Induction of micronuclei in human, rabbit and dog lymphocytes irradiated in vitro with gamma radiation (사람, 토끼 및 개 유래 말초혈액 림프구의 미소핵을 이용한 방사선 피폭의 생물학적 선량측정)

  • The frequencies of gamma-ray-induced micronuclei (MN) in cytokinesis-blocked (CB) lymphocytes at several doses were measured in three donors of three species (human, rabbit, dog). Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was : human : $y=0.1184D+0.01867D^2+0.01$, rabbit : $y=0.0387D+0.00528D^2+0.01$ (y = number of MN/CB cells and D = irradiation dose in Gy). The relative sensitivity of rabbit lymphocytes compared with human lymphocytes was estimated by best fitting linear-quadratic model based on the radiation-induced MN data over the range from 0 Gy to 4 Gy. In the case of MN frequency with 0.2, the relative sensitivities of rabbit lymphocytes was 0.39. These data indicate that the induction of MN in rabbit CB cells following irradiation was much less sensitive to the MN induction effects of gamma-irradiation than those from human. The MN assay with dog lymphocytes was very difficult and time-consumed because the dog PHA-stimulated lymphocytes yielded cultures with very low level of CB cells formation in the condition of this experiment. Our in vitro radiobiological study confirmed that the cytogenetic response obtained in blood from rabbit can be utilized for application in environmental studies.

  • To investigate the transplacental cytogenic effect of air pollutants the authors collected samples from Shinchon, Guro, Banpo and Jungnung-dongs in winter season. The air filters were extracted by mixture of benzene and ethanol, then a certain amount of extracted sustance was injected to pregnant mice at 16th day of gestation. From the fetal liver emulsion polychromatic erythrocytes were collected and stained with Giemsa solution. The cytogenic effect was evaluated by micronucleus test by which numbers of polychromatic erythrocytes containing microunclei (MNPCE) per 1, 000 polychromatic erythrocytes could be counted.

On the Unrecord Species of Paramecia in Korea (한국산 짚신벌레 분류에 관한 연구(I))

  • Ten species of paramecia have been recorded up to the present , but there has been no report on paramecium in Korea. With the purpose of taxonomical study, paramecia were collected as materials at the several areas(ponds and streams) in Seoul from the first of March to the late of October , 1962. From the characteristics of the body-shape and the type and number of micronuclei, the six unrecorded species are identified on this paper as follows : Paramecium caudatum P.aurelia P.multimicromucleaturm P.bursaria P.trichium P.calkinsi

In vivo Genotoxicity of Silver Nanoparticles after 90-day Silver Nanoparticle Inhalation Exposure

  • Objectives: The antimicrobial activity of silver nanoparticles has resulted in their widespread use in many consumer products. Yet, despite their many advantages, it is also important to determine whether silver nanoparticles may represent a hazard to the environment and human health. Methods: Thus, to evaluate the genotoxic potential of silver nanoparticles, in vivo genotoxicity testing (OECD 474, in vivo micronuclei test) was conducted after exposing male and female Sprague-Dawley rats to silver nanoparticles by inhalation for 90 days according to OECD test guideline 413 (Subchronic Inhalation Toxicity: 90 Day Study) with a good laboratory practice system. The rats were exposed to silver nanoparticles (18 nm diameter) at concentrations of $0.7\;{\times}\;10^6$ particles/$cm^3$ (low dose), $1.4\;{\times}\;10^6$ particles/$cm^3$ (middle dose), and $2.9\;{\times}\;10^6$ particles/$cm^3$ (high dose) for 6 hr/day in an inhalation chamber for 90 days. The rats were killed 24 hr after the last administration, then the femurs were removed and the bone marrow collected and evaluated for micronucleus induction. Results: There were no statistically significant differences in the micronucleated polychromatic erythrocytes or in the ratio of polychromatic erythrocytes among the total erythrocytes after silver nanoparticle exposure when compared with the control. Conclusion: The present results suggest that exposure to silver nanoparticles by inhalation for 90 days does not induce genetic toxicity in male and female rat bone marrow in vivo.
