A Fermentation Strategy for Anti-MUC1 C595 Diabody Expression in Recombinant Escherichia Coli

  • The development of fermentation conditions for the production of C595 diabody fragment (dbFv) in E. coli HB2151 clone has been explored. Investigations were carried out to study the effect of carbon supplements over the expression period, the comparison of C595 dbfv production in synthetic and complex media, the influence of acetic acid upon antibody production, and comparison of one-stage and two-stage processes operated at batch or fed-batch modes in bioreactor. Yeast extract supplied during expression yielded more antibody fragment than any other carbon supply. The synthetic medium presented higher specific productivity (0.066 mg dbFv $g^{-1}$ dry cell weight) when compared to the complex medium (0.044 mg dbFv $g^{-1}$ DCW). The comparison of fermentation strategies demonstrated that (1) one-stage fed-batch fermentation performed higher C595 dbFv production than that operated in batch mode which was significantly affected by acetate concentration; (2) a two-stage batch operation could enhance C595 dbFv production. It was found that a concentration of 12.3 mg $L^{-1}$ broth of C595 dbFv and a cell concentration of 10.8g $L^{-1}$ broth were achieved at the end of two-stage operation in 5-L fermentation.

High-frequency Approximate Formulation for the Prediction of Broadband Noise of Airfoil Cascades with Inflow Turbulence (유입 난류에 의한 에어포일 캐스케이드 광대역 소음장의 고주파 근사 예측식의 개발)

  • This paper investigates the noise radiated by a cascade of flat-plate airfoils interacting with homogeneous, isotropic turbulence. At frequencies above the critical frequency, all wavenumber components of turbulence excite propagating cascade modes, and cascade effects are shown to be relatively weak. In this frequency range, acoustic power was shown to be approximately proportional to the number of blades. Based on this finding at high frequencies, an approximate expression is derived for the power spectrum that is valid above the critical frequency and which is in excellent agreement with the exact expression for the broadband power spectrum. The approximate expression shows explicitly that the acoustic Power above the critical frequency is proportional to the blade number, independent of the solidity, and varies with frequency as ${\phi}_{ww}(\omega/W$), where ${\phi}_{ww}$ is the wavenumber spectrum of the turbulence velocity and W is mean-flow speed. The formulation is used to perform a parametric study on the effects on the power spectrum of the blade number stagger angle, gap-chord ratio and Mach number. The theory is also shown to provide a close fit to the measured spectrum of rotor-stator interaction when the mean square turbulence velocity and length-scale are chosen appropriately.

  • The crossover in fashion refers to the coordination of different items, each with a different atmosphere. In a broad sense, it is the mutual exchange across the existing areas, that is, the creation of a new style by disparate and opposite elements coexisting in a look. This paper studies typical features of the crossover shown in modern fashion based on sub-culture by examining the concept and formation factors of the crossover through social aspects rather than particular art modes to predict the current fashion and suggest a new direction. Crossover fashion appeared in social and cultural environments, has four characteristics : the visual discordance of sexes, the appearance of the lingerie look, the mixture of folk style, and the appearance of the reactionary mode. The crossover, appeared in sub-cultures and diffused into the public, may be considered the creation of a new sense of beauty and a possibility for the future. It is expected to keep expanding ifs range in fashion, since it is an expression of human will to restore liberal sensibility not confined to formality.

  • It is becoming progressively more understandable that between transcription and translation there lies another versatile regulator that quantitatively controls the expression of mRNAs. Identification of miRNAs as key regulators of wide ranging signaling cascades and modulators of different cell-type and context dependent activities attracted basic and clinical scientists to study modes and mechanisms in details. In line with this approach overwhelmingly increasing in vivo and in vitro studies are deepening our understanding regarding miR-421, mir-155 and miR-650 mediated regulation of cellular activities. We also attempt to provide an overview of long non coding RNAs.

  • The formal constructions of deconstructive architecture in the contemporary fashion expressed as intertexruality, difference and trace and the working of DIS and DE, and intertextuality in deconstructive has manifested itself as the intertextuality of material, hybridization of items, the blending of modes and the bi-sexuality of sexes. And the difference and trace found in fashion are fantastic and playful and come to exist as historicity. DIS and DE in fashion shows the retrogression of gravity through non-structural silhouette. Their disentring expression emerged as construction through mixture and repetition as well as overlapping of planes or spaces. And their disjuctive representation appeared in the form of mutual juxtaposition and substitution with the double-side of formal construction in fuctional terms.

A Study on Dynamic Characteristics of Rotor with Flywheel (慣性車 를 가진 回轉子 의 動特性 에 관한 硏究)

  • The purpose of this study is to determine the general frequency expression for a rotating shaft of uniform cross section, supported by two bearings, and carrying flywheel at the free end. The bearing spacing and the ratio of the weight of flywheel to the total distributed weight are used as parameters. The data have thus been reduced to dimensionless form so that the results are generally applicable for this type of rotor. Frequencies for the first three modes of vibration are determined. Experimental investigation with rotor/flywheel model confirmed the critical speed frequencies lie between analytical models with simply supported-simply supported boundary conditions and spring supported-spring supported boundary conditions.

  • The propagation characteristics of dielectric waveguides has been analyzed by finite element method. We have proposed the finite element formutation of the variational expression in the three-component magnetic field based on Galerkin's method which seek for the propagation constant by a given value of frequency. In this approach, the divergence relation for H is satisfied and spurious modes does not appear and finite element solustions agree with the exact solutions. In order to varify the validity of the present method the numerical results for a rectangular waveguide partilly filled with dielectric are compared with other results.

Vibration Characteristics of CD and DVD Disks (CD 및 DVD 디스크의 진동 특성)

  • The aerodynamically excited vibration and natural frequency of rotating CD and DVD disks are analytically and experimentally studied in this paper The theoretical analysis uses a fluid-structure model where the aerodynamic effects are represented in terms of elastic, lift and damping components. The explicit expression on natural frequency of the air coupled disk is obtained as functions of the three aerodynamic coefficients. The experiments performed using a vacuum chamber and CD/DVD disks rotating in vacuum, open air and enclosure give three main results. One is that the aerodynamic effect by the surrounding air reduces the natural frequencies and critical speeds of the vibration modes. The second is that natural frequency of disks rotating in open air is larger than that in enclosure. Finally, it is shown that the disk vibration is reduced as the gap between the disk and the rigid wall decreases.

Using of Transfer Matrix for Shaft Element and Vibration Analysis (회전축요소의 전달행렬의 이용과 진동해석)

  • Based on the analytic expression for the elasto-dynamic behavior of rotating shaft, the transfer matrix is formulated for the shaft element with uniform cross-section. Timoshenko beam theory is Introduced for modeling the behavior of shaft. Complex variables representing the displacement, slope, moment and shear force are used for deriving the transfer matrix between both ends of the shaft element. Simulation result obtained by applying the transfer matrix to a general rotor model is compared with the reference result and proved to be exact. Natural frequencies and the corresponding modes are analyzed with varying the bearing: stiffness. The generally used bearings are considered for discussions. and the bearing stiffness is shown to affect the vibration characteristics of rotor.

  • Modern museums introduce a so-called method of 'exhibitions that approach' apart from the scheme of exhibitions in a simple listing type, creating a series of stories based on the original forms of remains put on display, adopting various methods of media access, and enabling the spectators of the museums to find exhibits a little more convincing and understand them more in depth, which might look somewhat isolated from their everyday lives. The configuration of the exhibitions that approach can easily be found in the mode of narrative development of exhibition topics and scenarios, and in this sense, a study on the narrativity of exhibitions is effective for analyzing the exhibition spaces of the museums. Furthermore, an analysis on exhibition spaces may be conducted through the process of forming the messages of exhibition contents and interpreting the narrative structures of the modes of development, and allows people to think that the methods of interpreting the spaces established like this may form an organic complementary relationship with exhibition contents and have a more extended meaning. Thus, this study examines the narrativity of Jeon-gok prehistory museum and the narrative structure systems based on the structuralist narrative theory, approaches the modes of narrative development of the spaces based on semiotic judgment, and aims to understand the structures of the space narrative. In addition, It is another object of the present invention in order to verify the objectivity, throughout the course of additional case studies, to improve the efficiency of future exhibition design.
