영월과 신림에 있어서 소나무낙엽의 분해와 Microbial Population 의 소장 비교 (Comparison of the Pine Litter Decompositon and Microbial Population Change at Youngwal with Those at Sinlim)

  • The decomposition of litters of Pinus densiflora and the growth of microbial populations in a calcareous region, Youngwal were compared with those in a noncalcareous region, Sinlim. The decay rate of litter in Pinus densiflora in Youngwal was 0.128 and that in Sinlim was 0.096. The differences in the populations of soil bacteria and total microorganisms between the two regions were signficant at the 5% level, but that of fungi and actinomycetes was not at that level. The differences in the content of calcium and pH value of soil between the two regions were significant at the 1% level. The excessive content of calcium became to increase pH value, in turn the high pH decreased the content of available phosphorus in soil. The vertical distribution of the content of available phsophorus was consistent with that of the populations of fungi and actinomycetes in Youngwal.

Effects of Aspergillus Oryzae Culture and 2-Hydroxy-4-(Methylthio)-Butanoic Acid on In vitro Rumen Fermentation and Microbial Populations between Different Roughage Sources

  • An in vitro experiment was conducted to evaluate the effects of Aspergillus oryzae culture (AOC) and 2-hydroxy-4-(methylthio)-butanoic acid (HMB) on rumen fermentation and microbial populations between different roughage sources. Two roughage sources (Chinese wild rye [CWR] vs corn silage [CS]) were assigned in a $2{\times}3$ factorial arrangement with HMB (0 or 15 mg) and AOC (0, 3, or 6 mg). Gas production (GP), microbial protein (MCP) and total volatile fatty acid (VFA) were increased in response to addition of HMB and AOC (p<0.01) for the two roughages. The HMB and AOC showed inconsistent effects on ammonia-N with different substrates. For CWR, neither HMB nor AOC had significant effect on molar proportion of individual VFA. For CS, acetate was increased (p = 0.02) and butyrate was decreased (p<0.01) by adding HMB and AOC. Increase of propionate was only occurred with AOC (p<0.01). Populations of protozoa ($p{\leq}0.03$) and fungi ($p{\leq}0.02$) of CWR were differently influenced by HMB and AOC. Percentages of F. succinogenes, R. albus, and R. flavefaciens (p<0.01) increased when AOC was added to CWR. For CS, HMB decreased the protozoa population (p = 0.01) and increased the populations of F. succinogenes and R. albus ($p{\leq}0.03$). Populations of fungi, F. succinogenes (p = 0.02) and R. flavefacien (p = 0.03) were increased by adding AOC. The HMB${\times}$AOC interactions were noted in MCP, fungi and R. flavefacien for CWR and GP, ammonia-N, MCP, total VFA, propionate, acetate/propionate (A/P) and R. albus for CS. It is inferred that addition of HMB and AOC could influence rumen fermentation of forages by increasing the number of rumen microbes.

Functional Metagenomics using Stable Isotope Probing: a Review

  • The microbial eco-physiology has been the vital key of microbial ecological research. Unfortunately, available methods for direct identity of microorganisms and for the investigation of their activity in complicated community dynamics are limited. In this study, metagenomics was considered as a promising functional genomics tool for improving our understanding of microbial eco-physiology. Its potential applications and challenges were also reviewed. Because of tremendous diversity in microbial populations in environment, sequence analysis for whole metagenomic libraries from environmental samples seems to be unrealistic to most of environmental engineering researchers. When a target function is of interest, however, sequence analysis for whole metagenomic libraries would not be necessary. For this case, nucleic acids of active populations of interest can be selectively gained using another cutting-edge functional genomic tool, SIP (stable isotope probing) technique. If functional genomes isolated by SIP can be transferred into metagenomic library, sequence analysis for such selected functional genomes would be feasible because the reduced size of clone library may become adequate for sequencing analysis. Herein, integration of metagenomics with SIP was suggested as a novel functional genomics approach to study microbial eco-physiology in environment.

이산화염소수를 이용한 계사 내 깔짚의 미생물 수 저감화 (Microbial Inactivation of Chicken Cage Litter by Aqueous Chloride Dioxide)

  • 본 연구에서는 이산화염소수를 이용한 계사 내 깔짚의 미생물 수 저감화를 통한 깔짚의 미생물학적 안전성 확보를 위하여 연구를 수행하였다. 사용하기 전의 깔짚에는 coliform, E. coli, Listeria spp., yeasts and molds 및 total aerobic bacteria가 높은 수준으로 검출되었으며 사용기간이 늘어남에 따라 미생물 수가 증가하였다. 500 ppm 이산화염소수를 이용한 살균처리에서는 coliform, E. coli, Listeria spp., yeasts and molds, total aerobic bacteria 및 Salmonella spp.의 균수가 모든 깔짚 시료에서 처리횟수에 따라 유의적으로 감소하였다. 사용하지 않은 깔짚에서는 coliform, E. coli, Listeria spp., yeasts and molds 및 total aerobic bacteria 수가 각각 4.47, 1.29, 1.23, 3.24 및 5.2 log CFU/g로 감소하였고, 1주 및 5주 사용한 깔짚의 경우도 모든 미생물 수가 현저하게 감소하였다. 따라서 본 연구 결과, 이산화염소수를 이용한 계사 내 깔짚의 미생물 수 저감화 방법이 도계된 계육의 미생물학적 안전성을 향상시키기 위한 방법으로 적합하다고 판단된다.

  • The soil of the reclaimed tidal land, located in Chogi-ri, Is. Kanghwa, Korea was used in this experiment. The experimented soil samples were collected from 18 sites with its time elapsed after the shore-protection works, soil-depth and the vegetation of saline plants, and at each site samplings were conducted monthly from March through October, 1968, for the purposes of examining the changes of microbial populations for the microbes such as bacteria, actinomycetes and fungi, by using the dilution plate method. The numbers of the microbes in these soils generally showed lower levels comparing with those of other soils. The more time elapsed after the reclamation, the higher numbers of the microbes inhibited the soils. Higher populations were there in the surface soils than in the lower part of the area. The surface soils included comparatively better conditions in aeration and contents of organic matter than in the lower part, and this fact was. same as in general soils. However, not so was this in the case of March, April and October due to the higher soil temperatures in the lows. At the experimental sites where the halophytes such as Salicorniu were grown vigourously, the more densly the plants grew, the higher populations of actinomycetes and fungi were, but not in the case of bacterial population. This means, in this soil with dense Salicornia, it is difficult to obtain good-natured soils in short time without a higher population of bacteria. For the rapid utilization of the land soil, in this view of point, the methods increasing the number of bacteria in the soil are needed as well as the cultivation and harvesting Salicorniu which indicated in the privious paper(Hong, et al., 1969a). According to the results of this experiment, the changes of soil-microbial populations in the reclaimed tidal land soil containing high salinity depend deeply upon the interrelations of many environmental factors such as soil-salinity, soil-components and contents, concentration of organic matters, pH, aeration, and air and soil temperatures, as in the general soils.

Inactivation of Listeria monocytogenes and Campylobacter jejuni in Chicken by Aqueous Chlorine Dioxide Treatment

  • Aqueous chlorine dioxide $(ClO_2)$ treatment was used for the inactivation of Listeria monocytogenes and Campylobacter jejuni in chicken. Chicken breasts and legs were inoculated with $8{\sim}9log\;CFU/g$ of Listeria monocytogenes and Campylobacter jejuni, respectively, and then treated with 0, 50, and 100 ppm of $ClO_2$ solution. Aqueous $ClO_2$ treatment decreased the populations of the pathogenic bacteria on the chicken samples. One hundred ppm $ClO_2$ treatment on the chicken breast and leg reduced the populations of Listeria monocytogenes and Campylobacter jejuni by $0.61{\sim}1.93\;and\;0.99{\sim}1.21log\;CFU/g$, respectively. Aqueous $ClO_2$ treatment affected the microbial growth during storage at $4^{\circ}C$ by decreasing the initial microbial populations. These results clearly suggest that aqueous $ClO_2$ treatment should be useful in improving the microbial safety of chicken during storage and extending the shelf life.

FAME Analysis to Monitor Impact of Organic Matter on Soil Bacterial Populations

  • In order to assess the effects of organic fertilizer on soil microbial community structure and diversity in the greenhouse fields, fatty acid methyl ester (FAME) was analyzed by the MIDI (Microbial ID, Inc., Newark, DE, U.S.A.) system and enumerations were performed. In relation to bacterial division of each sample, low GC Gram-positive bacteria were predominant among bacteria cultured on aerobic bacteria media. On the other hand, alpha subdivision was predominant on proteobacteria of control and OM (organic matter) 1 treated plot, and Flavobacterium spp. existed in OM2 plot on crystal violet media of all samples. Shannon-weaver Index (H) of OM1 plot varied most by 1.9 and 5.0 among bacteria cultured on aerobic bacteria media and crystal violet media, respectively. Our results revealed that addition of the organic wastes to soil led to a highly diverse microbial community, but the excessive amounts of organic and mineral fertilizer applied in the greenhouse fields produced excess nutrients in soil and led to simplification on bacterial populations.

Effect of Electron Beam Irradiation on Microbial Growth and Qualities in Astragalus membranaceus

  • Electron beam irradiation was applied to examine the microbial growth and qualities of vacuum-packaged Astragalus membranaceus, a Korean medicinal herb. Samples were irradiated at dose of 2, 4, 8, 12, and 16 kGy, respectively. Microbiological data on A. membranaceus showed that populations of total bacteria, yeast and mold, total coliforms were significantly reduced with increase of irradiation dose. Populations of microorganisms in A. membranaceus were decreased by 2-3 log cycles at 8 kGy irradiation. Color measurements showed that electron beam treatment caused negligible changes in Hunter color L, a, and b values of A. membranaceus. Sensory evaluations showed that there were no significant changes among the samples. These results suggest that electron beam irradiated A. membranaceus have better microbial safety and qualities, compared with the non-irradiated control.

  • The balance of microbial populations in the gastrointestinal (GI) tract of all warm-blooded animals is critical to the maintenance of health and resistance to disease. The composition of the populations can be altered by diet and environment, making the host animal susceptible to disease, and reducing growth rate and feed efficiency. Some feed additives including antimicrobial agents, prohiotics or yucca extract have been used to promote growth and feed utilization. There is evidence that part of growth-promoting effect of those feed additives results from the suppression of microbial urease activity or ammonia production in the GI contents of animals. Over 200 microbial species have been known to produce urease and the product of urea hydrolysis, ammonia, is toxic to animals. Carefully tested probiotics or other urease-suppressing agents can be a possible alternative to antimicrobial agents including antibiotics as growth promotants used for animals feeds.

Effect of Aqueous Chlorine Dioxide Treatment on the Microbial Growth and Qualities of Iceberg Lettuce during Storage

  • Effects of aqueous chlorine dioxide ($ClO_2$) treatment on the microbial growth and the quality of iceberg lettuce during storage were examined. Lettuce samples were treated with 0, 5, 10, and 50 ppm of $ClO_2$ solution and stored at $4^{\circ}C$. Aqueous $ClO_2$ treatment significantly decreased the populations of total aerobic bacteria, yeasts and molds, and coliforms on the shredded lettuce. Fifty ppm $ClO_2$ treatment reduced the initial populations of total aerobic bacteria, yeasts and molds, and coliforms by 1.77, 1.34, 1.10 log CFU/g, respectively. Aqueous $ClO_2$ treatment caused negligible changes in the Hunter color L, a, and b values during storage. Sensory evaluations exhibited that there were no significant changes among treatments. These results indicate that the aqueous $ClO_2$ treatment can be useful in improving the microbial safety of the iceberg lettuce during storage and extending the shelf life.
