To maintain high quality and to extend the shelf life of intact and minimally processed apple cubes, the effect of heat shock on the texture and color of apple cubes (Fuji) was evaluated in this study. After peeled and cored, the apples were cut into cubes of 1.5 cm. The heat shock was immersion in heated water from 25$^{\circ}C$ to 95$^{\circ}C$ in 10$^{\circ}C$ increments for 2 min and cooling to storage temperature as soon as possible after heat treatment. The effect of heat treatment in terms of color and texture measurements with respect to time was investigated, respectively during 7 days storage at in LDPE film bags without sealing at 4$^{\circ}C$, and 95% relative humidity air. Results suggest that heat shock (55$^{\circ}C$, 2 min) may have effectively delayed browning, and there were significant changes in color of apple cube when the temperature of heat shock is above 75$^{\circ}C$. And hardness, stiffness and firmness, which were used to describe texture, behaved similarly in the textural qualities. It was shown that the texture of the apple cubes was decreased with temperature increase. The value of them were most decreased to 70% of the initial ones after treated with 85$^{\circ}C$ and 95$^{\circ}C$ on the lust day.

The national geodetic reference frame of Korea was adopted in 2003, which is referenced to ITRF (International Terrestrial Reference Frame) 2000 at the epoch of January 1, 2002. For precise positioning based on the satellites, it should be thoroughly maintained to the newest global reference frame. Other than plate tectonic motion, there are significant events or changes such as earthquakes, antenna replacement, PSD (Post-Seismic Deformation), seasonal variation etc. We processed three years of GNSS (Global Navigation Satellite System) data(60 NGII CORS stations, 51 IGS core stations) to produce daily solutions minimally constrained to ITRF. From the time series of daily solutions, the sites with unexpected discontinuity were identified to set up an event(mostly antenna replacement). The combined solution with minimum constraints was estimated along with the velocity, the offsets, and the periodic signals. The residuals show that the surrounding environment also affects the time series to a certain degree, thus it should be improved eventually. The transformation parameters to ITRF2014 were calculated with stability and consistency, which means the national geodetic reference frame is properly aligned to the global reference frame.
The bacterial density of partially processed horticultural crops in the commercial market was relatively high probably due to improper handing or processing. Between crops, the bacterial density was low in peeled garlic and ranged above $10^8cfu{\cdot}g^{-1}fw$ in other crops. Especially in brake fern, the bacterial density was the highest and non food materials such as packing strips were found. There was difference in the effects of ozone-treated water washing treatment between crops. In root crops such as burdock and lotus root, ozone showed positive effect on preventing discoloration as well as lowing bacterial density resulting in the increase of marketing period. Any positive effect was not found when sliced crops were treated over 5 min at $0.3mg{\cdot}L^{-1}$ of dissolved ozone. In lettuce slices, leaf tissues were severely discolored and rapidly soften at $0.3mg{\cdot}L^{-1}$ for 2 min dipping. Ozone was highly effective on inhibiting bacterium propagation and off-flavor. This effect was more stronger in Fischer ligulariata than brake fern, probably due to the difference of tissue thickness. Results indicated that ozone-treated water washing had a strong potential to improve the market quality of partially processed horticultural crops including dried wild greens and sliced root crops.
In this study, two duplex real-time PCR approach with melting curve analysis is presented for the detection of Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp. and Staphylococcus aureus, which are important food-borne bacterial pathogens usually present in fresh and/or minimally processed vegetables. Reaction conditions were adjusted for the simultaneous amplification and detection of specific fragments in the ${\beta}$-glucuronidase (uidA, E. coli), thermonuclease (nuc, S. aureus), hemolycin (hly, L. monocytogenes) and tetrathionate reductase (ttr, Salmonella spp.) genes. Melting curve analysis using a SYBR Green I real-time PCR approach showed characteristic $T_m$ values demonstrating the specific and efficient amplification of the four pathogens; $80.6{\pm}0.9^{\circ}C$, $86.9{\pm}0.5^{\circ}C$, $80.4{\pm}0.6^{\circ}C$ and $88.1{\pm}0.11^{\circ}C$ for S. aureus, E. coli O157:H7, L. monocytogenes and Salmonella spp., respectively. For all the pathogens, the two duplex, real-time PCR was equally sensitive to uniplex real-time PCR, using same amounts of purified DNA, and allowed detection of 10 genome equivalents. When our established duplex real-time PCR assay was applied to artificially inoculated fresh lettuce, the detection limit was $10^3$ CFU/g for each of these pathogens without enrichment. The results from this study showed that the developed duplex real-time PCR with melting curve analysis is promising as a rapid and cost-effective test method for improving food safety.

A brief general discussion of the nature and function of biosensors is presented. While the primary motivator for biosensor development has been the health-care industries, recent research efforts have spread to problems in agriculture and biological production systems. To illustrate some of the research from our laboratory, three example biosensors and their corresponding applications are presented. The first of these is an immunosensor for measurement of the hormone progesterone during milking as a method to improve reproductive management of dairy herds. The second example is an enzyme sensor for measurement of urea in milk as a menas to determine the efficiency of conversion of input protein to milk protein and, thus, improve nutritional management of dairy herds. The third example is a DNA sensor using polymerase chain reaction to detect pathogenic bacteria in the wash water of fresh and minimally processed fruits and vegetables. The potential for application of biosensors in agriculture, agrobiotechnology, food processing, and environmental monitoring has barely been realized.
Peeled garlic, soybean sprouts, cut onion and cut green pepper were treated by dipping in solutions of different antibrowning agents, and then stored at $10^{\circ}C$. Surface color and polyphenol oxidase activity of produce were measured through the storage. Tested antibrowning agents include ascorbic acid, citric acid and allyl isothiocyanate. The commodities showed different responses to the antibrowning agent solutions; 1% citric acid for peeled garlic, 1% ascorbic acid for soybean sprouts, 2% citric acid for cut onion, and 1% ascorbic acid for cut green pepper showed better retardation in browning than the other treatments for respective product. For peeled garlic, surface browning was concomitant with increase in polyphenol oxidase activity during storage. On the other hand, there was no positive correlation between surface browning and polyphenol oxidase activity for the other stored products.
The effect of alginate coating in combination with an anti-browning agent on increasing the post-cutting shelf life and improving the quality of minimally processed apple cubes was studied during storage at room temperature for 5 days. A simple coating technique involving the chemical cross-linkage of alginate by calcium was used. Statistical-based experimental designs were applied to improve the quality of the alginate-coated apple cubes (ACAC). Plackett-Burman design was first used to determine the main factors influencing the preservation of the original weight, color, and texture of ACAC. Among these variables, alginate concentration ($X_1$), dipping time ($X_2$), and dipping temperature ($X_3$) significantly influenced the ACAC weight and color (confidence levels above 90%). Subsequently, the effects of the 3 main factors were further investigated by a central composite design. The polynomial models developed by response surface methodology were adequate to describe the relationships between the studied factors and the responses. Overall optimization conducted by superimposing the curves of the responses enabled the determination of an optimal range of the independent variables in which the five responses were simultaneously optimized. The point chosen as representative of this optimal area corresponded to $X_1=2.98%$, $X_2=0.85\;min$, and $X_3=55^{\circ}C$ and under these conditions the model predicted weight loss=0.522%, relative hardness=1.517, ${\Delta}E=1.423$, browning inhibition=93.403%, and ${\Delta}L=0.158$.

This study examined the effects of ozone exposure alone (1, 3, and 5 ppm) as well as in combination with 1% acetic acid, citric acid, or lactic acid on the growth of indigenous microorganisms in enoki mushrooms. Populations of mesophilic bacteria, yeasts and molds in enoki mushrooms appeared to be decreased by stepwise increases in concentration (1 to 5 ppm) or exposure time (0.5 to 5 min) to ozone. Compared to untreated (control) enoki mushrooms, there were reductions of 1.03 to $2.61\;\log_{10}\;CFU/g$ in mesophilic bacteria and of 1.21 to $2.7\;\log_{10}\;CFU/g$ in yeasts and molds in all ozone- treated enoki mushrooms. Combination of 3 ppm ozone and 1% citric acid (p<0.05) synergistically brought about significant reductions in both mesophilic bacteria ($3.52\;\log_{10}\;CFU/g$) and fungi (yeasts and molds) ($2.77\;\log_{10}\;CFU/g$) from enoki mushrooms. The results of this study show that low concentrations of ozone inhibit indigenous microflora populations in enoki mushrooms. Combination treatments of 3 ppm ozone with 1% citric acid showed greater antimicrobial effectiveness than either 3 ppm ozone or 1% citric acid alone.

The aim of this study was to develop the appropriate packaging method for minimally processed sliced onions. The films of different gas permeabilities (LDPE $30{\mu}m$, PD900 and PD941) were used for packaging 1300g of onion slices cut into octuplicate pieces. Perforated LDPE package was prepared as control for comparison. The package atmosphere and onion quality were measured through storage at $1^{\circ}C$ for 38 days. PD900 package of the lowest gas permeability was the best in keeping the fresh-cut onions by maintaining MA conditions of 1-3% $O_2$ and 4-11% $CO_2$ concentrations. The benefits were reduced discoloration, decay and soakness.
This study was performed to evaluate the microbiological quality of fresh cut fruits in Korea. Forty freshly cut fruit samples were assessed for aerobic mesophilic count (AM), aerobic psychrophilic count (AP), total coliform, generic Escherichia coli, yeast and mold (YM), Bacillus cereus, Staphylococcus aureus, Salmonella spp., and E. coli O157:H7. The average value for AM, AP, and YM was 4.51, 5.35, and 4.31 log CFU/g, respectively. The average of the total coliform was 2.42 log CFU/g, and E. coli was not detected in all samples. For foodborne pathogen bacteria, B. cereus and S. aureus were detected in 2.5 and 7.5% samples, respectively, and Salmonella spp. and E. coli O157:H7 were not detected in all samples. Among the samples, pear generally had the highest contamination level. Therefore, to assure the safety of fresh cut fruits, low temperature and thorough hygiene management should be implemented.
