Vessel boundary detection and modeling is a difficult but a necessary task in analyzing the mechanics of inflammation and the structure of the microvasculature. In this paper we present a method of analyzing the structure by means of an active contour model(using GVF Snake) for vessel boundary detection and 3D reconstruction. For this purpose we used a virtual vessel model and produced a phantom model. From these phantom images we obtained the contours of the vessel by GVF Snake and then reconstructed a 3D structure by using the coordinates of snakes.
Introduction: 585 nm Pulsed dye laser (PDL) laryngeal surgery is based on the photodynamic characteristics of selective photothermolysis and photoangiolysis and recently considered to be the treatment for a variety of benign laryngeal disease. Objective: To review the indications and outcome of office-based 585nm PDL surgery and summarize new developments. Method: Retrospective study involving 402 patients was performed, The PDL surgery could be applied to various laryngeal diseases such as laryngeal papilloma, vocal fold dysplasia, laryngeal granuloma, vocal polyp, capillarectasia, scarred vocal fold and sulcus vocalis. Results : The physiologic properties of the vascular specificity of PDL provide many advantages and appear to be effective for laryngeal treatment. The PDL resulted in precise, selective coagulation of the microvasculature without damage to the surrounding tissue. Therefore PDL surgery is safe and effective for office-based treatment of benign laryngeal disease and for all patients regardless of their overall medical condition. Conclusion: PDL surgery provides potential benefits and advantage for treating common benign laryngeal disease.

Intravital video microscopy permits the observation of microcirculatory blood flow. This often requires fluorescent probes to visualize structures and dynamic processes that cannot be observed with conventional bright-field microscopy. Conventional light microscopes do not allow for simultaneous bright-field and fluorescent imaging. Moreover, in conventional microscopes, only one type of fluorescent label can be observed. This study introduces multispectral intravital video microscopy, which combines bright-field and fluorescence microscopy in a standard light microscope. The technique enables simultaneous real-time observation of fluorescently-labeled structures in relation to their direct physical surroundings. The advancement provides context for the orientation, movement, and function of labeled structures in the microcirculation.
진피 상층부 모세혈관은 젊은 사람에 비해 노인에서 그 수와 크기가 감소되어 있어 피부에서 모세혈관이 차지하는 비율이 나이에 따라 급격히 낮아진다. 연교는 물푸레나무과에 속하는 개나리 열매를 건조한 것으로 주로 염증성 또는 항균성 질환에 오랫동안 사용되어져온 약재로서 지금까지 피부 혈관신생과 관련된 효능은 보고되지 않았다. 따라서 본 연구에서는 연교 추출물이 혈관신생과 관련된 인자들에 미치는 영향을 피부 각질형성 세포주를 이용하여 조사하고자 하였다. 우리는 먼저 연교 추출물이 혈관신생과 관련된 인자들의 발현에 어떤 영향을 주는지 알아보고자 각질형성세포에 연교 추출물을 처리하고 혈관신생과 관련된 55개 단백질의 발현을 분석하였다. 발현 변화를 보인 인자들 중 혈관내피세포 성장인자(VEGF, vascular endothelial growth factor)는 강력한 혈관신생 촉진인자로서 연교 추출물에 의해 유의하게 발현이 증가되었다. 따라서 연교 추출물이 VEGF 생성에 미치는 영향에 대해 자세히 알아보고자 연교 추출물을 농도별로 세포에 처리하고 단백질 발현과 mRNA 발현 변화를 조사한 결과, 연교 추출물은 VEGF의 유전자 수준뿐만 아니라 단백질 수준의 발현을 2배이상 농도 의존적으로 증가시켰다. 다음으로 연교 추출물에 의한 VEGF 발현 증가에 관여하는 신호전달 기전을 밝히고자 MAPK 활성을 살펴본 결과, 연교 추출물을 세포에 처리하면 5 min 내 p38 MAPK의 활성이 관찰되었으며, 특이적 억제제 전처리를 통해 p38 MAPK 활성을 억제하면 연교 추출물을 처리하더라도 VEGF의 유전자 및 단백질 발현이 완전히 억제됨을 확인하였다. 이러한 결과로부터 연교는 피부 각질형성세포에 작용하여 p38 MAPK 활성을 통해 VEGF 생성을 유도함을 알 수 있었고, 피부에서 노화에 따른 표피아래 모세혈관 손상을 개선하는데 도움을 줄 수 있는 후보 물질로서 연교의 새로운 효능을 제안한다.
The dependence of the rheological properties of blood on shape, aggregation, and deformability of red blood cells (RBCs) has been investigated using hybrid systems by coupling fluid with solid models. We present a simple approach for simulating blood as a multi-component fluid, in which RBCs are modeled as droplets of acquired biconcave shape. We used lattice Boltzmann method (LBM) due to its excellent numerical stability as a simulation tool. The model enables us to control the droplet static shape by imposing non-isotropic surface tension force on the interface between the two components. The use of the proposed non-isotropic surface tension method is justified by the Norris hypothesis. This hypothesis states that the shape of the RBC is due to a non-uniform interfacial surface tension force acting on the RBC periphery. This force is caused by the unbalanced distribution of the lipid molecules on the surface of the RBC. We also used the same concept to investigate the dynamic shape change of the RBC while flowing through the microvasculature, and to explore the physics of the Fahraeus, and the Fahraeus-Lindqvist effects.

Microsurgery has advanced beyond its nascent stages reaching success rates of 90% to 95%. However, this means that even in the best circumstances, 5% to 10% of free flaps and replants fail. Almost all failures are due to vessel thrombosis, resulting in ischemia of the transplanted tissue. Many attemps have been undertaken to treat and reverse its effects. Zdeblick and colleagues noted an improvement in the viability of amputated limbs replanted after an extended period of ischemia following intraarterial infusion of urokinase. Subsequent studies have investigated many modalities of urokinase administration in various animal models by differing ischemic periods. These studies, however, have failed to establish a definitive, generally accepted protocol for administration of urokinase in the salvage of tissue subjected to prolonged ischemia. Our clinical observations suggest that a bolus of urokinase delivered under pressure may increase the thromoblytic effect of the drug, probably by means of increased delivery to microvasculature. We intend to investigate the role of selective pressure perfusion of ischemic flaps as a new means for increasing the effectiveness of urokinase in the treatment of the "no-reflow" phenomenon. A total of 32 male New Zealand rabbits were used and divided into the four groups according to the method of infusion. After 12 hours of ischemia the flaps were injected with Hartmann's solution or with urokinase and the percent survival of the flap was determined at 7 days following flap reperfusion. As the result, the flap survival rate was highest in the pressure injection of urokinase group.
Aquaporin 1 (AQP1) is expressed in most microvasculature endothelial cells and forms water channels that play major roles in a variety of physiologic processes. This study aimed to delineate the transcriptional regulation of AQP1 by Mef2c in endothelial cells. Mef2c cooperated with Sp1 to activate human AQP1 transcription by binding to its proximal promoter in human umbilical cord vein endothelial cells (HUVEC). Over-expression of Mef2c, Sp1, or Mef2c/Sp1 increased HUVEC migration and tube-forming ability, which can be abolished AQP1 knockdown. These data indicate that AQP1 is a direct target of Mef2c in regulating angiogenesis and vasculogenesis of endothelial cells.

Background Glomus tumors were first described by Wood in 1812 as painful subcutaneous tubercles. It is an uncommon benign neoplasm involving the glomus body, an apparatus that involves in thermoregulation of cutaneous microvasculature. Glomus tumor constitutes 1%-5% of all hand tumors. It usually occurs at the subungual region and more commonly in aged women. Its classical clinical triad consists of pain, tenderness and temperature intolerance, especially cold sensitivity. This study reviews 15 cases of glomus tumor which were analyzed according to its anatomic location, surgical approach and histologic findings. Methods Fifteen patients with subungual glomus tumors of the hand operated on between January 2006 and March 2013, were retrospectively reviewed. Patients were evaluated preoperatively with standard physical examination including ice cube test and Love's test. Diagnostic imaging consisted of ultrasonography, computed tomography, and magnetic resonance imaging. All procedures were performed with tourniquet control under local anesthesia. Eleven patients underwent excision using the transungual approach, 3 patients using the volar approach and 1 patient using the lateral subperiosteal approach. Results Total of 15 cases were reviewed. 11 tumors were located in the nail bed, 3 in the volar pulp and 1 in the radial aspect of the finger tip. After complete excision, patients remained asymptomatic in the immediate postoperative period. In the long term follow up, patients exhibited excellent cosmetic results with no recurrence. Conclusions Accurate diagnosis should be made by physical, radiologic and pathologic examinations. Preoperative localization and complete extirpation is essential in preventing recurrence and subsequent nail deformity.
Purpose: Autologous fat grafting is a popular procedure for the correction of the soft tissue depression and deformity. But there are several issues required to be carefully considered in relation to this procedure, mainly about the unpredictability and the low survival rate of the grafted fat due to absorption and partial necrosis. Sphingosine-1-phosphate (S1P) is a lysophospholipid mediator that has been proposed to promote angiogenesis and to regulate the differentiation of adipose derived stromal cells (ASCs). In this study, we analyzed the viability of the grafted fat tissue mixed with S1P into each 12 nude mice (cann.cg-fox1nu/crlori) compared to the group of mice grafted fat tissue only. Methods: Human aspirated fat was grafted subcutaneously into the backs of 8-week-old nude mice with or without S1P. Eight weeks later, the grafted fat was harvested and the weight and volume were checked. The fat was stained with hematoxylin-eosin and 4',6-diamidino-2-phenylindole. Results: S1P group weighed significantly more than the control group (p=0.009), and the volume from the S1P group was considerably larger than that of the control group (p=0.004) either. In histological features, the surviving layer of the S1P group was thicker than the control group and microvasculature appeared to be prominent in the S1P group, especially in the outer layers. Conclusion: These findings suggest that S1P plays a vital role in the soft tissue augmentation, potentially providing a novel point of the control in adipose tissue for microfat graft.
