Effects of Taeeumjowuitanggagam-bang on the Expression of Adipocytokines and SREBPs in Liver of db/db Mouse (태음조위탕가감방(太陰調胃湯加減方)의 db/db 마우스 간(肝)에 대한 아디포사이토카인 및 SREBPs의 발현에 미치는 영향)

  • The aim of this study was to investigate the effect of Taeeumjowuitanggagam-bang(TJV) on the mRNA expression of adipocytokines (TNF-${\alpha}$ and IL-6), and SREBPs. Mice were divided into 4 groups ; a normal group of db/+ mice, a control group of db/db mice, a group (db/db mice) treated with TJV 200 mg/kg, and a group (db/db mouse) treated with TJV 500 mg/kg. They were treated orally with TJV and measured their body weight every other day during 9 weeks. After that, we measured the mRNA expression of adipocytokines (TNF-${\alpha}$ and IL-6) and SREBPs (SREBE-1a and SREBP-1c) in liver, and blood concentration of total cholesterol, triglyceride, too. In addition, liver samples were fixed in 4% PFA for 2 hours and stored in $-70^{\circ}C$. Liver tissues were embedded in Optical cutting temperature(OCT) compound and 30 ${\mu}m$ sections were cut. Tissue sections were stained Oil-Red-O to visualize neutral lipids. Nuclei were stained with hematoxylin solution. In result, the TJV reduced the mRNA expressions of TNF-${\alpha}$ and IL-6 and SREBPs in liver and stained liver tissue less red than control group. However, there was no significant differences in total cholestreol and triglyceride blood concentration and body weight among groups. The TJV has inhibitory effect on the mRNA expression of adipocytokines and SREBPs. Therefore, it is assumed that the TJV is related to inhibiting lipogenesis in the liver

Scavenging Effects of Flavonoids on Paraquat Induced Pulmonary Toxicity (Paraquat 유도 폐독성에 대한 Flavonoid류의 독성경감 효과)

  • To investigate and evaluate the scavenging and antioxidative effects of various ftavonoids on paraquat induced pulmonary toxicity, in vivo and vitro tests of eight flavonoids(catechin, epicatechin, flayone, chrysin, apigenin, quercetin, morin and biochanin A) were carried out. In vitro test, inhibitory and antioxidative effects of lipoxygenase dependent lipidperoxidation, NADPH dependent cytochrome p-450 reductase to liver and lung microsome and superoxide anion production in rat peritoneal exudated macrophage were studied. In vivo test, biochemical parameters and cell population in bronchoalveolar lavage fluid(BALF) in mouse and rats after administration of paraquat and flavonoids were tested. The results are summerized as follows; 1. All flavonoids tested inhibited on NADPH dependent cytochrome p-450 reductase in liver and lung microsome. 2. All flavonoids tested showed the inhibitory effects on the superoxide anion production in rat peritoneal exudated macropharge. 3. Lactate dehydrogenase, acid phosphatase and total protein in BALF of mouse which increased by the administration of paraquat, decreased significantly by catechin, chrysin, morin and biochanin A. 4. Numbers of alveolar macropharge and PMN in BALF of rats which increased by the administration of paraquat decreased by all the tested flavonoids. Therefore, all flavonoids tested showed the useful compounds for scavenger and antioxidant on paraquat induced pulmonary toxicity.

  • Detoxication effect of red ginseng extract against toxicity of methly mercury on ultrastructure of mouse liver was studied by electron microscopy. The hepatocyte of methyl mercury treatment group showed partial rupture of nucleus, mitochondrial swelling, decrease of glycogen content, and appearance of a great number of large vacuoles. While, the hepatocyte of methyl mercury-red ginseng extract treatment group showed slight mitochondrial swelling and decrease of vacuoles in size and number than those of methyl mercury treatment group.

Effects of Zinc Deficiency on Immune Response in Mouse (식이 아연이 Mouse의 면역 반응에 미치는 영향에 관한 연구)

  • The purpose of this study was to investigate the effects of dietary zinc on immune response in mice. Weanling male mice was placed individually in stainless steel cages and fed a zinc dificient diet and control diet. All mice were given deionized water ad libitum. The introduction of extraneous zinc was minimized in all cage by washing feed jars and water bottles sequentially with 4mM EDTA and conc-nitiric acid followed by deionized water. After 4 and 5 weeks of the diets, mice were immunized with lx 106 Naegleria fowleri intraperitoneally. Mice were weighed once a week. The results from this study are summarized as followed ; 1) Mice fed the zinc dificient diet showed growth retardation. After 3 weeks of diets, mean body weight of zinc deficient mice was 21.4g and that of control was 25.0g. This difference is singnificant statistically (p<0.01). The more time passed, the more remarkable difference was found. 2) The weigth of organs were measured on liver, kidney, spleen, thymus, heart, lung, brain. Difference in weight were observed only in liver and spleen. 3) Proliferative response of spleen cells of zinc deficient mice to con A was lower than that of control mice after one week on immunization(p<0.005). 4) Stimulation index was lower in zinc deficient mice to phytohemagglutinin after two weeks on immunization (p<0.05). 5) Blastogenesis of speen cells of zinc deficient mice to Naegleria fowleric lysate was lower after 10 days on immunization (p<0.05). 6) Immunoglobulin G antribody titers of zinc deficient mice sera by ELISA was lowered to control mice after 5 weeks on immunization (p<0.005).

  • Objective : For various reasons, Obesity & Rhinitis are constantly rising. So Interest of treatment has been expanding. We want to verify The Chungyeol (fire extinguishing) Lisup (Draining) effect of Bangpungtongsungsan on obese model of allergy rhinitis.Material and Methods : BALB/c mouse were divided four groups: control(CON), allergic induction(ARE), Bangpungtongsungsan extract administration(BTT), Bangpungtongsungsan double concentration extract administration (BT2T). Every group except control group were caused allergic rhinitis by Ovalbumin. BTT & BT2T were orally administered the Bangpungtongsungsan for 21days. Since then we observed the liver tissue cell and the nasal mucous membrane.Results : In comparison with ARE, experimental groups show relief of the nasal mucous membrane damage(secretion of mucus decrease, Itching decrease), Th2 eruption control(IL-4 decline), effect of anti-inflamatory(reducing TNF-α creation, decreasing of iNOS through NF-κB activation-inhibition). In addition, experimental groups show a loss in weight, diminished accumulation of fat. (decreasing within liver tissue, reducing TNF-α creation) BT2T is more effective to BTT.Conclusion : Bangpungtongsungsan treat obese model on allergy rhinitis thereby control fat augmentation, relieving inflammation.

Antiviral Potential of the Silkworm Deoxynojirimycin against Hepatitis B Virus

  • Over 350 million people worldwide are chronic carriers of hepatitis B virus (HBV). Chronic viral infections of the liver can progress to cirrhosis, which may ultimately lead to hepatic failure or the development of hepatocellular carcinoma. There are two antiviral drugs on the market approved for clinical management of chronic HBV infections; interferon-alpha and the nucleoside analog lamivudine. However, they showed adverse side-effects. In the rational drug design for such therapies we would like to utilize antiviral drugs that inhibit the HBV replication in the liver. Investigation of natural extracts of silkworm exhibiting antiviral potential was held in the functional HBV polymerase activity and the release of virion particle in the HepG2.2.15 cell lines. HBV-producing transgenic mouse fed with silkworm DNJ molecule was shown as an inhibitor of serum HBV particles. We could represent this DNJ molecule as an antiviral potential complementing conventional therapies after preclinical tests against WHBV-infected animal model, woodchuck.

  • We have studied the mechanism by examing the effect of ginseng on the epoxide hydrolase which is catabolized the reactive intermetabolite of bromobenzene. and bromobenzene-induced hepatotoxicity. It was observed that ginseng saponin fraction protects against bromobenzene-induced hepatotoxicity in mice as evidenced 1. increased the epoxide hydrolase activity. 2. lower serum transaminase activity. 3. decreased the formation of lipid peroxide. These results suggested that the inducing effect of ginseng on the epoxide hydrolase is believed to be a possible detoxication mechanism for the bromobenzene toxicity in mice.

Free Redical Scavenging and Cytotoxicity Activitives of Soybean Germ Saponin (대두배아 사포닌의 유리기 생성 억제 및 세포독성)

  • This study was carried out to investigate functional activities of the free radical scavenging and germ of Glycin max. Merrill fur cytotoxicity toward P338 and L1210 cells derived from mouse. Effect of crude saponin were examined to oxygen radicals and their scavenger enzymes in liver fractions of Spragae-Dawley(SD) rats. Male rats were fed basic diets of control and experiment diets of 0.5∼1.0% crude saponin. There were no significant differences in hydroxy radical($.$OH) formation of liver mitochondria and microsomes in 1.0% group, while $.$OH formations were significantly decrease in 0.5% and 1.0% saponin compared with control group. Their oxygen radical(O$_2$$\^$$.$/) scavenging activities were significantly decrease in liver cytosol of 0.5% and 1.0% saponin group compared with control group. Soybean germ saponin was isolated purified by the method of HPLC to investigate the cytotoxicity of mouse cells by using the MTT assay. SA-1 saponin fraction of soybean germ showed to inhibit toward growth cell of P338 and L1210 cells and its showed less than 50% cytotoxicity These results suggest that the saponin may play a effective role in attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

Effects on Mammalian Tissues and Cells by Sulfur Containing Compounds (황함유 화합물이 동물의 조직과 세포에 미치는 영향)

  • To know the stress response and antioxidative effect of sulfur containing compounds, we observed the expression of the stress protein (heat shock protein; inducible protein) from mouse tissues and evaluated the protective effects to hydroxyl radical in mouse brain cell culture. Cysteine, methionine or sodium sulfide was fed by oral administration of 1 ml/per 6hr/three times with 1 mM, 2mM or 3mM to mouse, respectively. After that, the stress proteins were extracted from mouse tissues and analyzed the features of expression. The stress proteins by sulfur containing compounds were showed different aspects in the kinds and concentrations of their compounds, and in the tissues of mouse. In the liver, the stress proteins were appeared at different time on the concentration of sulfur containing compounds and had less than 20 KDa as small molecules. In general, the molecular weights of stress protein in liver, the stress proteins were appeared at different time on the concentration of sulfur containing compounds and had less than 20 KDa as small molecules. In general, the molecular weights of stress protein in the spleen were evaluated from 32KDa to 50KDA, and the induced times were relatively late at high concentration of cysteine, early at low concentration of methionine or sodium sulfide. The stress proteins in mouse muscle were detected mostly between 24hr after treatment of sulfur containing compounds. Their molecular weights were 15~24KDa. In the antioxidative effects of sulfur containing compounds to hydroxyl radical, cell viabilities were measured by 63.2% at 10 $\mu\textrm{M}$, 65.5% at 50 $\mu\textrm{M}$, 68.6% at 100 $\mu\textrm{M}$, 78.3% at 150 $\mu\textrm{M}$, or 83.0% at 200 $\mu\textrm{M}$ of cysteine, respectively. At addition of methionine, the cell viabilities were assessed as 58.1% at 10 $\mu\textrm{M}$, 62.8% at 50 $\mu\textrm{M}$, 75.7% at 100 $\mu\textrm{M}$, 78.6% at 150 $\mu\textrm{M}$, and 79.2% at 200 $\mu\textrm{M}$ after 4hrs exposure with 20mU/ml glucose oxidase (GO) system, while the numbers of live cells to hydroxyl radicals in treatment of sodium sulfide were showed 48.6% at 10 $\mu\textrm{M}$, 54.8% at 100 $\mu\textrm{M}$, 51.8% at 150 $\mu\textrm{M}$, and 51.6% at 200 $\mu\textrm{M}$ in the neuronal cells. In the inhibitory effects on the proliferation of tumor cells, percentages of dead cells of the CT-26 or HeLa cell were generally less than 30% even 48hr after addition of sulfur containing compounds. Conclusively, the results of these experiments indicate that stress protein by sulfur containing compounds can be used as physiological indicator for animal nutrition and for environment, and also that cysteine and methionine can play critical roles as an antioxidant.

  • The purpose of this study is to investigate the effects of bowontang on serum reactions of $CCl_4$- treated mouse. In this study, the experimental mouse were divided into three groups, normal, control, and sample. Under the same condition, the normal group was fed basal diet and water, the control group was injected with carbon tetrachloride ($CCl_4$, 0.6ml/kg) and fed a basal diet for 2 weeks, and the sample group was injected with carbon tetrachloride ($CCl_4$, 0.6ml/kg) and fed the bowontang extract (900mg/kg) for 2 weeks. It was shown that scavenging effect on DPPH radical was depended on concentration of the bowontang. And activities of GOT, GPT and ALP in blood serum of the sample group were significantly decreased as compared with the control group, respectively. Also, activities of LPO, GSH and catalase in the liver were significantly changed in experimental mice. In the liver of the sample groups, the activity of LPO was decreased as compared with control group, while activities of GSH and Catalase were significantly increased.
