Optimization of Fermentation Conditions for Production of Recombinant Human Interleukin-2 in Escherichia coli (대장균에서의 재조합 인체 인터루킨-2 생산을 위한 발효조건 최적화)

  • For optimal production of recombinant human interleukin-2 (IL-2) in E. coli the effect of fermentation conditions on cell growth, IL-2 production, and stability of recombinant cells were investigated. Among the complex nutrients tested in this work, yeast extract, peptone and corn steep liquor were found to be effective for recombinant cell growth. The recombinant cells were maintained stably under repression condition (3$0^{\circ}C$), but the stability of recombinant cells were drastically reduced upon induction of IL-2 expression (42$^{\circ}C$) even under the selection pressure. Addition of antibiotics to the culture medium resulted in the cell growth inhibition without significant improvement in recombinant stability. When the expression of IL-2 gene was induced at different growth phases, highest IL-2 production was achieved by the induction of IL-2 at the middle-exponential growth phase. It was found that the production of IL-2 significantly inhibited the cell growth and the ex-pression of other genes in the plasmid.

  • The effect of equivalence ratio and fuel/air mixing quality on the phase-resolved gas temperatures at different phases of the oscillating pressure cycle was experimentally investigated. An atmospheric pressure, optically accessible and laboratory-scale dump combustor operating on methane with heat release rate of 1.59kW was used. Temperature measurements were made using coherent anti-Stokes Raman spectroscopy (CARS) at several spatial locations for typical unstable combustion conditions. Analysis was conducted using parameters such as phase-resolved averaged temperature, normalized standard deviation and temperature probability distribution functions (PDFs). Also the probability on the occurrence of high temperature (over 1900K) was investigated to get the information on the perturbation of equivalence ratio and NOx emission characteristics. It was shown that most of temperature histograms exhibit Gaussian profile which has short breadth of temperature fluctuation at equivalence ratio of 0.6, while beta profile was predominant for the cases of other equivalence ratios (${\Phi}$=0.55, 0.50). The characteristics on the occurrence of high temperature also displayed periodic wave form which is very similar to the pressure signal. And the amplitude of this profile goes larger as the fuel/air mixing quality become poorer. These also provided additional information on the perturbation of equivalence ratio at flame as well as NOx emission characteristics.

STUDY OF FLARE-ASSOCIATED X-RAY PLASMA EJECTIONS : II. MORPHOLOGICAL CLASSIFICATION

  • X-ray plasma ejections often occurred around the impulsive phases of solar flares and have been well observed by the SXT aboard Yohkoh. Though the X-ray plasma ejections show various morphological shapes, there has been no attempt at classifying the morphological groups for a large sample of the X-ray plasma ejections. In this study, we have classified 137 X-ray plasma ejections according to their shape for the first time. Our classification criteria are as follows: (1) a loop type shows ejecting plasma with the shape of loops, (2) a spray type has a continuous stream of plasma without showing any typical shape, (3) a jet type shows collimated motions of plasma, (4) a confined ejection shows limited motions of plasma near a flaring site. As a result, we classified the flare-associated X-ray plasma ejections into five groups as follows: loop-type (60 events), spray-type (40 events), jet-type (11 events), confined ejection (18 events), and others (8 events). As an illustration, we presented time sequence images of several typical events to discuss their morphological characteristics, speed, CME association, and magnetic field configuration. We found that the jet-type events tend to have higher speeds and better association with CMEs than those of the loop-type events. It is also found that the CME association (11/11) of the jet-type events is much higher than that (5/18) of the confined ejections. These facts imply that the physical characteristics of the X-ray plasma ejections are closely associated with magnetic field configurations near the reconnection regions.

Isolation and characterization of Vitreoscilla mutant defective in catalase-peroxidase hydroperoxidase I

  • Mutants of an obligate aerobic bacterium, Vitreoscilla, that have deficiency in heat-labile catalase-peroxidase hydroperoxidase I (HPI) were created by EMS treatment. The catalase-peroxidase HPI-deficient mutant showed substantially lower peroxidase activity in exponential and mid-stationary phase compared with the wild type strain. In late stationary phase, the mutant exhibited no peroxidase activity. Peroxidase deficiency in the mutant was revealed by polyacrylamide gels stained for peroxidase activity. Characteristically, catalase levels in the mutant increased about 14- and 8-fold during growth in the exponential and stationary phases, respectively, compared to those in the wild type, suggesting a compensatory effect for protection from $H_2O_2$ toxicity. The mutant showed differences in physiology from the wild type: retardation in growth rate and decrease in oxygen consumption. Both the wild type and the catalase-peroxidase HPI-deficient mutant of Vitreoscilla had lower growth rates in media containing increasing $H_2O_2$ concentrations. However, the mutant exhibited an additionally decreased growth rate after 6 to 8 h of growth compared to the wild type. The wild type was resistent up to 20 mM $H_2O_2$, whereas the mutant was very sensitive to high concentrations of exogenous $H_2O_2$. Although elevated catalase levels would provide protection of the bacteria from the deleterious effect of $H_2O_2$, it did not appear to be complete. Cell-free extracts of the mutant showed decreased NADH oxidation rates and higher accumulation of $H_2O_2$ during this oxidation. These results may account for the impaired growth and earlier onset of death phase by the catalase-peroxidase HPI-deficient mutant of Vitreoscilla.

Effects of Methyl Gallate on Arachidonic Acid Metabolizing Enzymes: Cyclooxygenase-2 and 5-Lipoxygenase in Mouse Bone Marrow-Derived Mast Cells

  • Methyl gallate (MG) is a medicinal herbal product that is isolated from Paeonia lactiflora that inhibits cyclooxygenase-2 (COX-2) dependent phases of prostaglandin $D_2\;(PGD_2)$ generation in bone marrow-derived mast cells (BMMC) in a concentration-dependent manner with an $IC_{50}$ values of $17.0\;{\mu}M$. This compound also found inhibited the COX-2-dependent conversion of the exogenous arachidonic acid to $PGD_2$ in a dose-dependent manner with an $IC_{50}$ values of $190\;{\mu}M$, using a COX enzyme assay kit. However, at concentrations up to $80\;{\mu}M$, MG did not inhibit COX-2 protein expression in BMMC, indicating that MG inhibits COX-2 activity directly. Furthermore, MG consistently inhibited the production of leukotriene $C_4\;(LTC_4)$ in a dose dependent manner, with an $IC_{50}$ value of $5.3\;{\mu}M$. These results demonstrate that MG has a dual cyclooxygenase-2/5-lipoxygenase inhibitory activity, which might provide the basis for novel anti-inflammatory drugs.

A Loop Filter Size and Spur Reduced PLL with Two-Input Voltage Controlled Oscillator (두 개의 입력을 가진 VCO를 이용하여 루프필터와 스퍼 크기를 줄인 위상고정루프)

  • In this paper, a novel PLL has been proposed that reduces the size of the loop filter while suppressing spur by using a VCO with two inputs. Through the stability analysis according to the operating status, the PLL is designed to operate stably after the phase fixing. The capacitor of loop filter usually occupies larger area of PLL. It is a VCO that can reduce the size of the loop filter by increasing the effective capacitance of the capacitor through the simultaneous charge and discharge operation by two charge pumps and has two signals operating in opposite phases. The settling time of set to $80{\mu}s$ approximately by using a LSI(Locking Status Indicator) indicating the phase locking status. The proposed PLL is designed using a supply voltage of 1.8V and a $0.18{\mu}m$ CMOS process.

The Design and Characteristic Analysis of a Digital Signal Transmission System Based on Power Line Communications

  • The objective of this study is to share multimedia contents included in existing digital devices and to solve the problems of an increase in installation fees and non-environmentally friendly interiors. This study designed a new digital signal transmitter and receiver using power line transmission and HDMI in order to solve the problems in the existing systems. The transmitter and receiver designed in this study used an AD9867BCPZ PLC chip in which the transmission came from digital signals originating in a PC, and the system architecture was configured so that the outputs signals were connected to a TV from the receiver. The experiment was implemented by adding a Video Test Generator, a USBPre external sound card, and Smaart Live 6 for analyzing the characteristics of the configured system. In the video test results, it was verified that communication was actively implemented, and the image quality showed a constant level from the measurement of the captured video. In the case of the sound, it was verified that more than 90% of the sound signals were normally transmitted and received from the examination of their phases and magnitudes. Thus, the performance of the system designed in this study was verified, which leads to the resolution of some of the problems found in current digital devices.

Effects of Warming Rate and Degenerated Blastomere(s) on Development of Frozen and Thawed Mouse Embryos (냉동.해빙한 생쥐배아의 발생에 미치는 해빙속도와 퇴화할구의 영향)

  • The present experiments have been bone to verify the effects of the warming rate and the degenerated blastomere(s) on further development of the frozen and thawed 4- and 8-cell mouse embryos. The embryos obtained from the mouse superovulated and mated were frozen in the solution of 15M DMSO in PBS containing 10% FCS at a slowly cooling rate($0.3^{\circ}C/min$). Two methods of warming slowly($8^{\circ}C/min$) and quickly ($450^{\circ}C/min$) were applied for thawing embryos. The thawed embryos were grouped according to the number of healthy blastomere(s) in the embryos. Some of the embryos were eliminated their degenerated blastomere(s) by means of a micromanipulation technique. The embryos were examined their developmental phases after 48 or 72 hrs incubation. The rates of blastocyst development from the frozen and thawed 4- and 8-cell embryos were 72.7% and 73.5%, respectively in case of thawing slowly, and were 78.9% and 80.0%, respectively in case of thawing quickly. The rate in case of thawing quickly was significantly higher than that in case of thawing slowly. The rates of blastocyst development from the frozen and thawed 4- and 8-cell embryos eliminated their degenerated blastomere(s) increased 5.9% and 24.4%, respectively compared with those of control groups not eliminated. The more number of degenerated blastomere(s) were eliminated from the embryos, the higher rate of blastocyst development was shown. It may be concluded from the results that the quickly thawing method is better for increasing survival rate than the slowly thawing one, and that the degenerated blastomere(s) in the frozen and thawed embryos affects as an interfering factor for further development of the embryos.

Synthesis and Characterization of Ruthenium Doped TiO2 Nanofibers

  • Ruthenium(Ru)-doped $TiO_2$ nanofibers were prepared using electrospun Ru-$TiO_2$/poly(vinyl acetate) (PVAc) fibers and subsequent annealing for 1 h at temperatures in the range of $500^{\circ}C$ to $1000^{\circ}C$ in air. The properties of the Ru-$TiO_2$ fibers were characterized as a function of the Ru content and calcination temperature using X-ray diffraction, thermal gravimetry with differential scanning calorimetry, Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, and viscometer, pycnometer and dynamic tensiometer measurements. Although the diameter of the fiber decreased slightly with increasing calcination temperature, no dramatic changes were observed with respect to the ruthenium content. The XRD and FT-IR results revealed that anatase phase and ruthenium metal began to be formed after calcination at temperatures above $500^{\circ}C$. Anatase and rutile phases and ruthenium metal coexisted in the fibers calcined above $600^{\circ}C$. No anatase phase was detected in the fibers containing ruthenium when they were calcined at $1000^{\circ}C$. The morphology of the fibers changed from smooth and uniform to porous with increasing temperature. The experimental results suggest that the calcination temperature and Ru content were influential in determining the morphology and structure of the fibers.

A Study on the decision of Scattering distance by Shape of Fragments in LPG Tank lorry Explosion (LPG 탱크로리 폭발시 파편 형상에 따른 비산거리 산정에 관한 연구)

  • LPG is a substance that requires a lot of attention because it can cause fatal damage to people and environment when an accident occurs. LPG is frequently accidents in transportation facilities as well as fixed facilities, among which LPG tank lorries are the most frequent accidents. When the LPG tank is evacuated, the LP gas leaks into two phases, leaks mostly to the gas and leaks to some liquid. At this time, the leaked gas will also sink downward because it is heavier than air, and if it continues to leak, it may form an explosion and explode by the ignition source. The purpose of this study is to present the evacuation distance by analyzing the effect distance of the LPG liquefied petroleum gas in the event of explosion. As a result of calculation of the scattering radius of the fragment, the cylinder fragment was scattered up to 561 m. Therefore, it is appropriate to set the distance to be escaped when the LPG tanker leaks to 561m or more.
