Objectives: The purpose of this study is to determine the exposure risk to tattoo components by analyzing skin absorption using the in vitro method. Tattoos are commonly used for cosmetic purposes, and the skin of not only the operator but of the people who are undergoing the cosmetic procedure is continuously exposed to hazardous chemicals. Methods: Skin permeation risk determination was conducted by the in vitro Franz diffusion cell method according to the ingredient types of tattoo dyes, such as volatile organic compounds (VOCs), non-volatile organic compounds and heavy metals, using hairless mouse full skin and human cadaver epidermis. Results: The major components with good skin penetration for each type of tattoo dye ingredient were clarified. Among the tatto dye ingredients, 1,2-Dichlorobenzene, Zn, Al, Pb and Ti showed good skin penetration. Most of the skin transmission rates were higher in hairless mouse full skin than in human cadaver epidermis. Conclusion: A possible exposure risk to hazardous substances in tattoo dyes was confirmed from this study. These results are expected to provide a positive contribution to the establishment of management regulations for tattoo dyes.

Objective : Prolonged use of systemic corticosteroids negatively impacts skin barrier function. Corticosteroids have a major role in the practical management of many diseases, so it is necessary to find the drug or supplement which could keep the skin healthy during the systemic corticosteroids therapy. Seed of Trichossnthcs kiiilowii was commonly used for pulmonary disease in traditional Chinese medicine. Many studies have investigated and found that seed of Trichosanthes kirilowii has anti-bacterial, anti-HIV, anti-tumor and immunoregulatory functions. This study was performed to investigate the effect of seed of Trichosmthes kirilowii on the Skin Barrier Method : Triamcinolone 0.4mg was injected male hairless mice for 5 weeks ( 2 times a week, totally 10 times), Just before the first injection, hairless mice were divided into 3 groups; Control ( normal saline medicated group ), GroupA( seed of Trichosuuhes kirilowii qd/day medicated group ) and GroupB( seed of Trichosuuhes kiiilowii bid/day medicated group ), Body weight of all the groups were checked during the experiment, After $10^{th}$ injection, TEWL (Transeidermal water loss) of 3 groups were artificially increased by 9 times tape stripping and the changes of TEWL were checked at before stripping, right after, 2h, 4h, 6h, 24h, 48 and 72h later after stripping, AST, ALT and CBC were also checked. Indeed, stratum corneum of 3 groups were also examined and compared with that of normal hairless mouse. Results : 1. GroupB showed significantly lower body weight among three groups 2, There was no statistical difference at AST, ALT and CBC among three groups 3. GroupB was showed significantly lower TEWL than TEWL of Control at 48h later. 4 GroupB recovered it's TEWL of before tape stripping at 24h later, GroupA recovered that at 48h and Control recovered that at 72h later at the same condition 5. Stratum corneum of GroupA and GroupB were looked almost normal and healthy. On the contrary, stratum corneum of Control was looked thin and unhealthy. Conclusions : Seed of Ttichossnthes kirilowii has some effects on skin barrier function, especially TEWL of high dose corticosteroids injected hairless mouse and the efficacy seemed to be related with the dosage.
This study was intended to identify the effectiveness of Lithospermum erythrorhizon in the UVB-damaged mouse skin. The C57BL/6 mouse that weighted about 18 g were divided into three groups; the control group (A group), the UVB irradiated group (B group), and the group treated with Lithospermum erythrorhizon extracts after UVB irradiation (C group). In the results of superoxidase dismutant(SOD) activities, the C group was meaningfully lower at the 48hrs, 120 hrs, and 168 hrs group than B group (p<0.05). In the result of catalase(CAT) activities, the C group was decreased at the 120 hrs and 168 hrs group, but meaningless (p>0.05). In the result of light micrograph observation, the B group observed sunburn cells in the epidermis and acute inflammation in the dermis at the 24 hrs and 48 hrs. And proliferation of the epidermis and the stratum granulosum, and found the hyperkertosis at the 72 hrs, 120 hrs, and 168 hrs group than C group. The C group alleviated inflammation in the dermis than B group at the 24 hrs and 48 hrs. And inhibited the proliferation of the epidermis at the 72 hrs, 120 hrs, and 168 hrs groups than B group. In conclusion, Lithospermum erythrorhizon water extracts may protect the UVB-damaged mouse skin.
Antisense phosphorothioate oligodeoxynucleotide(PS-ODN) against $TGF-{\beta}$ was developed as scar formation inhibitor. The scar was caused collagen deposition due to overexpression of $TGF-{\beta}$ in wounded skin. The percutaneous absorption of partially modified PS-ODN(25 mer) was investigated for the purpose of its effective delivery. Though PS-ODN has high molecular weight (MW=8,000) and polyanionic charge, it was permeated through skin. The skin permeation of PS-ODN was markedly increased by the removal of stratum corneum and dermis. Moreover, the skin permeation of PS-ODN was decreased in the following order; hairless mouse skin>rat skin>human cadaver skin. Thus, PS-ODN represents a logical candidate for further evalution due to the potential for delivery into the wounded skin.
Extracellular interleukin 1 alpha (IL-$1{\alpha}$) released from keratinocytes is one of the endpoints for in vitro assessments of skin irritancy. Although cells dying via primary skin irritation undergo apoptosis as well as necrosis, IL-$1{\alpha}$ is not released in apoptotic cells. On the other hand, active secretion has been identified in interleukin-1 receptor antagonist (IL-1ra), which was discovered to be a common, upregulated, differentially-expressed gene in a microarray analysis performed with keratinocytes treated using cytotoxic doses of chemicals. This study examined whether and how IL-1ra, particularly extracellularly released IL-1ra, was involved in chemically-induced keratinocyte cytotoxicity and skin irritation. Primary cultured normal adult skin keratinocytes were treated with cytotoxic doses of chemicals (hydroquinone, retinoic acid, sodium lauryl sulfate, or urshiol) with or without recombinant IL-1ra treatment. Mouse skin was administered irritant concentrations of hydroquinone or retinoic acid. IL-1ra (mRNA and/or intracellular/extracellularly released protein) levels increased in the chemically treated cultured keratinocytes with IL-$1{\alpha}$ and IL-$1{\beta}$ mRNAs and in the chemically exposed epidermis of the mouse skin. Recombinant IL-1ra treatment significantly reduced the chemically-induced apoptotic death and intracellular/extracellularly released IL-$1{\alpha}$ and IL-$1{\beta}$ in keratinocytes. Collectively, extracellular IL-1ra released from keratinocytes could be a compensatory mechanism to reduce the chemically-induced keratinocyte apoptosis by antagonism to IL-$1{\alpha}$ and IL-$1{\beta}$, suggesting potential applications to predict skin irritation.
The usefulness of hypertermia for cancer therapy have well been established. The purpose of the present investigation was to ascess the effect of step-up $(42^{\circ}{\rightarrow}44^{\circ}C$ sequence) and step-down $(44^{\circ}{\rightarrow}42^{\circ}C$ sequence) heating on the skin of the hind foot of the mouse. Hyperthermic treatments were given by immersion the hind foot of the mouse in circulating water baths. Skin response was studied by the leg reaction, which was scored according to a numerical scoring system proposed by Urano et al (1980). The results were as follows 1. The skin damage of $44^{\circ}C$ control group was more severe than $42^{\circ}C$ control group (P<0.05), except for 15 min. heating group. 2. The Skin damage of step-down group was more severe than step-up group (P<0.05). 3. The skin damage of $44^{\circ}C$ control group was more severe than step-up group when there is no difference in $44^{\circ}C$ heating time of step-up group from $44^{\circ}C$ control group (P<0.05). 4. In step-down group, the skin damage was more severe than $44^{\circ}C$ control group after preheating 45 min at $44^{\circ}C$ (P<0.05). Therefore, the above findings suggest the normal tissue damage by step-up heating was correlated with heating time of post step-up. The dropping of heating temperature in late phase had more severe damage of the skin than that in early phase during hyperthermia, and so contineous control of satisfactory temperature should be considered as the one of the most important factor for prognosis, complications of clinical hyperthermia
The aim of this study was to investigate the effect of charge carrier lipid on the skin penetration, retention, and hair growth of topically applied finasteride-containing liposomes. Finasteride-containing liposomes were prepared by traditional thin film hydration method using Phospholipon$^{(R)}$ 85 G and cholesterol with or without charge carrier lipid (1,2 dimyristoyl-sn-glycero-3-phosphate or 1,2-dioleoyl-trimethylammonium-propane for anionic and cationic charge, respectively). Freshly prepared finasteride-containing liposome suspension was applied on the hairless mouse skin, and skin penetration and retention were measured using Keshary-Chien diffusion cell. Non-liposomal formulation (ethanol 10% solution containing 0.5 mg/ml of FNS) was also used as a control. The amount of finasteride in the diffusion cell and mouse skin was measured by HPLC. The hair growth was evaluated using depilated male C57BL/6N mice. Mean particle size of all finasteride-containing liposomes was less than a micron, and polydispersity index revealed size homogeneity. Skin penetration and retention studies showed that significantly less amount of finasteride was penetrated when applied as anionic liposome while more amount of the drug was retained. Specifically, in liposome prepared with 10% anionic charge carrier lipid, penetration was 12.99 ${\mu}g/cm^2$ while retention was 79.23 ${\mu}g/cm^2$ after 24 h of application. In hair growth study, finasteride-containing anionic liposomes showed moderate efficacy, but the efficacy was not found when applied as cationic liposomes. In conclusion, topical application of finasteride using anionic liposome formulation appears to be useful option for the treatment of androgenetic alopecia to avoid systemic side effects of the drug.

BACKGROUND/OBJECTIVES: Ultraviolet radiation (UV) is a major cause of skin photoaging. Previous studies reported that ethanol extract (PET) of Prunus persica (L.) Batsch flowers (PPF, peach flowers) and its subfractions, particularly the ethylacetate (PEA) and n-butanol extracts (PBT), have potent antioxidant activity and attenuate the UV-induced matrix metalloproteinase (MMP) expression in human skin cells. In this study, we investigated the protective activity of PPF extract against UV-induced photoaging in a mouse model. MATERIALS/METHODS: Hairless mice were treated with PET or a mixture of PEA and PBT either topically or orally along with UV irradiation. Histological changes and biochemical alterations of mouse skin were examined. Major phenolic compounds in PPF extract were analyzed using an ACQUITY UPLC system. RESULTS: The overall effects of topical and oral treatments with PPF extract on the UV-induced skin responses exhibited similar patterns. In both experiments, the mixture of PEA and PBT significantly inhibited the UV-induced skin and epidermal thickening, while PET inhibited only the UV-induced epidermal thickening. Treatment of PET or the mixture of PEA and PBT significantly inhibited the UV-induced MMP-13 expression, but not type I collagen expression. Topical treatment of the mixture of PEA and PBT with UV irradiation significantly elevated catalase, superoxide dismutase (SOD) and glutathione-peroxidase (GPx) activities in the skin compared to those in the UV irradiated control group, while oral treatment of the mixture of PEA and PBT or PET elevated only catalase and SOD activities, but not GPx. Thirteen phytochemical compounds including 4-O-caffeoylquinic acid, cimicifugic acid E and B, quercetin-3-O-rhamnoside and kaempferol glycoside derivatives were identified in the PPF extract. CONCLUSIONS: These results demonstrate that treatment with PET or the mixture of PEA and PBT, both topically or orally, attenuates UV-induced photoaging via the cooperative interactions of phenolic components having anti-oxidative and collagen-protective activities.

Highly toxic chemical warfare agents(CWA) could be used in chemical warfare and terrorism. The protection of skin is crucial for civilians and soldiers, because the primary routes of exposure to CWA are inhalation and skin absorption. Thus, topical skin protectants(TSP) have been studied and developed in many countries to complement protective equipments. In this study, in-vitro test procedure was optimized and established using a flow-through diffusion cell system containing excised hairless mouse skin in an attempt to assess the effectiveness of various TSP formulations against nerve agent simulants. In addition, the test results on the formulations including the ingredients used in SERPACWA(Skin Exposure Reduction Paste Against Chemical Warfare Agent) and IB-1(TSP of Israel) were included, indicating that the formulations with perfluorinated compounds were more effective than the glycerin-based formulations.
Chronic ultraviolet (UV) exposure is the major cause of photoaging that causes skin wrinkling, roughness, dryness, laxity, and pigmentation. Recently, increasing efforts are being made to understand the relationship between foods and skin health. Ginsenosides are present in ginseng (Ginseng Radix Rubra) extract, and are known to have biomedical properties, such as, anti-oxidant and anti-inflammatory effects. In this study, we investigated whether KTNG0345 prepared from red ginseng extracts delivered orally reduces skin wrinkling and ultraviolet B (UVB)-induced wrinkle formation in hairless mouse skin. KTNG0345 was administrated orally to the mice (5 times a week) during the period of UVB-irradiation (3 times a week) for 8 weeks at three different doses of 300 mg/kg, 500 mg/kg and 1000 mg/kg (w/v). UV doses were increased weekly by 1 MED (1MED = 75 $mJ/cm^2)$ up to 4 MED and then maintained at this level. After the 8-week administration period, it was found that orally administered KTNG0345 significantly inhibited UVB-induced wrinkle formation in a dose-dependent manner. Increases in skin thickness caused by UVB were prevented by KTNG0345. Moreover, it also significantly inhibited matrix metalloproteinases (MMP) -13 and MMP-9 expressional inductions by UVB. In addition, KTNG0345 was observed to prevent UVB-induced water loss of epidermis in hairless mouse skin. Our results demonstrate that orally administered KTNG0345 has anti-wrinkling effects in hairless mouse skin, and suggest that dietary red ginseng and herbal mixture may be considered a functional beauty food for preventing UVB-induced skin wrinkles.
