Evaluation of Mechanical and Electrical Properties of Bipolar Plate Made of Fiber-reinforced Composites for PEM Fuel Cell (섬유강화 복합재를 사용한 PEM 연료전지 분리판의 전기적.기계적 특성 평가)

  • The fuel cell is one of promising environment-friendly energy sources for the next generation. The bipolar plate is a major component of the PEM fuel cell stack, which takes a large portion of stack cost. In this study, as alternative materials for bipolar plate of PEM fuel cells, graphite composites were fabricated by compression molding. Graphite particles mixed with epoxy resin were used as the main substance to provide electric conductivity To achieve desired electrical properties, specimens made with different mixing ratio, processing pressure and temperature were tested. To increase mechanical strength, one or two layers of woven carbon fabric were added to the graphite and resin composite. Thus, the composite material was consisted of three phases: graphite particles, carbon fabric, and epoxy resin. By increasing mixing ratio of graphite, fabricated pressure and process temperature, the electric conductivity of the composite was improved. The results of tensile test showed that the tensile strength of the two-phase graphite composite was about 4MPa, and that of three-phase composite was increased to 57MPa. As surface properties, contact an91e and surface roughness were tested. Graphite composites showed contact angles higher than $90^{\circ}$, which mean low surface energy. The average surface roughness of the composite specimens was $0.96{\mu}m$.

A Study on Isolation of BTEX Degrading Microorganism and Variation of BTEX Removal Efficiency and Microorganism Growth Rate According to Co-Culture (BTEX 분해미생물의 순수분리와 혼합 배양에 따른 기질 분해율 및 미생물 성장률 변화에 관한 연구)

  • The isolated microorganisms, Pseudomonas stutzeri, Raoultella planticola (Klebsiella), Serratia fonticola from petroleum contaminated soil were enriched on benzene, toluene, ethylbenzene, o-xylene as carbon and energy sources, respectively. And the degradation characteristics of BTEX was observed in the mixed BTEX substrates. We found that the BTEX in mixed substrates were degraded more than 50% by three isolated microorganisms. Among three isolated microorganisms, the highest degradation rate was observed in Pseudomonas stutzeri, but the degradation rate was different according to microorganisms. In order to increase the degradation efficiency, we applied the co-culture of isolated three microorganisms. The mixture rate of pseudomonas stutzeri : Raoultella planticola (Klebsiella) : Serratia fonticola was follows ; 1:2:1, 1:1:2, and 2:1:1, respectively. In two co-culture of 1:2:1 and 1:1:2, degradation rate was lower than isolated microorganisms. However, degradation rate became higher than isolated microorganisms and the degradation rate of benzene, toluene, and ethylene was more than 95% in co-culture of 2:1:1. The degradation rate increased through the co-culture of isolated microorganisms, however, the growth rate decreased. This was resulted from the substrate competition between microorganisms. The co-culture of microorganisms is a effective method to increase the degradation efficiency of BTEX and the co-culture mixing rate is a important factor for determination of degradation efficiency.

  • This paper describes the use of a discrete mathematical model to represent the basic mechanisms of regulation of the bacteria E. coli in batch fermentation. The specific phenomena studied were the changes in metabolism and genetic regulation when the bacteria use three different carbon substrates (glucose, glycerol, and acetate). The model correctly predicts the behavior of E. coli vis-a-vis substrate mixtures. In a mixture of glucose, glycerol, and acetate, it prefers glucose, then glycerol, and finally acetate. The model included 67 nodes; 28 were genes, 20 enzymes, and 19 regulators/biochemical compounds. The model represents both the genetic regulation and metabolic networks in an integrated form, which is how they function biologically. This is one of the first attempts to include both of these networks in one model. Previously, discrete mathematical models were used only to describe genetic regulation networks. The study of the network dynamics generated 8 $(2^3)$ fixed points, one for each nutrient configuration (substrate mixture) in the medium. The fixed points of the discrete model reflect the phenotypes described. Gene expression and the patterns of the metabolic fluxes generated are described accurately. The activation of the gene regulation network depends basically on the presence of glucose and glycerol. The model predicts the behavior when mixed carbon sources are utilized as well as when there is no carbon source present. Fictitious jokers (Joker1, Joker2, and Repressor SdhC) had to be created to control 12 genes whose regulation mechanism is unknown, since glycerol and glucose do not act directly on the genes. The approach presented in this paper is particularly useful to investigate potential unknown gene regulation mechanisms; such a novel approach can also be used to describe other gene regulation situations such as the comparison between non-recombinant and recombinant yeast strain, producing recombinant proteins, presently under investigation in our group.

A study on the denitrification and microbial community characteristics by the change of C/N ratio of molasses and nitrate nitrogen (당밀과 질산성 질소의 C/N ratio 변화에 따른 탈질 및 미생물 군집 특성에 관한 연구)

  • To compare the denitrification efficiency, this study used molasses and methanol were used as external carbon sources. Specific experimental conditions were classified according to C/N ratio conditions. The batch test showed that the denitrification efficiency increased as C/N ratios of molasses and methanol rose. The most suitable C/N ratio of molasses turned out 4:1 considering the concentration of the residue chemical oxygen demand (COD) and the denitrification efficiency, which was 91.4%. Specific denitrification rate (SDNR) drawn as a kinetic factor demonstrated that molasses and methanol showed similar SDNR values as C/N ratios of molasses and methanol increased. Under the condition of C/N ratio 4:1, 0.0292 g $NO_3{^-}-N$ removal/g mixed liquor volatile suspended solid (MLVSS)/day (molasses), 0.0299 g $NO_3{^-}-N$ removal/g MLVSS/day (methanol) were found. Sludge adapted to molasses showed that Bacterium Pseudomonas sp. and Bergeylla sp. dominated through an analysis of microbial community. In addition, some bacteria were high convergences than the variety of microbial community. Accordingly, it was assumed that molasses focus on growing microorganisms specialized in denitrification and applied as a replaceable external carbon source that can enhance denitrification performance.

Development of Process for Village Scale Wastewater Treatment Using Biofilter and Sulfur-limestone (바이오필터와 황-석회석을 이용한 마을하수 처리 공정 개발)

  • This process which has a connection of biofilter and sulfur-limestone has been developed to treat organic substances including BOD, COD and SS etc. and to treat sulfur-limestone is for denitrification.. The whole process consists of chemical reaction tank, sedimentation tank, trickling filter, denitrification tank The trickling filter is equipped with a reactor filled with absorptive filter, and the sulfur denitrification tank is filled with sulfur-limestone mixed media. After setting up practical facilities whose capacity is 60 tons a day, we have observed the removal efficiencies of pollutants through 60 experiments during Summer and Winter seasons. The average concentration of polluted water was BOD for 3.6 mg/L, $COD_{Mn}$ for 11.3 mg/L, SS for 2.8 mg/L, T-N for 8.6 mg/L, and T-P for 0.8 mg/L, and the rate of treatment efficiencies 96.5%, 84.7%, 96.5%, 79.2%, and 80.8%, respectively was found through the experiments. The average treatment efficiency for BOD and $COD_{Mn}$ was 85.0% and 55.7%, respectively and the average removal efficiency for NH4+-N was 84.9% in the trickling filter. The removal efficiency in the denitrification tank is as follows; The removal rate of $NO_3^--N$ was as high as 93.2% within the compass of pH 6.3 to 7.3 through $16.8{\sim}37.0mg/L$ flown into $NO_3^--N$ and $0.1{\sim}8.3mg/L$ outflown. It had observed that this process has implemented highly efficient and advanced treatment without external carbon sources and internal recycle during its process. In conclusion, this process is suitable for a sewerage in a small village due to the merits of low power consumption and easy maintenance.

Biological Nitrogen Removal Using a Single Biofilter Packed with Granular Sulfur (황 충진상 단일 생물막여과 공정을 이용한 생물학적 질소제거)

  • This study was performed to advance nitrogen removal efficiency by employing an single biofilter packed with granular sulfur, which consists of nitrification occurring at upper part and denitrification at lower part of the reactor. Continuos nitrification/denitrification was carried out with different alkalinity sources, which were $NaHCO_3$ and $CaCO_3$(limestone). In the downflow nitrification/denitrification biofilter packed with granular sulfur, first, terms for nitrogen removal was decided. As results, nitrification and denitrification rate with NaHCO3 at 0.85 kg $NH_4^+-N/m^3{\cdot}d$ were accomplished $0.80kg\;N/m_3{\cdot}d$, $0.43kg\;N/m^3{\cdot}d$, respectively. In the sulfur/limestone packed downflow nitrification/denitrification biofilter, sulfur and limestone were mixed packed, preliminary test showed sulfur/limestone mixing ratio was 3:1 and that was ideal. In the result, nitrification and denitrification rate at $0.7kg\;NH_4^+-N/m^3{\cdot}d$ were accomplished$0.65kg\;N/m^3{\cdot}d$, $0.34kg\;N/m^3{\cdot}d$, respectively. In general, employing granular sulfur can be implemented for only denitrification, but this system can accomplish nitrification as well as denitrification in a single reactor even though low carbon concentration was present in influent limiting to nutrient removal process. This biofilter system of limestone and granular sulfur packed together can successfully apply for nutrient removal effectively.

Dissolved Organic Matter (DOM) Leaching from Microplastics under UV-Irradiation and Its Fluorescence P roperties: Comparison with Natural P articles (UV 광풍화에 의한 미세플라스틱 기원 유기물 용출과 형광 특성: 자연유래 유기성 입자와의 비교)

  • Numerous studies have investigated the occurrence and fate of microplastics in the environment; however, only limited effort has been devoted to exploring the characteristics of dissolved organic matter (DOM) leached from microplastics. In microplastic (MP)-contaminated environment, MPs are typically mixed with naturally-occurring particles, which interferes with their detection in the environment. Thus, it is necessary to distinguish between the DOM leached from MPs and those leached from natural particles and also to characterize their properties. This study investigated DOM leaching behavior from MPs (polystyrene: PS, polyvinylchloride: PVC) and natural particulates (forest soil: FS, litter leaves: LL) under light, which is considered one of the main weathering processes that affect MPs in the environment. The leached DOM concentrations and fluorescence characteristics were compared under dark versus light conditions. Regardless of the origins, UV light promoted DOM release from all the particulates. More DOM was released from natural particles than from MPs under both conditions. However, the effect of promoting DOM release by UV was more pronounced for MPs than for natural particles. It was observed from fluorescence spectra that the intensity of the humic-like region was substantially reduced when MP-derived DOM was exposed to UV light, whereas the change of intensity was very little for natural particles. Under light conditions, the ratio of protein-like to humic-like fluorescence of MP-derived DOM was higher than that of DOM from natural particles. This study implies that a substantial amount of DOM could be leached from MPs even in MP-polluted environment under UV irradiation. Protein/humic fluorescence ratio could be utilized as a fast probing indicator to separate the two sources of particles under light.

Condition of ex situ Bioremediation of Polycyclic Aromatic Hydrocarbons in Marine Sediments (해양퇴적토내 다환방향족탄화수소 생분해 증진 조건 연구)

  • Polycyclic aromatic hydrocarbons (PAHs) are a kind of toxic environmental pollutants and has been accumulated usually in marine sediments. Due to their potential hazardous to human, removal of PAHs from environments has been great concern. In the present study, the effect of microbial inoculation and the supplementation of mixed form cyclodextrin (M-CD) was assessed in the pre-sterilized or nonsterilized microcosms for optimizing operational conditions for ex situ bioremediation of sediments contaminated by PAHs. Activity of electron transport system (ETSA) was increased by the addition of M-CD regardless of inoculation of microorganisms in microcosms without sterilization. The degradation rate of PAHs in sterilized microcosms was app. 9-20% by the inoculation of single strain and 24-37% by the inoculation of microbial consortium supplemented with 1% M-CD, respectively. The degradation was not observed in microcosms without sterilization under the same conditions. The proportion of inoculated microorganisms also decreased in nonsterilized microcosms. Signals of inoculated bacteria were decreased to detection limit after 2 days in the microcosms without M-CD. In conclusion, microbial inoculation with appropriate carbon sources and removal of natural flora and grazers are required for the efficient ex situ bioremediation of sediments contaminated by PAHs in bioslurry reactor.

Cometabolism of $\omega$-Phenylalkanoic Acids with Butyric Acid for Efficient Production of Aromatic Polyesters in Pseudomonas putida BM01

  • Poly(3-hydroxy-5-phenylvalerate) [P(3HPV)] was efficiently accumulated from 5-phenylvalerate (5PV) in Pseudomonas putida BM01 in a mineral salts medium containing butyric acid (BA) as the cosubstrate. A nove aromatic copolyester, poly(5 mol% 3-hydroxy-4-phenylbutyrate-co- 95 mol% 3-hydroxy-6-phenylhexanoate) [P(3HPB-co-3HPC)] was also synthesized from 6-phenylhexanoate (6PC) plus Ba. The two aromatic polymers, P(3HPV) and P(3HPB-co-3HPC), were found to be amorphous and showed different glass-transition temperatures at $15^{\circ}C$ and $10^{\circ}C$, respectively. When the bacterium was grown ina medium containing 20 mM 5PV as the sole carbon source for 140 h, 0.4 g/l of dry cells was obtained in a flask cultivation and 20 wt% of P(3HPV) homopolymer was accumulated in the cells. However, when it was grown with a mixture of 2 mM 5PV and 50 mM BA for 40 h, the yield of dry biomass was increased up to 2.5 g/l and the content of P(3HPV) in the dry cells was optimally 56 wt%. This efficient production of P(3HPV) homopolymer from the mixed substrate was feasible because BA only supported cell growth and did not induce any aliphatic PHA accumulation. The metabolites released into the PHA synthesis medium were analyzed using GC or GC/MS. Two $\beta$-oxidation derivatives, 3-phenylpropionic acid and trans-cinnamic acid, were found in the 5V-grown cell medium and these comprised 55-88 mol% of the 5PV consumed. In the 6PC-grown medium containing Ba, seven ${\beta}$-oxidation and related intermediates were found, which included phenylacetic acid, 4-phenylbutyric acid, cis-4-phenyl-2-butenoic acid, trans-4-phenyl-3-butenoic acid, trans-4-phenyl-2-butenoic acid, 3-hydroxy-4-phenylbutyric acid, and 3-hydroxy-6-phenylhexanoic acid. Accordingly, based on the metabolite analysis, PHA synthesis pathways from the two aromatic carbon sources are suggested.

Effects of Various Light Sources on the Carotenoid and Glucosinolate Contents in Chinese Cabbage (Brassica rapa L. ssp. pekinensis) (다양한 광원이 배추 내 Carotenoid와 Glucosinolate 함량에 미치는 영향)

  • BACKGROUND: Chinese cabbage biosynthesizes various phytochemicals including carotenoids and glucosinolates. Environmental stress has a major effect on the growth and yields of vegetables, and can significantly affect nutritionally important phytochemicals. Phytochemicals of plants are influenced by light, temperature, carbon dioxide, and growing conditions. The aim of this study was to investigate the effect of various light sources on carotenoid and glucosinolate contents in Chinese cabbage. METHODS AND RESULTS: [Experiment I] Set the control (field control, FC) on the ground. Using acrylic sunlight, experiments were set up transparency box (field transparency, FT), red box (field red, FR) and blue box (field blue, FB). [Experiment II] Set the control (chamber control, CC) in the greenhouse. Using plant growth chamber with artificial light, experiments were set up LED red (chamber red, CR), LED blue (chamber blue, CB), LED mixed red+blue (chamber red+blue, CRB) and fluorescent (chamber fluorescent, CF). After plant growth, Chinese cabbage was harvested at 110 days after sowing (DAS). The status of plants growth (leaf length, width, fresh weight etc.) was immediately investigated. Carotenoid and GSL contents were analyzed by HPLC. [Experiment I] Results documented that the ranges of total carotenoid contents were 25.39 ~ 58.80 mg/kg dry wt for lutein, 0.84~ 4.22 mg/kg dry wt for zeaxanthin, and 3.85~18.71 mg/kg dry wt for ${\beta}$-carotene. Lutein was the highest for the content and the largest for the variation as well. [Experiment II] Results documented that the ranges of total carotenoid contents were 24.66~137.96 for lutein, 2.51~20.65 for zeaxanthin, and 8.40~49.80 mg/kg dry wt for ${\beta}$-carotene. The total carotenoid contents of CR (156.62) and CB (115.90) were 1.6~2.3 times larger than the other treatments, and ${\beta}$-carotene content was about twice as high as that of the other treatments on the CR (38.74 mg/kg dry wt.). [Experiment I] Total GSL content was the highest in FT (19.76) that was higher 1.7 times than the lowest treatment ($11.39{\mu}mol/g\;dry\;wt$.). [Experiment II] The total content of GSL was highest in CRB (4.19) and lowest in CF ($2.88{\mu}mol/g\;dry\;wt$.). In the CRB, total GSL contents ($4.19{\mu}mol/g\;dry\;wt$.) was the highest. CONCLUSION: Total and individual carotenoid and GSL contents in Chinese cabbage show significant differences under different light sources. Red and blue lights contribute to significant carotenoids expression and antioxidant activity for nutrition and health benefits. These results concluded that the introduction of varying lights affected the synthesis of important nutrient compounds in Chinese cabbage. It is predicted that the application of good light source enhances the accumulation of functional compounds.
