가미육군자탕(加味六君子湯)이 호흡기 뮤신분비 및 뮤신 유전자 발현에 미치는 영향 (Effect of Gamiyukgunja-tang on Secretion and Gene Expression of Airway Mucin)

  • In the present study, the author intended to investigate whether Gamiyukgunja-tang (Jiaweiliujunzi-tang, GYGT) significantly affect both mucin release from and MUC5AC gene expression in cultured hamster tracheal surface epithelial (HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of GYGT to assess the effect on 3H-mucin release. Possible cytotoxicity of the agent was assessed by measuring lactate dehydrogenase (LDH) release. Total elution profiles of control spent media and treatment sample through Sepharose CL-4B column were analysed and effect of GYGT on MUC5AC gene expression in cultured HTSE cells were investigated. GYGT did not affect mucin release from cultured HTSE cells. GYGT did not show significant cytotoxicity. GYGT also did not affect the secretion of the other releasable glycoproteins with less molecular weight than mucin. GYGT increased the expression level of MUC5AC gene. We suggest that the effect of GYGT with their components should be further investigated through ongoing research.

  • Objectives : This study was done with intend to investigate whether two oriental medical prescriptions, daecheongryong-tang (DCRT) and prescription A (P-A) significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. Methods : Confluent HTSE cells were metabolically radiolabeled with $^3H$-glucosamine for 24 hrs and chased for 30 min in the presence of DCRT or P-A to assess the effect of each agent on $^3H$-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase (LDH) release. Also, the effects of DCRT and P-A on contractility of isolated tracheal smooth muscle were investigated. Results were as follows : 1. DCRT significantly inhibited mucin release from cultured HTSE cells, with significant cytotoxicity. 2. P-A significantly increased mucin release from cultured HTSE cells, with significant cytotoxicity. 3. DCRT inhibited Ach-induced contraction of isolated tracheal smooth muscle. 4. P-A also inhibited Ach-induced contraction of isolated tracheal smooth muscle. Conclusion: Results suggest that DCRT and P-A have regulating effects on mucin secretion from goblet cells. Further investigation is needed, because of the value in finding novel agents to this purpose, and these oriental medical prescriptions have potential for this role.

  • Objectives : In the present study, the authors intended to investigate whether two oriental medical prescriptions named chwiyeon-tang and chihyosan-gamibang significantly affect much release from cultured hamster tracheal surface epithelial HTSE cells. Methods : Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of chwiyeon-tang or chihyosan-gamibang to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase LDH release. Also, the effect of chwiyeon-tang and chihyosan-gamibang on contractility of isolated tracheal smooth muscle were investigated. Results : (1) Chwiyeon-tang significantly inhibited mucin release from cultured HTSE cells, with significant cytotoxicity ; (2) Chihyosan-gamibang significantly stimulated mucin release from cultured HTSE cells, with minute cytotoxicity ; (3) Chwiyeon-tang and Chihyosan-gamibang did not affect contractility of isolated tracheal smooth muscle. Conclusions : We suggest that the effects of Chwiyeon-tang and Chihyosan-gamibang with their components should be further investigated and it is of great value to find, from oriental medical prescriptions, noel agents which might regulate mucin secretion from airway goblet cells.

  • Objective : To investigate the effectiveness of two oriental medical prescriptions named Sigoungcheongpyetang(SCPT) and Tonggyutanggamibang(TGT) for mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Method : Confluent HTSE cells were metabolically radiolabeled with $^{3}H-glucosamine$ for 24hrs and chased for 30 min in the presence of SCPT or TGT to assess the effect of each agent $^{3}H-mucin$ release. Possible cytotoxicities of each agent were assessed by measuring lactate dehy drogenase(LDH) release. Also, the effects of SCPT and TGT on contrectility of isolated tracheal smooth muscle were investigated. Results : (1) SCPT and TGT significantly increased mucin release from cultured HTSE cells, with significantly cytotoxicity ; SCPT did not affect contractility of isolated tracheal smooth muscle and TGT inhibited Ach-induced contraction of isolated tracheal smooth muscle. Conclusion : We suggest that the effects of SCPT and TGT with their components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which might regulate mucin secretion from airway goblet cells.

Conclusions : The author suggests that the effects of SCTS and P-D with their components should be further investigated and it is valuable to find, from oriental medical prescriptions, novel agents which might regulate mucin secretion from airway goblet cells.

Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presen+ce of GSGT or GCPT to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed dy measuring lactate dehydrogenase(LDH) release. Also, the effects of GSGT and GCPT on contractility of isolated tracheal smooth muscle were investigated. (1) GSGT did not affect mucin release without cytotoxicity ; (2) GCPT significantly stimulated mucin release from cultured HTSE cells, with significant cytotoxicity ; (3) GSGT and GCPT did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of GCPT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have potent expectorant effects on mucin secretion from airway goblet cells.

  • To elucidate the ichthyotoxic mechanisms of a harmful dinoflagellate Cochlodinium polykrikoides, biochemical and hematological responses of fish exposed to blooms were investigated. Particularly, based on our finding that oxidative damages of gill were associated with fish mortality, dysfunction of ion-transporting enzymes and secretion of gill mucus of fish exposed to this bloom species were examined.   • The purpose of this study was to report and discuss the diagnosis and treatment of obstructive atelectasis secondary to pus obstruction in a patient who had developed a maxillofacial abscess, and to review the literature on similar cases. Persistently discharging pus within the oral cavity can act as an aspirate, and may lead to obstructive atelectasis. Additionally, maxillofacial surgery patients should be carefully assessed for the presence of risk factors of obstructive atelectasis, such as, epistaxis after nasotracheal intubation, oral bleeding, and mucus secretion. Furthermore, patients with these risk factors should be continuously followed up by monitoring $SPO_2$, breath sounds, and chest x-ray.

Siphonochilus aethiopicus, a traditional remedy for the treatment of allergic asthma

  • Asthma is a chronic inflammatory disease of the lungs, characterized by increased sensitivity to bronchoconstriction associated with infiltration of immune cells and mucus hyper secretion. In South Africa, the indigenous plant Siphonochilus aethiopicus, is used by traditional health practitioners to treat colds and flu, wheezing of the chest, coughs, influenza, sinus problems and mild asthma. In this study we aimed to investigate the potential anti-inflammatory and immune-modulating properties of S. aethiopicus in vitro. The dried and powdered S. aethiopicus plant material was extracted with organic solvents. The dried extracts were screened in vitro in the transcription response, NF-${\kappa}B$ and a cytokine assay. Significant activity was observed for organic extracts of the plant in these assays. This study provides evidence that S. aethiopicus has anti-inflammatory and immune-suppressing properties in vitro. These findings may support anecdotal accounts of its effectiveness against allergic asthma.

선천면역 및 적응면역에서 비만세포의 기능 (The Role of Mast Cells in Innate and Adaptive Immunity.)

  • The function of mast cells as effector cells in allergy has been extensively studied. Mast cells activated through high affinity IgE-receptor ($Fc{\varepsilon}RI$) release diverse mediators, and lead to smooth muscle constriction, vasodilation, increase of vascular permeability, leukocyte recruitment and activation, mucus secretion, and tissue proliferation and remodeling. However, various other immunological and non-immunological signals can lead to the activation of mast cells. In resent years, mast cells have been identified to be involved in a complex range of immune functions. Mast cells can be important as key players in the regulation of innate as well as adapted immune responses, and may influence the development of allergy, autoimmune disorder and peripheral tolerance. This review summarizes the recent advances in the understanding of effector functions of mast cells in immune responses.
