  • Cytochrome P-450(CP-450) is one of the three components of the liver microsomal enzyme system which hydroxylates fatty acids, hydrocarbons and a variety of drugs and other foreign compounds. Female Balb/c mice were immunized with purified polychlorinated bipheny(PCB)-induced CP-450 LMII. The spleen cells derived from immunized mice were fused with $SP^2$ myeloma cells using polyethylene glycol(PEG 3500). The hybrid cells were selected by hypoxanthine-aminopterine and thymidine(HAT) medium and the culture fluid were screened by enzyme-linked immunosorbent assay to CP450 LMII. The hybrid cess(${\times}10^7$) were innoculated into intraperitoneal cavity of Balb/c mice for the purpose of production of ascitic fluids. Monoclonal antibody(Mab) was purified from ascitic fluid by DEAE cellulose ion exchange chromatography and $I^{125}$-labeled Mab was also confirmed by autoradiography and SDS-polyacrylamide gel electrophoresis (MW : 55,000 and 110,000).

A Monoclonal Antibody That Specifically Binds Chitosan In Vitro and In Situ on Fungal Cell Walls

  • We report the generation of the first monoclonal antibody that specifically binds to the polysaccharide chitosan. Mice were immunized with a mixture of chitosans, and hybridoma clones were screened for specific binders, resulting in the isolation of a single clone secreting a chitosan-specific IgM, mAbG7. In ELISAs, the antibody could bind to chitosans of varying composition, but demonstrated the highest affinity for chitosans with lower degrees of acetylation (DA) and very poor binding to chitin. We tested the ability of the antibody to bind to chitosan in situ, using preparations of fungal cell walls. Immunofluorescence microscopy confirmed that the antibody bound strongly to the cell walls of fungi with high levels of chitosan, whereas poor staining was observed in those species with cell walls of predominantly chitin or cellulose. The potential use of this antibody for the detection of fungal contamination and the protection of plants against fungal pathogens is discussed.

  • These experiments were carried out to investigate of the molecular characteristics of porcine zona pellucidae and to examine the reactivity of anti-zona antibody by SDS-PAGE, Immunoblotting and Immunoprecipitation. The results obtained in these experiments were summarized as follows : 1. The result obtained by SDS-PAGE of porcine zona pellucidae indicated that it composed of several units with molecular weight ranging 55,000-110,000. 2. In order to see the reactivity of antibodies to zona pellucidas, immunoblotting was applied. The results indicated that polyclonal antibodies to porcine and mouse zona reacted with porcine zona. While monoclonal antibody to porcine did not react with the procine zona enough to show a clear band on a gel. 3. Labelling of porcine zonae with 125I was performed using the Iodogen method, the radioactivity and the percent incorporation of 125I into porcine zonae were approximately 26,000 cpm/10${mu}ell$ and 16, respectively. 4. Measurements of radioactivity and O.D value for Immunoprecipitates produced by reaction of 125I-porcine zona with anti-zona antisera were confirmed that existence of reactivity of monoclonal antibody to porcine zona although its reactivity was lower than that of polyclonal antibodies, and reconfirmed that cross-reactivity of polyclonal antibody of mouse zona with porcine zona.

  • When monoclonal antibodies are used in radioimmunoassay or immunoscintigraphic studies, post-labelling immunoreativity is a critical parameter. $^{125}I$ was incorporated to CEA-79 (anti CEA monoclonal antibody developed in Korea) by chloramine T and iodogen method with variable specific activities from $0.1{\mu}Ci/{\mu}g$ to $100{\mu}Ci/{\mu}g$. We used a new method to evaluate the immunoreactivites of modified antibody relative to the unlabelled native antibody from competitive binding assay. The effect of immunoreactivity and specific activity to the sensitivity of radioimmunometric assay was also evaluated. As a result, chloramine T method was better than iodogen method in radioiodination of CEA-79, because the immunoreactivity of antibody was relatively well reserved and more stable. New competitive binding assay was simple and effective to evaluate the change of immunoreactivity in radiolabelling. Antibody with high immunoreactivity and high specific activity improved the sensitivity of radioimmunometric assay, whereas antibody with high specific activity but low immunoreactivity didn't. The immunoreactivity and specific activity should be optimized according to the clinical un, and competitive binding method is useful in selection of optimal radiolabelling assay.

  • A new chemiluminescence immunochromatographic analysis system with high sensitivity and high reproducibility was developed for the determination of microcystins (MCs) in water. Horse radish peroxidase (HRP) labeled microcystin monoclonal antibody was used for the sensitive chemiluminescence detection. The chemiluminescence immunochromatographic analysis system was composed of microcystin LR (MCLR)-monoclonal antibody (mAb)-Horse Radish Peroxidase (HRP) conjugate, MCLR-BSA conjugate, luminol, hydrogen peroxide mixture solution, an immunochromatographic assay strip and luminometer. To detect the concentration of microcystins in water, we utilized one spot analysis of the strip instead of flow type analysis. We could detect the microcystins in water at a concentration as low as 9.45 pg/mL with the chemiluminescence (CL) detection.

  • Two clones of monconal antibodies(Co-1 and Co-2) against BSA-benzoylecgonine(BSABE) were produced. Both monoclonal antibodies showed high binding affinity to BSA-BE. Observing from ELISA inhibition assay, Co-1 reacted only weakly with soluble benzoylecgonine, while Co-2 showed considerable reactivity with soluble benzoylecgonine.

  • 본 연구자들은 근원세포를 면역시켜 얻은 hybidoma들을 검색하여. 계배 근원세포의 분화와 관련된 단백질을 인지하여 분화를 억제하는 대과가 있는 monoclonal antibody 3H35를 선별하여 그 항원을 확인한 바 있다(Kim et af.. (1992), Korean J. Zool 35 29-36) 본 연구에서는 λZAP에 cloning된 chicken muscle CDNA library들을 lacZ fusion protein으로 발현시켜 항체 3H35로 검색하여 그 유전자를 찾아내었다. 선별한 CDNA clone 중 C59의 삽입 절편은 1.6 kb이었고, 발현시킨 facE fusion protein 은 60 kDa로, f-galactosidase에 대한 항체에 반응하며 3H35와도 반응함을 immunoaffinitv adsorbant와 immunoblot으로 확인하였다 Clone C59의 삽입 절편의 염기서열을 분석한 결과, 실제 유전자는 1.6 kb 이상이며, 알려진 어느 다른 유전자와도 관련이 없는 새로운 근특이 유전자로 판단되었다. 아미노산으로 전환시켰을 때 31개의 특이한 서열이 7차례 반복된 부분이 나타났으며 이 서열의 23개가 일정하게 보존되어있고 나머지 서열의 아미노산의 polarity도 매우 유사하게 효존되어있다. 이들의 보존성이 극히 높은 것으로 보아 독특한 기능을 수행하는 domain으로 추정된다.

  • Clonorchis sinensis is a common parasite of man in Korea. Researches on the specific antigens of C. sinensis would be valuable not only because those elucidate the molecular characteristics of this fluke but also because it is applicable to immunodiagnosis. Although many monoclonal antibodies have been used in the field of parasite immunology, few articles on monoclonal antibodies against C. sinensis have been published so far. The aim of this study was to analyse C. sinensis antigens recognized by monoclonal antibodies, and to set up ELISA-inhibition test using C. sinensis specific monoclonal antibodies for improved specificity of immunodiagnostic tests. By fusion between spleen cells of the mice immunized with C. sinensis water-soluble crude adult worm antigens and plasmacytoma cells of mouse origin, 29 hybridoma clones secreting anti-C. sinensis monoclonal antibodies were made, and 8 clones among those were found specific. After cell cloning, isotypes of 6 selected specific monoclonal anti- bodies were determined to be IgGl, IgG2b and IgA. Four exposed antigenic determinants of natural infection were recognized by different specific monoclonal antibodies. By enzyme-immunoelectrotransfer blot, 10 KD, 34 KD antigenic determinants were found to be reacted with CsHyb 0714-20, CsHyb 0605-10 monoclonal antibodies, respectively, The antigenic determinant recognized by CsHyb 0714-20 monoclonal antibody was revealed to be located at the surface and parenchyme of a parasite by indirect immunoauorescent antibody technique, and those reacted with CsHyb 0605-10, CsHyb 0714-25 monoclonal antibodies were found at the parenchyme and intestine. The antigenic determinant reacted with CsHyb 0605-23 monoclonal antibody was found mainly around the uterine eggs. Four antigenic determinants recognized by specific monoclonal antibodies were all found to be present in the early eluted fractions of C. sinensis antigens separated by Sephadex G-200 gel filtration. By conventional ELISA, 75% of clonorchiasis cases were found positive, but 7.1% of normal controls and 37.5% of paragonimiasis cases showed false positives. However, by ELISA-inhibition test using C. sinensis specific monoclonal antibody (CsHyb 0605-23), 77.1% of clonorchiasis cases were found positive, and there were no false positives in normal controls or paragonimiasis cases, indicating 100% specificity. The ELISA- inhibition test using monoclonal antibodies was found to have same sensitivity and definitely high specificity in comparison with conventional ELISA for serodiagnosis of human clonorchiasis.

  • Avian infectious bronchitis virus(IBV) was propagated in SPF eggs and purified by sucrose density gradient centrifugation in order to prepare the antigen. Several fusions were made between mouse myeloma cells and spleen cells from BALB/c mouse immunized with IBV antigen and two hybridoma clones producing specific monoclonal antibody(MCA) against the IBV were established. The MCAs were classified as IgG type and revealed no neutralizing and hemagglutination inhibition activity. Using the MCA IBV antigen was detected by IFA method in tracheal smears made from chickens infected with IBV during the experimental period of 10 days.

  • The role of each supplement in serum-free medium KM3 for the growth of hybridoma and the production of monoclonal antibody was investigated. Transferrin, ethanolamine and bovine serum albumin were shown to be indispensable for the growth of four kinds of hybridoma tested in this work, especially transferrin for Alps 25-3, and ethanolamine for A4W and KW hybridoma. The addition of $\beta$-mercaptoethanol to the culture medium of HCGK showed a good influence of both the cell growth and the production of monoclonal antibody. Upon the experimental results, we suggested a serum-free medium containing a minimum composition for the culture of hybridoma.
