Genetic Diversity of Seven Strawberry mottle virus Isolates in Poland

  • The studies on detection of the Strawberry mottle virus (SMoV) have been conducted in Poland for breeding programme purpose and for producers of strawberry plant material. Leaf samples collected from infected strawberry plants were grafted on Fragaria sp. Indicators which were maintained in greenhouse for further study. Seven Fragaria vesca var. semperflorens 'Alpine' indicators infected by SMoV were used for the study aimed on molecular characterization of virus isolates. Partial RNA2 was amplified from total nucleic acids using the RT-PCR method. The obtained amplicons separately digested with BfaI, FauI, HaeIII, HincI, and TaqI enzymes showed different restriction profiles. The nucleotide sequences analysis of RNA2 fragment confirmed the genetic diversity of the SMoV isolates as their similarity ranged from 94.7 to 100%. Polish isolates shared 75.7-99.2% identity with sequence of the virus strains from the Czech Republic, the Netherlands, and Canada. Phylogenetic analysis resulted in grouping of the isolates found in Poland together with one of the Czech strain whereas two other from the Czech and the strains from the Netherlands and Canada created the separate cluster.

  • 우리나라 파프리카에 발생하는 주요 바이러스는 오이모자이크바이러스(Cucumber mosaic virus, CMV), 고추모틀바이러스(Pepper mottle virus, PepMoV), 잠두위조바이러스2(Broad bean wilt virus2, BBWV2), 고추약한모틀바이러스(Pepper mild mottle virus, PMMoV), 토마토반점위조바이러스(Tomato spotted wilt virus, TSWV) 및 사탕무황화바이러스(Beet western yellows virus, BWYV) 등 6종이다. 생산량 감소 및 고품질 규격품 생산의 가장 큰 제한요인이 되고 있는 바이러스병의 피해를 최소화하기 하기 위하여 바이러스별 병징 및 발생원인에 따른 예방법을 알아본다.

다층도공층의 설계에 의한 인쇄적성 개선 (제1보) - 프리코팅층의 바인더 물성이 탑코팅층의 특성에 미치는 영향 - (Improving printability by designing a multi-layered coating structure (I) - The effect of binder properties in the pre-coating layer on the characteristics of the top-coating layer -)

  • This study was carried out in order to investigate the effect of binder properties in pre-coating layer on the characteristics of top-coating layer and print mottle of coated paper. Four different latices were used as a binder in pre-coating layer, and coating color prepared with a same formulation was applied for top-coating. The properties and print mottle of coated paper were evaluated. It was found that glass transition temperature (Tg) was the important factor to control the properties of pre- and top-coating layer. PPS roughness of coated paper was decreased and paper gloss was increased with applying binder which has lower Tg. Properties of top-coating layer were affected by the binder used in pre-coating layer. Print mottle of coated paper was improved with using binder which has lower Tg in pre-coating layer. These results indicate that final printability and properties of top-coating layer can be improved with using suitable binder in pre-coating layer.

Soybean yellow mottle mosaic virus의 자연기주와 병환 (Natural Hosts and Disease Cycle of Soybean yellow mottle mosaic virus)

  • 2006년에서 2007년까지 콩연구센터 육종포장 주변에서 잡초 서식상황을 조사한 결과 31과 74속 96종의 식물이 확인되었다. SYMMV의 자연기주 또는 중간기주를 동정하기 위하여 26과 84종의 식물체가 포함되는 495점의 시료를 RT-PC R로 분석한 결과, 단지 콩과 식물에서(돌콩, 새팥, 토끼풀, 비수리)만 SYMMV가 검출되었다. SYMMV를 가지고 있는 콩과 식물 중에서 분석한 돌콩의 3분의 1이상이 SYMMV를 보독하고 있는 것으로 보아 돌콩은 SYMMV의 감염원으로써 중요한 역할을 담당하는 것으로 생각된다. 돌콩은 SYMMV에 7월 중순경에 감염되는 것으로 추정되었다. 돌콩에서 SYMMV의 빠른 감염시기, 높은 종자감염율 그리고 종자전염과 같은 결과로 볼 때, 돌콩은 SYMMV의 생활사 완성에 큰 역할을 담당하고 있는 것으로 판단된다.

  • This study was carried out in order to investigate the effect of pigment properties in a pre-coating layer on the characteristics of a top-coating layer and the print mottle of the coated paper. Five different pigments were applied for this study as raw materials for the pre-coating layer. The properties and print mottle of the coated paper samples were evaluated according to the coating color formulation. Type of pigments appliied in a pre-coating layer was one of the most important factor to control the properties of pre and top coating layer. Surface properties of pre and top coated paper were improved by blending GCC which had smaller particle size, with clay. Properties of a top-coating layer was affected by the pigment properties used in the pre-coating layer. It was found that print mottle of coated paper can be improved by replacing part of GCC with smaller particle size GCC or clay in pre-coating layer.

Biological, Physical and Cytological Properties of Pepper mottle virus-SNU1 and Its RT-PCR Detection

  • A strain of Pepper mottle virus (PepMov) was isolated from chili pepper plants in Korea. In host range study, this virus, designated PepMoV-SNU1, shared most characteristics with PepMoV isolates reported previously. Thermal inactivation point ($45^{\circ}C\;to\;75^{\circ}C$) and dilution end point ($10^{-1}\;to\;10^{-4}$) of PepMoV-SNU1 showed differences depending on the propagation hosts. Cylindrical and pinwheel-shaped inclusions were always observed in pepper leaf tissues infected with the virus alone. Unexpectedly, a special structure of pinwheel shaped inclusion surrounded with unknown small spots was also observed in the leaf section when co-infected with a strain of pepper mild mottle virus. The partial sequence of coat protein gene and 3' untranslated region of PepMoV-SNU1 showed 98% identity with those of other PepMoV isolates. A primer pair derived from 3' end of the coat protein gene and poly A tail regions were designed. Optimal detection condition of PepMoV-SNU1 by RT-PCR was tested to determine appropriate annealing temperature and additional volumes of oligo-dT (18-mer), dNTP, and Taq polymerase. Under the optimized condition, an expected 500 Up PCR-product was detected in pepper leaves infected with PepMoV-SNU1 but not in healthy plants.

  • 박과는 스쿼시, 호박, 애호박, 조롱박, 오이 및 수박 등 100여 속이 넘는 속이 포함된 식물과이며, 종자의 수입량이 매우 많은 작물이다. 이들이 우리나라로 수입될 때, 규제바이러스인 Squash mosaic virus (SqMV), Cucumber green mottle mosaic virus (CGMMV) 및 Kyuri green mottle mosaic virus (KGMMV)에 대하여 검역을 수행한다. 본 연구에서는 SqMV, CGMMV 및 KGMMV를 신속·높은 감도로 검출할 수 있는 특이적 RT-PCR 및 nested PCR 프라이머 조합과 유전자변형-양성대조구 플라스미드를 개발하였다. 또한 본 연구에서 개발한 진단시스템을 2010년부터 2014년 상반기까지 현장적용하여, SqMV 47건, CGMMV 67건 및 KGMMV 17건을 검출하였다.

  • Transgenic Nicotiana benthamiana plants harboring coat protein (CP) gene of Zucchini green mottle mosaic virus (ZGMMV) were generated for virus-resistant screening and complementation analysis of related viruses for environmental safety assessment (SA) of living modified organism (LMO) purposes. Transformation of leaf disc of N.benthamiana was performed by using Agrobacterium-mediated method and the pZGC-PPGA748 containing the ZGMMV CP and NPTII genes. Two kinds of transgenic homozygous groups, virus-resistant and virus-susceptible N.benthamiana lines, were obtained by screening of challenging homologous virus for Tl generations. These two pathologically different lines can be useful for host-virus interactions and LMO environmental SA.

Production and Evaluation of Monoclonal Antibodies Against Recombinant Coat Protein of Lily mottle virus for Western Blotting and Immono-blot Analysis

  • Lily mottle virus (LMoV) causes flower quality reduction in Lilium spp. The coat protein gene was RT-PCR-amplified from total RNA extracted from infected lily leaves and the amplified fragment was cloned into the pRSET expression vector tagged with a His-MBP. The plasmid of recombinant coat protein was used to transform an Escherichia coli strain pLysS and was expressed. The coat protein was purified by affinity chromatography using a Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE. The in vitro-expressed protein was used for immunization of mice. The polyclonal and monoclonal antibodies reacted specifically for the detection of LMoV in lily extracts in Western blot. Moreover the monoclonal antibodies reacted with lily extracts in DAS-ELISA with no unspecific or heterologous reactions against other non-serologically related viruses, but the polyclonal antibodies revealed a weak reaction against both infected lily and healthy control.

Pathological and molecular comparisons of five distinct species of pepper-infecting Potyviruses (oral)

  • Five pepper-infecting potyviruses, Pepper mottle virus (PepMoV), Chilli veinal mottle virus (CVMV), Pepper veinal mottle virus (PVMV), Pepper severe mosaic virus (PSMV) and Tobacco each virus (TEV), are known filamentous virus and can be infected pepper crops systemically. To understand pathology and genome information of the five viruses on pepper plants, host reactions and sequences were compared to the 5 viruses. Five potyviruses were inoculated onto some typical cultivars of hot peppers and compared their symptoms, and virus accumulations. A set of degenerate primers for potyviruses were applied to 5 viruses and RT-PCR was performed. RT-PCR products containing partial nuclear inclusion b and coat protein (CP) genes were cloned. Then, oligo dT primer and species-specific primer were redesigned to amplify the C-terminal part of CP and 3' noncoding regions of each viruses. Sequences of the viruses were analyzed and compared to serological relationships among the viruses. The data can be useful for screening of potyviruses in pepper plants and pathogen-derived transgenic pepper plant development.
