Processing of Convenient Rice Gruels with Sericultures (쌀과 양잠 산물을 활용한 즉석 죽 제조)

  • The purpose of this study was to develop traditional Korean rice gruels using sericultures. The SOD-like activities of the sericultures were in the order of 39.56%(Paecilomyces tenuipes: PT)>26.91%(Mulberry leaf: ML)>7.68%(Mulberry fruit: MF). The total phenolic acid contents were ML(0.21 mg/ml)>PT(0.16 mg/ml)>MF(0.07 mg/ml). The ML was the highest in all the groups. In the sensory evaluation, for overall quality, the rice gruel with ML was scored higher than the other samples, The brightness(L) of the rice gruels with sericultures were PT>MF>ML, and redness(a) was highest for MF(3.70) and yellowness(b) was highest for PT(17.74). The moisture contents of the gruels were 0.38%, 0.19%, and 0.07%, for ML, MF, and PT, respectively. The ash contents of the gruels were in the order of MF>PT>ML. The crude fat and protein contents were highest in the PT gruels.

Effects of mulberry fruit juice powder on inflammation and microRNA-132/143 regulation in 3T3-L1 adipocytes (뽕나무 열매 착즙 분말이 3T3-L1 지방세포의 염증 및 microRNA-132/143 조절에 미치는 영향)

  • Purpose: Mulberry (Morus alba L.) fruit is widely grown in Asia and consumed as fresh fruit, jam, and juices. The fruit has beneficial health effects, including anti-diabetic, anti-tumor, and anti-obesity properties. However, the mechanisms by which mulberry fruit juice powder (MJ) regulates inflammatory microRNAs (miRs) are not yet known. This study investigated the effect of mulberry fruit juice powder on the regulation of inflammation and miR-132/143 during 3T3-L1 adipocyte differentiation. Methods: The 3T3-L1 cells were induced to differentiate for 2 days and then treated with various concentrations of MJ for 7 days. Cytotoxicity was determined by evaluating cell viability using a water-soluble tetrazolium salt-8 assay kit. Intracellular lipid accumulation was evaluated by oil-red O staining. The levels of the expression of genes involved in adipogenesis and inflammation, and miR-132/143 were measured by quantitative real-time polymerase chain reactions. Results: MJ showed no cytotoxic effect on 3T3-L1 adipocytes at concentrations below 100 ng/mL. Intracellular lipid accumulation was reduced by MJ treatment at concentrations of 100 ng/mL. The messenger RNA (mRNA) levels of proliferator-activated receptor-γ, cytosine-cytosine-adenosine-adenosine-thymidine/enhancer-binding protein-α, and adipocyte protein 2, which are involved in adipogenesis, were suppressed by MJ. A reduction was also seen in mRNA levels of genes related to the inflammatory response, such as tumor necrosis factor-α, interleukin-6, monocyte chemoattractant protein-1, and inducible nitric oxide synthase. The expression of the inflammatory miR-132 and miR-143 was also decreased by MJ. Conclusion: These results suggest that MJ may suppress adipogenesis and inflammation through the regulation of miR-132/143 expression in 3T3-L1 adipocytes. Thus, MJ may be useful as a food agent that prevents obesity-associated inflammation.

Alleviating Effects of Mulberry Fruit Extract on Postprandial Hyperglycemia in Streptozotocin-induced Diabetic Mice (STZ으로 유도된 당뇨 마우스에서 오디열매추출물의 식후 고혈당 완화 효과)

  • Postprandial hyperglycemia is an early defect of type 2 diabetes and one of primary anti-diabetic targets. The alpha-glucosidase inhibitors regulate postprandial hyperglycemia by impeding the rate of carbohydrate (such as starch) digestion in the small intestine. This study was designed to investigate the inhibitory actions of mulberry fruit extract (MFE) on α-glucosidase and α-amylase activities, and its alleviating effect on postprandial hyperglycemia activities in vitro and in vivo. Male four-week old ICR mice and streptozotocin (STZ)-induced diabetic mice were treated with mulberry fruit extract. MFE showed strong inhibitory effects against α-glucosidase and α-amylase activities, with half-maximal inhibitory concentration (IC50) values of 0.16 and 0.14 mg/ml, respectively, and was more effective than acarbose, which was used as a positive control. The increase in postprandial blood glucose levels was more significantly attenuated in the MFE-administered group mice than in the control group mice of both STZ-induced diabetic and normal mice. Moreover, the area under the glucose response curve significantly decreased following MFE administration in diabetic mice. These results indicate that MFE may be a potent inhibitor of α-glucosidase and α-amylase, and helpful in suppressing postprandial hyperglycemia in diabetic mice. The mulberry fruit extracts may be considered as a potential candidate for the management of diabetes.

Quality characteristics of functional Nokdujuk prepared with optimum mixing ratio of mulberry leaf and fruit powder by response surface method (반응표면분석법을 이용한 최적 비율의 뽕잎과 오디 분말 첨가 기능성 녹두죽의 품질특성)

  • This study was performed to develop and evaluate functional prepared with optimum mixing of mulberry leaf and fruit powder using response surface method (RSM). In order to develop the optimized functional Nokdujukr using RSM, mulberry leaf powder (MLP:X1) and mulberry fruit powder (MLF:X2) were set as independent variables, and pH (Y1), sweetness (Y2), viscosity (Y3), L (Y4), a (Y5), b (Y6), color (Y7), flavor (Y8), taste (Y9), overall quality (Y10), TPC (Y11), and DPPH radical scavenging ability ($IC_{50}$)(Y12) were set as dependent variables. The optimum mixing ratio of MLP and MLF was determined to be 3.88 g of MLP and 6 g of MLF. The values of color, flavor, taste, overall quality, TPC, and DPPH radical scavenging ability ($IC_{50}$) of optimized Nokdujuk were 5.20, 5.85, 6.00, 6.22, 330.99 mg TAE/g and 650.10 g/mL, respectively. In conclusion, this study has led to the development of an improved version of Nokdujuk that has antioxidative properties and good sensory evaluation and, will likely serve as a functional meal replacement for the busy modern world.

Characteristics of mulberry cultivar 'Sangmaru' (Morus alba L.) for mulberry fruit production (오디 생산용 뽕나무 품종 '상마루'의 특성)

  • We bred a mulberry cultivar named Sangmaru, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at three places (Suwon, Cheongju, and Jangseong) for seven years from 2008. This is tetraploidy variety belonging to Morus alba L. made by colchicine treatment on growing point of 'Chungil (Morus alba L.)' winter buds. Sangmaru was high yielding cultivar in fruit productivity by 6% compared to control cultivar 'Chungil (Morus alba L.)' for five years. Although Sangmaru was similar in sugar content of mulberry fruits, it was bigger than that of 'Chungil'. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.

Inhibitory Effects of Complex of Mulberry Extract on Degenerative Arthritis (상심자추출물 등 복합물의 퇴행성관절염 억제효과)

  • Complex of mulberry extract (CME) is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). In this study, we investigated prevention effects of CME on degenerative arthritis. The $SC_{50}$ value of DPPH radical scavenging by CME was $158.49{\pm}11.35{\mu}/mL$. We found that CME significantly reduced the production of nitric oxide (NO) and protein expression of cyclooxygenase-2 (COX-2) in RAW 264.7 cells which were activated by LPS. Experiments using animal model of degenerative arthritis showed that CME (400 mg/kg body weight) inhibited the production of TNF-${\alpha}$ (77.5%) and IL-$1{\beta}$ (95.0%). Furthermore, it was observed that CME reduced to 85.9% of paw edema induced by carrageenan. These results suggest that CME could improve degenerative arthritis.

Characteristics of mulberry cultivar 'palchung' (Morus Lhou(Ser.) Koidz.) for mulberry breeding as pollen parent (오디 생산용 뽕 품종 육성을 위한 교배모본 '팔청'의 특성)

  • We bred a mulberry cultivar named Palcheong, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at four places (Suwon, Kongju, Wanju, and Jinju) for seven years from 2007. This is belonging to (Morus Lhou(Ser.) Koidz.) selected among mulberry genetic resources preserved in Suwon. Its sex expression was hermaphrodite, which can be used for the production of seedling for fruit production by artificial crossing as pollen parent Palchung showed lower yielding in fruit productivity by 16% compared to control cultivar 'Chungil (Morus alba L.)' for five years. Although Palchung was higher in sugar content of mulberry fruits, it was bigger than that of 'Chungil'. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.

A Study on the Antidiabetic Effect of Mulberry Fruits (오디의 항당뇨 효능에 관한 연구)

  • This study was designed to evaluate the antidiabetic effect of mulberry fruits using insulin-dependent and/or non-insulin-dependent diabetes mellitus animal models. The administration of mulberry fruit did not affect either body wight or blood glucose level in the normal ICR mice and streptozotocin induced-type I diabetic mice group. In second experiment, prolonged mulberry fruits treatment did not significantly attenuate the blood glucose level in type I diabetes induced by streptozotocin. In third experiment, the antidiabetic effect of mulberry fruits have been investigated using type II diabetes animal model that was induced by administration of streptozotocin to 2-day-old rats. Significant decrease in blood glucose level was observed in prolonged mulberry fruits treated group. In these treated group, the weight of liver significantly decreased than that of control group. In fourth experiment using KK mice showing genetical type II diabetes mellitus, glucose tolerance has been significantly recovered in mulberry fruits treated group but not in control group. In conclusion, prolonged administration of mulberry fruits significantly reduced the blood glucose level in type II diabetic animals. However, the blood glucose level was not significantly reduced by prolonged mulberry treatment. These data suggest that mulberry fruits can be developed as functional food that has effect on the insulin-independent diabetus mellitus(type II daibetus mellitus).

Quality Characteristics of Sikhye added with Mulberry (Morus alba L.) Fruit Concentrate (오디농축액 첨가에 따른 식혜의 품질 특성)

  • This study compared the physicochemical characteristics, proximate composition, taste compounds, and antioxidant properties of Sikhye prepared with mulberry fruit concentrate. Analysis of the physicochemical characteristics of Sikhye added with mulberry indicated that sugar content and titratable acidity increased significantly with increasing mulberry concentration, whereas pH decreased significantly. The whiteness index (L) was 36.77~51.40, which significantly decreased with increasing mulberry concentrate. The redness index (a) was -0.90~1.97 and highest in Sikhye sample containing 4% mulberry concentrate. The yellow index (b) was 0.03~1.90 and highest in Sikhye sample containing 1% mulberry concentrate. Analysis of the antioxidant properties of Sikhye added with mulberry indicated that total polyphenol content and flavonoid content increased significantly as the amount of mulberry concentrate increased above 1%. Total anthocyanin color also increased significantly with increasing mulberry concentrate. The mulberry Sikhye sample containing 8% extract showed the strongest antioxidant properties based on DPPH radical scavenging activity, reducing power, and FRAP assay. Evaluation of the sensory properties of Sikhye added with mulberry revealed that the most preferred flavor, color, and taste were observed in Sikhye samples containing 2%, 4%, and 8% extract, respectively. However, the highest overall preference was observed in Sikhye sample containing 2% extract, indicating that 2% concentration was most suitable for Sikhye and that flavor and aftertaste were more critical than taste. Analysis of the storage characteristics of Sikhye added with mulberry indicated that total bacteria count increased across all samples with increased storage period. However, total bacteria count in the added mulberry concentration group decreased in comparison to the control group as the amount of added mulberry increased.

Isolation and Development of Quantification Method for Cyanidin-3-Glucoside and Cyanidin-3-Rutinoside in Mulberry Fruit by High-Performance Countercurrent Chromatography and High-Performance Liquid Chromatography

  • Cyanidin-3-glucoside (C3G) and cyanidin-3-rutinoside (C3R) were isolated by high-performance countercurrent chromatography (HPCCC) using a two-phase solvent system composed of tert-butyl methyl ether/n-butanol/acetonitrile/water/trifluoroacetic acid (1 : 3 : 1 : 5 : 0.01, v/v) to give pure C3G (34.1 mg) and C3R (14.3 mg) from 1.5 g crude mulberry fruit extract. Using the pure C3G and C3R, a reliable high-performance liquid chromatography (HPLC) method was developed and validated to determine the C3G and C3R contents in mulberry fruit. C3G and C3R were separated simultaneously using an Eclipse XDB-C18 column ($4.6{\times}250mm$ I.D., $5{\mu}m$) coupled with a photodiode array detector (PDA). The gradient elution of the mobile phase consisting of acetonitrile (0.5% formic acid) and water (0.5% formic acid) was applied (1.0 mL/min), and the detection wavelength was 520 nm. The calibration curves of C3G and C3R showed good linearity (both with $r^2=0.9996$) in the concentration range $15.625-500{\mu}g/mL$, and the relative standard deviations (RSD%) of intra- and inter-day variability were in the ranges 2.1 - 8.2% and 4.1 - 17.1%, respectively. The accuracies were ranged 96.5 - 102.6% for C3G and C3R, respectively. The developed HPLC method was used to determine the contents of C3G and C3R in newly harvested mulberry from eight different provinces of Korea.
