Synthesis of 18F-labeled Novel Phosphonium cations as PET Myocardial Perfusion Imaging Agents: Pilot Imaging Studies

  • The development of myocardial perfusion imaging (MPI) agents has been motivated because coronary artery disease has been one of the leading causes of death worldwide since the 1960s. Several positron emission tomography (PET) MPI agents were developed, and 18F-labeled phosphonium cations were reported actively among them. In this study, we synthesized novel 18F-labeled phosphonium cations, (5-[18F]fluoropentyl)diphenyl(pyridin-2-yl)phosphonium and (2-(2-[18F]fluoroethoxy)ethyl)diphenyl(pyridin-2-yl)phosphonium, and evaluated potential as MPI agents. Two labeled compounds were synthesized via nucleophilic substitution reactions of 18F-fluoride with the appropriate tosylate precursor in the presence of Kryptofix 2.2.2 and K2CO3. MicroPET studies were performed in normal rats to evaluate in vivo distribution of radiolabeled phosphonium cations for 60 min. The radiolabeled compounds were synthesized with 5%-10% yield. The radiochemical purity of labeled compounds was > 98% by analytical HPLC, and the specific activity was > 11.8 GBq/µmol. The result of microPET studies of these labeled compounds in rats showed intense uptake in the myocardium at 30 and 60 min. The results suggest that these 18F-labeled novel phosphonium cations would have potential as promising candidates for myocardial perfusion imaging.

Auranofin accelerates spermidine-induced apoptosis via reactive oxygen species generation and suppression of PI3K/Akt signaling pathway in hepatocellular carcinoma

  • Auranofin is a US Food and Drug Administration (FDA)-approved anti-arthritis medication that functions as a thioredoxin reductase inhibitor. Spermidine, a polyamine present in marine algae, can exert various physiological functions. Herein, we examined the synergistic anticancer activity of auranofin and spermidine in hepatocellular carcinoma (HCC). Combined treatment with auranofin and spermidine suppressed cell viability more efficiently than either treatment alone in HCC Hep3B cells. The isobologram plotted by calculating the half maximal inhibitory concentration (IC50) values of each drug indicated that the two drugs exhibited a synergistic effect. Based on the analysis of annexin V and cell cycle distribution, auranofin and spermidine markedly induced apoptosis in Hep3B cells. Moreover, auranofin and spermidine increased mitochondria-mediated apoptosis by promoting mitochondrial membrane potential (Δψm) loss. Auranofin and spermidine significantly increased reactive oxygen species (ROS) production in Hep3B cells, and the blocking ROS suppressed apoptosis induced by spermidine and auranofin. In addition, auranofin and spermidine reduced the expression of phosphorylated phosphatidylinositol-3 kinase (PI3K) and protein kinase B (Akt), and PI3K inhibitor accelerated auranofin- and spermidine-induced apoptosis. Using ROS scavenger and PI3K inhibitor, we revealed that ROS acts upstream of auranofin- and spermidine-induced apoptosis. Collectively, our study suggests that combination treatment with auranofin and spermidine could afford synergistic anticancer activity via ROS overproduction and reduced PI3K/Akt signaling pathway.

First Record of Bothus pantherinus (Bothidae, Pleuronectiformes) from Korea (한국산 둥글넙치과(Bothidae) 어류, Bothus pantherinus의 첫기록)

  • A single specimen of the genus Bothus (family Bothidae) was collected for the first time at the intertidal zone of Moseulpo Port, Daejeong-eup, Seogwipo-si, Jejudo Island, on 14 August 2022. The specimen was identified as Bothus pantherinus in having following morphological traits: dark spots and ring-shaped patterns near eyes and the pectoral fins, one distinct spot in the center of the lateral line, 75 lateral line scales, and seven hourglass shaped supracranial pterygiophores. As a result of analyzing 603 bp of mitochondrial DNA COI sequences, our specimen was perfectly matched to those of B. pantherinus registered in NCBI. It has been known that the species is widely distributed throughout the Indo-Pacific Ocean from Red Sea to Hawaiian (32 degrees north to 32 degrees south), but this study revealed that its distribution expanded to the waters of Jejudo Island (33 degrees north), Korea. We propose its new Korean name "Beot-kkoch-mu-nui-dung-geul-neob-chi".

First Record of the Eared Blacksmelt, Lipolagus ochotensis (Bathylagidae, Osmeriformes) Larvae from the Southern Coastal Waters of Jejudo Island, Korea (제주도 남부해역에서 채집된 Bathylagidae (바다빙어목) Lipolagus ochotensis 자어의 한국 첫기록)

  • Four individuals of Lipolagus ochotensis larvae (13.4~21.3 mm SL), belong to the family Bathylagidae, were collected by a Bongo net from the southern waters off Jejudo Island, Korea in February to March 2018. L. ochotensis is characterized by a elongated and compressed body, the eye stalks, series of melanophores on posterior of body, dorsal fin origin above the middle of the body. A molecular analysis based on 625 base pairs sequences in the mitochondrial DNA cytochrome c oxidase subunit I region shows that specimens were closely matched to adult L. ochotensis (genetic distance=0.024). We report the first record of family Bathylagidae, genus Lipolagus, L. ochotensis in Korean waters, and suggest their new Korean names, "Sim-hae-bing-eo-gwa", "Geom-eun-bing-eo-sok", and "Geom-eun-bbyam-bing-eo", respectively.

  • This is the third series of catalogs reporting on Korean species discovered through the 'Discovery of Korean Indigenous Species'. This catalog includes 22 species of invertebrates, excluding insects. The catalog includes the scientific name, an abridged list of synonyms, collection sites, distribution, diagnosis, and figures for each species. Additionally, we provide the newly assigned Korean name, specimen voucher, and, if available, mitochondrial CO1 or 16S gene sequences of the species listed. All species identified and documented here will be officially listed on the 'National Species List of Korea', a database maintained by the National Institute of Biological Resources(NIBR).

Changes of the Ultrastructure and $Ca^{2+}$ Distribution after Transient Ischemia and after Reperfusion in the Myocardial Cells of Isolated Perfused Guinea Pig Hearts (일과성 허혈 및 허혈후 재관류가 기니픽 심실심근세포의 미세구조 및 칼슘 분포에 미치는 영향에 관한 연구)

  • It has been debated whether postischemic reperfusion is necessarily beneficial to salvage the myocardium after ischemic insult or not. Therefore, this study was undertaken to compare the ultrastructural changes as well as the distribution of $Ca^{2+}$ in the ventricular myocardial cells after transient ischemia and after postischemic reperfusion, and to suspect to what extent the postischemic reperfusion is beneficial. After 10 minutes of ischemia, the heart developed wide I bands, glycogen depletion, intramyofibrillar edema, mitochondrial swelling, clumping and migration of chromatin, ghosts of lipid droplets, disintegration of cell junctions, sarcolemmal disruption, and loss of $Ca^{2+}$ binding capacity of the sarcolemma and the mitochondria. In spite of reperfusion, in a large number of cells, the ultrastructure was more severely damaged, however, $Ca^{2+}$ binding capacity of the sarcolemma and the mitochondria restored. These results suggest that postischemic reperfusion may help the myocardial cells to restore their function to control $Ca^{2+}$ to a certain extent, but that it could aggravate the ischemic insult.

Effects of epigallocatechin-3-gallate on bovine oocytes matured in vitro

  • Objective: Epigallocatechin-3-gallate (EGCG) is a major ingredient of catechin polyphenols and is considered one of the most promising bioactive compounds in green tea because of its strong antioxidant properties. However, the protective role of EGCG in bovine oocyte in vitro maturation (IVM) has not been investigated. Therefore, we aimed to study the effects of EGCG on IVM of bovine oocytes. Methods: Bovine oocytes were treated with different concentrations of EGCG (0, 25, 50, 100, and $200{\mu}M$), and the nuclear and cytoplasmic maturation, cumulus cell expansion, intracellular reactive oxygen species (ROS) levels, total antioxidant capacity, the early apoptosis and the developmental competence of in vitro fertilized embryos were measured. The mRNA abundances of antioxidant genes (nuclear factor erythriod-2 related factor 2 [NRF2], superoxide dismutase 1 [SOD1], catalase [CAT], and glutathione peroxidase 4 [GPX4]) in matured bovine oocytes were also quantified. Results: Nuclear maturation which is characterized by first polar body extrusion, and cytoplasmic maturation characterized by peripheral and cortical distribution of cortical granules and homogeneous mitochondrial distribution were significantly improved in the $50{\mu}M$ EGCG-treated group compared with the control group. Adding $50{\mu}M$ EGCG to the maturation medium significantly increased the cumulus cell expansion index and upregulated the mRNA levels of cumulus cell expansion-related genes (hyaluronan synthase 2, tumor necrosis factor alpha induced protein 6, pentraxin 3, and prostaglandin 2). Both the intracellular ROS level and the early apoptotic rate of matured oocytes were significantly decreased in the $50{\mu}M$ EGCG group, and the total antioxidant ability was markedly enhanced. Additionally, both the cleavage and blastocyst rates were significantly higher in the $50{\mu}M$ EGCG-treated oocytes after in vitro fertilization than in the control oocytes. The mRNA abundance of NRF2, SOD1, CAT, and GPX4 were significantly increased in the $50{\mu}M$ EGCG-treated oocytes. Conclusion: In conclusion, $50{\mu}M$ EGCG can improve the bovine oocyte maturation, and the protective role of EGCG may be correlated with its antioxidative property.

  • Cytochrome-c-oxidase in mitochondria membrane is one of the most important factors for energy generation in the cell. As well as it is electron transfer enzyme, it is also heavily related to the apoptosis and other pathologic conditions. Meanwhile, porin is a protein located in inner and outer membranes of mitochondria, which is assumed to be functionally correlated with cytochrome-c-oxidase. It functions as forming electron transfer chain and conveying ATP. Therefore, using the immune-microscopy, It compared the distribution of cytochrome-c-oxidase and porin to figure out the formation and changes on cytochrome-c-oxidase in mitochondrial cristae. The sarcroplasm of cardic muscle tissue has many mitochondria. They are classified into two groups: the mitochondria with many cytochrome-c-oxidase and the mitochondria with only porins. The mitochondria with porins had few cytochrome-c-oxidases in their membrane; in contrast, the other mitochondria with rich cytochrome-c-oxidase had few porins in their walls. In addition, according to the location of the tissue in bovine heart, distribution of those kind of mitochondria had been clearly separated. As a result, it could be assumed that immature mitochondria has many porins to transfer the protein materials from sarcroplasm through the porins, and they made cytochrome-c-oxidase until it is enough, and then they decreased the porin and maintained minimum number of the porin.

College Students' Attitude toward Body Weight Control, Health-related Lifestyle and Dietary Behavior by Self-perception on Body Image and Obesity Index (대학생의 체형인식과 비만도에 따른 체중조절 태도, 건강관련 생활습관 및 식행동)

  • The Purpose of this study was to investigate college students' attitude toward body weight control, health-related lifestyle and dietary behavior by their perception on body image and obesity index. The subjects were 871 nation- wide college students (330 male and 541 female students) participating in a nutritional education program via internet- A cross-sectional survey was conducted using a self-administered questionnaire via internet. The data were analyzed by a SPSS 10.0 program. Height and weight (p < 0.05), and the distribution of obesity index (p < 0.001) showed significant differences between male and female college students. As for self perception on body image, the necessity of body weight control and the experience of efforts to change the body weight, there were significant differences between male and female college students (p < 0.001). There were significant differences among groups by obesity index in the hours of exercise (p < 0.01), drinking (p < 0.01) and smoking (P < 0.001). As for methods of body weight control, all the groups by obesity index thought that program of body weight control by diet company is the most desirable method. Therefore, we must recognize that it is hard to correct health problems resulting from wrong lifestyle and dietary behavior fixed during college life and prepare a system for college students to obtain correct lifestyle and dietary behavior.

Detection and Differentiation of Intentional and Unintentional Mixture in Raw Meats Using Real-time PCR (Real-time PCR을 이용한 식육원료의 의도적, 비의도적 혼입 판별법 개발)

  • In this study, the detection method was developed using real-time PCR to distinguish 4 species (bovine, porcine, horse, and chicken) of raw meats. The genes for distinction of species about meats targeted at 12S rRNA and 16S rRNA parts in mitochondrial DNA. Probes were designed to have a 5' FAM and a TAMRA at the 3' end. This study is to develop 4 species-specific primer and probes about raw materials and real-time PCR on 10 strains to observe the products of non-specific signal for similar species. As a result, any non-specific signal were not detected among each other. Real-time PCR method was developed for quantitation and identification of intentional and unintentional mixture in ground mixed meat (The difference of $C_T$ value between intentional mixture and 100% meat: $${\leq_-}$$ cycles, The difference of $C_T$ value between unintentional mixture and 100% meat: $${\geq_-}$$ cycles). The detection and differentiation of intentional and unintentional mixture in this study would be applied to food safety management for eradication of adulterated food distribution and protection of consumer's right.
