Effect of Xylitol on various Oral bacteria

  • Xylitol is a five-carbon sugar alcohol that reduces the incidence of caries by inhibiting the growth of oral streptococci, including Streptococcus mutans. Since xylitol is transported via the fructose phosphotransferase system, we hypothesized that it could also affect the growth of other oral bacteria strains. We tested the effects of xylitol against non-periodontopathogenic oral bacteria frequently found in healthy subjects as well as periodontopathogens including Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia. With 5% xylitol, Streptococcus vestibularis and Gemella morbillorum showed marked growth inhibition. With 10% xylitol, all of the tested periodontopathogens and Actinomyces naeslundii showed marked growth inhibition, whereas the growth inhibition of Neisseria mucosa, Neisseria sicca and Veillonella parvula was mild only. Xylitol is a widely used sweetener and the concentration used in our experiment is easily achieved in the oral cavity. If xylitol reduces the growth of periodontopathogens more preferentially, it could also reduce the prevalence of these pathogens and have clinical utility in the prevention or treatment of periodontal disease.

Isolation of vibrio species from cultured flounders (paralichthys olivaceus) with uicers and ascites in the southern coast of Korea during the winter season (동절기 한국 남해안의 궤양증 및 복수증 양식 넙치로부터 vibrio종의 분리)

  • this study was performed for the purpose of surveying the distribution and characteristics of the Vibrio species in the diseased farm flounders (Paralichthys olivaceus) during the winter season in Korea. Samples were collected along the southern coast in the Sinam, Keoje Island, and Teosu areas from November '90 to Feburary '91. Nine species of Vibrio were identified as V. tubiashii (85 strains), V. damsela (78 strains), V. anguillarum (21 strains), V. compbelli (9 strains), V. fluvialis (9 strain), V. costicola (1 strain), V. alginolyticus (1 strain), V. gazogenes (1 strain), V. marinus (1 strain), and unidentified 3 groups of bacteria (154 strains) which may have related with V. tubiashii were isolated. Eleven kinds of discs (BBL Co.) were used for antibiotic susceptibility test and 116 strains were selected for the experiment. Most bacteria were powerfully inhibited their growth by 4 antibiotics such as cephalothin, chloramphenicol, colistin, and tetracycline, but they were poorly inhibited by 4 antibiotics such as carbenicillin, kanamycin, penicillin G, and streptomycin.

  • We isolated Streptomyces spry. from Korean soil, which showed high cytotoxicity against tumor cell lines, L1210 and P388Dl. Among 30 strains, three strains (DKM 104, DKM 128, DKM 409) were appeared to possess plasmid. Strain DKM 104 and DKM 128 had two CCC(Covalently Closed Circular) form plasmid, about 20 Kb in size and about 1 Kb compared with λ Hind III DNA size marker. And strain DKM 409 had three plasmids, among which two plasmic were CCC form about 20 Kb and about 20 Kb compared with same size marker. To find out whether plasmid involved in production of antitumor agent or not, we performed to curing experiment. Comparing cytotoxicity between culture filtrate of plasmid-containing strains and cured strains, we knew that only the cytotoxic activity of the strain DKM 128 was involved in plasmid.

  • Pseudomonas sp. EL-041 was previously isolated from phenol-acclimated activated sludge. This bacterium was capable of degrading phenol and cometabolizing trichloroethylene (TCE). In this study, we report the identification of trichloroethylene- degrading enzyme in Pseudomonas sp. EL-041 by the investigation of enzyme activity and DNA sequencing of specific phenol oxygenase gene. As the results of experiment, trichloroethylene-degrading enzyme in Pseudomonas sp. EL-041 was monooxygenase and suspected to phenol hydroxylase.

  • A portion (7.4 kb) of ribosomal DNA tandem repeat unit from a genome of the mushroom T. matsutake has been cloned. A 1.75 kb EcoRI fragment was cloned first using S. cerevisiae 255 rRNA gene as a probe, and this was then used for further cloning. A chromosomal walking experiment was carried out and the upstream region of the 1.75 kb fragment was cloned using SmaI/BamHI enzyme, the size was estimated to be 5.2 kb in length. Part of the downstream region of the 1.75 kb fragment was also cloned using XbaI/BamHI enzymes. Restriction enzyme maps of three cloned DNA fragments were constructed. Northern hybridization, using total RNA of T. matsutake, and the restriction fragments of three cloned DNAs as probes, revealed that all four ribosomal RNA genes (large subunit[LSU], small subunit [SSU], 5.85 and 5S rRNA genes) are present in the cloned region. The gene organization of the rDNA are regarded as an intergenic spacer [IGS]2 (partial) - SSU rRNA - internal transcribed spacer [ITS]1 - 5.8S rRNA - ITS2 - LSU rRNA - IGS1 -5S rRNA - IG52 (partial).

The Biological Characteristics and Drug Resistance of Vibrio Species (Vibrio균속의 생물학적 성상 및 약제내성에 관하여)

  • In the present experiments, isolated Vibrio species from marine and clinical specimens from July, 1985 to October, 1986, had the results as follows: 1. The 55 strains of Vibrio were isolated and identified; Vibrio parahaemolyticus was 35 strains, Vibrio vulnificus was 10 strains, Vibrio alginolyticus was 10 strains. 2. In the K-serotyping of Vibrio parahaemolyticus, fourteen serotypes identified but three were not strains typable by the availble K-antisera. 3. In the Kanagawa phenomenon experiment of Vibrio parahaemolyticus, it proved positive reaction, 14 of 15 strains(93%) isolated from the patient and 13 of 20 strains(65%) isolated from the nature. 4. In twelve antibiotic resistance experiments, Vibrio parahaemolyticus and Vibrio alginolyticus showed 100% resistance on ampicilline, but Vibrio vulnificus showed 100% sensitivity. But all of them proved 100% sensitivity on chloramphenicol, tetracycline, nalidixic acid. 5. In the antibiotic resistance patterns, Vibrio parahaemolyticus proved that 15 strains(43%) resisted on 4 antibiotics and 5 strains(14%) resisted on 7 antibiotisc and. Vibrio vulnificus proved that 1 strain(10%) resisted on 2 antibiotics and 6 strains(60%) without resistance, Vibrio alginolyticus proved that 7 strains(70%) resisted on 3 antibiotics and 2 strains(20%) resisted on 8 antibiotics.

  • Japanese encephalitis(JE) shows its explosive epidemicity in the temperate zone of Asia but little is known on the overwintering mechanism. One of the hypotheses on the overwintering mechanism is that the virus overwinters in the hibernating animals. There has been no report on the proliferation of JE virus(JEV) or antibody formation in the snakes. The purpose of this experiment is to explore the mutual relationship between JEV and snake and to clarify whether JEV proliferates and induce antibody formation in snakes. Three species of non-poisonous common snakes were employed. Precipitation test was carried out after injecting calf serum and, HI and neutralization tests were done by injecting JEV into the snakes. The gamma globulin fraction of pre- and post-injection serum were compared by paper chromatography. According to the results, precipitating antibody reaction to calf serum could be observed only at $4^{\circ}C$. It was failed to demonstrate HI antibody formation but neutralizing antibody could be detected in one of the 9 snakes. Although antibody could not be detected in test-tube, tile result of paper chromatography shows the remarkable increase of gamma globulin fraction after the injection. Above results are strongly indicating the antibody formation in the snakes.

Determination of Mineral Components in the Cultivation Substrates of Edible Mushrooms and Their Uptake into Fruiting Bodies

  • The mineral contents of the cultivation substrates, fruiting bodies of the mushrooms, and the postharvest cultivation substrates were determined in cultivated edible mushrooms Pleurotus eryngii, Flammulina velutipes, and Hypsizigus marmoreus. The major mineral elements both in the cultivation substrates and in the fruiting bodies were K, Mg, Ca, and Na. Potassium was particularly abundant ranging 10${\sim}$13 g/kg in the cultivation substrates and 26${\sim}$30 g/kg in the fruiting bodies. On the contrary, the calcium content in the fruiting bodies was very low despite high concentrations in the cultivation substrates, indicating Ca in the cultivation substrates is in a less bio-available form or the mushrooms do not have efficient Ca uptake channels. Among the minor mineral elements determined in this experiment, Cu, Zn, and Ni showed high percentage of transfer from the cultivation substrates to the fruiting bodies. It is noteworthy that the mineral contents in the postharvest cultivation substrates were not changed significantly which implies that the spent cultivation substrates are nutritionally intact in terms of mineral contents and thus can be recycled as mineral sources and animal feeds.

  • This experiment was carried out to investigate the yeasts for the brewing of red pepper paste during the fermentation periods. The yeasts in the red pepper paste were isolated and identified, and they were classified by coloring with the treatment of TTC(2, 3, 5, triphenyltetrazolium chloride) agar and counted in process of time.

  • A simple and effective procedure is described for semi-purification of viable Myco. leprae from biopsied lepromatous nodules by trypsinization and high-speed centrifugation. An unique characteristic of this method is a complete omission of conventional grinding or homogenization of minced lepromatous tissues prior to purification. Inoculation of trypsin-purified preparation of Myco. leprae into foot pads and ear lobes of Korean chipmunk(Tamias sibiricus asiaticus, Gmelin) resulted in apparent increases in total number of acid-fast bacilli per inoculated tissue 8 and 12 months after inoculation.
