Age Dating and Paleoenvironmental Changes of the Kunang Cave Paleolithic Site

  • The Kunang cave paleolithic site is located at Tanyang [$N37^{\circ}2'$, $128^{\circ}21'E$], Chungbuk Province, which is in the Central part of the Korean peninsula. The cave is developed at 312 amsl in a karstic mountainous area. The South Han River flows across this region and other caves can also be found near the river. The site was discovered in 1986 and excavated 3 times by the Chungbuk National University Museum until now. The cave was wellpreserved from modem human activities until the first discovery. The full length of the cave is estimated to be ca. 140 m. However, a spacious part up to 11 m from the entrance has been excavated. Eight lithological units are divided over the vertical profile at a depth of 5 m. Each unit is deposited in ascending order as follow: mud layer (Unit 9), lower complex (Unit 8) which is composed of angular blocks and fragments with a muddy matrix, lower travertine layer (Unit 7; flowstone), middle complex (Unit 6; cultural layer) which is composed of fragments with a muddy matrix, middle travertine layer (Unit 5; flowstone), yellowish muddy layer (Unit 4), upper complex (Unit 3; cultural layer) which has a similar composition to Unit 8. the upper travertine layer (Unit 2; flowstone), and finally surface soil layer (Unit 1). The most abundant vestiges in the cultural layers are the animal bones. They are small fractured pieces and mostly less than 3 cm in length. About 3,800 bone pieces from 25 animal species have been collected so far, 90 percent of them belonging to young deers. Previous archaeological study of these bone pieces shows thatprehistoric people occupied the cavenot for permanent dwelling but for temporary shelter during their seasonal hunting activity. More extensive studies of these bones together with pollen analysis are in progress to reconstruct the paleoenvironment of this cave. Only a single date (12,500 BP) obtained from a U-Th measurement of the upper travertine layer was previously available. In spite of the importance of the cave stratigraphy, there was no detail chronological investigation to establish the depositional process of the cultural layers and to understand the periodic structure of the cave strata, alternating travertine floor and complex layers. We have measured five 14C age dating (38900+/-1000, 36400+/-900, 40600+/-1600, more than 51000 and 52000 14C BP) using Seoul National University 14C AMS facility, conducted systematic process of the collagen extraction from bone fragments samples. From the result, we estimate that sedimentation rate of the cave earth is constant, and that the travertine layers, Unit 2 and Unit 3, was formed during MIS 5a(ca. 80 kBP) and MIS 5c (ca. 100 kBP) respectively. The Kunang Cave site is located at Yochonli of the region of Danyang in the mid-eastern part of Korea. This region is compased of limestones in which many caves were found and the Nam-han river flows meanderingly. The excavations were carried out three times in 1986, 1988, and 1998.

Study on the Manufacturing of Chitosan Using Krill(Euphausia superba Dana) and Quality Characteristics (크릴을 이용한 키토산 제조 및 품질 특성)

  • For the use of Antartic krill(Euphausia superba Dana) as food resource, general composition, extracting condition of chitin and quality characteristics of chitosan were investigated. General composition of frozen krill(Euphausia superba Dana) was consisted of moisture 79.0%, protein 13.1%, lipid 4.0%, VBN 7.7mg%, ash 2.7%, others 1.2% and that of dried krill powder was moisture 5.6%, protein 56.1%, lipid 18.8%, ash 11.4%, others 8.1%. The condition of chitin extraction from krill powder was treated with 1N NaOH at $40^{\circ}C$ for removing protein, 1N HCl for excepting mineral substances and methanol for decoloring. The yield of chitin by new procedure developed was 3.7%. The composition of extracted chitin contents was moisture 7.1%, ash 0.4%, protein 3.5%, lipid 3.1%. The results of degree of deacetylation in chitosan at 50% NaOH, $121^{\circ}C$, for 2 hrs was showed 82%. At the same alkali concentration and reaction concentration, a longer reaction time gave a decreased degree of deacetylation. The apparant viscosity was 0.09241 Pa in 1% chitosan from krill and 0.13826 Pa in standard chitosan.

STUDY ON MUTATION OF RAS GENE IN DMBA INDUCED CARCINOMA OF HAMSTER BUCCAL POUCH (DMBA로 유도된 햄스터 협낭암종에서 ras 유전자 변이에 관한 연구)

  • Alterations in the cellular genome affecting the expression or function of genes controlling cell growth and differentiation are considered to be the main cause of cancer. Over 30 oncogenes can be activated by insertional mutagenesis, single point mutations, chromosomal translocations and gene amplification. The ras oncogenes have been detected in $15{\sim}20%$ of human tumors that include some of the most common forms of human neoplasia and are known to acquire their transforming properties by single point mutations in two domains of their coding sequences, most commonly in codons 12 and 61. The ras gene family consists of three functional genes, N-ras, K-ras and H-ras which encode highly similar proteins of 188 or 189 amino acid residues generically known as P21. ras proteins have been shown to bind GTP and GTP, and possess intrinsic GTPase activity. Experimental study was performed to observe the mutational change of the ras gene family and apply the results to the clinical activity. 36 Golden Syrian Hamster each weighing $60{\sim}80g$ were used and painted with 0.5% DMBA by 3 times weekly on the right buccal cheek(experimental side) for 6, 8, 10, 12, 14 and 16 weeks. Left buccal cheek (control side) was treated with mineral oil as the same manner of the right side. The hamsters were sacrificed on the 6, 8, 10, 12, 14 & 16 weeks. Normal and tumor tissues from paraffin block were completely dissected by microdissection and DNA from both tissue were isolated by proteinase K/phenol/chloroform extraction. Segments of the K-ras and H-ras gene were amplified by PCR using the oligonucleotide primers corresponding to the homologous region (codon 12 and 61) of the hamster gene, and then confirmational change of ras genes was observed by SSCP and autosequencing analysis. The results were as follows : 1. Malignant lesion could be found in the experimental side from the experimental six weeks. 2. One hamster among six showed point mutation of the H-ras codon 12($G{\rightarrow}A$ transition) at the experimental 10 and 14 weeks. 3. One of six at 6 weeks, two of six at 8 weeks and one of six at 12 weeks revealed the confirmational change of the H-ras codon 61($A{\rightarrow}T$ transversion). 4. The incidence of point mutation of H-ras codon 12 and 61 were 5.5%(2 of 36) and 11%(4 of 36) respectively. 5. Point mutation of the K-ras could not be seen during the whole experimental period. Form the above results, these findings strongly support the concept that H-ras oncogenes may have the influence of the DMBA induced carcinoma of hamster buccal pouch.

Postoperative irradiation after implant placement: A pilot study for prosthetic reconstruction

  • PURPOSE. On maxillofacial tumor patients, oral implant placement prior to postoperative radiotherapy can shorten the period of prosthetic reconstruction. There is still lack of research on effects of post-implant radiotherapy such as healing process or loading time, which is important for prosthodontic treatment planning. Therefore, this study evaluated the effects of post-implant local irradiation on the osseointegration of implants during different healing stages. MATERIALS AND METHODS. Custom-made implants were placed bilaterally on maxillary posterior edentulous area 4 weeks after extraction of the maxillary first molars in Forty-eight Sprague-Dawley rats. Experimental group (exp.) received radiation after implant surgery and the other group (control) didn't. Each group was divided into three sub-groups according to the healing time (2, 4, and 8 week) from implant placement. The exp. group 1, 2 received 15-Gy radiation 1 day after implant placement (immediate irradiation). The exp. group 3 received 15-Gy radiation 4 weeks after implant placement (delayed irradiation). RESULTS. The bone mineral density (BMD) was significantly lower in the immediate irradiation groups. BMD was similar in the delayed irradiation group and the control group. The irradiated groups exhibited a lower bone-to-implant contact ratio, although the difference was not statistically significant. The irradiated groups also exhibited a significantly lower bone volume and higher empty lacuna count than the control groups. No implant failure due to local irradiation was found in this study. CONCLUSION. Within the limits of this study, the timing of local irradiation critically influences the bone healing mechanism, which is related to loading time of prostheses.

Comparative Studies on K2CO3-based Catalytic Gasification of Samhwa Raw Coal and Its Ash-free Coal (삼화 원탄과 무회분탄의 촉매(K2CO3) 가스화 반응성 비교 연구)

  • Catalytic gasification of raw coals at mild condition is not realized yet mainly due to deactivation of catalysts via their irreversible interaction with mineral matters in coal. In this work, the gasification behavior of ash-free coal (AFC) was compared with that of the parent raw coal. In order to modify the gasification conditions, the raw coal gasified with fixed variables (water supply, space velocity, temperature, catalysts) in a fixed bed reactor. When catalysts are added by physical mixing method with coal, $K_2CO_3$ was the most effective additives for steam gasification of coal. However, the activity of ash-free coal (AFC) was much less reactive than raw coal due to high temperature extraction in a 1-methylnaphthalene under 30bar at $370^{\circ}C$ for 1 h, almost removed oxygen functional groups, and increased carbonization. The addition of $K_2CO_3$ in AFC achieved higher conversion rate at low temperature ($700^{\circ}C$). At that time, the molar ratio of gases ($H_2/CO$ and $CO_2/CO$) was increased because of water-gas shift reaction (WGSR) by addition of catalysts. This shows that catalytic steam gasification of AFCs is achievable for economic improvement of gasification process at mild temperature.

Development of In Vitro Porcine Oocytes Following Intracytoplasmic Injection of Sperm-Mediated GFP Gene

  • Transgenic animals production tools have been valuable for research and purpose. The current methods of gene transfer, microinjection and nuclear transfer, which are widely used in transgenic animal production, but all most methods has only had limited success in production of larger species. Here, we report the possibility of a sperm-mediated gene transfer method in porcine embryos. Oocytes were collected from ovaries harvested at a local slaughterhouse were matured in 500${mu}ell$ drops of TCM-199 under mineral oil at 38.5$^{\circ}C$ in a humidified atmosphere of 5%CO2 in air. After 42-43h of in vitro maturation oocytes were denuded. for sperm injection into the cytoplasm of the porcine oocytes, sperm suspension in NIM medium are subjected extraction with TritonX-100 before mixing with a green fluorescent gene (GFP). Sperm with Tritonx-100 were prepared by adding TritonX-100 to a final volume of 0.05% in the sperm suspension and mixing by trituration for 60s before two wishes in NIM medium at 2$^{\circ}C$. A(ter wishing, sperm were mixed with TritonX-100 at $25^{\circ}C$ followed by washes at 2$^{\circ}C$. Sperm were resuspended in ice cold NIM to a final volume of 400${mu}ell$ and 2-20ng/${mu}ell$ DNA were triturated on ice for 60s. All microinjection was performed in HEPES-buffered CZB medium at room temperature within 2h. After culture in NCSU-23 for 72h, percent of porcine embryos transfected GFP gene are 20.7%(6/29) in 20ng/${mu}ell$ sperm-DNA mixed group and other groups were 3.7 %(2/54)and 4.7%(3/67). These data suggests that sperm-mediated gene transfer method should be used to the production tool of transgenic pig efficiently.

Nutritional Component and Anticancer Properties of Various Extracts from Haesongi Mushroom (Hypsizigus marmoreus) (해송이버섯(Hypsizigus marmoreus)의 영양성분과 추출용매에 따른 암세포 생장억제 효과)

  • This study was aimed to analyze the nutritional components and anticancer properties of Haesongi mushroom (Hypsizigus marmoreus), which has been recently available in Korea, to estimate its nutritional and functional values. Fruit body of Haesongi mushroom was investigated for its proximate components and mineral contents. Its water and ethanol extracts were compared for nutritional components such as $\beta$-glucan, protein, and total sugar. Anticancer effects of both extracts were measured against human cancer cell lines in vitro. This mushroom contained high protein (22.63%), total dietary fiber (30.80%), and K (3383.3 mg/100 g). The water extract contained more nutritional components such as $\beta$-glucan (9.32 mg/g), protein (17.71%), and total sugar (39.93%), compared with the ethanol extract. Moreover the extraction yield of the water extract was higher than the ethanol extract. The growth inhibitory effects of the water extract (5 mg/mL) on AGS, HepG2, and SW480 human cancer cells were 90.61, 75.43, and 58.49%, respectively. However, the ethanol extract showed 81.79, 49.90, and 25.71% growth inhibition, respectively. In this study, it is demonstrated that water is a more efficient solvent than ethanol for extracting nutritional and functional components from Haesongi mushroom.

Physicochemical Properties of Black Doraji (Platycodon grandiflorum) (흑도라지의 이화학적 품질특성)

  • The physicochemical properties of raw and black doraji were examined with the goal of increasing the utilization of doraji. Raw and Black doraji contained similar amounts of crude ash, crude fat, and crude protein, but raw doraji had higher level of fiber. Arginine was the major free amino acid in both types of doraji. Free sugar and organic acid contents of doraji increased after steaming heat treatment. Potassium was a prominent mineral in both raw and black doraji, constituting 85% of total minerals. Black doraji contained almost twice as much crude saponin as did raw doraji. Black doraji showed lower values of L (lightness), a (redness), and b (yellowness). The hardness and chewiness of raw doraji were higher than those of black doraji. Black dorai showed a higher amount and extraction rate of saponin, even though nutrient value of doraji slightly decreased after steaming heat treatment. Therefore, black doraji can be a valuable ingredient in functional foods.

Simultaneous analysis of ethylene glycol and glycolic acid in bio-specimens by GC/MS (생체시료에서 GC/MS에 의한 에틸렌글리콜 및 대사체인 글리콜산 동시분석)

  • Mistaking pink colored thermal oil for grape wine, a victim drank the oil to death which was analyzed to contain 39% of ethylene glycol. Thermal oil could be used for heat transfer to prevent the malfunction due to the high pressure in the boiler operated at high temperature when using water. Main component of thermal oil is known to be mineral oil or ethylene glycol. From the blood and other tissue of the victim from autopsy, ethylene glycol and its metabolite were simultaneously analyzed by GC/MS after extraction under acidic condition with acetonitrile followed by derivatization with BSTFA. About 0.2 g of the specimens were pretreated with 50 uL of 0.5 M HCl solution to keep acidic condition, then dehydrated with anhydrous sodium sulfate followed by concentration under nitrogen stream. Ethylene glycol and glycolic acid concentration in blood was measured to be $2,755\;{\mu}g/mL$ and $174\;{\mu}g/mL$ respectively. In other specimen, the concentration of ethylene glycol and glycolic acid was $860\;{\mu}g/g\sim1,290\;{\mu}g/g$ and $93\;{\mu}g/g\sim134\;{\mu}g/g$. Especially, crystal appeared in kidney which was supposed xalate from the metabolite of ethylene glycol.

Petrology and Geochemistry of Peridotite Xenoliths from Miocene Alkaline Basalt Near the Mt. Baekdu Area (백두산 지역의 마이오세 알칼리 현무암에 포획된 페리도타이트의 암석학적/지화학적 특성)

  • Peridotite xenoliths in middle Miocene alkaline basalt from the Mt. Baekdu area are mainly anhydrous spinel lherzolites, displaying coarse-grained protogranular texture. These xenoliths have late-stage secondary orthopyroxene replacing olivine as the metasomatic mineral and glass formed along the grain boundaries. The studied xenoliths are characterized by the high $Mg{\sharp}[=100{\times}Mg/(Mg+Fe_{total})$ atomic ratio] of olivine, orthopyroxene and clinopyroxene (89~92) and the $Cr{\sharp}[=100{\times}Cr/(Cr+Al)$ atomic ratio] of spinel (10~29). Based on major-element data, the studied xenoliths are similar to those from the abyssal peridotites. Clinopyroxenes of the xenoliths are mostly enriched in incompatible trace elements, exhibiting two types of REE patterns: (1) LREE-depleted with $(La/Yb)_N$ of 0.1~0.2 and $(La/Ce)_N$ of 0.4~0.8. (2) LREE enriched with $(La/Yb)_N$ of 2.2~3.8 and $(La/Ce)_N$ of 1.2~1.6. The calculated equilibrium temperatures and oxygen fugacities resulted in $920{\sim}1050^{\circ}C$ and ${\Delta}fO_2(QFM)=-0.8{\sim}0.2$, respectively. It is suggested that the Mt. Baekdu peridotite xenoliths represent residues left after variable degrees of melt extraction(less than 15 vol%), which was subsequently subjected to different degrees of modal/cryptic metasomatism by silica- and LREE-enriched fluids (or melts).
