$_L$ =lysine에 의한 Microcystis sp.의 선택적 성장억제 (The Effect of $_L$=lysine on Growth Inhibition of Microcystis sp.)

  • 본 연구는 $_L-lysine \; (_{-2}$, 6-Diaminohexanoic acid)의 영향을 받는 남조류와 남조류에 영향을 주는 $_L-lysine $의 농도를 파악 하고자 20종의 Microcystis.를 실험에 사용하였다. 남조류의 생장 억제 능력은 double-layered agar method와 microplate method를 이용하여 측정하였다. L-lysine의 농도를 100-${\mu}g\; ml-1~300 ${\mu}g\; ml^{-1}$이상에서 처리한 경우 Microcystis ichthyoblabe NIER-10021외 7종의 Microcystis sp.에서 투명대가 형성되었다. Microplate method서는10~500${\mu}g\; ml^{-1}$의 농도에서 생장억제 및 lysis를 나타내어TEk. 그리고 Microcystis viridis NIER-10020 외 3종은 10${\mu}g\; ml^{-1}$이하의 낮은 농도에서도 생장억제 및 분해를 나타내는 높은 활성을 보였다.

유해 남조류 Microcystis와 Dolichospermum에 대하여 선택적 제어가 가능한 생물유래 살조물질 (Naphthoquinone 계열) (Novel Algicidal Substance (Naphthoquinone Group) from Bio-derived Synthetic Materials against Harmful Cyanobacteria, Microcystis and Dolichospermum)

  • 유해 남조류 Microcystis와 Dolichospermum의 친환경적인 제어를 위해 생물유래 물질을 기반으로 한 Naphthoquinone(NQ) 계열의 녹조제어 물질을 개발하였다. NQ의 기본구조를 바탕으로 치환기 ($R_n$)를 변화시켜 총 92종의 NQ 유도체를 합성 확보하였고, 대상 남조류 Microcystis와 Dolichospermum에 대하여 농도별 살조효과를 평가하여 $1{\mu}M$의 농도에서 80% 이상의 제어가 가능한 NQ 화합물을 각각 22종을 선정하였다. 그 중에서, NQ 40 물질은 최적 적용 농도인 $1{\mu}M$에서 Microcystis와 Dolichospermum 종에 각각 99.6%과 100%의 높은 제어효과가 관찰되었다. NQ 40 물질 ($1{\mu}M$)은 규조류 Stephanodiscus, Cyclotella와 와편모조류 Peridinium에는 살조효과가 없었으며, 대상 유해 남조류 Microcystis와 Dolichospermum에만 선택적으로 작용하였다. 또한, NQ 40 물질의 급성독성을 평가 결과, Selenastrum capricornutum, Daphnia magna 및 Danio rario 종에 대하여 $EC_{50}$/$LC_{50}$ 값이 각각 3.2, 14.5 및 $15.7{\mu}M$로 측정되었다. 더불어, D. magna를 이용한 만성독성 평가 결과, NQ 40 물질 $1{\mu}M$에서 생존, 성장, 번식에 대한 영향은 없었다. 따라서, 최종 선정된 NQ 40 물질을 현장에서 $1{\mu}M$로 적용한다면, 다른 수생생물에 미치는 영향 없이 대상 조류인 Microcystis와 Dolichospermum 종만 선택적으로 제어 가능한 친환경적인 살조물질로 사용될 것으로 판단된다.

  • 생물조절기법(Biomanipulation)을 이용한 수질개선 방법으로서, 인공식물섬을 이용한 부유성 수생식물(I. pseudoacorus)은 그 표면적에 따라서 수층으로의 광량 투과를 제어함으로써 미세조류의 광합성 효율을 저해한다. 또한, 수층에서 영양염을 흡수하여, 미세조류의 성장을 억제함으로서 남조류(M. aeruginosa) 대량발생을 제어할 수 있다는 것을 $^{13}C$, $^{15}N$ 추적자 실험을 통하여 정량적으로 확인하였다. 이와 같이 부유수생식물을 이용하여 저수지내 남조류 번성을 억제함으로서 수질 개선에 중요한 역할을 하지만, 동물플랑크톤의 종 조성 변화를 야기할 수 있다는 것을 고려해야 할 것이다. 본 연구 결과는 향후 인공 식물섬을 사용한 남조류 제어 기법에 중요한 자료로 활용될 것으로 사료된다.

Draft Genome Database Construction from Four Strains (NIES-298, FCY-26, -27, and -28) of the Cyanobacterium Microcystis aeruginosa

  • Microcystis aeruginosa is a cyanobacterium that can form harmful algal blooms (HABs) producing toxic secondary metabolites. We provide here draft genome information of four strains of this freshwater cyanobacterium that was obtained by the Next Generation Sequencing approach to provide a better understanding of molecular mechanisms at the physiological and ecological levels. After gene assembly, genes of each strain were identified and annotated, and a genome database and G-browser of M. aeruginosa were subsequently constructed. Such genome information resources will enable us to obtain useful information for molecular ecological studies with a better understanding of modulating mechanisms of environmental factors associated with blooming.

조류기인 유기물질의 제브라피쉬에 대한 뇌파측정기반 독성평가 (Electroencephalography (EEG) based Toxicity Test of Algae Organic Matter on Zebrafish)

  • Harmful algae blooms have become a serious environmental problem in major river basins in Korea. They are known to produce various algal organic matters (AOMs) including intracellular organic matters (IOMs) and extracellular organic matters (EOMs). Generally AOMs cannot be easily removed by coagulation/flocculation process in conventional drinking water plants. AOMs produced by blue-green algae also include various toxins such as Microcystins, Anatoxin-a, and Saxitoxin known to have harmful effects on living organisms in aquatic environment. In this study, toxic effects of EOMs produced by three different algae species (Microcystis sp., Anabaena sp., and Oscillatoria sp.) on zebrafish were investigated using electroencephalography (EEG) recording method, a technology for recording brain activity. Electroencephalographic changes in zebrafish revealed that a low EOM had a negative effect on zebrafish compared to both Anabaena sp. and Oscillatoria sp. at 30 ppm EOM exposures. This result might be due to Microcystins present in EOMs produced by Microcystis sp. As a result of power spectrum density anallysis, exposure to EOMs produced by Microcystis sp. caused a state of vigilance in zebrafish. This EEG based toxicity test can be used to examine effects of harmful materials at low levels on living organisms in an aquatic system.

낙동강 하류에서 식물플랑크톤 생산력과 수질 변화와의 관계 (The Relationship between Phytoplankton Productivity and Water Quality Changes in Downstream of Nakdong River)

  • The relationship between primary productivity and changes in water quality was investigated at Mulgum station, a site downstream of the Nakdong River, Korea. Phytoplankton production was characterized by blooms of Microcystis aeruginosa during the summer and Stephanodiscus hantzschii during the winter. Primary production and secondary production by bacterioplankton ranged from 1.5~53.5 mg-C/ι day and 0.1~0.3 mg-C/ι day, respectively. Distribution of total organic carbon appeared to be highly correlated with phytoplankton biomass, especially during blooms of M. aeruginosa, when particulate organic carbon was 81% of total organic carbon and the main source of organic materials supplied into the water. The correlation coefficient between chlorophyll-a and BOD was 0.86. Thus it was concluded that autochthonous phytoplankton mostly affected the BOD level. Total bacterial numbers were also highly correlated with chlorophyll-a ($r^2$= 0.84) and the bacterial community appears to be regulated by phytoplankton biomass in this area.

Classification of algae in watersheds using elastic shape

  • Identifying algae in water is important for managing algal blooms which have great impact on drinking water supply systems. There have been various microscopic approaches developed for algae classification. Many of them are based on the morphological features of algae. However, there have seldom been mathematical frameworks for comparing the shape of algae, represented as a planar continuous curve obtained from an image. In this work, we describe a recent framework for computing shape distance between two different algae based on the elastic metric and a novel functional representation called the square root velocity function (SRVF). We further introduce statistical procedures for multiple shapes of algae including computing the sample mean, the sample covariance, and performing the principal component analysis (PCA). Based on the shape distance, we classify six algal species in watersheds experiencing algal blooms, including three cyanobacteria (Microcystis, Oscillatoria, and Anabaena), two diatoms (Fragilaria and Synedra), and one green algae (Pediastrum). We provide and compare the classification performance of various distance-based and model-based methods. We additionally compare elastic shape distance to non-elastic distance using the nearest neighbor classifiers.

수종 담수적조 원인종들의 형광특성과 적용연구 (The study on the Fluorescence Characteristics of Several Freshwater Bloom Forming Algal Species and Its Application)

  • The freshwater blooms mainly blue-green algal blooms occur frequently in the lower Naktong River in summer, which provoke many socio-economical problems; therefore, the early detection of bloom events are demanding through the quantitative and qualitative analyses of blue green algal species. The in vivo fluorescence properties of cultured strains of Microcystis aeruginosa, M. viridis, M. wesenbergii, M. ichthyoblabe, Anabaena cylindrica, A. flos-aquae, and Synedra sp. were investigated. Wild phytoplankton communities of the lower Naktong River were also monitored at four stations in terms of their standing stocks, biomass and fluorescence properties compared with its absorption spectram. The 77K fluorescence emission spectra of each cultured strains normalized at 620 nm was very specific and enabled to detect of blue green algal biomass qualitatively and quantitatively. The relative chlorophyll a concentration determined by chlorophyll fluorescence analysis method showed significant relationship with chlorophyll a concentration determined by solvent extraction method ($R^2$ = 0.906), and the blue-green algal cell number determined by microscopic observation ($R^2$ = 0.588), which gives insight into applications to early detection of blue green algal bloom.
