Agronomic Performance of G. max x G. soja Hybrid Progenies for Crop Improvement in Soybean

  • Genetic improvement of the cultivated soybean [Glycine max (L.) Merr] may be possible through hybridization with its wild progenitor, G. soja Sieb. & Zucc. Interspecific cross between G. max (Hwangkeumkong) and G. soja (IT.182932) was made in the summer of 1997. In F$_2$ the percentage of plant height, nodes per plant, and pods per plant were high but gradually reduced from F$_2$ to F$_4$. In contrast pod length, seeds per pod, and 100-seeds weight were increased gradually through generations advanced. Wild variation as evident in F$_2$ in plant height, number of branches, pods per plant, and 100-seeds weight. Twenty six percent of the F$_2$, 44 % of the F$_3$ and 60% of the F$_4$ segregants showed more G. max traits. The combination of useful traits from both species is possible through interspecific hybridization. The characters that could be transferred from wild species to cultivated species are more pod number, better capacity, and resistance to disease and insects. The interspecific derivatives offer scope for selection for high grain yield. Therefore, introducing genes from G. soja to G. max could be contribute to greater genetic diversity of future cultivars. And semicultivated soybean had some desired characteristics including tolerance to adverse environments and multi-seed characters. It means the infusing of semicultivated germplasm to the cultivated soybean could increase number of seeds and pods per plant significantly, and consequently could enhance selecting potential on yield.

Genetic Diversity of Curcuma Genus Collected Germplasm using Analysis of AFLP (AFLP 분석을 통한 Curcuma속 수집종간 유전적 다양성 분석)

  • Collected germplasms of five representative species belonging to Curcuma genus (C. longa, C. aromatica, C. zedoaria, C. phaeocaulis and C. kwangsiensis) were 52 samples from different farmhouse in Korea and China. To elucidate the genetic diversity among the species, 52 samples were analyzed by genomic fingerprinting method using amplified fragment length polymorphism (AFLP). AFLP results of 6 primer combinations were revealed 643 total DNA fragments and 349 polymorphic bands with the 54.3% ratio of polymorphism. In the analysis of coefficient similarity using unweight pair group method with arithmetic averages (UPGMA), 52 Curcuma germplasm lines were ranged from 0.60 to 0.99 and clustered distinct five groups according to the species and collected geographical levels. However, the result of principal coordinate analysis (PCA) by multi-variate analysis was shown significantly greater differences among species than geographical origins based on AFLP profiling data of these samples.

  • Cytospora is a genus including important phytopathogens causing severe dieback and canker diseases distributed worldwide with a wide host range. However, identification of Cytospora species is difficult since the currently available DNA sequence data are insufficient. Aside the limited availability of ex-type sequence data, most of the genetic work is only based on the ITS region DNA marker which lacks the resolution to delineate to the species level in Cytospora. In this study, three fresh strains were isolated from the symptomatic branches of Elaeagnus angustifolia in Xinjiang Uygur Autonomous Region, China. Morphological observation and multi-locus phylogenetic analyses (ITS, LSU, ACT and RPB2) support these specimens are best accommodated as a distinct novel species of Cytospora. Cytospora elaeagnicola sp. nov. is introduced, having discoid, nearly flat, pycnidial conidiomata with hyaline, allantoid conidia, and differs from its relatives genetically and by host association.

Preliminary Environmental Impact Assessments on Fish Compositions and the Ecological Health of Jeokbyeok River on the Road Construction of Muju-Geumsan Region (무주-금산간 도로건설에 따른 적벽강의 어류 종 조성 분석 및 생태건강도 사전환경성평가)

  • The objectives of the study were to evaluate fish compositions, endangered species, community structure, physical habitat, and general water quality for a preliminary environmental impact assessment of Jeokbyeok River on the road construction between two regions. Total number of species and total number of individuals, based on CPUE, were 23 and 1186, respectively. The endangered species (I, II) as the legal protection species were Pseudopungtungia nigra (79 samples) Gobiobotia brevibarba) (5) Gobiobotia macrocephala (2), indicating a requiring of endangered species conservation. In the meantime, exotic species and ecological disturbing species such as Micropterus salmoides and Lepomis macrochirus, were not present, indicating a well conserved area. According to fish community analysis, values of species diversity index were high (range: 0.788 - 1.030), and the dominance index were low (range: 0.097 - 0.183), indicating that the fish community in this area was maintained well without high dominacne by specific species. Also, fish analysis on tolerance guilds and trophic guilds showed that the proportions of sensitive species were largely exceeded the proportions of the tolerant species, while the proportions of insectivore species were largely exceeded the proportions of the omnivore species. This outcome suggests that the ecosystem was well maintained in terms of tolerance and trophic compositions (food chain). Ecological health, based on the multi-metric fish model of Fish Assessment Integrity (FAI), reflected those fish conditions. In other words, values of FAI model averaged 82.4, which means a "good condition" in the criteria of ecological health by the Minstry of Environment, Korea. In addition, general water quality and physical habitat analyses showed that the system was in good condition. Under these conditions, if the road constructions between the two regions happen in the future, inorganic suspended solids may increase in the waterbody, and this may result in indirect or direct influences on the physical habitats and food chain as well as fish compositions, so the ecological protections and prevention strategy from the soil erosion are required in the system.

Effect of Feeding Direct-fed Microbial as an Alternative to Antibiotics for the Prophylaxis of Calf Diarrhea in Holstein Calves

  • The objective of this study was to determine the effect of feeding direct-fed microbials (DFM) on the growth performance and prophylaxis of calf diarrhea during the pre-weaning period as an alternative to antibiotics. A multi-species DFM was formulated including three lactic acid bacteria (Lactobacillus salivarius Ls29, Pediococcus acidilactia Pa175, and L. plantarum Lp177), three Bacillus strains (B. subtilis T4, B. polymyxa T1 and SM2), one yeast, Saccharomyces boulardii, and a nonpathogenic E. coli Nissle 1917. Lactic acid bacteria and Bacillus strains were selected based on the antibacterial activity against various animal pathogens, especially pathogenic E. coli using agar diffusion methods in vitro. Test and control groups were fed milk replacer and calf starter supplemented with DFM ($10^9$ cfu each of eight species/d/head, n = 29) or with antibiotics (0.1% neomycin sulfate in milk replacer and Colistin 0.08% and Oxyneo 110/110 0.1% in calf starter, n = 15), respectively. Overall fecal score and the incidence rate of diarrhea were reduced in the DFM group compared to the antibiotics one. About 40% of calves in antibiotic group suffered from diarrhea while in DFM group only 14% showed diarrhea. There was no difference in the average daily gain and feed efficiency of two groups. The hematological levels of calves were all within the normal range with no significant difference. In conclusion, the feeding of multispecies DFM during the pre-weaning period could reduce calf diarrhea and there was no difference in the growth performance between the groups, thus showing the potential as an alternative to antibiotics.

  • Korean goral (Nemorhaedus caudatus raddeanus) is one of the endangered species in Korea, and the rugged terrain of the Soraksan National Park (373㎢) is a critical habitat for the species. But the goral population is threatened by habitat fragmentation caused by roads and hiking trails. The objective of this study was to develop a potential habitat suitability model for Korean goral in the park, and the model was based on the concepts of fuzzy set theory and multi-criteria evaluation. The process of the suitability modeling could be divided into three steps. First, data for the modeling was collected by using field work and a literature survey. Collected data included 204 points of GPS data obtained through a goral trace survey and through the number of daily visitors to each hiking trail during the peak season of the park. Second, fuzzy set theory was employed for building a GIS data base related to environmental factors affecting the suitability of the goral habitat. Finally, a multiple-criteria evaluation was performed as the final step towards a goral habitat suitability model. The results of the study were as follows. First, characteristics of suitable habitats were the proximity to rock cliffs, scattered pine (Pinus densiflora) patches, ridges, the elevation of 700∼800m, and the aspect of south and southeast. Second, the habitat suitability model had a high classification accuracy of 93.9% for the analysis site, and 95.7% for the validation site at a cut off value of 0.5. Finally, 11.7% of habitatwith more than 0.5 of habitat suitability index was affected by roads and hiking trails in the park.

Performance characteristics of a multi-directional underwater CCTV camera system to use in the artificial reef survey (인공어초 조사용 다방향 수중 CCTV 카메라 시스템의 성능 특성)

  • Underwater CCTV camera systems are increasingly replaced the traditional net approach of assessing the species, numbers and aggregation patterns of marine animals distributing around the artificial reefs installed in the inshore fishing grounds, in particular, in relation to the biological investigation of behavior and distribution patterns of target fishes. In relation to these needs, we developed a multi-directional underwater CCTV camera system to use in detecting and tracking marine animals in the artificial reef ground. The marine targets to be investigated were independently tracked by using a camera module toward the bottom and four camera modules installed in the interval of $90^{\circ}$ in horizontal plane and inclination of $45^{\circ}$ in vertical plane of the CCTV system without the overlap of video frames by each camera module. From the results of several field tests at sea, we believe that the developed multi-directional underwater CCTV camera system will contribute to a better understanding in evaluating the effect of artificial reefs installed in the inshore fishing grounds.

Study on Biological Effect of Multi-Herbal Drug KOCO-P1 on Human Hepatocytes (HepG2) (인간 간조직세포(HepG2 Cells)에 대한 한약조성물 KOCO-P1의 효과 연구)

  • Objectives : The purpose of this study is to investigate the biological Effect of multi-herbal drug 'KOCO-P1' on human hepatocyte HepG2 cells. Methods : Multi-herbal drug 'KOCO-P1' was composed of Ginseng Radix, Astragali Radix, Polygonati Rhizoma, Liriopis Tuber, and Scrophulariae Radix. Cytotoxicity and cytoprotective activity of KOCO-P1 was verificated by MTT assay. And antioxidative effect of KOCO-P1 against EtOH, Nicotine was inspected by Hydroperoxide assay. Results : KOCO-P1 showed no cytotoxicity on HepG2 cells for 24, 48 hours. KOCO-P1 at 50 ${\mu}g/mL$ reduced the production of H2O2 in HepG2 cells by EtOH. KOCO-P1 at 50 ${\mu}g/mL$ reduced the production of $H_2O_2$ in $HepG_2$ cells by Nicotine. Conclusions : KOCO-P1 at the low concentration could be supposed to have antioxidative effect on human hepatocyte with no cytotoxicity.

Cancer Treatment Using Multiphoton Photodynamic Therapy

  • Photodynamic therapy (PDT), a newly established treatment for solid tumors, involves the systemic administration of a tumor localizing photosensitizer that is only activated when exposed to light of appropriate wavelength. Photoactivation of photosensitizer in the presence of oxygen results in the formation of highly cytotoxic molecular species, which precipitates necrosis. PDT has now become a promising means for the treatment of cancer due to its specificity, relatively minimal side effects, and inexpensive. However, the application of PDT has been restricted to the treatment of superficial lesions or the use of interstitial light delivery. A single photon generally activates the photochemical reaction in traditional PDT. However the use of multi photon excitation, where two or more photons simultaneously excite a photosensitizer, allows for the use of wavelengths twice as long. Such wavelengths exhibit better transmittance through tissue and thereby deeper penetration is achieved. This paper will review theoretical principles of multi photon excitation, challenges associated with multi photon PDT and update the current and future role of multi photon PDT in cancer.

The Bacterial Colonization of Burn Wound and the Changes of Antibiotic Susceptibility in Childhood Burn (January, 1999~December, 2002) (소아 화상의 화상부위 세균 집락화와 항균제 감수성 변화(1999년 1월~2002년 12월))

  • Purpose : Children occupy a large proportion of burn victims. So we want to aid to pediatric burn care through the understanding of the bacterial distribution in burn wounds and antibiotic susceptibility against isolated microorganisms from burn wounds. Methods : We analysed the medical records of 213 pediatric burn patients(0~15 years), 406 samples that grew bacteria in burn wound sites. Results : Of the total 213 patients, male were 59.6% and female were 40.4%. Scalding burn was the most common(78.4%), flame burn was the second(16.4%). Pathogens were isolated in 406 samples. The most common was Pseudomonas aeruginosa(58.1%). Next were Enterococcus species, Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus species, Acinetobacter species. P. aeruginosa was resistant to trimethoprim-sulfamethoxazole in 100%, cephalothin in 98.1%, ampicillin-sulbactam in 96.2%, ampicillin in 95.3%, ceftriaxone in 95.2%, tobramycin in 93.7%, cefoperazone in 68.9%, ceftazidime in 67.7%. Enterococcus species were resistant to tetracycline in 63.9%, streptomycin in 45.5%, gentamicin in 36.1%, penicillin G in 13.7%. S. aureus was resistant to gentamicin in 89.7%, tetracycline in 86.2%, ciprofloxacin in 86.2%, penicillin G in 84.3%, oxacillin in 78.4%, erythromycin in 76.5%. Acinetobacter species were resistant to ampicillin-sulbactam in 100%, gentamicin in 85.7%, ampicillin in 83.3%, piperacillin in 61.5%. Conclusion : P. aeruginosa was highly resistant to drugs like cefoperazone in 68.9%, ceftazidime 67.7%. S. aureus was highly resistant to penicillin G in 84.3%, oxacillin in 25.9 %, but none to vancomycin in 0%, teicoplanin in 2.2%. According to the study, Acinetobacter species turned out to be multi-resistant strains, so careful attention must be paid to the choice of antibiotics.
