The Structure of Visual Cells in the Retina of the Pond Loach, Misgurnus anguillicaudatus (Pisces; Cobitidae)

  • A histological study on the retina of Misgurnus anguillicaudatus was carried out by light microscopy and scanning electron microscopy as part of getting information about relation between its habitat and visual cells. The visual cells of the retina in M. anguillicaudatus, a bottom-dwelling freshwater pond loach in stagnant or slow waters such as swamps, reservoirs and paddy fields, consists of double cones and large rods. The cones form a row mosaic pattern in which the partners of double cones are linearly oriented with a large rod. In a double cone, the two members are unequal such that one cone may be longer than the other.

  • This study was carried out to clarify the egg, larvae and juveniles development of Misgurnus anguillicaudatus, and relationships of M. anguillicaudatus and M. mizolepis, Cobitididae Fishes. The adult fishes were collected in Samsan-cheon, Haenam-gun, Jeollanam-do, Korea and their spawning inducement was carried by ovaprim injections. The egg shape was circular and the size was average 1.12 mm. The eggs were hatched at 61 to 72 h after fertilization. The newly hatched larvae had an average 3.23 mm in total length (TL). At 5 days after hatching, the larvae reached to post larval stage and they were 10.3 mm in TL. At 19 days after hatching, it reached to juvenile stage and was 25.3 mm in TL. The egg size of M. anguillicaudatus was almost same as M. mizolepis but the hatching period of M. anguillicaudatus has taken longer. It was possible for interspecific distinguishability of M. anguillicaudatus and M. mizolepis when their larvae reached to juvenile stage by the development of keel-like ridges.

  • 미꾸리(Misgurnus anguillicaudatus)에 대한 4가지 농약(diazinon, carbaryl, fenpropathrin, myclobutanil)의 급성독성을 평가하기 위하여, 치어 및 자어 단계(postlarva I, postlarva II, 8 WPH(weeks posthatch), 16 WPH, 20 WPH, 32 WPH, 40 WPH)의 미꾸리를 사용하여 급성독성실험을 실시하였다. 각 생육단계 별로 구해진 각 실험농약의 96-hr $LC_{50}$값을 비교하여 미꾸리에 대한 급성독성이 높은 순서를 정리한 결과는 Fenproparthri>Diazinon>Carbaryl, Myclobutanii 순으로 나타났다. 또한, 본 연구에서 사용된 4가지 농약의 독성은 미꾸리의 생육단계에 따라 차이를 나타냈다. 농약에 대한 감수성이 민감한 미꾸리의 생육단계 순으로 정리하면 Postlarva I>Postlarva II, 8 WPH>16 WPH, 20 WPH, 32 WPH, 40 WPH 나타났다. 그러므로 환경관리 및 규제목적으로 미꾸리 성체를 사용하여 독성실험을 할 경우에는 독성을 과소평가 할 수도 있음을 보여주었다.

Changes of Morphometric Traits in the Cultured Cyprinid Loach (Misgurnus anguillicaudatus) to Starvation

  • The experiment was conducted to investigate the effects of various dietary conditions on external morphometric traits, and sectioned morphometric traits in the cultured cyprinid loach, (Misgurnus anguillicaudatus) for 28 days. For the fed group there was an increase in body weight, standard length, and morphometric dimensions of the head and body cavity regions compared with the starved and initial groups. Sectioned morphometric trait analysis revealed that relative to the starved group, the fed group had greater body circumference, cross-sectional area, and total height (P<0.05). Our results provide data on external and sectioned morphometric changes under starvation conditions, and can be used as a guide to assist in the regulation and scheduling of feeding, and as indices of the nutritional status of cyprinid loach.

The Effects of Starvation on Physiological Changes and Stress Response in Cultured Cobitid Loach (Misgurnus anguillicaudatus) Exposed to Sodium Nitrite

  • To investigate effects of starvation on physiological changes, stress response, and survival of cobitid loach (Misgurnus anguillicaudatus) exposed to sodium nitrite (NaNO2), a 4-week experiment was conducted. Fewer fish survived in the starved group than those in the fed group during the experiment. Starvation resulted in growth retardation, leading to differences in body length and body depth between fed and starved groups. The fed gorup continued to grow and remained in good condition. Blood chemical analysis (plasma cortisol and glucose) showed significant differences in stress response to nitrite exposure between fed and starved groups (p < 0.05). These results suggest that all parameters employed in this study to assess effects of starvation with NaNO2 stress are useful information for researching nutritional status in cobitid loach.

  • BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oriza sativa and Streptomyces hygroscopicus into the genome of rice (Nakdongbyeo). With the potential problems of safety, the non-target organism evaluation is required as an essential element for the environmental risk assessment of genetically modified (GM) crops. In present study, we studied the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. METHODS AND RESULTS: The M. anguillicaudatus and C. carpio were fed on disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice (Nakdongbyeo) to 0, 10, 100, 1,000 and 5,000 mg/L, as treatment concentration respectively. The OsCK1 rice used for the test was confirmed to have the OsCK1/PAT gene expression by the PCR and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between OsCK1 rice and non-GM rice. The 96hr-$LC_{50}$ values showed no difference between OsCK1 rice (>5,000 mg/L) and non-GM rice (>5,000 mg/L). CONCLUSION(S): The results of this study suggested that there was no significant difference in toxicity for M. anguillicaudatus and C. carpio between OsCK1 rice and non-GM counterparts.

  • We developed insect-resistant GM rice(Bt transgenic rice) by inserting the mCry1Ac1 a modified gene from the soil bacterium, Bacillus thuringiensis. The Bt transgenic rice expressing the Bttoxin mCry1Ac1 was tested for the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. M. anguillicaudatus and C. carpio fed 100% ground rice in suspension, using either Bt rice or non-GM counterpart rice(Nakdong). The Bt rice used for the test were confirmed to have the mCry1Ac1 gene expression by the immuno-strip and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between Bt rice and non-GM counterpart rice. The 96hr-LC50 values showed no difference between Bt rice(>1,000mg/L) and non-GM rice(>1,000mg/L). We concluded that there was no significant difference in toxicity for non-target organisms(M. anguillicaudatus and C. carpio) between Bt rice and non-GM counterparts.

Genetic Species Identification by Sequencing Analysis of Nuclear and Mitochondrial Genes for Albino Misgurnus Species from Korea (우리나라 미꾸리속(genus Misgurnus) 알비노 개체의 미토콘드리아 및 핵 유전자 염기서열 분석에 의한 유전적 동정)

  • The spontaneous color mutant, albino individuals of genus Misgurnus, are rarely discovered in Korea and there are difficult to identify morphological species due to lack melanin pigmentation. In this study, we developed a genetic identification method for the species of albino Misgurnus individuals based on phylogenetic analysis by using recombination activating gene 1 (rag1) and cytochrome b (cytb) region of mitochondrial DNA. As a result of molecular phylogenetic analysis, three clades were identified as Misgurnus mizolepis, M. anguillicaudatus and M. mohoity. The homology of the cytb sequences of M. mohoity was best match to that of M. mohoity sequences in GenBank database. As a result of species identification of 25 albino Misgurnus individuals based on the phylogenetic tree, the red-eye type was identified as 16 M. anguillicaudatus and one M. mizolepis. The remaining three individuals were identified as one M. mizolepis ♀${\times}$M. anguillicaudatus ♂, and two M. mohoity ♀${\times}$M. anguillicaudatus ♂, respectively. In addition, the five black-eye type individuals were identified as one M. anguillicaudatus, three M. mizolepis and one M. mohoity. Therefore, this genetic identification method will be an useful techniques for species or hybrid identification in genus Misgurnus.

  • The structure of skin of a mud loach Misgurnus anguillicaudatus was described in relation with their histochemical nature from four regions of the skin. The epidermis has a strongly thick layer of two glandular cells, consisting of a elongate mucous cell and club cell, and a thin layer of superficial layer. The secretion of the elongated mucous cell was acid mucopolysaccharides in nature but the club cell did not give any histochemical reaction. A well defined lymphatic system, comprising small lymphocytes was present in the stratum germinativum layer of the epidermis. A pit organ of a pear-shaped structure was present below the epithelial cells and lie directly on the basement membrane. The organ has blood vessels serving the sense organs of the epidermis. There was a definite area showing acid mucopolysaccharides in the stratum laxum layer of the dermis. Small scales are present deep in the dermis except the top of the head. A great number of blood capillaries were found just under basement membrane. These structural features of skin in M. anguillicaudatus seem to be closely related with cutaneous respiration using air.

Comparison of Some Tissues in Diploid and Triploid Hybrid between Mud Loach, Misgurnus mizolepis and Cyprinid Loach, M. anguillicaudatus (미꾸라지, Misgurnus mizolepis와 미꾸리, M. anguillicaudatus간 잡종 및 잡종 3배체 일부 조직에서의 세포 및 핵 크기)

  • Diploid and triploid hybrid between female mud loach Misgurnus mizolepis and male cyprinid loach, M. andguillicaudatus were produced, using artificial propagation. The influence of hybridization and triploidization on characteristics of the some tissues of retina optic tectum and trunk kidney in loach was determined histologically. Diploid hybrid have almost size similarities in nucleus and the cell size of some tissues to both parents. In the triploid hybrid the nucleus and/or the cell size in erythrocyte, horizontal cell in inner nuclear layer of retina ganglion cell in ganglion cell layer of retina, neuronal cell in granular layer of optic tectum, and secondary proximal tubule cell of trunk kidney, are much larger than those of the corresponding cells in the diploid hybrid individuals. However, triploid hybrid showed fewer number of cells in outer nuclear layer in retina and nuclei in secondary proximal tubule of trunk kidney than in diploid hybrid. Results of this study suggest that some characteristics in the triploid hybrid exhibiting larger cell and nucleus size with fewer number of cell than diploid hybrid can be useful criteria for the distinction between diploid hybrid and triploid hybrid, and the ploidy level in loach.
