A Study on Successional Direction of Robinia pseudoacacia for the Vegetation Restoration in Waste Landfill of Noeul Park (노을공원 쓰레기매립지 식생복원을 위한 아까시나무 천이방향 연구)

  • The waste landfill that has been used as a park through a stabilization project to provide green space to local residents. Vegetation restoration is necessary for the landfill park project, but it is difficult to restore vegetation due to various disturbances in the landfill. This study analyzed the successional dynamics and ecological characteristics of Robinia pseudoacacia communities from the slopes of Noeul Park by applying the 7-stage successional hypothesis. As a result of the study, there was almost no intermediate successional stage. There are only the early successional stage which Robinia pseudoacacia has an absolute dominance of 100% in the crown and middle layers, and the degeneration successional stage which formed by the introduction of Morus alba. This result showed that the succession of Robinia pseudoacacia communities were not able to proceed to the climax forest due to various disturbances in the waste landfill. Therefore, it was analyzed that it is necessary to induce the succession through intermediate steps such as Morus alba, since it is difficult to transition from Robinia pseudoacacia community to the native Quercus spp. community.

Studies on the Changes with Time of the Reserve Substances in the Hardwood Cutting of Mulberry Varieties (뽕나무 품종별 고조삽목에 있어서 삽수내 저장물질의 경시적 변화)

  • To investigate changes of dry weight, C/N ratio, carbohydrate, ratein and nucleic acids in the barks of the hardwood cutting of mulberry plants, the barks of the hardwood cutting were taken every 10days from the cutting day to 40days after cutting. Results were summarized as follows : Growth of shoot and root in Keomseolppong (Morus bimbycis KoIDZ.) after cutting was better than that in kaeryangppon (Morus alba L.) and Daeryugppong(Morus Lhou(SER.) KoIDZ.). Dry weight in the barks of the hardwood cutting decreased up to 40days after cutting (decrease percentage of dry weight : about 14%) and Keomiseolppong showed the highest decrease precentage of dry weight. Change of C/N ratio decreased in the initiation, whereas that from 10days after cutting increased with the growth of new shoot, C/N ratio was the highest in Keomseolppong and the lowest in Kaeryangppong. Total sugar showed decreasing tendency up to 40 days after cutting the increase from 10 days to 20 days. Reducing sugar increased in the frist 20days, followed gradually decrease up to 40 days after cutting Starch levels decreased up to 40 days. Change in the content of starch closely paralleled to that of dry weight of bark of the hardwood cutting. Content of total sugar and starch in Keomseolppong was the highest, and was consumed more than that in the rest of varieties. Crude protein and soluble protein increased up to 10 days after cutting, followed decrease up to 40 days. Protein content of Keomseolppong was smaller than that in the rest of varieties. RNA content showed the same tendency with that of protein content, but Keomseolppong showed always the highest. Content of DNA did not show any change and difference among three varieties.

Morphological and Chemical Characteristics of Mulberry(Morus) Fruit with Varietes (몇 가지 뽕품종에 따른 오디의 형태 및 화학적 성분의 특성)

  • The morphological and chemical characteristics of leaves and fruits were observed in the three mulberry varieties, including Daeryugppong(Morus Lhou(Ser.) koidz), Kugsang 20(Morus Lhou(Ser.) koidz) and Cataneo(Morus alba L.). The leaf development in spring was earliest in Cataneo and latest in Kugsang. Content of total nitrogen and Mg in leaf was the highest in Cataneo while that of P2O5, K and Ca in Daeryukppong. Flowers started to bloom from May 8 and in full bloom around May 15. Mature fruits began to set from June 10 to 15 and lasted by July 10 in Cataneo. Average fruit weight was heaviest in Kugsang 20(3.52 g/fruit), while lowest in Daeryukppong(1.61 g/fruit). In fruits, glucose and fructose were the major sugars. Citric acid was the most abundant organic acid in three varieties with its average content from 0.8 to 0.14%. The major pigment in fruit was anthocyanin and its content varied among varieties. The stability of anthocyanin was evaluated under various pH, temperature, and sugar concentrations. Rutin was the major flavonol glycoside present in fruits and its content varied from 0.92 to 3.36 mg/gDW. Other flavonol glycosides such as isoquercitrin and quercitrin were also detected in fruit.

Effect of Stem Bark Extract from Morus alba and Cudrania tricuspidata on the Concentrations of Lipid and Tissue Lipid Peroxidation in the Cholesterol-Fed Rats (뽕나무와 꾸지뽕나무의 수피 수용성 추출물이 콜레스테롤 함유식이 투여 흰쥐의 지질농도 및 과산화지질 농도에 미치는 영향)

  • Male Sprague-Dawley rats received either a cholesterol diet(Control group) or cholesterol diets supplemented with the water-soluble extract of stem bark from Morus alba(M group) or Cudrania tricuspidata(C group) at the level of 1% for 2 weeks. Concentrations of total cholesterol and phospholipid in serum of C group and triglyceride in serum of M group were lower than those of control group. Concentration of cholesterol in liver of M and C groups has a tendency to be lower than that of control group. Antioxidative activities of water-soluble extracts from stem bark of Morus alba and Cudrania tricuspidata on the peroxidation of lipid in tissues of rats were also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Concentration of TBARS in kidney of M and C groups was significantly lower than control group. However, concentration of TBARS in liver and brain of C and M groups was significantly higher than in control group. The result that concentration of nonheme ion was significantly increased in liver of the mulberry supplemented groups comparision to control group, suggested that enhanced concentration of nonheme ion was associated with enhanced peroxidation of lipid in this group. Concentration of TBARS in microsomes of liver and brain in control group induced with $Fe^{2+}$/ascorbate increased by reaction time at $37^{\circ}C$, whereas this observation in liver did not occurred in C and M groups. This study suggested that water-extract from stem bark of Morus alba and Cudrania tricuspidata exert hypotriglycerolemic effect as well as antioxidative effect in kidney and liver microsomes in rats fed a cholesterol diet.

Comparison of Nutritional and Functional Constituents, and Physicochemical Characteristics of Mulberrys from Seven Different Morus alba L. Cultivars (뽕나무(Morus alba L.) 품종별 오디의 영양 및 기능성 성분과 이화학적 품질 특성 비교)

  • Nutritional and functional components, and physicochemical characteristics of mulberry fruits from Morus alba L. cultivars, including Daejappong, Iksuppong, Daesungppong, Yongppong, Cheongilppong, Gwasang 1 and Gwasang 2, were investigated and compared. Among the 7 mulberry cultivars examined, Iksuppong, Cheongilppong and Gwasang 2 had higher contents of nitrogen free extract, total sugar and total reducing sugar, than other mulberry cultivars, while contents of protein, fat, and fiber were lowest. Gwasang 1 and Daesungppong had the highest content of total phenolic, total flavonoid, and total anthocyanin. Daesungppong, Gwasang 1 and Gwasang 2 had higher contents of C3G and C3R than other mulberry cultivars, although C3R content of Cheongilppong was the highest. Four flavonoid contents varied among of 7 mulberry cultivars. Contents of two major flavonoids, rutin and quercetin, were the highest in Gwasang 2, while Iksuppong and Cheongilppong were the lowest. Contents of piceid and 4-prenylmoracin were the highest in Daesungppong and Cheongilppong, respectively. Cheongilppong of mulberrys had the highest content of GABA. Iksuppong and Gwasang 2 with moderate weight and dark color had higher contents of total soluble solid than other mulberry cultivars, while contents of titratable acidity were lower. Our results provide useful information on nutritional and functional constituents of mulberry fruits according to cultivars, and their physicochemical and processing properties.

Effect of the extracts from Schisandra chinensis Fruit and Morus alba Leaf on Insulin Secretion in Glucose-induced HIT-T15 Cells (오미자와 뽕잎 추출물이 glucose에 의해 유도된 HIT-T15세포의 인슐린 분비능에 미치는 영향)

  • This study aimed to examine the effect of the Schizandra chinensis fruit and Morus alba leaf on insulin expression in HIT-T15 cells, which is exposed by glucose. The total polyphenol contents of the S. chinensis fruit ethanol extract and the M. alba leaf hot-water extract were $20.11{\pm}0.35$ mg/g and $50.02{\pm}0.62$ mg/mL, respectively. The S. chinensis fruit ethanol extract and the M. alba leaf hot-water extract contained $2.85{\pm}0.15$ and $8.76{\pm}0.43$ mg/g flavonoids, respectively. The antioxidant ability of the M. alba leaf hot-water extract was found to be superior to that of the S. chinensis fruit ethanol extract. Compared to the HIT-T15-treated 10 mM 2-deoxy-D-glucose, the $100{\mu}g/mL$ S. chinensis ethanol extract was found to have a two fold increase in insulin productivity. Moreover, the $100{\mu}g/mL$ M. alba leaf hot-water extract promoted the insulin secretion of high-glucose-damaged HIT-T15 almost ten fold. The above results showed that the S. chinensis fruit ethanol extract and M. alba leaf hot-water extract may improve the insulin productivity of the beta cell with glucose-induced oxidative damage. These data suggest that the S. chinensis fruit ethanol extract and the M. alba leaf hot-water extract can be used as food materials for the regulation of insulin secretion.

Interaction Between time of Nodal Explant Collection and Growth Regulators Determines the Efficiency of Morus alba Micropropagation

  • The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.

Studies on the Growth Rate of Silkworm Bombyx mori (L.) (Lepidoptera: Bombycidae) Fed with Control and Silver Nanoparticles (AgNps) Treated MR2 Mulberry Leaves

  • To evaluate the growth rate of larval and pupal parameters of silkworm Bombyx mori fed with Silver Nanoparticles (AgNps) treated $MR_2$ mulberry leaves, the following works have been considered. The AgNp was synthesized by chemical reduction method, it was diluted by different concentrations such as 25%, 50%, 75% and 100% (without dilution). Fresh mulberry leaves (Morus alba L.) were sprayed by each concentration and were fed to silkworms, from $3^{rd}$, $4^{th}$ and $5^{th}$ instar, five feedings/day. Group $T_1$ larvae received $MR_2$ mulberry leaves sprayed with distilled water and served as control, group $T_2$, $T_3$, $T_4$ and $T_5$ larvae received 25%, 50%, 75% and 100% AgNps sprayed mulberry leaves, respectively. Silkworm larvae fed on M. alba ($MR_2$) leaves sprayed with 25% concentration of AgNps (group $T_2$) was significantly increased the larvae and cocoon length, width and weight as compared to those fed on control (group $T_1$) $MR_2$ mulberry leaves and other groups ($T_3$, $T_4$ and $T_5$). Hence, 25% AgNps dose was fixed as an effective dose. It has been observed from the present study that 25% AgNps treated (group $T_2$) leaves fed by silkworms have enhanced the larval and pupal growth and quantity of silk production than control.

  • Anatomical and chemical properties of three mulberry species (Morus spp) were analyzed as an alternative row material for the paper mulberry (Broussonetia kazinoki) . The pulping and papermaing characteristics of bast fiber and whole stalks by three different pulping processes, conventional alkali, alkali-hydrogen peroxide, and sulfomethylated, for the Hanji were investigated. The fiber dimension of M.bombycis was the biggest of the species. The fiber length of upper part and the fiber width and cell wall thickness of lower part were bigger than the others. The extractives and ash contents of bast fiber were higher than those of whole stalk , and holocellulose and lignin contents of whole stalk were higher than those of bast fiber. The pulp yields of M.alba bast fiber, M.bombycis whole stalk, and sufomethylated pulping were higher than the others.

Correlation between Air Temperature Factors and Leafing Date of Mulberry Tree (뽕나무 출엽일과 기온 요인 간 상관성)

  • This study was conducted to identify the air temperature factors that affect leafing date of mulberry (Morus alba) in Buan, Jeonbuk province. Over the period from 2009 to 2014, the leafing date averaged April 24 (114 Julian days). The earliest leafing date was April 12, in 2014, and the latest leafing date was May 2, in 2011. Yearly variations of daily mean temperature (DMT) and daily maximum temperature (DMxT) were high in the last part of March. There were highly negative correlations between leafing date and degree days of accumulated DMT above 0, 5, and $10^{\circ}C$ in the period from March 1 to April 10. Linear regression equations for estimating the leafing date were y = 153.8 - 0.1886 ${\times}$ ($r^2=0.965^{**}$) and y = 126.2 - 0.2246 ${\times}$ ($r^2=0.825^*$) for the degree days of accumulated DMT above $5^{\circ}C$ in the period from March 1 to April 10 and the last part of March, respectively. Therefore, leafing date of mulberry, which is important for popcorn disease control, is 96.5% predicted by degree days of accumulated DMT above $5^{\circ}C$ in the periods from March 1 to April 10.
