salmonella isolates are widely distributed in duck farms, further epidemiological studies should be carried out.

  • Three-hundred and sixteen Escherichia coli strains from seawater, and a variety of farmed fishes, including oliver flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major) and sea bass (Lateolabrax japonicus) between May to October in 2004, were tested by agar dilution method to determine their susceptibility patterns to 17 antimicrobial agents. Overall, 92.1% of Escherichia coli isolates from samples showed antimicrobial resistance to at least one antimicrobial agent and the multiple resistance was seen in 173 isolates (54.7%). The resistance of E. coli isolates to tetracycline (74.1%) was highest, followed by cephalothin (69.9%), doxycycline (66.5%), streptomycin (47.2%), ampicillin (46.2%), cefazolin (31.6%), enrofloxacin (31.0%). norfloxacin (28.2%). The most frequent resistance pattern was TE-D-CF-CIP-ENO-NOR-AM-S-C-SXT-AmC-CZ (14.7%), followed by CF (6.2%), TE (5.1%), TE-CF (4.5%) in 177 isolates from fishes and TE-D-CF (7.2%) followed by TE-D-CF-S (5.8%), CF and TE-D-S (3.6%) in 139 isolates from seawater.
