Plant proteomics is the large-scale studies of proteins, particularly on their structures and functions, governed by the physiological metabolism of plant cells. With the development of techniques and strategies in proteomics, proteomics approach is moving forward in systems biology handling sophisticated components of major signaling and biochemical pathways in plants responding to their environment. In Korea, pioneers in plant proteomics are trying to catch up with global trends in plant proteomics; these researchers are not only improving existing techniques in protein extraction but also developing new techniques in proteomics context. In the hot field of abiotic and biotic stress proteomics, 29 and 9 out of 74 papers have been published during the review period from 2005 to 2010, respectively. This present review article provides an overview on the output of Korean plant proteomers while paying special attention to both abiotic and biotic stress proteomics.

Chitosan-based nanoparticles (CSNP) were prepared through ionic cross-linking and gelation of chitosan (CS) by tripolyphosphate (TPP). CS properties such as molecular weight, and preparation conditions were screened and the resulting nanoparticles were examined by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The obtained particles were consistently spherical with an overall diameter of approximately $107{\pm}20$ nm. They were successfully used as a carrier for Zidovudine, an anti-human immunodeficiency virus (HIV) which, to our knowledge, is novel. The encapsulation ability, loading capacity, and controlled release behavior for these CSNP was evaluated. Results indicated that their intrinsic properties were strongly affected by properties inherent to CS such as molecular weight, and by the preparation condition, such as cross-linking density, which depends on the concentration of the cross-linker. In vitro release tests for the entrapped zidovudine showed that the CNNP provided a continuous release that can last upwards 20 h.
Orchidaceae species account for one-tenth of all angiosperms including more than 30,000 species having significant ecological, evolutionary, and economic importance. Despite Orchidaceae being one of the largest families among flowering plants, crucial cytogenetic information for studying species diversification, inferring phylogenetic relationships, and designing efficient breeding strategies is lacking, except for 10% or less of orchid species cases involving mostly chromosome number or karyotype analysis. Also, only approximately 1.5% of the identified orchid species from less than a hundred genera have genome size data that provide crucial information for breeders and molecular geneticists. Various molecular cytogenetic techniques, such as fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH), have been developed for determining ploidy levels, analyzing karyotypes, and evaluating hybridity, in several ornamental crops including orchids. The estimation of genome size and the determination of nuclear DNA content using flow cytometry have also been employed in some Orchidaceae subfamilies. These different techniques have played an important role in supplementing beneficial knowledge for effective plant breeding programs and other related plant research. This review focused on orchid breeding summarizes the status of current cytogenetic tools in terms of background, advancements, different techniques, significant findings, and research challenges. Principal roles and applications of cytogenetics in orchid breeding as well as different ploidy level determination methods crucial for breeding are also discussed.
Ground cherry (Physalis pubescens) is the most prominent species in the Solanaceae family due to its nutritional content, and prospective health advantages. It is grown all over the world, but notably in northern China. In 2019 firstly bacterial leaf spot (BLS) disease was identified on P. pubescens in China that caused by both BLS pathogens Xanthomonas euvesicatoria pv. euvesicatoria resulted in substantial monetary losses. Here, we compared whole genome sequences of X. euvesicatoria to other Xanthomonas species that caused BLS diseases for high similarities and dissimilarities in genomic sequences through average nucleotide identity (ANI) and BLAST comparison. Molecular techniques and phylogenetic trees were adopted to detect X. euvesicatoria on P. pubescens using recQ, hrpB1, and hrpB2 genes for efficient and precise identification. For rapid molecular detection of X. euvesicatoria, loop-mediated isothermal amplification, polymerase chain reaction (PCR), and real-time PCR techniques were used. Whole genome comparison results showed that the genome of X. euvesicatoria was more closely relative to X. perforans than X. vesicatoria, and X. gardneri with 98%, 84%, and 86% ANI, respectively. All infected leaves of P. pubescens found positive amplification, and negative controls did not show amplification. The findings of evolutionary history revealed that isolated strains XeC10RQ, XeH9RQ, XeA10RQ, and XeB10RQ that originated from China were closely relative and highly homologous to the X. euvesicatoria. This research provides information to researchers on genomic variation in BLS pathogens, and further molecular evolution and identification of X. euvesicatoria using the unique target recQ gene through advance molecular approaches.
Soil is a dynamic biological system, in which it is difficult to determine the composition of microbial communities. Knowledge of microbial diversity and function in soils are limited because of the taxonomic and methodological limitations associated with studying the organisms. In this review, approaches to measure microbial diversity in soil were discussed. Research on soil microbes can be categorized as structural diversity, functional diversity and genetic diversity studies, and these include cultivation based and cultivation independent methods. Cultivation independent technique to evaluate soil structural diversity include different techniques such as Phospholipid Fatty Acids (PLFA) and Fatty Acid Methyl Ester (FAME) analysis. Carbon source utilization pattern of soil microorganisms by Community Level Physiological Profiling (CLPP), catabolic responses by Substrate Induced Respiration technique (SIR) and soil microbial enzyme activities are discussed. Genetic diversity of soil microorganisms using molecular techniques such as 16S rDNA analysis Denaturing Gradient Gel Electrophoresis (DGGE) / Temperature Gradient Gel Electrophoresis (TGGE), Terminal Restriction Fragment Length Polymorphism (T-RFLP), Single Strand Conformation Polymorphism (SSCP), Restriction Fragment Length Polymorphism (RFLP) / Amplified Ribosomal DNA Restriction Analysis (ARDRA) and Ribosomal Intergenic Spacer Analysis (RISA) are also discussed. The chapter ends with a final conclusion on the advantages and disadvantages of different techniques and advances in molecular techniques to study the soil microbial diversity.

To understand molecular and cellular mechanisms of many gene products in the female reproductive organs including the ovary and uterine endometrium as well as during embryo development, researchers have developed and utilized many effective methodologies to analyze gene expression in cells, tissues and animals over the last several decades. For example, blotting techniques have helped to understand molecular functions at DNA, RNA and protein levels, and the reverse transcription-polymerase chain reaction (RT-PCR) method has been widely used in gene expression analysis. However, some conventional methods are not sufficient to understand regulation and function of genes expressed in very complex patterns in many organs. Thus, it is required to adopt more high-throughput and reliable techniques. Here, we describe several techniques used widely recently to analyze gene expression, including annealing control based-PCR, differential display-PCR, expressed sequence tag, suppression subtractive hybridization and microarray techniques. Use of these techniques will help to analyze expression pattern of many genes from small scale to large scale and to compare expression patterns of genes in one sample to another. In this review, we described principles of these methodologies and summarized examples of comparative analysis of gene expression in female reproductive organs with help of those methodologies.
남조류에 대한 오랜 연구로 많은 사실을 알게 되었음에도 여전히 미지의 영역으로 남아있는 부분이 많은데, 분자 생물학에 기반한 오믹스 기술의 발전으로 새로운 도구를 이용한 다른 관점에서의 연구가 최근 활발해지고 있다. 일차적으로는 유전체 염기서열 분석기술을 사용하여 다양한 남조류의 유전체 비교분석과 유전자의 발현 양상을 연구함으로써, 독소 합성의 조절 기작 등 생리적 특성이 나타나는 원리 규명에 많은 노력이 기울여지고 있다. 또한 남조류 유전형의 다양성과 이들이 밀접하게 상호작용하는 박테리아 군집이 계절적 및 환경적 요인에 어떻게 반응하여 변화하고, 이러한 변화가 생태계에는 어떤 영향을 미치는지에 대한 연구가 생물정보학 분석기법과 결합하면서, 생태계의 복잡한 작동방식에 대한 이해도 늘어나고 있다. 특히 다양한 오믹스 기법을 복합 적용함으로써 생태계 안에서 일어나는 모든 층위의 생물학적 반응에 대한 총체적 그림을 그리는 것이 현실화되고 있으며, 이렇게 그려진 설계도로부터 녹조를 효과적으로 제어하고 건강한 수생태계를 유지할 수 있는 새로운 통찰의 가능성에 대한 기대가 고조되고 있다.
The internal standard method, which is widely used in chromatographic techniques, is applied to the size exclusion chromatography for the on-line determination of dn/dc value of the sample. This method is found to provide the dn/dc value with suitable accuracy in determining the absolute molecular weight and the molecular weight distribution of the polymers when a light scattering detector is used.
Clinical and genetic heterogeneity in association with overlapping spectrum is characteristic in pediatric neuromuscular disorders, which makes confirmative diagnosis difficult and time consuming. Considering evolution of molecular genetic diagnosis and resultant upcoming genetically modifiable therapeutic options, rapid and cost-effective genetic testing should be applied in conjunction with existing diagnostic methods of clinical examinations, laboratory tests, electrophysiologic studies and pathologic studies. Earlier correct diagnosis would enable better clinical management for these patients in addition to new genetic drug options and genetic counseling.
Small animal models are extensively utilized in the study of biomedical sciences. Current animal experiments and analysis are largely restricted to in vitro measurements and need to sacrifice animals to perform tissue or molecular analysis. This prevents researchers from observing in vivo the natural evolution of the process under study. Imaging techniques can provide repeatedly in vivo anatomic and molecular information noninvasively. Small animal imaging systems have been developed to assess biological process in experimental animals and increasingly employed in the field of molecular imaging studies. This review outlines the current developments in nuclear medicine imaging instrumentations including fused multi-modality imaging systems for small animal imaging.
