Effects of Milk Production, Season, Parity and Lactation Period on Variations of Milk Urea Nitrogen Concentration and Milk Components of Holstein Dairy Cows

  • The study was conducted to assess the effect of milk production, parity, stage of lactation, season and individual milk components themselves on milk urea nitrogen (MUN) concentration and other milk components of 3,219 Holstein dairy cows in Korean dairy farms. The MUN concentrations in Korean dairy cows were estimated to 16.68$\pm$5.87 mg/dl. Milk yield was negatively correlated with fat and protein contents and somatic cell counts (SCC) in milk (p<0.01). The increasing MUN concentration has positive correlation with yield and fat content. By increasing somatic cell, milk yield was reduced and MUN level was increased. Cows in spring and winter produced more milk over 1.43 and 0.93 kg/day, respectively, than cows in summer (p<0.01). Milk urea nitrogen concentrations of milk produced in summer and fall were significantly lower (p<0.01) than those in spring and winter. Both MUN concentration and somatic cell counts were highest in winter. Milk yield was lower (p<0.01) in the first calving than other calving time and was tended to increase until the fifth parity and then decrease. Milk urea nitrogen and SCC were not related to parity of cows in this study. Milk yield and SCC were positively related to lactation period while MUN concentrations and milk fat and protein contents were negatively influenced by stage of lactation. In the present study, the relationship between MUN and reproduction of dairy cows was also investigated. Cow produced milk in high MUN concentrations (greater than 18 mg/dl) had more open days than cows in MUN concentrations less than 18 mg/dl. However, no significant difference between MUN concentration levels and frequency of artificial insemination was found in this study. It is suggested that although MUN values for nutritional management and measures of production or reproduction are used, non-nutritional factors should be considered.

Genetic parameter estimation for milk β-hydroxybutyrate and acetone in early lactation and its association with fat to protein ratio and energy balance in Korean Holstein cattle

  • Objective: The objective of this study was to estimate the genetic parameters for milk ${\beta}$-hydroxybutyrate (BHBA), acetone (Ac), fat protein ratio (FPR), and energy balance (EB) using milk test day records and investigate the effect of early lactation FPR and EB on milk ketone body concentrations. Methods: Total 262,940 test-day records collected from Korea Animal Improvement Association during the period of 2012 to 2016 were used in this study. BHBA and Ac concentrations in milk were measured by Fourier transform infrared spectroscopy (FTIR). FPR values were obtained using test day records of fat and protein percentage. EB was calculated using previously developed equation based on parity, lactation week, and milk composition data. Genetic parameters were estimated by restricted maximum likelihood procedure based on repeatability model using Wombat program. Results: Elevated milk BHBA and Ac concentrations were observed during the early lactation under the negative energy balance. Milk FPR tends to decrease with the decreasing ketone body concentrations. Heritability estimates for milk BHBA, Ac, EB, and FPR ranged from 0.09 to 0.14, 0.23 to 0.31, 0.19 to 0.52, and 0.16 to 0.42 respectively at parity 1, 2, 3, and 4. The overall heritability for BHBA, Ac, EB and FPR were 0.29, 0.32, 0.58, and 0.38 respectively. A common pattern was observed in heritability of EB and FPR along with parities. Conclusion: FPR and EB can be suggested as potential predictors for risk of hyperketonemia. The heritability estimates of milk BHBA, Ac, EB, and FPR indicate that the selective breeding may contribute to maintaining the milk ketone bodies at optimum level during early lactation.

  • Milk data may be increasingly used as indicators of the protein-energy balance and actual farm feeding practices. It was related to milk production, nutritional and reproductive disorders. The purpose of this study was to investigate the relationship between level of fat, protein or milk urea nitrogen (MUN) from the first test within 35 days in milk and displaced abomasum (DA) in a large dairy herd with high yielding Holstein cows. Milk data from forty-five DA cases were compared to those from 90 healthy cows. Higher odds of DA diagnosis was found with higher 5.0% milk fat, lower 3.0% milk protein. Therefore, cows with a fat to protein ratio of>1.5 had higher risks for DA. Also, incidence rates of DA was higher in the cows which the level of MUN was lower than 12.0 mg/dl or higher than 25.0 mg/dl relative to healthy cows. These results indicate that cows diagnosed with DA were energy deficient prior to DA diagnosis. We conclude that level of fat, protein or MUN serve as a monitoring tool of protein and energy nutritional balance in early lactation cows and also as a significant predictor of risk for DA.

  • The purpose of this study was to develope a computer program to help with gross diagnosis of protein-energy balance and feeding management practice and with the prediction about the risk possibility of productive disease such as reproductive and metabolic disorders by evaluating fat, protein, and milk urea nitrogen (MUN) from individual cow milk in dairy herd Somatic cell counts also represent the condition of udder health. The principal flow charts of this program was to check on herd management, sampling the composite milk, analysis the milk composition, conversion of data from milking equipment to program, input and analysis of data in program, and report. This program is compatible with window 95/98 system. The major analytical elements of this program were presented as; the profile of herd lactation curve analysis of the test-day milk production level, the distribution of somatic cell count, the fat to protein ratio to evaluate body energy balance, and the interpretation of dietary protein-energy balance by milk protein and MUN contents for individual cows. This program using milk fat, protein, MUN, and somatic cell counts will serve as a monitoring tool for the protein-energy balance and the feeding management practice, and for distribution of mastitis in individual cows. It will also be used to manage the nutritional and reproductive disorders and mastitis at the farm level.

Effects of Soybean Oil or Rumen Protected Conjugated Linoleic Acid Supplementation on Accumulation of Conjugated Linoleic Acid in Dairy Cows' Milk

  • The effects of feeding soybean oil (SBO) or rumen protected conjugated linoleic acid (RP-CLA) on CLA accumulation in milk, and performance of lactating dairy cows were studied. Twenty four Holstein Friesian crossbred lactating dairy cows, averaging $126{\pm}45days$ in milk, $15.6{\pm}2.43kg$ of milk and $452{\pm}51kg$ body weight were stratified randomly and assigned in a randomized complete block design (RCBD) to three treatments of 8 cows each. The treatments were control, 150 g of SBO and 150 g of RP-CLA supplementation. Performance parameters showed that DM intake, NELP intake and body weight change were similar across treatments, while CP intake was decreased by SBO and RP-CLA supplementation. Milk yield and milk composition were not significantly different among treatments, except for milk fat percentage and fat yield which were significantly decreased by 27% (p<0.05) and by 28% (p<0.01), respectively, by RP-CLA supplements compared with control treatment. Feeding RP-CLA reduced 3.5% FCM compared with the other treatments (p<0.003). Both SBO and RP-CLA supplementation reduced ${\geq}C18:0$ and CLA concentration in milk fat.

  • The relationship between level of milk composition and conditions of ovary and uterus were analyzed in Holstein cows at seven farms participating in a reproductive herd health management program. Milk data were taken from 503 early lactating cows between 30 and 60 days in milk with reproductive examination with ultrasonography from september 1999 to August 2000. Milk fat, protein and solid-not-fat concentration in the herds were $3.70{\pm}1.08%$, $2.97{\pm}0.35$, and $8.41{\pm}0.61%$, respectively. The reproductive disorder relative to normal cows had higher risk in the cows that the level of protein was lower than 2.70%. Also, the higher milk fat than 4.50% were associated with a higher risks in the uterine disease and follicular cysts. Therefore, the cows with the fat to protein ratio of > 1.30 had higher risks for reproductive disorder such as cystic ovarian diseases, inactive ovaries and endometritis. These results indicated that cows diagnosed with reproductive disorder were energy deficient prior to reproductive disorder diagnosis. Consequently, milk fat and protein analyses may be used serve as a monitoring tool for condition of ovary and uterus in early lactating cows

Variance Component Estimates with Dominance Models for Milk Production in Holsteins of Japan Using Method R

  • Fractions of herd-year-season, sire by herd interaction, additive genetic and dominance genetic variances were estimated for milk production traits in Holsteins of Japan using Method R. Inbreeding depressions for milk production traits were also estimated. Estimated fractions of herd-year-season variances ranged from 0.056 to 0.074 for yield traits and from 0.033 to 0.035 for content traits. Estimated fractions of additive genetic variances to phenotypic variances (heritabilities across a herd in the narrow sense) were 0.306, 0.287, 0.273, 0.255, 0.723, 0.697 and 0.663 for milk, fat, SNF and protein yields, and fat, SNF and protein contents, respectively. Estimated fractions of dominance genetic variances ranged from 0.019 to 0.022 for yield traits and from 0.014 to 0.018 for content traits. Fractions of variances for sire by herd interaction were estimated to range from 0.020 to 0.025 for yield traits and 0.011 to 0.012 for content traits. Estimates of inbreeding depression for milk, fat, SNF and protein yields were -36.16 kg, -1.42 kg, -3.24 kg and -1.15 kg per 1% inbreeding for milk, fat, SNF and protein yields, respectively. Estimates of depression per 1% inbreeding for content traits were positive at $0.39{\times}10^{-3}%$, $0.31{\times}10^{-3}%$ and $0.82{\times}10^{-3}%$ for fat, SNF and protein contents, respectively.

The cooperative regulatory effect of the miRNA-130 family on milk fat metabolism in dairy cows

  • Objective: There is a strong relationship between the content of beneficial fatty acids in milk and milk fat metabolic activity in the mammary gland. To improve milk quality, it is therefore necessary to study fatty acid metabolism in bovine mammary gland tissue. In adipose tissue, peroxisome proliferator-activated receptor gamma (PPARG), the core transcription factor, regulates the fatty acid metabolism gene network and determines fatty acid deposition. However, its regulatory effects on mammary gland fatty acid metabolism during lactation have rarely been reported. Methods: Transcriptome sequencing was performed during the prelactation period and the peak lactation period to examine mRNA expression. The significant upregulation of PPARG drew our attention and led us to conduct further research. Results: According to bioinformatics prediction, dual-luciferase reporter system detection, real-time quantitative reverse transcription polymerase chain reaction and Western blotting, miR-130a and miR-130b could directly target PPARG and inhibit its expression. Furthermore, triglyceride and oil red O staining proved that miR-130a and miR-130b inhibited milk fat metabolism in bovine mammary epithelial cells (BMECs), while PPARG promoted this metabolism. In addition, we also found that the coexpression of miR-130a and miR-130b significantly enhanced their ability to regulate milk fat metabolism. Conclusion: In conclusion, our findings indicated that miR-130a and miR-130b could target and repress PPARG and that they also have a functional superposition effect. miR-130a and miR-130b seem to synergistically regulate lipid catabolism via the control of PPARG in BMECs. In the long-term, these findings might be helpful in developing practical means to improve high-quality milk.

수유부의 식품 및 영양소 섭취와 유즙 생성량 및 조성 (Composition and Yield of Korean Breast Milk and Maternal Intakes of Foods and Nutrients)

  • This study was conducted to investigate the relationship between lactational capacity and intakes of energy and energy-yielding nutrients. Food consumption, intakes of carbohydrate, protein, fat and energy and quantity and proximate composition of milk of 11 lactating Korean mothers were determined at 1, 2 and 3 months postpartum longitudinally. Food consumption was estimated using a 24-hour recall method ; intakes of energy and energy-yielding nutrients were calculated according to the Food Composition Table. Daily milk production was estimated using a 72-hour test-weighing method ; protein, fat, lactose and energy concentrations were analyzed. Average intakes of energy and protein were 1974$\pm$386㎉/day and 67.0$\pm$12.3㎉/day, these were 73% and 74% of the Korean Recommended Allowances, respectively. Average milk yield was 720.1$\pm$123.3ml/day containing energy 59.6$\pm$9.5㎉/dl, protein 1.1$\pm$0.1g/dl, fat 2.7$\pm$0.8g/dl, and lactose 6.3$\pm$0.3g/dl. No relationship existed between the intakes of carbohydrate, protein, fat and energy and the quantity and proximate composition of the milk. However, the intakes of energy, carbohydrate and vegetable protein were inversely related to the concentrations of energy and lactose in the milk. This result indicates that lactational capacity may be affected by the other factors excluding intakes of energy and energy-yielding nutrients and a high intake of energy may not guarantee optimal lactational capacity.

초임계이산화탄소에 의한 유지방의 연속 병류식 추출 (Continuous Cocurrent Extraction of Milk Fat by Supercritical Carbon Dioxide)

  • Performance of a continuous cocurrent supercritical fluid extraction column for processing of anhydrous milk fat (AMF) was determined. The extract loading increased and the extraction yield decreased as the superficial velocity of AMF increased. The maximum solubility of AMF in $CO_2$ at 4$0^{\circ}C$/3,500 psig was 0.0195g/g. The increase of the carbon dioxide density enhanced the solubility of milk fat and decreased the cholesterol concentration in the extract. Three operation conditions, such as recycle , reflux, and temperature gradient of extraction column, were tested. More short-chain fatty acvids were extracted by the reflux operation . Longchain fatty acids and thehighest ratio fo long-chain unsaturated to saturated fatty acids were also obtained in the raffinate phase with refluex.
