This study was carried out to know the variation of soybean seed proteins, 11S and 7S globulins, and their amino acid compositions among different colored soybean varieties, 'Danbaegkong' (yellow), 'Pureunkong' (green) 'Jinyulkong' (brown), and 'Geoumjeongkong l' (black). Soybean seed proteins showed a wide range in molecular size, but the electrophoresis patterns of total seed protein subunits showed a similarity among different colored soybean varieties. Amino acid compositions of total seed proteins were similar for all soybean varieties tested. However, soybean varieties showed low composition rates in sulfur containing amino acids. The composition rates of cysteine and methionine in the 11S globulins were higher than those of total seed proteins and 7S globulins. Glutamic acid and glycine were higher in the 11S and 7S globulins than those of total seed proteins. However, the levels of methionine and phenylalanine are high in the 11S globulins, but those of valine and lysin are slightly lower than the 7S globulins. By using HPLC, we tried to analyse the soybean seed proteins. The 11S globulin was composed of 10 major peaks whereas the 7S globulin was composed of 4 major peaks. The composition rates of 11S related proteins have a tendency to increasing during the maturing whereas those of 7S related proteins have a tendency to decreasing. Composition rates of each peaks among different colored soybean varieties suggested that soybean seed proteins are varied, although they showed similarity in the electrophoresis patterns, and understanding of this characteristics is important for the utilization of soybeans.
This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components in the MDBK(Mardin-Darby Bovine Kidney) cells. For the In vitro test, MDBK cells in ${\alpha}$-MEM(Minimum Essential Medium) were divided into 4 treatment groups according to drug types and dosages as follows; the control(group A), group B was treated with 0.3mM caffeine, group C was treated with 0.3mM caffeine and 0.3mM ferric chloride, group D was treated with 0.3mM caffeine and 0.3mM vitamin E. Those groups were further divided into 5 subgroups according to the time lapsed(control, 4hrs, 8hrs, 24hrs and 48hrs lapsed group). The concentrations of the carbonyl group and malondialdehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis) and fatty acid compositions were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the MDBK cells. The results obtained from this study were summarized as follows; 1. The concentrations of carbonyl group and malondialdehyde in MDBK cells of group C were significantly higher(p<0.01) in comparison to the control, and increased according to the time lapsed. But the results of groups B and D were little different in comparison to the group C. 2. As the analytical results of fatty acid compositions in MDBK cells, the proportions of palmitoleic acid and linoleic acid in groups B, C and D were lower in comparison to the control, while the proportion of arachidonic acid in groups B, C and D were significantly higher(p<0.01) in comparison to the control. 3. In order to determine the oxidative damages to the protein in MDBK cells, the patterns of the SDS-PAGE were examined and the patterns of SDS-PAGE in groups C and D were significantly different between 43kd and 200kd of molecular weight.
In recent years, research into medicinal herbs with antioxidative activities has increased. Chrysanthemum morifolium and Chrysanthemum indicum are aromatic herb plants and that have long been used in traditional Vietnamese medicine. This study aims to evaluate the chemical compositions and antioxidative activities of essential oils hydrodistilled from the flower heads of C. morifolium and C. indicum. The chemical compositions of the essential oils were compared using gas chromatography/mass spectrometry (GC/MS) analysis. The antioxidative activity was determined and evaluated spectroscopically by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, metal chelating activity, reducing power, and total antioxidant capacity assays. According to the GC/MS results, chrysanthenone was predominant in the essential oils of both C. morifolium (64.14%) and C. indicum (32.02%). This is the first report of the identification of chrysanthenone as a major constituent of the essential oil of C. morifolium. Both Chrysanthemum oils were also revealed to possess antioxidant potential, exhibiting high antioxidative activities. In particular, the DPPH radical scavenging activities of the C. morifolium and C. indicum oils at a concentration of 100 mg/mL were 76.9 and 83.2%, respectively. The metal chelating values of C. morifolium and C. indicum were 0.85 and 0.76, whereas the reducing power values of that at 100 mg/mL were 0.76 and 0.71, respectively. This study provides the chemical properties of the essential oils of both C. morifolium and C. indicum grown in Vietnam and their potential antioxidant capacity.

A unicellular red microalga was isolated from environmental freshwater in Korea, and its morphological, molecular, and biochemical properties were characterized. Morphological analysis revealed that the isolate was a unicellular biflagellated green microalga that formed a non-motile, thick-walled palmelloid or red aplanospore. To determine the taxonomical position of the isolate, its 18S rRNA and rbcL genes were sequenced and phylogenetic analysis was performed. We found that the isolate was clustered together with other related Haematococcus strains showing differences in the rbcL gene. Therefore, the isolated microalga was classified into the genus Haematococcus, and finally designated Haematococcus sp. KORDI03. The microalga could be cultivated in various culture media under a broad range of pH and temperature conditions. Compositions of the microalgal cellular components were analyzed, and its protein, carbohydrate, and lipid compositions were estimated to be 21.1 ± 0.2%, 48.8 ± 1.8%, and 22.2 ± 0.9%, respectively. In addition, D-glucose and D-mannose were the dominant monosaccharides in the isolate, and its amino acids were composed mainly of aspartic acid, glutamic acid, alanine, and leucine. Moreover, several polyunsaturated fatty acids accounted for about 80% of the total fatty acids in Haematococcus sp. KORDI03, and the astaxanthin content in the red aplanospores was estimated to be 1.8% of the dry cell weight. To the best of our knowledge, this is the first report of an Haematococcus sp. isolated from Korea, which may be used for bioresource production in the microalgal industry.
Microalgae hold promise as producers of sustainable biomass for the production of biofuels and other biomaterials. However, the selection of strains with efficient and robust production of desirable resources remains challenging. In this study, we isolated a green microalga from Korea and analyzed its morphological, molecular, and biochemical characteristics. Microscopic and phylogenetic analyses demonstrated that the isolate could be classified into the genus Chlamydomonas, and we designated the isolate Chlamydomonas sp. KIOST -1. Compositions of protein, lipid, and carbohydrate in the microalgal cells were estimated to be 58.8 ± 0.2%, 22.7 ± 1.2%, and 18.5 ± 1.0%, respectively. Similar to other microalgae belonging to Chlorophyceae, the dominant amino acid and monosaccharide in Chlamydomonas sp. KIOST-1 were glutamic acid and glucose. On the other hand, the proportions of saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids clearly differed from other species in the genus Chlamydomonas, and monounsaturated fatty acids accounted for a large portion (41.3%) of the total fatty acids in the isolate. Based on these results, Chlamydomonas sp. KIOST-1 has advantageous characteristics for biomass production.
One of the biggest challenges for the animal feed industry in the coming years will be to meet the growing demand in animal protein in light of increased cost of feed ingredient as well as tougher restrictions on the use of antimicrobial growth promoters imposed by consumers and governments. A key focus area will be to maximise feed efficiency and minimise nutrient waste. It has been widely acknowledged that the composition of the intestinal microflora is closely related to intestinal health and performance of animals. Advanced microbial techniques have shown a close relationship between bacterial communities and their ability to modulate nutrient absorption and processing. In addition it has been recognised that modulating the immune response has significant impact on overall health as well as overall nutrient demand. Molecular techniques are a useful tool to gain an understanding of the impact of dietary interventions including the use of functional feed additives on specific changes in microbial communities or the immune system. Most these techniques however focus on the evaluation of large changes in bacterial compositions and often underestimate or neglect to recognise small changes in microbial diversity or behaviour changes without any measurable immune response. The key to understanding the relationship between specific nutritional intervention and the impact on health and performance lies in a deeper understanding of the impact of these nutrients on the expression of specific genes or specific metabolic pathways. The development of molecular tools as a result of developments in the field of Nutrigenomics has enabled researchers to study the effects of specific nutrients on the whole genome or in other words, the effect of thousands of genes simultaneously, and has opened a completely different avenue for nutritional research.
Most, if not all, Basidiomycetes mushrooms have biologically active polysaccharides showing potent antitumor activity with immunomodulating properties. These polysaccharides have various chemical compositions and belong primarily to the $\beta$-glucan group. In this study, the crude water-soluble polysaccharide HEF-P, which was obtained from the fruiting body of Hericium erinaceus by hot water extraction and ethanol precipitation, was fractionated by DEAE-cellulose and Sepharose CL-6B column chromatographies. This process resulted in four polysaccharide fractions, named HEF-NP Fr I, HEF-NP Fr II, HEF-AP Fr I, and HEF-AP Fr II. Of these fractions, HEF-AP Fr II was able to upregulate the functional events mediated by activated macrophages, such as production of nitric oxide and expression ofcytokines (IL-1${\beta}$ and TNF-${\alpha}$). The molecular mass of HEF-AP Fr II was estimated by gel filtration to be 13 kDa. Its structural characteristics were investigated by a combination of chemical and instrumental analyses, including methylation, reductive cleavage, acetylation, Fourier transform infrared spectroscopy (FT-IR), and gas chromatography-mass spectrometry (GC-MS). Results indicate that HEF-AP Fr II is a low-molecular-mass polysaccharide with a laminarin-like triple helix conformation of a ${\beta}$-1,3-branched-${\beta}$-1,6-glucan.

Ion implantation has been widely developed during the past decades to become a standard industrial tool. To comply with the growing needs in ion implantation, innovative technology for the control of ion beam parameters is required. Beam current, beam profile, ion fractions are of great interest when uniformity of the implant is an issue. Especially, it is important to measure the spatial distribution of beam power and also the energy distribution of accelerated ions. This energy distribution is influenced by the proportion of mass for ion in the plasma generator(ion source) and by charge exchange and dissociation within the accelerator structure and also by possible collective effects in the neutralizer which may affect the energy and divergence of ions. Hydrogen atom has been the object of a good study to investigate the energy distribution. Hydrogen ion sources typically produce multi-momentum beams consisting of atomic ion ($H^+$) and molecular ion ($H_2^+$ and $H_3^+$). In the beam injector, the molecular ions pass through a charge-exchanges gas cell and break up into atomic with one-half (from $H_2^+$) or one-third (from $H_3^+$) according to their accelerated energy. Burrell et al. have observed the Doppler shifted lines from incident $H^+$, $H_2^+$, and $H_3^+$ using a Doppler shift spectroscopy. Several authors have measured the proportion of mass for hydrogen ion and deuterium using an ion source equipped with a magnetic dipole filter. We developed an ion implanter with 50-KeV and 20-mA ion source and 100-keV accelerator tube, aiming at commercial uses. In order to measure the proportion of mass for ions, we designed a filter system which can be used to measure the ion fraction in any type of ion source. The hydrogen and helium ion species compositions are used a filter system with the two magnets configurations.
The rhamnan sulfat extracted from green algae seaweed, Monostroma nitidum was characterized on sugars, sulfate compositions and molecular structure. Rhamnan sulfate was extracted with boiling water, and purified with two steps of cetylpyridinium chloride and ion exchange chromatography. The yield of crude rhamnan sulfate was about 2% from raw material. Rhamnan sulfate fraction, F-4 was composed of 30% rhamnose, 0.9% arabinose, 2.5% xylose, 2% glucose and 32.6% sulfate. Rhamnan sulfate F-4-3 obtained from F-4 fraction was composed of 36.8% rhamnose, 3.6% xylose, 2.7% glucose, 1.4% galactose and 30.8% sulfate. The molecular weight of F-4-3 fraction was estimated as 10,000-10,300 dalton with Sephacryl S-200 gel filtration chromatography.
This study was performed to investigate the effects of high pressure/high temperature (HPHT) treatment on the recovery efficiency and characteristics of porcine placenta hydrolysates. The placenta hydrolysates were characterized by solubility, free amino acid contents, gel electrophoresis, gel permeation chromatography (GPC) and amino acid composition. Placenta was treated at 37.5 MPa of pressure combined with various temperatures (150, 170, and $200^{\circ}C$) or various holding times (0, 30, and 60 min at $170^{\circ}C$). Insoluble raw placenta collagen was partially solubilized (> 60% solubility) by the HPHT treatment. Free amino group content of placenta collagen was increased from 0.1 mM/g collagen to > 0.3 mM/g collagen after HPHT treatment, reflecting partial hydrolysis of collagen. The molecular weight ($M_w$) distribution showed evidence of collagen hydrolysis by shifting of $M_w$ peaks toward low molecular weight when treated temperature or holding time was increased. Alanine (Ala), glycine (Gly), hydroxyproline (Hyp), and proline (Pro) contents increased after the HPHT treatments compared to a decrease in the others. In particular, the increase in Gly was obvious, followed by Hyp and Pro, reflecting that placenta hydrolysates were mainly composed of these amino acids. However, increasing temperature or holding time hardly affected the amino acid compositions. These results indicate that the HPHT treatment is advantageous to hydrolyze collagen derived from animal by-products.
