Effect of Iron Activators on the Persulfate Oxidation of Polycyclic Aromatic Hydrocarbons (PAHs) in Contaminated Soils (다환방향족 탄화수소(PAHs) 오염토양의 과황산 산화 시 철 활성화제의 영향)

  • PAHs commonly found in industrial sites such as manufactured gas plants (MGP) are potentially toxic, mutagenic and carcinogenic, and thus require immediate remediation. In-situ chemical oxidation (ISCO) is known as a highly efficient technology for soil and groundwater remediation. Among the several types of oxidants utilized in ISCO, persulfate has gained significant attention in recent years. Peroxydisulfate ion (S2O82-) is a strong oxidant with very high redox potential (E0 = 2.01 V). When mixed with Fe2+, it is capable of forming the sulfate radical (SO4) that has an even higher redox potential (E0 = 2.6 V). In this study, the influence of various iron activators on the persulfate oxidation of PAHs in contaminated soils was investigated. Several iron sources such as ferrous sulfate (FeSO4), ferrous sulfide (FeS) and zero-valent iron (Fe(0)) were tested as a persulfate activator. Acenaphthene (ANE), dibenzofuran (DBF) and fluorene (FLE) were selected as model compounds because they were the dominant PAHs found in the field-contaminated soil collected from a MGP site. Oxidation kinetics of these PAHs in an artificially contaminated soil and the PAH-contaminated field soil were investigated. For all soils, Fe(0) was the most effective iron activator. The maximum PAHs removal rate in Fe(0)-mediated reactions was 92.7% for ANE, 83.0% for FLE, and 59.3% for DBF in the artificially contaminated soil, while the removal rate of total PAHs was 72.7% in the field-contaminated soil. To promote the iron activator effect, the effects of hydroxylamine as a reducing agent on reduction of Fe3+ to Fe2+, and EDTA and pyrophosphate as chelating agents on iron stabilization in persulfate oxidation were also investigated. As hydroxylamine and chelating agents (EDTA, pyrophosphate) dosage increased, the individual PAH removal rate in the artificially contaminated soil and the total PAHs removal rate in the field-contaminated soil increased.

  • The bovine fatty acid binding protein 4 and 5 (FABP4 and 5) is a major positional and physiological candidate gene for the bovine marbling and carcass weight. The aim of this study was to evaluate the association between economic traits of Korean cattle (Hanwoo) and genetic variation in fatty acid binding protein 4 and 5 (FABP4 and 5) genes within carcass/meat quality traits and the before/after of fatting in breed Hanwoo. Here, we characterized the nucleotide polymorphism of FABP4 and 5 in 86 cattle. We were detected the variability of three types (GG, AG, and AA) by PCR, and economic traits were analyzed by the mixed regression model implemented in the ASReml program. As the result of statistical and supersonic analysis, FABP4 gene was highly showed significant effect (p<0.006) on marbling score (MS), in contrast FABP5 gene was lowed (p<0.084) on MS before fatting. But, FABP4 gene was highly showed significant effect (p<0.0054) on MS, in contrast FABP5 gene lowest (p<0.0899) on MS in the after of fatting. Compare to supersonic result before fatting in FABP4 gene, it was detected type GG: (p<7.18), AG: (p<8.50), and AA: (p<10.50) (n=50), showed type GG: (p<4.88), AG: (p<2.33), and AA: (p<0.00) after weed out (n=20). Futhermore, it was detected type GG: (p<9.30), AG: (p<7.95), and AA: (p<7.40) (n=50) before fatting in the FABP5 gene. It was shown type GG: (p<2.67), AG: (p<3.50), and AA: (p<5.00) after weed out (n=50). Our results indicate that FABP4 and 5 gene transcription is regulated by the environment of feeding and management, and suggest that feeding and management could be potential key in determining FABP4 and 5 genes transcription for carcass/meat quality traits in breed Hanwoo.

Association of UDP-galactose-4-epimerase with milk protein concentration in the Chinese Holstein population

  • Objective: An initial RNA-Sequencing study revealed that UDP-galactose-4-epimerase (GALE) was one of the most promising candidates for milk protein concentration in Chinese Holstein cattle. This enzyme catalyzes the interconversion of UDP-galactose and UDP-glucose, an important step in galactose catabolism. To further validate the genetic effect of GALE on milk protein traits, genetic variations were identified, and genotypes-phenotypes associations were performed. Methods: The entire coding region and the 5'-regulatory region (5'-UTR) of GALE were re-sequenced using pooled DNA of 17 unrelated sires. Association studies for five milk production traits were performed using a mixed linear animal model with a population encompassing 1,027 Chinese Holstein cows. Results: A total of three variants in GALE were identified, including two novel variants (g.2114 A>G and g.2037 G>A) in the 5'-UTR and one previously reported variant (g.3836 G>C) in an intron. All three single nucleotide polymorphisms (SNPs) were associated with milk yield (p<0.0001), fat yield (p = 0.0006 to <0.0001), protein yield (p = 0.0232 to <0.0001) and protein percentage (p<0.0001), while no significant associations were detected between the SNPs and fat percentage. A strong linkage disequilibrium (D' = 0.96 to 1.00) was observed among all three SNPs, and a 5 Kb haplotype block involving three main haplotypes with GAG, AGC, and AGG was formed. The results of haplotype association analyses were consistent with the results of single locus association analysis (p<0.0001). The phenotypic variance ratio above 3.00% was observed for milk protein yield that was explained by SNP-g.3836G >C. Conclusion: Overall, our findings provided new insights into the polymorphic variations in bovine GALE gene and their associations with milk protein concentration. The data indicate their potential uses for marker-assisted breeding or genetic selection schemes.

Effect of Experience, Education, Record Keeping, Labor and Decision Making on Monthly Milk Yield and Revenue of Dairy Farms Supported by a Private Organization in Central Thailand

  • The objective of this research was to assess the effect of experience, education, record keeping, labor, and decision making on monthly milk yield per farm (MYF), monthly milk yield per cow (MYC), monthly milk revenue per farm (MRF), and monthly revenue per cow (MRC) of dairy farms supported by a private organization in Central Thailand. The dataset contained 34,082 monthly milk yield and revenue records collected from January 2004 to December 2008 on 497 farms, and information on individual farmer experience and education, record keeping, and decision making obtained with a questionnaire. Farmer experience categories were i) no experience, ii) one year, iii) two to five years, iv) six to ten years, v) eleven to fifteen years, vi) sixteen to twenty years, and vii) more than twenty years. Farmer education categories were i) no education or primary school, ii) high school, and iii) bachelor or higher degree. Record keeping categories were: i) no records and ii) kept records. Labor categories were: i) family, ii) hired people, and iii) family and hired people. Decision making categories were: i) decisions made by farmers themselves, ii) decisions made with help from government officials, and iii) decisions made with help from organization staff. The mixed linear model contained the fixed effects of year-season, farm location-farm size subclass, experience, education, record keeping, labor, and decision making on sire selection, and the random effects of farm and residual. Results showed that longer experience increased (p<0.05) monthly milk yield (MYF and MYC) and revenue (MRF and MRC). Farms that hired people produced the highest (p<0.05) monthly milk yield (MYF and MYC) and revenue (MRF and MRC), followed by farms that used family, and the lowest values were for farms that used both family and hired people. Better educated farmers produced more MYC and MRC (p<0.05) than lower educated farmers. Farms that kept records had higher MYF and MRF (p<0.05) than those without records. Although differences among farms were non-significant, farms that received help from the organization staff had higher monthly milk yield (MYF and MYC) and revenue (MRF and MRC) than those that decided by themselves or with help from government officials. These findings suggested that dairy farmers needed systematic training and continuous support to improve farm milk production and revenues in a sustainable manner.

A Study on the Effect of Direct Government R&D Funding on SMEs: A Comparative Analysis between SMEs and Large Companies (정부의 중소기업에 대한 연구개발투자 효과: 대기업과의 비교분석을 통하여)

  • This study is intended to demonstrate how different factors induce the Korean government's direct investment in research and development according to the type of business enterprise in Korea. We analyzed that what factors made the government directly invest in research and development by using a total of 18,272 company panel data, which are not limited to specific industries or government-led research and development (R&D) programs. The results showed that the direct investment for R&D by the government led to the increase in the number of researchers in SMEs. For major companies, the direct investment resulted in the increase of sales and company's own R&D expenses. Moreover, this study found that government's direct support for R&D even led to the decrease of SMEs' own R&D expenses; however, this result was not statistically significant. In addition, the most significant factor to increase both SMEs' and major company's own R&D expenses was the sales amount of the company, rather than government's direct investment for R&D. The factor that increases sales was the company's own R&D expenses, rather than government's direct investment for R&D. Through the analysis using Mixed Effects Model, this study suggested the policy should be changed to make SMEs invest in its own R&D expenses, rather than to secure researchers of SMEs by government's direct investment for R&D.

Association between the Polymorphism of the Fatty acid binding protein 5 (FABP5) Gene within the BTA 14 QTL Region and Carcass/Meat Quality Traits in Hanwoo (한우 14번 염색체 QTL 영역내 Fatty acid binding protein 5 유전자의 다형성과 도체 및 육질 형질과의 관련성 분석)

  • The aim of this study was to evaluate the association between economic traits of Korean cattle (Hanwoo) and genetic variation in fatty acid binding protein 5 (FABP5) gene within QTL region of carcass weight and marbling score traits on BTA 14. We sequenced for detection of single nucleotide polymorphism (SNP) with 24 unrelated Hanwoo samples and identified four SNPs (-1141A>G, 949A>G, 969A>G and 1085C>G). Relationship between the genotypes of 583 Hanwoo individuals by PCR-RFLP and economic traits were analyzed by the mixed regression model implemented in the ASReml program. As the result of statistical analysis, SNP1 (-1141A>G) showed significant effect (p<0.003) on marbling score (MS) and SNP2 (949A>G) showed significant effect (p<0.034) on eye muscle area (EMA). Further studies are required to validate the significant SNPs in a bigger population, but the SNPs (-1141A>G and 949A>G) of FABP5 could be a genetic marker to estimate molecular breeding value (MEBV) for carcass traits in Hanwoo.

Effects of Three Lumbar Stabilization Exercises on the Thickness of Deep and Superficial Fibers of the Lumbar Multifidus (다열근 심부 및 표면 근섬유 두께에 대한 세 가지 요추안정화 운동의 효과)

  • The lumbar multifidus muscle, which can be separated into deep fascicles (DM) and superficial fascicles (SM), is important for lumbar segmental stability. However, no previous studies have investigated the effect of lumbar stabilization exercises on the thickness of DM and SM. Thus, the purpose of this study was to assess DM thickness after three different lumbar segmental stabilization exercises. In total, 30 healthy male participants were recruited and randomly assigned to one of three exercise groups: hollowing in the quadruped position (H-Quad), contralateral arm and leg lift (CALL), and bilateral arm and leg lift (BALL). Each lumbar segmental stabilization exercise was conducted over 4 weeks. Ultrasonography was used to compare the DM and SM thickness before and after the 4 weeks of exercise. A mixed-model analysis of variance using Scheffe's post-hoc test was used for statistical analysis. The results showed a significant effect for the measurement time (before vs. after 4 weeks of exercise) in the DM (F=31.26, p<.05) and SM (F=4.56, p<.05). At the end of the 4 weeks, the DM thickness had increased significantly in the H-Quad exercise group, and the SM thickness had increased significantly in the CALL and BALL exercise groups. Also in the BALL exercise group, the SM thickness was greater compared with that in the H-Quad exercise group. These findings suggest that the thickness of the DM and SM were increased by different types of lumbar segmental stability exercise after 4 weeks.

Essential oils as growth-promoting additives on performance, nutrient digestibility, cecal microbes, and serum metabolites of broiler chickens: a meta-analysis

  • Objective: The purpose of this meta-analysis was to evaluate the effect of dietary essential oils (EOs) on productive performance, nutrient digestibility, and serum metabolite profiles of broiler chickens and to compare their effectiveness as growth-promoting additives against antibiotics. Methods: Peer-reviewed articles were retrieved from Web of Science, Science Direct, PubMed, and Google scholar and selected based on pre-determined criteria. A total of 41 articles containing 55 experiments with 163 treatment units were eligible for analyses. Data were subjected to a meta-analysis based on mixed model methodology considering the doses of EOs as fixed effects and the different studies as random effects. Results: Results showed a linear increase (p<0.001) on body weight gain (BWG) where Antibiotics (FCR) and average daily feed intake decreased (p<0.001) linearly with an increasing dose of EOs. Positive effects were observed on the increased (p<0.01) digestibility of dry matter, crude protein, ether extract, and cecal Lactobacillus while Escherichia coli (E. coli) population in the cecum decreased (p<0.001) linearly. There was a quadratic effect on the weight of gizzard (p<0.01), spleen (p<0.05), bursa of fabricius (p<0.001), and liver (p<0.10) while carcass, abdominal fat, and pancreas increased (p<0.01) linearly. The dose of EOs linearly increased high density lipoprotein, glucose, protein, and globulin concentrations (p<0.01). In comparison to control and antibiotics, all type of EOs significantly reduced (p<0.001) FCR and tended to increase (p<0.1) BWG and final body weight. Cinnamaldehyde-compound was the only EOs type showing a tendency to increase (p<0.1) carcass weight, albumin, and protein of serum metabolites while this EOs together with EOs-Blend 1 decreased (p<0.01) E. coli population. Low density lipoprotein concentration decreased (p<0.05) with antibiotics and carvacrol-based compound when compared to the control group. Conclusion: This evidence confirms that EOs are suitable to be used as growth promoters and their economical benefit appears to be promising.

Protective Effect of Nitroglycerin on the Ischemia-Reperfusion Model of the Isolated Rat Lung (흰쥐의 분리 폐장 관류 모델에서 Nitroglycerin의 폐장 보존 효과)

  • Protection against ischemia-reperfusion injury is crucial for successful transplantation of the lung. It has been known that nitric oxide has many favorable effects on the donor lungs but at the same time, has some potential side effects of cytotoxicity. In this regards, we investigated whether the administration of nitroglycerin could decrease ischemia-reperfusion injury in isolated rat lung reperfusion model for the confirmation of the effect of nitroglycerin, a donor of nitric oxide, on lung transplantation. Material and Method: 35 Sprague-Dawley species male white rats were used for this experiment. For nitroglycerin group (n=18), nitroglycerin was administered intravenously followed by mixed in flushing solution for preservation. As a control group (n=17), we used the same amount of normal saline. To evaluate the effect of nitroglycerin on the lung, heart-lung block was obtained, weighed and stored in University of Wisconsin Solution at 1$0^{\circ}C$ for 24 hours. In each group of the isolated lungs, reperfusion was carried out with Krebs-Hensleit-diluted human blood for 60 minutes. As parameters of the state of the isolated lung, peak inspiratory and pulmonary arterial pressures were continuously recorded. Oxygen and carbon dioxide tension of reperfusing blood were measured before and after 30, 60 minutes of reperfusion. After sixty minutes of reperfusion, protein content in bronchoalveolar lavage fluid was measured also for the evaluation of the degree of alveolar flooding. Lung myeloperoxidase activity was determined to verify the accumulation of neutrophils. Results: Although statistically significant differences were not noted in peak inspiratory and pulmonary arterial pressure between control and nitroglycerin group, latter group showed lowering tendency of pulmonary arterial pressure during the entire reperfusion period. Oxygen tension was higher (p<0.05) in nitroglycerin group compared with that of the control group, in contrast, there were no differences in carbon dioxide tension, protein content in bronchoalveolar lavage fluid and myeloperoxidase activity between the groups. In the examination of ultrastructural changes, nitroglycerin denoted the protective effect on the pulmonary architecture compared with that of control group. Conclusion: Collectively, on the bases of these experimental results, prior treatment of donor lung with nitroglycerin could result in better preservation of the lung. Consequently, these nitroglycerin preserved lungs are thought to be more suitable for successful transplantation of the lung.

Application of 'Sponge Model' with Disinfectants for the Inhibition of Listeria monocytogenes (Listeria monocytogenes의 증식억제를 위한 살균제 'Sponge model'의 응용)

  • The antimicrobial effects of two disinfectants commonly used in food industry on Listeria monocytogenes ATCC 15313 were studied. The two disinfectants tested were commercial benzalkonium chloride (BAC) and sodium hypochlorite (NaOCl). Their effects were studied on cells suspended in disinfectants (in vitro) and in the sponge model with the disinfectants (in vivo). When cells were exposed to $0\~0.1\%$ BAC and $0\~150\;ppm$ NaOCL for 20 minutes, BAC and NaOCl concentration more than $0.25\%$ and 100 ppm showed the antimicrobial effects respectively. This organism decreased rapidly during the first $0.5\~1$ minute followed by a slower decrease during the rest of the exposure time. Fifteen ml of cell solution $(about\;10^7\;CFU/ml\;in\;the\;TSB)$ was mixed with 15 ml of disinfectants in the sponge $(6.0{\times}4.0{\times}4.0cm)$, BAC and NaOCl concentration more than $0.1\%$ and 300 ppm showed the antimicrobial effects, and at $0.25\%$ and 800 ppm diminished in cell numbers 3-log cycles during the first 20 minutes. In the case of sponge model, 15 ml of cell solution and 15 ml of disinfectants $(0.25\%\;of\;BAC,\;800\;ppm\;of\;NaOCl)$ were suspended in the sponge during 20 minutes, washing with 200 ml of sterilized distilled water, and this sponge was transfered in the 100 ml TBS, and then incubated at various temperature. The cells were increased about 1-log cycle during 24 hrs at $5\~15^{\circ}C$. And the others temperature, the cells growth was in proporation to storage tepmerature and the cells were about $10^9\;CFU/ml$ after $1\~3$ days incubations.
