The Principle and Application of Bioremediation (생물학적 복구법(Bioremediation)의 원리와 응용)

  • The efficiency of bioremedation can be measured by the enumeration of microorganism, respiration rate and decomposition rate. The side-effect can be measured by using Daphnia, oyster larvae and rainbow trout. Oxygen transfer could be a problem in the on-site treatment. For these, hydrogen peroxide can be used for solvents such as benzenes. Oleophilic nitrogen and phosphorus can be added for the treatment of oil pollution. Mixed microbial population or pure culture can be used for the inoculum. The pure culture used is Pseudomonas and Phanerochate. Sometimes enzymes are added and Photodegadation is coupled to increase the efficiency. For the treatment of oil pollution residue on soil such as waste lubrication oil and machine oil sludges, top soil of 15cm∼20cm depth is plowed and oil residue with approximately 5% concentration is applied. The optimum pH range is 7∼8, the ratio of phosphorus to hydrocarbon is 1:800. Appropriate drainage is necessary. For the treatment of marine oil pollution residue, addition of oleophilic fertilizer is effective. Air pollutiant such as oder can be treated by bioremediation. In this case, biofilters or biosrubbers are used for the reactor.

Development and Evaluation of Protected Fat in Wheat Straw Based Total Mixed Ration

  • Ca salt of soybean oil (PSO) and that of mustard oil plus mahua oil (PMOMO) (50:50) were prepared using double decomposition method, and further tested for their fatty acid composition and degree of saponification. Furthermore, the different levels of protected fat of PSO and PMOMO were evaluated in wheat straw based total mixed ration (TMR) in vitro. Results indicated that capric, lauric, myristic, palmitic, steric, oleic, linoleic, leinolenic acids were traces, traces, traces, 10.00, 2.00, 25.00, 58.50, 5.0% in PSO while the corresponding values in PMOMO were 1.08, 0.28, 0.45, 16.9, 12.95, 44.38, 17.46 and 6.50%, respectively. The degree of saponification of both protected fat supplements was more than 80%. Six treatment combinations were tested I.e., blank without feed and fat supplement (T1); control diet with out fat supplement (T2); control diet plus bypass fat supplement (PSO) so that diet contain 5% fat (T3); control diet plus bypass fat supplement (PSO) so that diet contain 7.5% fat (T4); two more diets viz. T5 and T6 were formulated using bypass fat supplement from PMOMO containing 5 and 7.5% fat respectively. TMR was prepared using 50% concentrate mixture and 50% wheat straw. Result indicated that TVFA, $NH_3-N$,TCA-N, total-N and total gas production were increased in treatment diets at 7.5% level of supplementation, however, fermentation pattern remain similar at 5.0% level of supplementation with respect to control diet. Nevertheless, IVDMD and IVOMD values remained unchanged, rather non-significant at both fat levels and with the both fat sources. On the basis of results it was concluded that Ca-salt of Soybean oil or Mustard plus Mahua oil did not show any negative effect either on digestibility or on microbial protein synthesis in rumen, hence the dietary fat upto 7.5% level in total mixed ration based on wheat straw, could be safely used without any adverse effect on rumen fermentation.

  • Alakline peroxide pretreatment for the delignification of poplar wood was performed. sinceit is a simple and efficent method for enhancing the enzymatic digestibility of wood residues. Approximately one-half of their lignin and most of the hemicellulose present in poplar wood were removed when the wood sawdust was reacted at 25$^{\circ}C$ for 100 hrs in an alkaline solution (pH 11.5) of 1% peroxide. The rate of decomposition as well as the saccharification efficiency were enhanced up to 350% and 260% respectively in comparision with those of the controll. This enhancement is comparable with that pretreated with 1% sodium hydroxide and 20% peracetic acid successively. The advantages of alkaline peroxide as delignifying agents against other chemicals were also discussed.

Characteristics of digestive enzyme activity, antibiotic resistance, and pathogenicity of bacteria inhabited in animal feed resources (사료자원에 서식하는 세균의 소화효소활성, 항생제내성 및 병원성에 관한 특성)

  • Among different types of spoilage, microbial contamination can cause feed decomposition, which results in decreases in feed intake and productivity, infection, and breeding disorder. During the storage time, various microbes have a chance to inoculate with depreciation of feed and to infect the animals. We investigated bacteria that inhabit diverse feed ingredients and complete feed which have been stored for a few months. We isolated and identified 30 genera and 62 species of bacteria. Among these 62 species, 21 species were of non-pathogenic bacteria, 18 species were of pathogenic bacteria, 9 species were of opportunistic pathogens, and 14 species were of unknown bacteria. Pantoea allii and 24 species showed proteolytic enzyme activity. We also confirmed that 6 species including Pseudomonas psychrotolerans showed ${\alpha}$-amylase activity, and 29 species including Burkholderia vietnamiensis showed cellulase activity. Microbacterium testaceum and 3 species showed resistance to Ampicillin, Kanamycin, Streptomycin, Gentamicin, Carbenicillin, and Erythromycin ($50{\mu}g/mL$). Using mealworm larvae (Tenebrio molitor L.) as a model for pathogenicity, we confirmed that 8 species including Staphylococcus xylosus had pathogenicity for mealworm larvae. Especially, Enterobacter hormaechei, Staphylococcus xylosus, and Staphylococcus hominis were reported as being pathogenic for humans. This research suggests that hygienic management of animal feed is essential because beneficial and harmful bacteria can inhabit animal feed differently during storage and distribution.

Effect of Crop Rotation System on Soil Chemical Properties and Ginseng Root Rot after Harvesting Ginseng (인삼 연작지에서 윤작물 작부체계가 토양화학성 및 인삼뿌리썩음병 발생에 미치는 영향)

  • Background: The application of crop rotation systems may reduce the occurrence of soil-borne diseases by releasing allelochemicals and by subsequent microbial decomposition. Methods and Results: For reduction of ginseng root rot by the crop rotation system, after harvesting 6-year-old ginseng, fresh ginseng was grown along with continuous cultivation of sweet potato, peanut, and bellflower. Growth of 2-year-old ginseng was significantly inhibited in the continuous cultivation than in the first cultivation. Sweet potato, peanut and bellflower cultivations assisted in obtaining normal yields of ginseng in the first year after the harvest of 6-year-old ginseng. Salt concentration, potassium and sodium contents were gradually decreased, and, organic matter was gradually increased through cirp rotation. Phosphate, calcium and magnesium contents were not altered. The density of the root rot fungus was gradually decreased by the increase in crop rotation; however it was decreased distinctly in the first year compared to the second and third year. The severity of root rot disease tended to decrease gradually by the increase of crop rotation. Conclusions: Short-term crop rotation for three years promoted the growth of ginseng, however root rot infection was not inhibited significantly, although it was somewhat effective in lowering the density of the root rot pathogen.

Microbial Decomposition of s-Triazine Herbicides, Atrazine and Simazine by a TNT-degrading Bacterium (TNT-분해세균에 의한 s-Triazine계 제초제인 Atrazine과 Simazine의 미생물학적 분해)

  • The purpose of U7is work was to iilvestigate the degradation of s-h~azine hel-hicidcs, ahilzine and simazine by TNT-degrader under several relevaut physicochemical environ~nental parameters. TNT-degrader showed effective degradability of atrazine and snnazine as well. Both atrazme (GO 1i1~11) and simazine ( 4 5 rng//) were completely degraded within 30 hrs and 4 days of incubation, respectively. As d ~ e concentrations of atrazine and sunazine increased in the media, the degradation ofthose compounds were delayed. Additional caubans were essential to degrade atrazine and simazule, and no degradation was achieved in the absence of additional carbons. The effect of supplemented nitrogens on the degradation of atrazine and sunazine was evalualed. Addition of a suppleinented nitrogen in he growth medium containing ah-azine or siinazine showed partial degr-adation olihose herbicides duriug the incubation period. However, complete degradation of atrazine and simazu~e was examined ul the absence or any supplemented nitrogens. Addltion of yeast extract in this study was inhibilory to atrazine aud siinazine degradations, respectively. TNT-degrader was a small Gram-negative cocco-bacillus. Physiological analysis using BIOLOG sysleln revealed that this strain was Ste~~ol~~opl~orno~~ns rrialtophilia.

Effect of Hydraulic Loading Rate of a Continuous Stirred Tank Reactor(CSTR) on the Removal Efficiency of Pollutants of Pig Slurry (CSTR의 수리학적 부하율이 돈슬러리의 유기물 및 영양염류 저감효율에 미치는 영향)

  • Anaerobic digestion is a naturally occuring microbial process involving the decomposition of organic materials such as livestock manure. This study explores the effect of the operating conditions, HRT (Hydraulic Retention Time) and feeding frequency on treatment efficiency for digestion of pig slurry, which has been one of most difficult organic waste for proper treatment in livestock production industry in Korea at the present time. The pilot-scale CSTR of 5 m3 in volume was designed. manufactured, and operated at the temperature of 35$\pm$1$^{\circ}C$. The digester was designed to hydraulically stir for complete mixing and to supply heat from the water bath to maintain mesophilic temperature. The HRT of the digester for Test 1 and Test 2, and Test 3 was set for 17 days and 13 days respectively and pig slurry was fed once a day with 300$\ell$ each for Test 1 and Test 3, while twice with 150$\ell$each for Test 2. Test 2 showed better performance by increase of 4% in VS removal efficiency and 5% in biogas production rate. This is mainly attributed to smaller temperature drop by feeding frequently with half amount, which eventually led to lesser impact on anaerobic mocrobes in the digester. Test 2 maintained optimum pH 7.8 which uplifted the activaton of sulfur-reduction bacteria, alkalinity of around 4,000mg/$\ell$, VA of over 3,000mg/$\ell$ for whole period of experiment. Further research may require to provide the practical operation strategy of anaerobic treatment system for treatment of pig slurry.

Changes of Bacterial Population during the Decomposition Process of Red Tide Dinoflagellate, Cochiodinium polykrikoides in the Marine Sediment Addition of Yellow Loess (황토첨가 해양퇴적물에서 적조생물 Cochiodinium polykrikoides 분해중 세균군집의 변동)

  • To investigate the effects of yellow loess on the microbial community after applying into C. polykrikoides as a red tide centrol method during decomposition process, we conducted this study using microcosm experiments, which consisted of sediment collected from Jinhae and Masan bay. The composition, number of bacteria and respiratory electron transport system activity (ETSA) were analyzed. The number of heterotrophic bacteria examined in the samples of both stations reached maximum value within 12 hrs with $10^7$ cells/dry g, independent with the yellow loess applied. In addition, a differenee in the variation of heterotrophic bacterial composition was not observed by adding the yellow loess, and Vibrio spp. always appeared during the culture periods, However, in day 8 culture, the sulfate reducing bacteria was $3.8\times10^7$ cells/dry g in Masan bay and $5.5\times10^6$ cells/dry g in Jinhae bay samples without yellow loess, and these were 120, 350 fold-and 160, 420 fold-increased when yellow loess was added (1 : 1, 1 : 2). The average ETSA was 6.8$\~$7.6 $\mu$g formazan $h^{-1}$ dry $g^{-1}$ independently with yellow loess in aerobic condition for both samples, but activity was decreased by addition of yellow loess in anaerobic. Thus the addition of yellow loess to marine sediment seems to have an effect to inhibit the anaerobic decomposition process and growth of sulfate reducing bacteria which lead to the bad condition of marine environments.

Effects of Cover Plants on Soil Microbial Community in Organic Apple Orchards (피복작물이 유기 사과과원 토양미생물상에 미치는 영향)

  • Organic fruit production has increased due to consumer's interest and government's political support for environmentally-friendly agriculture. The aim of this study was to investigate the effects of cover plants on soil microbial community and establish the fruit cultivation method by organic farming techniques. Cover plants used as an organic nutrient source in an apple orchard were rye and barley, the Gramineae and red clover and hairy vetch, the Leguminosae. In the effects of cover plants on the soil chemical characteristics, the soil pH values were higher than that of conventional organic pear orchard. The content of P showed no significant difference between control and cover plant plots. Organic matter level was similar in control and Gramineae cover plant plots, while organic matter content in cover plants belong to Leguminosae was lower than that of control plot. K content was lower in the plots treated with rye and red clover than control plot, while K content in hairy vetch treated plot was higher than control plot. Ca content was lower in control plot than in cover plant treated plots. Concentrations of Mg in the plots treated with barley and hairy vetch was lower than control plot. In August rye and red clover covered soil showed higher bacterial community density than that of control soil and barley treated soil showed highest Actinomycetes community density among treatments. Barley and hairy vetch soils showed higher level of fungi community density than that of control soil in August. In pyrosequencing analysis barley treated soil showed highest distribution ratio of Actinomycetes among treatment. Our findings might be used as basic data for choosing cover plant with effective organic matter decomposition and nutrition supply capacity.

Quality Stability of High Pressure Boiled Extract of Ogol Chicken during Storage Periods (오골계 증탕액의 저장 및 관능 특성)

  • This study was conducted to examine quality stability during storage periods, herb high pressure boiled extract(HPBE)(T$_1$), Korean Ogol chicken HPBE(T$_2$), cross-bred Ogol chicken HPBE(T$_3$), cross-bred Ogol chicken meat hydrolyzed with flavourzyme(T$_4$) were pouch packaged and stored at 37$\^{C}$. After each period, TBARS, VBN, pH, total microbial counts and sensory properties were determined and the results were as follows. There was no noticeable difference in TBARS value until 42 days at the ambient environment among the treatments, but T$_4$ showed a significantly higher TBARS value at 56 days. There was a tendency for a higher protein decomposition as storage time increased, and in particular at 56 days, T$_1$ group showed a significantly higher values than other groups. Given to the sensory properties in which overall sensory preference decreased after 42 day, it was considered that the maximum storage time for the extract was less than 42 days at 37$\^{C}$.
