Objectives : Prunus mume has been used for the folk medicine from old times. The purpose of the study was to investigate the antimicrobial activity of prunus mume extract to various oral microbes. Methods : This study was carried out to examine the antimicrobial effect of Prunus mume extract against oral microbes. Data were collected using a Dentocult SM Strip mutans and Dentocult LB Strip mutans from April 5 to May 4, 2013. A total of 36 experimental and 32 control group were selected for this study. Results : The MIC of Prunus mume extract was tested for 0.39% in S. mutans, S. salivarius and S. auerus, 0.78% in S. mitis, S. equi and E. coli. In vivo, experimental group showed significantly the lower Streptococcus mutans levels by the use of the gum contained Prunus mume extract from 15 days compared with control group(p=0.012). The reduction was more significant in Lactobacilli level of the experimental group than the control group(p=0.022). Conclusions : These findings suggest that the oral products containing Prunus mume extracts is effective in preventing oral diseases.
This study was investigated the antibacterial activities of Cirsium japonicum from extracts five regions(Chungnam, Gyeonggi, Gangwon, Jeju and Jeonnam) extract against six food-borne pathogenes(Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, Salmonella enterica and Vibrio vulnificus) using the broth dilution and agar diffusion method. At concentrations between 0 and $750{\mu}g/mL$ the extracts showed an antibacterial effect against all tested bacteria. The antibacterial activities of Cirsium japonicum from Jeju and Gangwon are stronger than others. The minimum inhibitory concentration(MIC) values against the six bacteria ranged from 93.75 to $750{\mu}g/mL$. In time killing assay(cell growth curves), the tested bacteria inactivated upon exposure to the extracts for 24h. At the 24h exposure to the extracts, all bacteria were inhibited to over 70% for growth. In particular, Bacillus subtilis, Salmonella enterica and Vibrio vulnificus conveyed an inhibition of growth to almost complete. It is anticipated that Cirsium japonicum extracts may have greater potential as natural food preservatives.
This study was performed to investigate the anti-microbial activity of extract from Korean fermented soybean paste (doen-jang) against 16 types of oral microbes, and to determine the minimum inhibition concentration (MIC) of the extract for three major microbes causing human oral diseases (Streptococcus mutans, Porphyromonas gingivalis, and Candida albicans). The extract was prepared using ethyl acetate and it was treated with the oral microbes at a concentration of 5.00 mg/ml (0.5%). The anti-microbial activity and MIC were measured using broth dilution method. Significant reduction of microbial activities of 16 types of oral microbes occurred when the soybean paste extract was added to the broth compared to the control (p<0.01), and striking inhibition (more than 99%) was observed in ten types. S. mutans, which causes dental caries, showed MIC at a concentration of 1.25 mg/ml for the extract. P. gingivalis, which causes adult periodontal disease, showed MIC at a concentration of 2.50 mg/ml for the extract. C. albicans, which causes denture stomatitis and angular stomatitis, showed MIC at a concentration of 20 mg/ml for the extract. These results indicate that ethyl acetate extract of doen-jang showed strong anti-microbial effect against 16 types of oral microbes, and the anti-microbial effect of the extract against oral microbes was stronger against bacteria than against fungi. The anti-microbial effect might be possibly enhanced by the fermentation of soybeans.
This experimental study was performed to investigate the effect of herbal eye drops, Tangpo-san and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. The following results were obtained by using Minimum inhibition Concentration(MIC) and inhibition Zone. 1. MIC on Staphylococcus aureus in Tangpo-san was 100%, in Coptidis rhizoma was 100% and in Cravit was 0.1% 2. MIC on Staphylococcus epidermidis in Tangpo-san was 100%, in Coptidis rhizoma was 10% and in Cravit was 0.1%. 3. MIC on Pseudomonas aeruginosa in Tangpo-san, Coptidis rhizoma was not showing and in Cravit was 0.1%. 4. The size of inhibition zone on Staphylococcus aureus for Tangpo-san was 13.3mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 26mm in $50{\mu}{\ell}$ and for Cravit was 31mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 5. The size of inhibition zone on Staphylococcus epidermidis for Tangpo-san was 16mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 25mm in $40{\mu}{\ell}$ and for Cravit was 34mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 6. The size of inhibition zone on Pseudomonas aeruginosa for Tangpo-san, Coptidis rhizoma was not and for Cravit was 24.7mm in $50{\mu}l$, showing the antibacterial effect. In addition, the results shows that the herbal eye drops, Tangpo-san and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphylococcus epidermidis keratitis and if further study is performed, the use of herbal eye drops will be valuable and beneficial in the clinical medicines.
Nine hundred and ninety-two isolates of Botrytis cinerea were obtained from infected strawberries, tomatoes and cucumbers in Taejon, Gongju, Puyo, Nonsan and Kimhae in Korea. Six hundred forty-two (64.7%) isolates were benomyl resistant (BR), 245 (24.7%) were procymidone resistant (PR), and 105 (10.6%) were dichlofluanid resistant (DR). In the minimum inhibitory concentration (MIC) test, DR isolates showed mycelial growth on the PDA incorporated with 100 or 500 $\mu\textrm{g}$/ml of dichlofluanid, while dichlofluanid sensitive (DS) isolates did not grow on the PDA incorporated even with 10 $\mu\textrm{g}$/ml of dichlofluanid. Chemical concentrations for inhibition of spore germination were much lower than those for inhibition of mycelial growth. IC50 values, the effective concentrations for 50% inhibition of spore germination, for DR were 0.11~0.29 $\mu\textrm{g}$/ml, whereas they were 0.04~0.09 $\mu\textrm{g}$/ml for DS isolates. In comparison of fitness-related characteristics such as virulence, sclerotial formation, and sporulation, DR isolates were inferior to DS isolates. However, mycelial growth was little different between DR isolates and DS isolates.
Objectives : This experimental study was performed to investigate the efficacy of Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution manufactured with freezing dryness method to use eye drop. Methods : After administering Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : There was no response to MIC on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution was medicated. Conclusions : The present study suggests that Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution dosen't have anti-bacterial effects on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.
This study analyzed the antibacterial activity of a Rubus coreanum (Bokbunja) ethanol extract. The antimicrobial activity was determined by disc diffusion, minimum inhibitory concentration (MIC), and growth inhibition methods with seven kinds of bacteria related to foodborne illness (Bacillus cereus, Escherichia coli, Escherichia coli O157:H7, Listeria monocytogenes, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhimurium). In the results, disc diffusions of the ethanol extract from R. coreanum (9.8-17.5 mm at $4,000{\mu}g/disc$) clearly showed the antimicrobial activity of the extract against all tested microorganisms. Rubus coreanum promoted an inhibitory effect as follows: E. coli O157:H7 > P. aeruginosa > L. monocytogenes > E. coli > S. aureus > B. cereus ${\geq}$ S. typhimurium. In the MIC test, R. coreanum showed high antimicrobial effect against L. monocytogenes at 500 ppm. Moreover, the R. coreanum ethanol extract showed strong growth inhibition against microorganisms, similar to the MIC results. These results show that a R. coreanum ethanol extract has powerful antimicrobial activity against all tested microorganisms, suggesting that R. coreanum will be useful as a potential natural preservative.
Objectives : This experimental study was performed to investigate the effect of Fel Ursi & Bovis Calculus pharmacopuncture solution(FUBCPS) manufactured by using alcohol/water extraction method for identify ing the use of it as eyedrops. Methods: FUBCPS was manufactured by using alcohol/water extraction method. Measure the size of inhibition zone and MIC(Minimum Inhibition Concentration) after administering FUBCPS on bacterial species. Staphylococcus aureus. Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans, which cause keratitis. Administering cravit(Levonoxacin medicine) on bacterial species also performed to compare the anti-bacterial potency of this material, measured by using the size of inhibition zone Results : After administering FUBCPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans). there was no response to MIC and there was no anti -bacterial potency also. Conclusions : This study suggests that FUBCPS dose not have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that we need to research more on herbal medicines of eye drop which have anti-bacterial effects on keratitis
본 논문은 국내 적하수오 및 백하수오 추출물의 항산화능 및 항균활성을 구명하기 위한 목적으로 진행하였다. 각 하수오의 열수, 에탄올, 부탄올 추출물에 대하여, 일반성분을 분석한 후, 항산화능에 대해서는 총 페놀함량 및 DPPH 라디칼 소거능을 측정하였으며, 항균활성에 대해서는 Staphylococcus aureus 및 Propionibacterium acne에 대한 최소생육저해농도(minimum inhibitory concentration, MIC) 및 생육저해환을 통하여 평가하였다. 적하수오 및 백하수오 부탄올 추출물의 총 페놀함량은 각각 1,212.6 및 1,454.5 mg/g GAE로 다른 유기용매 추출물에 비해 유의적으로 높은 함량을 보였다. 이에 비해 DPPH 라디칼 소거능은 열수추출물에 비해 에탄올 추출물(89.0%), 부탄올 추출물(88.9%)이 유의적으로 높은 값을 나타내었다. MIC 측정 결과, 적 백하수오 에탄올 추출물만이 0.8 mg/mL로 S. aureus와 P. acne에 대해서 항균효과를 나타냈다. 디스크 확산법 측정 결과, 모든 유기용매 추출물이 5 mg/disc 농도부터 생육저해환을 형성하였으며, 농도가 증가함에 따라 생육저해환의 크기도 증가하였다. 적하수오 및 백하수오 에탄올 추출물의 조사균주에 대한 생육저해완의 직경은 10 mg/mL 농도에서 각각 8.9 및 9.2 mm를 나타내, 추출물 중 가장 우수한 항균활성을 나타냈다. 본 연구는 하수오 추출물이 항산화 효과 뿐만 아니라 피부 건강과 관련된 세균에 대해 항균활성을 보여줌으로써, 하수오 추출물이 피부 건강을 위한 천연 유래의 기능성 화장품 소재로도 활용될 수 있는 잠재적 가능성을 제시하였다.
요소가수분해효소는 요소를 암모니아와 이산화탄소로 가수분해하는 효소로 요로감염을 일으키는 세균의 주요 병원성 인자이다. 따라서 요소가수분해효소는 세균이 요로감염증을 유발하는데 중요한 역할을 한다. 본 연구에서는 요소가수분해효소를 생성하는 6가지 종류의 세균에 (K. oxytoca, K. pneumoniae, M. morganii, P. mirabilis, P. vulgaris, S. saprophyticus) 대한 제럼본의 억제효과와 요소가수분해효소 활성능을 평가하였다. 최소억제농도와 최소살균농도 실험에서 제럼본은 요소가수분해효소를 생성하는 6가지 종류의 세균에 대해 억제효과를 보였으며, 최소억제농도는 0.5-2 mM, 최소살균농도는 1-4 mM를 나타내었다. 요소가수분해 활성억제 실험에서 제럼본은 요소가수분해효소의 억제제로 사용하는 아세토히드록사민산 보다 뛰어난 요소가수분해 활성억제효과를 보였다. 그러나 제럼본은 요소가수분해효소를 이루는 소단위체의 발현양에는 영향을 주지 않았다. 이러한 결과들을 종합하여 볼 때, 제럼본은 요소가수분해효소를 생성하는 세균에 대한 살균력을 가질 뿐 만 아니라 훌륭한 요소가수분해 활성억제력도 보유하고 있는 것으로 사료된다.
