Development of Automatic Rearing System of Silkworm

  • Decrease in the cocoon production of Japan is drastic because of low price of cocoon, scarcity of successors and so on. To tide over the difficulty, the automation system in the sericulture was discussed and some trials have been conducted by the Ministry of Agriculture, Forestry and Fisheries of Japan. The attempts are based on a low cost artificial diet which does not rely on mulberry leaves. Automatic machines developed until now are a rearing machine constituted with repeated belt conveyor, an reformation type of former rearing machine "Bombyx" and a mounting machine. Running parallel with them, utilization of 20-hydroxyecdysone extracted from a plant to the mounting was also studied to use their machines efficiently in the fields. In conclusion, 10 tons of law cocoon will come to be produced by two persons labor. At present, an automatic rearing system on low cost artificial diet has been developing for the future sericulture.

Fluorescent Properties of Daehwangjam, Golden Silk, and Juhwangjam and Their Diminishing upon HCl Vapor Exposure

  • For over five millennia, humans have benefited from the valuable byproducts of Bombyx mori silkworms nourished on mulberry leaves and a multitude of potential applications remains available due to the diverse array of silkworm varieties. In this work, we discuss the utilization of Daehwangjam (DHJ), golden silk (GS), and Juhwangjam (JHJ), distinctive colored silks found in Korea, as chemosensors. These novel silks emit fluorescence under external stimuli and show a diminishing fluorescence intensity when exposed to HCl vapor. The considerable surface-to-volume ratio of these cocoons allows for the identification of 5 ppm, 300 ppm, and 3000 ppm HCl vapors through decreased fluorescence intensity. The results show the suitability of natural DHJ, GS, and JHJ for applications in biosensing applications.

Uptake and Content of Ions on Several Organs of Mulberry Tree(Morus alba L.) in Relation to Different Nitrogen Sources in Water Culture (수경액중(水耕液中)의 공급질소형태(供給窒素形態)에 따른 상수(桑樹)의 이온 흡수와 기관별(器官別) 분포(分布)에 관한 연구)

  • Water culture experiment with mulberry (Morus alba L.)was carried out to investigate the ionic composition in the exudate and the ionic content in the organs (leaves, petioles, stem cortex, and roots) of mulberry tree with three different nitrogen sources ($NO_3-N$, $NH_4-N$, and $NH_4NO_3$). 1. Amount of exudate was much lower for $NH_4-N$ than for $NO_3-N$. 2. Total nitrogen content in the exudate was the highest in the NH4-N, the order of 2.2 times than in the $NO_3-N$. However, total nitrogen amount absorbed by tree per hour was proportional with the exudate amount. As the result, total nitrogen amount was higher 1.6 times in $NO_3-N$ ($289.6\;me\;plant^{-1}\;hr^{-1}$ than in $NH_4-N$. 3. Ammonium nitrogen depressed $Ca^{2+}$ uptake critically and even all of the anions, whereas $Mg^{2+}$, $K^+$ and $Na^+$ affected little. 4. Reduction of $NO_3$ may occured both in root and in leaves. 5. Content of cations and anions was highest in petioles, except $Ca^{2+}$ which was highest in leaves. As the result, petioles may be the storage organ of nutrients. 6. By increasing $NH_4-N$, ionic balance (C-A) decreased proportionally. 7. Nutrients amount in the exudate were approximatelly proportional with the amount in the roots. This suggested that roots may be the part diagnosed for nutrients. Being the sum of anions (${\Sigma}A$) higher than that of cations (${\Sigma}C$) in the roots, the ionic balance showed negative value.

Monitoring of the residues of some pesticides in mulberry leaves and their safety evaluation (뽕잎중 잔류농약의 monitoring 및 안전성 평가)

  • In order to evaluate the safety of mulberry leaves for sericultural purpose to the contamination by pesticide residues, the analyses of dichlorvos, acephate, phenthoate, pyrazophos, deltamethrin, and cypermethrin were done for 24 samples collected in spring and fall over two consecutive years (1996 and 1997), each, from 8 provinces across the country, by selecting three representative farmhouses in each province. In spring samples of 1996, the residual amounts of dichlorvos, acephate, and phenthoate were $0.018{\sim}0.032$, $0.013{\sim}0.072$, and 0.051 ppm, respectively, whereas, pyrazophos was not detected, and the detection frequencies were 29, 58, and 4%, respectively. In fall samples, on the other hand, those of the above pesticides were $0.012{\sim}0.048$, $0.020{\sim}0.156$, and $0.018{\sim}0.050$ ppm, respectively, and pyrazophos was not detected either and the detection frequencies were 42, 17, and 13%, respectively. While in spring samples of 1997, the residual amounts of dichlorvos, acephate, and cypermethrin were $0.014{\sim}0.064$, $0.033{\sim}0.061$, and $0.019{\sim}0.068$ ppm, respectively, and deltamethrin was not detected. The detection frequencies were 75, 13, and 8%, respectively. In fall samples, on the other hand, those of the above pesticides were $0.013{\sim}0.062$, 0.015, and $0.009{\sim}0.013$ ppm, respectively, and deltamethrin was not detected either. The detection frequencies were 88, 4, and 17%, respectively. Almost all of the samples turned out to be non-toxic to silkworms, except that one sample (No. 10, spring 1997) contaminated by a high concentration of cypermethrin (0.068 ppm) was judged to do harm to silkworms. In the GC analyses, the selection of columns, the change in column temperature, and the use of MSD made the separation and identification of the vague compounds possible.

Experiment of the formulation for the viral pesticide of nuclear polyhedrosis virus of the fall webworm, Hyphantria cunea Drury. (흰불나방 핵다각체병 바이러스의 제제화에 관한 시험)

  • The intent of this research is to acquire some basic informations about formulation of the viral pesticide, Hyphantria cunea nuclear polyhedrosis virus and its virulence under field condition. The nuclear polyhedrosis virus was formulated as wettable powder using spreader, sticker and U.V. protector. The formulated product and aqueous virus were diluted with water at the concentration of 1${\times}$106PIB/ml and sprayed on mulberry leaves in the field. The leaves were fed with 3rd instar larvae of H. cunea to determine the inactivation period of the viral pesticides. The aqueous virus was completely inactivated on 5th day after spray, while the formulated one showed a spare mortality to the larvae even on 20th day after spray. In field application test, The fromulated and aqueous virus were sprayed on individual mulberry tree and 3rd instar laevae of H. cunea were fed on the trees. The mortality of the larvae one day after spray of the formulated and aqueous virus were about 50% and 40%, respectively. The formulated virus exhibited a persistent virulence to the larvae up to 9th day after spray, which the mortality was approximately 30%. The residual virulence of the formulated and aqueous virus was extended up to 4th day and 2nd day after spray, respectively.

The toxicity of an IGR class insecticide, Diflubenzuron on silkworm, Bombyx mori and abnormal symptoms (IGR계 농약 diflubenzuron의 독성과 누에이상증상)

  • Surveying, laboratory and field trial were performed to elucidate the causes of non-spinning syndrome of silkworm, Bombyx mori, abnormal symptoms of silkworm instars observed in certain sericultural fanning region. Mortality and growth of silkworm was still influenced by feeding the mulberry leaves sprayed with 10,000 fold diluted solution of standard spray of diflubenzuron WP (25%). Mortality and growth of silkworm fed with diflubenzuron-sprayed mulberry leaves with recommended spray solution were entirely affected even six weeks after spraying irrespective to the instars. Larval period of 5th instar and cocooning duration of silkworm was not affected in case 50m of buffer zone has maintained in near-by orchard field sprayed with diflubenzuron WP (25%). However, survival rate of pupae was not influenced in case 100 m of buffer zone has kept. Insect growth regulating insecticides were required to keep a certain distance of buffer zone when sprayed with water-diluted solution, 100 m from the application site at least in order to prevent contamination of the spraying drift by the chemicals.

Quality Characteristics of Cookies Fortified with Soymilk Cake Fermented by Bacillus subtilis GT-D (고초균에 의한 비지 발효물을 첨가한 쿠키의 품질특성 연구)

  • Soymilk cake (SMC) fortified with defatted soy flour was fermented by a novel Bacillus subtilis GT-D in the solid-state fermentation with chlorella, mulberry leaves or green tea as food ingredient. The fermentation of SMC resulted in higher content of tyrosine and higher activities of protease, amylase and fibrinolytic enzymes. Fermented SMC including mucilage indicated six-fold higher content of dietary fiber compared to that of non-fermented SMC. The SMC fermented without additional ingredient allowed the highest spread ratio and width of cookies. Cookies prepared without SMC showed the lowest value of spread ratio and width compared to those of fermented SMC. In addition, these cookies showed the highest hardness value of 8,975 g, and thickness value of 5.97 and slightly higher bright color. Cookies fortified with SMC fermented with chlorella or mulberry leaves showed higher sensory evaluation value. Fortification of SMC fermented without additional ingredient resulted in the highest preference in the sensory evaluation of cookies.

Effect of Ultraviolet Light on Survival and Pathogenicity of Entomopathogenic Nematodes against an Alternative Host Silk Worm, Bombyx mori (자외선이 곤충병원성선충의 생존과 대체기주 누에에 대한 병원성에 미치는 영향)

  • This study was conducted to find a useful alternative herbivore system with which to study the effects of ultraviolet exposure on the pathogenicity and survival of Korean isolated entomopathogenic nematodes (Heterorhabditis sp. 202 strain, Heterorhabditis sp. Gyeongsan strain, Steinernema sp. 223 strain, S. carpocapsae Pocheon strain, S. glaseri Dongrae strain and S. longicaudum Nonsan strain). Pathogenicity of entomopathogenic nematodes against silkworm, Bombyx mori, differed depending on the nematode species and strain and instar of silkworm challenged. Steinernematidae were relatively more pathogenic to the 5th instar silkworms, whereas Heterorhabditidae were more pathogenic to the 1st instars. UV-C exposure was harmful to entomopathogenic nematodes. All nematodes were killed within 60 minutes of exposure of UV-C, and after 10 min of exposure they caused only 6.7% corrected mortality of silkworms. Silkworms were not infected when they were fed S. carpocapsae Pocheon strain treated mulberry leaves which were exposed to UV intensity of about $2.3mW/cm^2$ for 4 hours in the field. Pathogenicity of S. carpocapsae was not significantly reduced after 1 hour of exposure to $4.0mW/cm^2$ UV intensity on the mulberry leaves against silkworms.

Some Ecological Characteristics of the Mulberry Scale, Pseudaulacaspis pentagona T., and Its Control with Insecticides (뽕나무깍지벌레(Pseudaulacaspis pentagona T.)의 생태적 특성 및 약제방제에 관한 연구)

  • A study was conducted to provide some basic data for controlling the Mulberry scale insect(Pseudaulacaspis pentagona T.). Experiments were carried out with checking their field-life, and a few selected insecticides were tested when mulberry leaves were not in use. About 57% of female were survived from the overwintering attached to the bark. Over 80% of the scale were distributed within the range of 30cm from the base of each stem. One hundred percent and 96.7% of eggs were hatched under 25$^{\circ}C$ and 27$^{\circ}C$ respectively. Eighty percent of pupae emerged both under 25 and 27$^{\circ}C$. The Mulberry scale require about 33 days under 25$^{\circ}C$ and 30 days under 27$^{\circ}C$. This species had 2 generations each year in mulberry field in Suwon. Overwintered as a mated female only. Oviposited about 170 eggs under their scale starting from the late April (in 1994) through the mid May (in 1993). Phenthoate EC sprayed in the mid June against the larvae was very effect with over 96% of control value.

STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. NOTICE: A part of this treatise was presented at the annual Korea Sericultural Society meeting held in 1961.
