Mon1 Is Essential for Fungal Virulence and Stress Survival in Cryptococcus neoformans

  • Mon1 is a guanine nucleotide exchange factor subunit that activates the Ypt7 Rab GTPase and is essential for vacuole trafficking and autophagy in eukaryotic organisms. Here, we identified and characterized the function of Mon1, an ortholog of Saccharomyces cerevisiae Mon1, in a human fungal pathogen, Cryptococcus neoformans. Mutation in mon1 resulted in hypersensitivity to thermal stress. The mon1 deletion mutant exhibited increased sensitivity to cell wall and endoplasmic reticulum stress. However, the mon1 deletion mutant showed more resistance to the antifungal agent fluconazole. In vivo studies demonstrated that compared to the wild-type strain, the mon1 deletion mutant attenuated virulence in the Galleria mellonella insect model. Moreover, the mon1 deletion mutant was avirulent in the murine inhalation model. These results demonstrate that Mon1 plays a crucial role in stress survival and pathogenicity in C. neoformans.

Multiplex PCR Detection of the MON1445, MON15985, MON88913, and LLcotton25 Varieties of GM Cotton

  • A multiplex polymerase chain reaction (PCR) method was developed to simultaneously detect 4 varieties of genetically modified (GM) cotton. The event-specific primers were used to distinguish the 4 varieties of GM cotton (MON1445, MON15985, MON88913, and LLcotton25) using multiplex PCR. The acyl carrier protein 1 (Acp1) gene was used as an endogenous reference gene of cotton in the PCR detection. The primer pair Acp1-AF/AR containing a 99 bp amplicon was used to amplify the Acp1 gene and no amplified product was observed in any of the 13 different plants used as templates. This multiplex PCR method allowed for the detection of event-specific targets in a genomic DNA mixture of up to 1% GM cotton containing MON1445, MON15985, MON88913, and LLcotton25.

A kinematic study of young stars in Monoceros OB1 and R1 associations

  • The Gaia mission opens a new window to study the kinematics and dynamics of young stellar systems in detail. The kinematic properties of young stars provide vital constraints on the formation process of their host systems. Here, we present a kinematic study of the two associations Monoceros OB1 (Mon OB1) and R1 (Mon R1). Member candidates are first selected from the published list of member candidates, a compilation of OB star catalogues, and the classification of young stellar objects with the AllWISE data. According to the conventional wisdom, we selected a total of 728 members with similar proper motions at almost the same distance. Mon OB1 and Mon R1 have high levels of substructures that are also kinematically distinct. We identify six stellar groups in these associations, of which five show a pattern of expansion. In addition, the signature of rotation is found in two stellar groups of Mon OB1. Star formation history is inferred from a color-magnitude diagram. As a result, star formation in Mon OB1 has been sustained for several million years, while Mon R1 formed at almost the same epoch as the recent star formation in Mon OB1. Some old members in the outskirt of Mon OB1 have outward motions, which rules out the previously proposed outside-in star formation scenario. Star-forming regions including Mon OB1 and Mon R1 are found along a large arc-like gas structure. Hence, the formation of these two associations may originate from the hierarchical star formation along filaments in a turbulent molecular cloud.

  • This study is for estimate the amount of water supplied data to get a database for economic plan of water supply system by survey for the amount of water supplied of primary, middle and highschool. For this, 54 of primary school, middle school, high school in 6 cities of Kangwon for 5 years. As a result, average of the amount of water supplied per month is $600.7m^3/mon$ and primary, middle schools are $484.2m^3/mon$ and high schools are $833.8m^3/mon$. Average of the amount of water supplied per class is $27.0m^3/mon/class$ and primary, middle schools are $24.4m^3/mon/class$ and high schools are $32m^3/mon/class$. Average of the amount of water supplied per person per day is 55.8l/c/d and primary school, middle schools are 54.4l/c/d and high schools are 58.6l/c/d.

Conserved Roles of MonA in Fungal Growth and Development in Aspergillus Species

  • MonA is a subunit of a guanine nucleotide exchange factor that is important for vacuole passing and autophagy processes in eukaryotes. In this study, we characterized the function of MonA, an orthologue of Saccharomyces cerevisiae Mon1, in the model fungus Aspergillus nidulans and a toxigenic fungus A. flavus. In A. nidulans, the absence of AnimonA led to decreased fungal growth, reduced asexual reproduction, and defective cleistothecia production. In addition, AnimonA deletion mutants exhibited decreased spore viability, had reduced trehalose contents in conidia, and were sensitive to thermal stress. In A. flavus, deletion of AflmonA caused decreased fungal growth and defective production of asexual spores and sclerotia structures. Moreover, the absence of monA affected vacuole morphology in both species. Taken together, these results indicate that MonA plays conserved roles in controlling fungal growth, development and vacuole morphology in A. nidulans and A. flavus.

Effects of Sucrose Stearate Addition on the Quality Improvement of Ready-To-Eat Samgyetang During Storage at 25℃

  • The effects of sucrose stearate at various concentrations (0.1%, 0.2%, and 0.3%, w/v) on the physico-chemical characteristics of ready-to-eat (RTE) Samgyetang were investigated during storage at $25^{\circ}C$ for 12 mon. Over the storage duration, the addition of sucrose stearate had no significant effects on the proximate composition of Samgyetang, including meat, broth, and porridge, or the hardness and spreadability of the porridge, although it resulted in significantly higher CIE $L^*$ values for the porridge. The CIE $L^*$ values of Samgyetang porridge with added sucrose stearate increased until 9 mon, while the control decreased until 6 mon, and the values for both changed insignificantly thereafter. The breast meat of Samgyetang treated with sucrose stearate showed higher percentages of polyunsaturated fatty acid after 3 mon and lower percentages of monounsaturated fatty acid after 6 mon compared to the control (p<0.05), while no significant differences were observed with the different sucrose stearate concentrations (p>0.05). The overall sensory acceptability scores were higher at sucrose stearate concentrations of 0.2% or 0.3% after 6 mon and at 0.1% after 9 mon compared to those of the control.

Effect of Feed Additives in Growing Lambs Fed Diets Containing Wet Brewers Grains

  • The study was conducted to evaluate and compare the effects of feed additives on ruminal fermentation, nutrient digestibility and performance of lambs fed diets containing 60% wet brewers grains (WBG). In Experiment 1, two simultaneous trials were conducted. Fifty intact ($20.2{\pm}0.8kg\;BW$) lambs were used in a feedlot trial and 10 (rumen cannulated; $32{\pm}1kg\;BW$) in a digestion trial. The pH, volatile fatty acids (VFA) and ammonia-N in lambs were also estimated. Lambs were randomly assigned to one of five diets: i) without additives (Con), ii) with 1% bicarbonate (Bic), iii) with 1% bentonite (Ben), iv) with 33 mg/kg monensin (Mon) and v) with 200 mg/kg fibrolityc enzymes (Enz). In Experiment 2, 120 RambouilletPelibuey intact male lambs ($19.5{\pm}1.5kg\;BW$) were used in a feedlot trial and randomly assigned to four diets: i) without additives (control), ii) with 1% Bic, iii) with 33 mg/kg Mon and iv) with 1% Bic and 33 mg/kg Mon. In Experiment 1, lambs fed diets containing Bic or Mon had significantly higher final weight, DMI, ADG than other lambs. However, apparent DM, OM, CP, NDF and ADF digestibilities and ruminal individual VFA content were similar (p>0.05) among treatments. Conversely, treatmentcollection period interaction was significant for ruminal pH and NH3. In Experiment 2, lambs fed diets containing a Bic and Mon combination had significantly higher final weight, DMI and ADG. It is concluded lambs fed Bic or Mon or Bic and Mon combination had better performance characteristics than lambs on Ben or Enz.

Characteristics of New Shiitake Strain "Sanlim No. 7" Produced by Di-mon Hybridization Method (Di-mon교배법에 의하여 육성한 표고 신품종 "산림7호"의 특성)

  • New shiitake(Lentinula edodes) strain "Sanlim No. 7" was produced by Di-mon hybridization method. Bed-log cultivation of Sanlim No. 7 was performed. Fruit-body production was most at autumn and it is suitable to use as fresh shiitake rather than dried one. The fruit body is hemispherical shape, brown coloured and diameter of pileus is ca. 47 mm. Row of gill is straight, and its width is narrow. Optimum temperature of fruit-body formation was $11{\sim}23^{\circ}C$, and the fruiting is sporadic. The total amount of fruit-body production during 4 years(one generation) was ca. 1.4 kg/log.

  • MOnCa2 is a framework for building intelligent smartphone applications based on smartphone sensors and ontology reasoning. In previous studies, MOnCa determined and inferred user situations based on sensor values represented by ontology instances. When this approach is applied, recognizing user space information or objects in user surroundings is possible, whereas determining the user's physical context (travel behavior, travel destination) is impossible. In this paper, MOnCa2 is used to build recognition models for travel behavior and routes using smartphone sensors to analyze the user's physical context, infer basic context regarding the user's travel behavior and routes by adapting these models, and generate high-level context by applying ontology reasoning to the basic context for creating intelligent applications. This paper is focused on approaches that are able to recognize the user's travel behavior using smartphone accelerometers, predict personal routes and destinations using GPS signals, and infer high-level context by applying realization.

Meat Quality Traits of Longissimus Muscle of Hanwoo Steers as a Function of Interaction between Slaughter Endpoint and Chiller Ageing

  • Carcass characteristics and meat quality traits as a function of endpoint months of slaughter age (26 vs 32 mon) and chiller ageing (1 vs 10 d) were evaluated for m. longissmus of 26 Hanwoo steers fed with commercial diets including whole crop barley silage. Totally twenty six Hanwoo steers for 6 mon of age that were fed until 26 mon of age constituted the short term-fed group and fed until 32 mon of age constituted long-term fed group. Carcasses were chilled for 24 h and were graded. Strip loin samples were divided into two age groups (1 d and 10 d). Long-term feeding increased carcass weight, rib-eye area, yield grade, marbling score, firmness and quality grade of the meat. The feeding for 32 mon produced tender, juicy meat (p<0.01) with lower cooking loss and higher rating score (p<0.05) than short term feeding, while other quality traits were not influenced by the length of feeding. Intramuscular fat content and oxidative stability (TBARS value) were significantly (p<0.05) higher in beef from long-term feeding however the length of feeding did not alter the fatty acid composition. Chiller aging reduced instrumental tenderness (WBSF value), improved color, sensory tenderness, acceptability and rating of beef. The results of the present study mirrors that Hanwoo steers until 32 mon of age overall improved carcass traits and palatability compared to that for 26 mon. However, from the viewpoints of economical and environmental aspects, cost of the additional feeding for 6 mon for value-adding of eating quality was relatively high and the effects in turn were limited.
