Morphological and Molecular Phylogenetic Data of the Chinese Medicinal Fungus Cordyceps liangshanensis Reveal Its New Systematic Position in the Family Ophiocordycipitaceae

  • A cordycipitoid fungus infecting Hepialidae sp. in Nepal was supposed to be identical to Cordyceps liangshanensis, originally described from southwestern China, and thus, transferred to the genus Metacordyceps or Papiliomyces in previous studies. However, our multi-gene (nrSSU-nrLSU-tef-1α-rpb1-rpb2) phylogenetic and morphological studies based on the type specimen and additional collections of C. liangshanensis revealed that the fungus belongs to the genus Ophiocordyceps (Ophiocordycipitaceae). Therefore, a new combination O. liangshanensis was made, and a detailed description of this species was provided.

Consideration of primary molar's root and root canal for pulpectomy treatment (치수절제술 시 고려해야 할 유구치 치근과 근관)

  • The purpose of the pulp treatment is to make the function of the primary molar in the oral cavity possible for as long as possible until the exfoliation of the tooth or the development of the occlusion is as long as possible. The pulpectomy is a relatively common procedure for the pediatric dentist in the clinic with dental care of the children nowadays. Primary molar is morphologically more susceptible to dental caries than permanent tooth, and the dental pulp treatment is frequently performed. Furthermore, unlike permanent teeth, the root canal has a large degree of curvature and morphological diversity and complexity, careful consideration is needed. Therefore, it is very important to comprehensively understand the morphological characteristics and diversity of the root and root canals for the successful pulp treatment of the primary molars.

Current Taxonomical Status of Korean Auricularia Species

  • Auricularia (Auriculariaceae, Basidiomycota) is well-known jelly fungi, and most species are cultivated or utilized medicinally around the world. A total of 127 Auricularia specimens were collected during Korean mushroom forays by the Korea National Arboretum from 2008 to 2020. In this investigation, fungal barcode analysis and morphological examinations of 33 specimens were conducted and five Korean Auricularia species were identified. Of the species identified, A. americana, A. heimuer, and A. villosula were new to Korea, while A. auricula-judae which was previously identified in Korea, was not found. Herein, we discuss the Korean Auricularia species and provide a taxonomic key for their morphological identification.

  • Psalidoremus is one of the subgenus which consists of five species and eleven subspecies through Taiwan, Japan, Korea and north eastern part of China which present strong endemism to their distribution. While most of advance studies focus on the new species or subspecies from this taxon, this paper suggests paleogeographical assumption and hypothesis of how they diverged from common ancestors. Phylogeographical patterns illustrated by the distribution of five species, which including Prosopocoilus inclinatus, Prosopocoilus hachijoensis, Prosopocoilus dissimilis, Prospocoilus pseudodissimilis, Prosopocoilus motschulskii respectively, has been assessed in this research. Speciation history is suggested by morphological tendencies especially in male mandibles with assumption from geographical patterns of under sea level.

Morphological characteristics of Neural Tissue and Corazonin Neurons of Central Nervous System in Larval Stage of Scuttle Fly

  • Through previous studies, the central nervous system (CNS) was collected by dividing the scuttle fly into larval, pupa, and adult stages by developmental stage, and the morphological characteristics were observed. In situ hybridization (ISH) using the collected central nervous system, it was possible to confirm the location and extent of expression of the neurotransmitter corazonin (Crz) at each stage of development. In this study, paraffin specimens were prepared using central nervous system tissues of 3rd instar larval stage of scuttle fly, which had completed in situ hybridization, and general histochemical staining (hematoxylin-eosin, H-E) and special histochemical staining (luxol fast blue-cresyl violet) was performed to observe the histological and cytological morphology characteristics of corazonin neurons. As a result, a variety of nerve cell body existed between many myelin sheath. The corazonin neurons compose cortex of central nervous system with other neurons congregating in this tissue and show larger cell body relatively in neurohistochemical analysis.

  • 본 논문에서 이슈 키워드 추출을 위해 텍스트 마이닝(Text Mining) 기술을 요구한다. 사회적 이슈 키워드를 추출하기 위해 키워드 수집 모델이 되는 사이트에서 크롤링(crawling)을 수행한 뒤, 형태소 단위 의미있는 단어를 수집하기 위해 형태소 분석(morphological analysis)을 수행한다. 한국어 형태소 분석을 위해 파이썬의 코엔엘파이(KoNLPy) 패키지를 활용한다. 형태소 분석을 통해 나뉘어진 단어에서 통계를 내어 이슈 키워드 추출한다. 이슈 키워드를 뒷받침할 연관 단어를 분석하기 위해 단어 임베딩(Word Embedding)을 수행한다. 단어 임베딩 수행을 위해 Word2Vec 모델 중 Skip-Gram 방법론을 적용하여 연관 단어를 분석하도록 개발하였다. 웹 소켓(Web Socket) 통신을 통한 채팅 프로그램의 상단에 분석한 이슈 키워드와 연관 단어를 출력하도록 개발하였다.

Study on Development Effect on Zebrafish Embryo by Alacholr, Butachlor and Fipronil (농약 alacholr, butachlor 및 fipronil이 제브라피쉬 배아 발생에 미치는 영향)

  • This study is aimed to search the possibility of developmental toxicity test using the zebrafish from the pesticide. We selected herbicides alachlor and butachlor, reported for fish toxicity, and insecticide fipronil reported for the high fish toxicity and the honey bee risk among the pesticides with high usability for the examples of the pesticides in this experiment. In this study, we showed those effects on the zebrafish embryo development by exposing different kinds of pesticide with different concentration and exposed time periods. As a result, the rates of hatching and abnormality of the zebrafish embryo after treatments of alachlor were increased in 24-48 hpf group, and the juvenile fishes in every group exposed to $40{\mu}M$ or more of alachlor displayed sever morphological changes such as bent tails, edema and activity failures. In case of the butachlor, the rates of hatching and the abnormality in 24-48 hpf group were higher than the other groups exposed in different time periods. The fatality before hatching was high in $40{\mu}M$ or more of butachlor treatment, and entire zebrafish embryos in 48 hpf group died before hatching. All the living juvenile fishes showed morphological changes as like as the treatment of alachlor. The rate of hatching and the survival of the zebrafish embryo by the fipronil were higher than other pesticides. However, morphological changes such as bent tails were observed from the most of living juvenile fishes. Therefore, the effects of three different pesticides with different concentrations and exposing time periods on the development of zebrafish embryos showed that all the pesticides effects were proportional to the concentration, and exposing time periods may cause the morphological abnormality.

  • Two spiny lobsters of the genus Panulirus, P. stimpsoni Holthuis, 1963 from Jeju Island and P. versicolor (Latreille, 1804) from Busan, are briefly described with colorations. These lobsters are new members of the Korean decapod crustacean fauna. Morphological and distributional accounts of these species are discussed.

  • The teratogenic potential of the anticonvulsant drug phenytoin (PHT) has been well documented both in the human and in the experimental animals. However there are few reports on the effects of PHT on embryonic development in rats in vitro. The present study was performed to evaluate the teratogenic effects of PHT using whole-embryo culture system in rats. Sprague-Dawley rat embryos were explanted on gestational day (GD) 9.5 and cultured for 48 hrs in the immediately centrifuged and heat-inactivated rat serum containing 0,25,50, or $100{\mu}g$ PHT/mL. At the end of culture period the embryos were scored for morphological development according to the procedure of Van Maele-Fabry, and their total protein contents were determined. At 100 ${\mu}$g/mL of culture medium. PHT caused significant reduction in developmental score and protein content of embryos and a high incidence morphological abnormalities (100%). Characteristic malformations included altered yolk and embryonic circulation, craniofacial hypoplasia, neural tube schisis, branchial arch defects, abnormal ratation, and limb bud hypoplasia, among others. There were no adverse effects on embryonic growth and development at concentrations of 25 and 50 ${\mu}$g /mL of culture medium. The results indicated that the dysmorphogenic effect of PHT on cultured embryos is due to a direct interference with embryonic development.

Systematic Relationships of Korean Freshwater Snails of Semisulcospira, Koreanomelania, and Koreoleptoxis (Cerithiodiea; Pleuroceridae) revealed byMitochondrial Cytochrome Oxidase I Sequences

  • Many freshwater snail taxa are difficult to identify using morphological traits due to phenotypic plasticity. However, using of molecular DNA marker in combination with morphological traits can provide a reliable means for discriminating among freshwater snail taxa including cryptic species. To discriminate among Korean freshwater snail taxa and resolve their systematic relationships, wesequenced a fragment of mtDNA cytochrome oxidase I (COI) gene from 82 specimens collected from ten different sites distributed along the Korean peninsula. We identified more than seven freshwater snail taxa including cryptic species in Korea. Whereas traditional shell morphology of freshwater snails offers only weak discriminatory power for recognizing 'good' taxa, DNA sequence data provided positive and reliable identification. In addition, a major Semisulcospira clade was clearly separated from the remaining lineages observed including cryptic species. However, a phylogenetic tree inferred from the COI gene data did not fully resolve systematic relationships among pleurocerid taxa in Korea. Establishing more robust shell characteristics for identifying taxa unambiguously and hence improving traditional key shell morphology characters for freshwater snail species is an urgent requirement and will require more rigorous examination of all nominal taxa. While molecular data generated here will be useful for species identification and for describing the systematic relationships among Korean freshwater snails, further analysis will be required.
