Morphological Characterization of small, dumpy, and long Phenotypes in Caenorhabditis elegans

  • The determinant factors of an organism's size during animal development have been explored from various angles but remain partially understood. In Caenorhabditis elegans, many genes affecting cuticle structure, cell growth, and proliferation have been identified to regulate the worm's overall morphology, including body size. While various mutations in those genes directly result in changes in the morphological phenotypes, there is still a need for established, clear, and distinct standards to determine the apparent abnormality in a worm's size and shape. In this study, we measured the body length, body width, terminal bulb length, and head size of mutant worms with reported Dumpy (Dpy), Small (Sma) or Long (Lon) phenotypes by plotting and comparing their respective ratios of various parameters. These results show that the Sma phenotypes are proportionally smaller overall with mild stoutness, and Dpy phenotypes are significantly stouter and have disproportionally small head size. This study provides a standard platform for determining morphological phenotypes designating and annotating mutants that exhibit body shape variations, defining the morphological phenotype of previously unexamined mutants.

Morphological Variation Classification of Red Blood Cells using Neural Network Model in the Peripheral Blood Images (말초혈액영상에서 신경망 모델을 이용한 적혈구의 형태학적 변이 분류)

  • Recently, there have been researches to automate processing and analysing images in the medical field using image processing technique, a fast communication network, and high performance hardware. In this paper, we propose a system to be able to analyze morphological abnormality of red-blood cells for peripheral blood image using image processing techniques. To do this, we segment red-blood cells in the blood image acquired from microscope with CCD camera and then extract UNL fourier features to classify them into 15 classes. We reduce the number of multi-variate features using PCA to construct a more efficient classifier. Our system has the best performance in recognition rate, compared with two other algorithms, LVQ3 and k-NN. So, we show that it can be applied to a pathological guided system.

Morphological study on effect of radiation in developing mouse brain after fetal exposure (태아시기의 방사선 노출에 의한 마우스 뇌 발달에 미치는 영향에 관한 형태학적 연구)

  • The present study analyze the morphological aspects of the cerebrum of mice with prenatal exposure to high and low dose (0.5, 1, 2 Gy) of $\gamma$-radiation on gestation day 12 or 16. The animal were allowed to give birth and the offspring were sacrificed at postnatal days 28 for gross and microscopic examination of cerebrum. Their body weight, brain weight, brain length, brain width, cortical thickness and area of cingulum bundle were examined. The histological and planimetric analysis were performed observing coronal sections. The gross malfomation (microcephaly) and abnormality of cortical architecture were prominent after exposure to 2 Gy on day 12 of gestation. significant dose-related reductions in body weight, brain weight, brain size were found in all irradiated groups. A significant change was found in thickness of the cerebral cortex and area of the cingulum bundle in the groups exposed to 0.5 Gy or more. There was no difference a lamina patter of six layers in cerebral cortex between the control and irradiated groups, but cell packing density increased significantly in the group exposed to 1 Gy or more. These results suggested that dose as low as 0.5 Gy could cause a morphologically reduce change in developing mouse cerebrum and exposure on day 12 of gestation to $\gamma$-irradiation is a particularly sensitive phase in causing malformation and abnormality of central nerve system.

Effects of Storage Time on Bacteria Concentration and Sperm Parameters in Boar Semen (돼지 액상정액 보존 일수에 따른 정액내 세균과 정자 기능의 변화)

  • This study was conducted to determine the relationship between elapsed time after semen preservation on the changes of bacteria and semen quality. Semen was diluted with BTS(Beltsville Thawing Solution) extender without antibiotic for 7 days and sperm parameter and fertility were measured. Sperm motility was measured by CASA and total bacteria number was counted after 22~24 hr incubation from counting agar plate in which sperm dilute to $10{\sim}10^6$ in 0.9% saline solution and inoculate to agar. Acrosomal integrity was measured by Chlortetracycline (CTC) staining. CTC patterns were uniform fluorescence over the whole head (pattern F), characteristic of incapacitated acrosome-intact spermatozoa; fluorescence-free band in the post-acrosomal region (pattern B), characteristic of capacitated acrosome-intact spermatozoa; and almost no fluorescence over the whole head except for a thin band in the equatorial segment (pattern AR), characteristic of acrosome reacted spermatozoa. Total number of bacteria was significantly increased (p<0.0001) 3 days after preservation. Sperm motility, viability, and morphological abnormality on elapsed time after preservation were lower from 5 ($77.24{\pm}6.47$, p<0.001) and 7 days ($77.24{\pm}6.47$, p<0.001) after preservation compared to 1 ($15.71{\pm}7.18$) and 3 days($18.39{\pm}7.22$) after preservation, respectively. Sperm viability was significantly lower ($53.25{\pm}35.03$, p<0.0001) at 7 days after preservation. Morphological abnormality of sperm was lower (p<0.001) at 1 ($15.71{\pm}7.18$) and 3 ($18.39{\pm}7.22$) days compared to 5 ($21.84{\pm}7.91$) and 7 ($22.59{\pm}9.93$) days after preservation. Acrosomal integrity and capacitation rate (pattern F) were significantly lower (p<0.001) from 5 days after preservation. Based on the data we obtained from this study suggested that semen preserved more than 5 days without antibiotic would not recommend use for artificial insemination.

Skeletal and Morphological Development of Larvae and Juveniles in Cultured Threeline Grunt Parapristipoma trilineatum (Perciformes: Haemulidae) from Korea (양식산 벤자리(Parapristipoma trilineatum) (농어목, 하스돔과) 자치어의 형태 및 골격발달)

  • Morphological and skeletal development in the cultured threeline grunt Parapristipoma trilineatum was studied from 1.96 to 61.39 mm notochord length (NL) or standard length (SL) specimens (n=360). After two days post-hatching (dph, 2.13 mm NL), the mouth opened, and melanophores formed in the eyes and egg yolk were absorbed. Oli globules were absorbed at 3 dph (2.71 mm NL). The total number of fin rays reached that of adults at 28 dph (10.58 mm SL). During skeletal development, the premaxillaray, dentary, opercle, preopercle, and cleithrum first began to ossify at 6 dph (3.51 mm NL). Ossification of all skeletal elements was completed at 28 dph (14.57 mm SL). The vertebrae began to ossify at 17 dph (5.63 mm SL) and were completed at 28 dph (8.48 mm SL), progressing from cranium to caudal bone. The centrum ossified from the dorsal to ventral direction. Vertebral fusion and separation were found to be common skeletal abnormalities.

Inadvertent Dural Puncture during Caudal Approach by the Introducer Needle for Epidural Adhesiolysis Caused by Anatomical Variation

  • There have been reports of abnormalities in the lumbosacral region involving a lower-than-normal termination of the dural sac, which is caused by disease or anatomical variation. Inadvertent dural puncture or other unexpected complications can occur during caudal epidural block or adhesiolysis in patients with these variations, but only a small number of case reports have described this issue. We report a case of dural puncture by the introducer needle before attempting caudal epidural adhesiolysis, which occurred even though the needle was not advanced upward after penetrating the sacrococcygeal ligament. Dural puncture was caused by a morphological abnormality in the lumbosacral region, with no pathological condition; the dural sac terminal was located more distally than normal. However, dural puncture could have been prevented if we had checked for such an abnormality in the magnetic resonance imaging (MRI) taken before the procedure.

  • The morphological change of ECG is the important diagnostic parameter to finding the malfunction of a heart. Generally ST segment deviation is concerned with myocardial abnormality. The aim of this study is to detect the change of ST in shape using a polynomial approximation method and the reference ST type. The developed algorithm consists of feature point detection, ST level detection and ST shape classification. The detection of QRS complex is accomplished using it's the morphological characteristics such as the steep slope and high amplitude. The developed algorithm detects the ST level change, and then classifies the ST shape type using the polynomial approximation. The algorithm finds the least squares curve for the data between S wave and T wave in ECG. This curve is used for the classification of the ST shapes. ST type is classified by comparing the slopes of the specified points between the reference ST set and the least square curve. Through the result from the developed algorithm, we can know when the ST level change occurs and what the ST shape type is.

Study on Development Effect on Zebrafish Embryo by Alacholr, Butachlor and Fipronil (농약 alacholr, butachlor 및 fipronil이 제브라피쉬 배아 발생에 미치는 영향)

  • This study is aimed to search the possibility of developmental toxicity test using the zebrafish from the pesticide. We selected herbicides alachlor and butachlor, reported for fish toxicity, and insecticide fipronil reported for the high fish toxicity and the honey bee risk among the pesticides with high usability for the examples of the pesticides in this experiment. In this study, we showed those effects on the zebrafish embryo development by exposing different kinds of pesticide with different concentration and exposed time periods. As a result, the rates of hatching and abnormality of the zebrafish embryo after treatments of alachlor were increased in 24-48 hpf group, and the juvenile fishes in every group exposed to $40{\mu}M$ or more of alachlor displayed sever morphological changes such as bent tails, edema and activity failures. In case of the butachlor, the rates of hatching and the abnormality in 24-48 hpf group were higher than the other groups exposed in different time periods. The fatality before hatching was high in $40{\mu}M$ or more of butachlor treatment, and entire zebrafish embryos in 48 hpf group died before hatching. All the living juvenile fishes showed morphological changes as like as the treatment of alachlor. The rate of hatching and the survival of the zebrafish embryo by the fipronil were higher than other pesticides. However, morphological changes such as bent tails were observed from the most of living juvenile fishes. Therefore, the effects of three different pesticides with different concentrations and exposing time periods on the development of zebrafish embryos showed that all the pesticides effects were proportional to the concentration, and exposing time periods may cause the morphological abnormality.

The Association between Morphological and Functional Characteristics of the Bicuspid Aortic Valve and Bicuspid Aortopathy

  • Objective: To identify the association between morphological and functional characteristics of the bicuspid aortic valve (BAV) and bicuspid aortopathy and to identify the determinants of aortic dilatation using transthoracic echocardiography (TTE) and cardiac computed tomography (CCT). Materials and Methods: This study included 312 subjects (mean [SD] age, 52.7 [14.3] years; 227 males [72.8%]) who underwent TTE and CCT. The BAVs were classified by anterior-posterior (BAV-AP) or right-left (BAV-RL) orientation of the cusps and divided according to the presence (raphe+) or absence of a raphe (raphe-) based on the CCT and intraoperative findings. The dimensions of the sinus of Valsalva and the proximal ascending aorta were measured by CCT. We assessed the determinants of aortic root and proximal ascending aortic dilatation (size index > 2.1 cm/m2) by Univariable and multivariable logistic regression analyses. Results: Of the 312 patients, BAV-AP was present in 188 patients (60.3%), and 185 patients (59.3%) were raphe+. Moderate-to-severe aortic stenosis (AS) was the most common hemodynamic abnormality (54.8%). The most common type of aortopathy was the combined dilated root and mid-ascending aortic phenotype (62.5%). On multivariable analysis, age and AS severity were significantly associated with aortic root dilatation (p < 0.05), and age, sex, and AS severity were significantly associated with ascending aortic dilatation (p < 0.05). However, the orientation of the cusps, presence of a raphe, and severity of aortic regurgitation were not associated with aortic root and ascending aortic dilatation. Conclusion: BAV morphological characteristics were not determinants of aortic dilatation. Age, sex, and AS severity were predictors of bicuspid aortopathy. Therefore, age, sex, and AS severity, rather than valve morphology, need to be considered when planning treatment for BAV patients.

Esthetic Rehabilitation of Anterior Dentition by All Ceramic Crowns Using IPS e.max CAD : A Case of Tetracycline Discoloration (전치부 전부도재관을 이용한 테트라싸이클린 착색의 심미보철 회복 증례 (IPS e.max CAD 블록의 이용))

  • The purpose of this case report was to present an example of an esthetic and functional rehabilitation of anterior teeth with tetracycline discoloration and minor morphological abnormality of a 39-year old female. A chairside computer-aided design/computer-aided manufacturing (CAD/CAM) system with CEREC AC was applied for the prosthetic procedure and all ceramic crowns made with lithum disilicate (IPS e.max CAD) restored the esthetic and functional features of sixteen anterior teeth successfully.
