Biological deinking process of used papers was studied by the polymer modified cellulase. Cellulase was modified with copolymers which consist of polyoxyethylene derivative and maleic anhydride(MA). The MA functional groups of copolymer can react with amino acids groups of the cellulase without much loss of activity. Modified degree of amino acids was controlled by the added copolymer. The maximum modified degree was about 60% and it was obtained when the weight ratio of copolymer and cellulase was 4. The remained activity of the maximum modified cellulase(MMC) was higher than 80% of native cellulase. The MMC's concentration was 0.05-2.0 wt% relative to the dry paper. In mechanical pulping process, cellulase enhanced the detachment of the ink particle from the used paper by partial hydrolysis of the fiber. The polyoxyethylene of modified cellulase produced the forms which can float the separated ink particle. Compared to the convention deinking method with NaOH or organic chemicals, the new biological deinnking process improved the physical properties such as freeness, tearing strength and whiteness.
In the study, we have obtained modified cellulase with higher cellulose degradation activity by molecular evolution method. Cellobiohydrolase(CBH I ) gene of Trichorderma reerri has been used. Cellulase mutant 228-G2 was selected and the activity of cellulase mutant 228-G2 was increased by 300% compared to original CBH I The 17 among 1542bases were found to be modified with mutant 228-G2.

Cellulase was modified with copolymer with polyethylene(PE)/polypropylene(PP) oxide and maleic anhydride(MA) by maleylation reaction, and modified cellulase was applied to the reprocessing of old newsprint (ONP). Cellulase of modified cellulase enhanced the detachment of ink particles by fibrillation of fiber. The copolymer, which acted as the surfactant formed bubbles and removed the ink particles in the floatation process. Modified cellulase showed the same deinking ability without excess dosage compared with the conventional method. And, it improved the physical properties including tensile strength, brightness, and whiteness compared with the conventional deinking process. The bond between the ink and fiber got stronger as the storage time increased, and it became very difficult to remove the ink particle. But, modified cellulase increased the deinking ability by 41% compared with the conventional process at the experiment of the ONP for 1 year storage time. It removed the yellowing and increased the whiteness and brightness as well as tensile strength and internal bond strength.

The effect of crosslinking on hand of the cellulase treated Tencel fabrics was investigated. Tencel fabrics were crosslinked with DMDHEU, mechanically prefibrillated, hydrolyzed by cellulase, and treated with softener. The treated fabrics were characterized by add-on, weight loss, DP rating, WRA, strength, SEM analysis and hand measurement. As DMDHEU concentration increased, weight loss of DMDHEU/cellulase treated fabrics decreased. However, cellulase treatment decreased DP properties and strength retention. Less fibrils were observed in the cellulase treated fabrics after DMDHEU treatment than the cellulase treated ones. It was confirmed that crosslinking with DMDHEU treatment was effective to control fibrillation. At 5% of DMDHEU concentration, DMDHEU/cellulase treated fabrics showed softer, smoother and bulkier hand compared with other treated fabrics. Among mechanical properties, bending and shearing properties were decreased progressively through DMDHEU, cellulase, and softener treatment. DMDHEU treatment contributed to impart resilience, cellulase treatment to bulkiness and softener treatment to smoothness. As the treatment of DMDHEU, cellulase, and softener progressed NUMREI, FUKURAMI, and THV increased with the exception of KOSHI.
Cellulase was modified with synthetic copolymers of polyoxyethylene derivative and maleic acid anhydride. The saccharification characteristics and enzymatic reaction kinetic mechanism of modified and native cellulases were observed. In modification reaction of cellulase, degree of modification(DM) increased, as mass ratio of copolymers to enzyme increased. Maximum DM was 55% at mass ratio of 4 and remained activity was 75%. In saccharification experiment modified enzyme had maintained higher stability than native enzyme over all the reaction and the final conversion yield of modified enzyme was greater than that of native enzyme. Numerical simulation based on the reaction mechanism considering enzymatic deactivation was performed. Modified enzyme had kept higher free enzyme concentration over all the reaction than that of native enzyme. Comparing calculation values with experimental data, calculation values were in accordance with experimental data.

To make a basepaper for multipurposed seed paper, old news print (ONP) and mixed office waste paper (MOW), modified and unmodified with a commercial cellulase, were investigated. Each handsheet was applied with different chemicals such as insecticides, germicides and herbicides. The interactions behaviors of chemicals used with base papers were evaluated by means of the contents of chemicals impregnation and dissolving behaviors in water and soil. The ONP and MOW treated with the cellulase had higher impregnation and dissolving capacities in both water and soil than untreated ONP and MOW. However, the modified ONP showed lower impregnation and dissolving capacities compared to the modified MOW. The content of impregnation of chemicals would be affected with the degree of microfibrils produced by the modification treatment. Otherwise, dissolving capacities of chemicals depend on the affinity of the residual lignin in the paper.
Although Energy demands of modern society increase rapidly, current energy would be exhausted shortly. Therefore development of bio-ethanol production process from cellulose containing materials was extremly demanded. Therefore development of highly functional cellulase is requisite for this purpose. In this study cellobio-hydrolase (CBH1) gene from Trichorderma reesei was used to increase cellulase activity by directed evolution and highly functional cellobio-hydrolase was obtained and characterized.

Cellulase and xylanase are main hydrolysis enzymes for the degradation of cellulosic and hemicellulosic biomass, respectively. In this study, our aim was to develop and test the efficacy of a rapid, high-throughput method to screen hydrolytic-enzyme-producing microbes. To accomplish this, we modified the 3,5-dinitrosalicylic acid (DNS) method for microwell plate-based screening. Targeted microbial samples were initially cultured on agar plates with both cellulose and xylan as substrates. Then, isolated colonies were subcultured in broth media containing yeast extract and either cellulose or xylan. The supernatants of the culture broth were tested with our modified DNS screening method in a 96-microwell plate, with a $200{\mu}l$ total reaction volume. In addition, the stability and reliability of glucose and xylose standards, which were used to determine the enzymatic activity, were studied at $100^{\circ}C$ for different time intervals in a dry oven. It was concluded that the minimum incubation time required for stable color development of the standard solution is 20 min. With this technique, we successfully screened 21 and 31 cellulase- and xylanase-producing strains, respectively, in a single experimental trial. Among the identified strains, 19 showed both cellulose and xylan hydrolyzing activities. These microbes can be applied to bioethanol production from cellulosic and hemicellulosic biomass.

The morphological optimization of Trichoderma harzianum was carried out using several surfactants to achieve increased cellulase production. Addition of the surfactants to the culture medium successfully modified the fungal morphology from an aggregated form to a dispersed form. Optimization of the fungal morphology increased cellulase activity up to 177%. The morphologically optimized conditions enhanced the accessibility of the fungus to substrates and thus promoted cellulase production.
Mycelial growth of ectomycorrhizal fungi (27 strains of 8 species) collected from Korean forests was observed on various culture conditions (media, temperature, pHs). After 60 days of incubation, all strains grown on potato dextrose agar (PDA) and modified Melin-Norkran's agar (MMNA), whereas no mycelial growth was observed on malt extract agar (MEA) or sabouraud dextrose agar (SDA) in some strains including Tricholoma matsutake. Mycelial growth on PDA was poor at high temperature ($30^{\circ}C$) than the low temperature ($10^{\circ}C$). The optimal temperature on PDA and pH in potato dextrose broth (PDB) for mycelial growth in most strains were $20-25^{\circ}C$ and pH 4-5, respectively. All strains tested showed the carboxymethyl cellulase (CM-cellulase) activity and the maximal cellullase activity was expressed by the mycelium of T. matsutake (KFRI 1266) on the CMC agar plate with pH 5.0.
