Quantitative Analysis of Cyanidin-3-O-rutinoside and Quercetin-3-O-rutinoside from Extracts of Morus alba Fruit (오디 추출물로부터 Cyanidin-3-O-rutinoside와 Quercetin-3-O-rutinoside의 함량분석)

  • Morus alba fruit contains several anthocyanins and flavonoids like cyanidin-3-O-rutinoside and quercetin-3-O-rutinoside. Extractions from Morus alba fruit were performed with five different compositions of ethanol/water [v/v]. High-Performance Liquid Chromatography (HPLC) was used for the validation and content determination of cyanidin-3-O-rutinoside and quercetin-3-O-rutinoside. The contents of cyanidin-3-O-rutinoside in ethanol extracts (100%, 70%, 40%, 20% and 0%, ethanol/water [v/v]) were 1.150%, 0.822%, 0.749%, 0.663% and 0.597%, respectively. Quercetin-3-O-rutinoside contents of ethanol extracts (100%, 70%, 40%, 20% and 0%, ethanol/water [v/v]) were 0.167%, 0.161%, 0.159%, 0.155% and 0.096%, respectively. The highest contents of cyanidin-3-O-rutinoside and quercetin-3-O-rutinoside were in the ethanol extract with 100% ethanol/water [v/v]. These HPLC analysis method could be used as basic data for standardization of functional food from Morus alba fruit.

Growth Characteristics of Listeria monocytogenes Scott A under High Osmotic Condition and Antibacterial Effect by Morus alba L. Leaf Extract (고삼투압조건하에서 Listeria monocytogenes Scott A의 생육특성과 상엽(桑葉)추출물에 의한 증식억제효과)

  • Growth rate and osmolyte accumulation of L. monocytogenes were measured at the varying concentrations of NaCl. L. monocytogenes accumulated glycine betaine and glutamate intracellularly when grown under osmotic stress by NaCl, and the amounts of them increased as the concentration of NaCl was increased. They were 685 and 345 nmol/mg protein, respectively, when grown in the BHI supplemented with 4% NaCl. In order to inhibit L. monocytogenes effectively, both NaCl and Morus alba L. leaf extract were supplemented in TSB, and antibacterial effect of those supplements on L. monocytogenes was tested. Growth of L. monocytogenes grown in TSB supplemented with 2% NaCl and 100 ppm M. alba leaf extract decreased by 10 times in CFU/ml unit comparing to the growth of control. When grown in TSB, supplemented with 2% NaCl plus 500 ppm M. alba leaf extract and 2% NaCl plus 1,000 ppm M. alba leaf extract, growth of L. monocytogenes decreased by $10^5\;and\;10^8$ times in CFU/ml unit, respectively.

Anti-stress and Promoting Effect of the Fruit of Morus alba (상심자(Morus alba)의 운동능력 향상과 스트레스 개선효과)

  • Effects of Morus alba fruit extracts on monoamine oxidase (MAO) activity were examined in rats during and after physical exercise. Oral administration of M. alba extract (0.3 g/kg body weight) significantly increased brain MAO-A activity but decreased liver MAO-B activity when they were measured using serotonin and benzylamine as substrates. Type of physical exercises had significant effect on MAO activity. Brain MAO-A activity markedly decreased with physical activity-related stress compared to normal group, whereas Liver MAO-B activity increased up to 60 min after exercise. Lactate dehydrogenase (LDH) activity and lactate concentration in blood, clinical indices of physical exercise activities, were also determined for correlation to MAO activities. MAO-A activity of rats subjected to oral administration of M. alba extract and physical exercise increased whereas MAO-B and LDH activities, and lactate level decreased, All indices eventually recovered normal levels, These results suggest M. alba may increase capability of physical activities by modulating MAO activities during exercise.

Effects of Water-Soluble Extract from Leaves of Morus alba and Cudrania tricuspidata on the Lipid Peroxidation in Tissues of Rats (뽕나무(Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎의 수용성 추출물이 흰쥐 각 조직중의 지질 과산화물 함량에 미치는 영향)

  • 뽕나무 (Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎으로부터 추출한 수용성 물질을 0.5% 콜레스테롤 투여 식이에 1%(w/w) 수준으로 첨가하여 각 조직에서의 지질 과산화물 생성정도를 나타내는 TBARS 측정법으로 항산화 활성을 검토하였다. 흰쥐의 체중 및 식이 섭취량은 실험군 간의 유의적인 차이는 없었다. 간장 및 뇌 조직 중량은 뽕잎 추출물군에서 증가하였다. 신장의 지질 과산화물 함량은 대조군에 비해서 뽕잎 추출물군에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화물 함량은 대조군에 비교해서 꾸지뽕잎 추출물군에서 유의적으로 증가하였다. 간장 및 비장의 경우에는 각 실험군 간에 유의적인 차이가 없었다. 각 조직의 microsomes 분획으 지질 과산화물은 심장과 비장에서 뽕잎 및 꾸지뽕잎 추출물군에서 감소하였으나, 다른 조직에서는 현저한 차이가 없었다. 한편, 각 조직의 microsomes 분획을 얻어서 Fe2+/ascorbate로 지질 과산화물을 유도하기 위하여 37$^{\circ}C$에서 한시간 반응을 시킨 결과, 뽕잎 및 꾸지뽕잎 추출물군에서 약 절반 정도 감소하였으며, 상대적 증가량은 뇌조직 microsomes에서 현저하였다. 본 실험의 결과 신장조직 및 간장 microsome막 지질 과산화물 함량은 뽕잎 및 꾸지뽕잎 수용성 추출물에 의해 감소되었으며, 뇌 및 심장조직의 지질 과산화물 함량은 꾸지뽕잎 수용성 추출물에 의해 증가됨으로써 봉잎 추출물간의 차이를 나타내었다.

  • The fruits of Morus alba L. were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned into EtOAc, n-butyl alcohol, and water fractions. The repeated silica gel ($SiO_2$) and octadecyl silica gel column chromatographies for the EtOAc and n-butyl alcohol fractions led to isolation of two phenolic compounds. The chemical structures of the compounds were determined as Diels-Alder type adducts, mulberrofuran E (1) and chalcomoracin (2) based on spectroscopic data analyses including nuclear magnetic resonance, mass spectrometry, and infrared spectrometry. Compounds 1 and 2 were isolated for the first time from the fruits of M. alba L. in this study.

  • The MeOH extract of the twig of Morus alba L. (Moraceae) inhibited strong lipid peroxidation activity. In order to find out active principle from the plant, acivity-guided fractionation was performed and five antioxidant compounds were isolated. Their chemical structures were identified as 6-geranylapigenin (1), 6-geranylnorartocarpetin (2), resveratrol (3), oxyresveratrol (4) and quercetin (5) by physicochemical and spectrometric methods. Compounds 1-5 significantly inhibited lipid peroxidation in rat brain homogenate $(IC_{50}\;values\;of\;3.37,\;3.74,\;0.23,\;0.29\;and\;0.06\; {\mu}M,\;respectively)$.

  • Morus alba extract(MAE) was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in serum, liver, kidney and heart were measured by the TBA method. ICR mice receiving alloxan at a dose of 6mg/kg intraperitoneally after a 24hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in serum, liver, kidney of alloxan-diabetic mice were decreased by the treatment of MAE at the dose of 50mg/kg, 100mg/kg for 7 days.

  • The crude extract of Morus alba Linne bark which showed strong antimicrobial activity on Listeria monocytogenes was fractionated by some solvents and applied column chromatography for purification of active component. By silica gel column chromatography, active fraction (F-5') was purely isolated and confirmed their strong bactericidal effect on Listeria monocytogenes tested at level of 100 and 300 ppm in tryptic soy broth. The cell wall observed by SEM was abnormal after treatment of F-5'.

  • The root bark of Morus alba L. (MA) used in traditional oriental medicine for the treatment of pulmonary diseases exerts various pharmacological activities including anticancer effects. In the current study, we investigated the effects of MA on the migration and invasion of colorectal carcinoma cells. Results from a transwell assay showed that the methylene chloride extract of MA (MEMA) suppressed the migration and invasion of HCT116 human colorectal carcinoma cells in a concentration-dependent manner. MEMA reduced both mRNA and protein levels of matrix metalloproteinase (MMP)-9, but did not suppress the expression of MMP-2 in HCT116 cells. As a molecular mechanism, MEMA inhibited the phosphorylation of mitogen-activated protein kinases (MAPKs), including ERK, JNK and p38, in a dose-dependent manner. In addition, MEMA dephosphorylated both Src and signal transducer and activator of transcription 3 (STAT3) in HCT116 cells. Taken together, we demonstrate that MEMA suppressed the migration and invasion capacity of HCT116 human colorectal cancer cells by downregulation of MMP-9 and inactivation of both MAPKs and Src/STAT3 signaling pathway.
