Polynuclear aromatic hydrocarbon (PAH) compounds are highly carcinogenic chemicals and common groundwater contaminants that are observed to persist in soils. The adherence and slow release of PAHs in soil is an obstacle to remediation and complicates the assessment of cleanup standards and risks. Biological degradation of PAHs in soil has been an area of active research because biological treatment may be less costly than conventional pumping technologies or excavation and thermal treatment. Biological degradation also offers the advantage to transform PAHs into non-toxic products such as biomass and carbon dioxide. Ample evidence exists for aerobic biodegradation of PAHs and many bacteria capable of degrading PAHs have been isolated and characterized. However, the microbial degradation of PAHs in sediments is impaired due to the anaerobic conditions that result from the typically high oxygen demand of the organic material present in the soil, the low solubility of oxygen in water, and the slow mass transfer of oxygen from overlying water to the soil environment. For these reasons, anaerobic microbial degradation technologies could help alleviate sediment PAH contamination and offer significant advantages for cost-efficient in-situ treatment. But very little is known about the potential for anaerobic degradation of PAHs in field soils. The objectives of this research were to assess: (1) the potential for biodegradation of PAH in field aged soils under denitrification conditions, (2) to assess the potential for biodegradation of naphthalene in soil microcosms under denitrifying conditions, and (3) to assess for the existence of microorganisms in field sediments capable of degrading naphthalene via denitrification. Two kinds of soils were used in this research: Harbor Point sediment (HPS-2) and Milwaukee Harbor sediment (MHS). Results presented in this seminar indicate possible degradation of PAHs in soil under denitrifying conditions. During the two months of anaerobic degradation, total PAH removal was modest probably due to both the low availability of the PAHs and competition with other more easily degradable sources of carbon in the sediments. For both Harbor Point sediment (HPS-2) and Milwaukee Harbor sediment (MHS), PAH reduction was confined to 3- and 4-ring PAHs. Comparing PAH reductions during two months of aerobic and anaerobic biotreatment of MHS, it was found that extent of PAHreduction for anaerobic treatment was compatible with that for aerobic treatment. Interestingly, removal of PAHs from sediment particle classes (by size and density) followed similar trends for aerobic and anaerobic treatment of MHS. The majority of the PAHs removed during biotreatment came from the clay/silt fraction. In an earlier study it was shown that PAHs associated with the clay/silt fraction in MHS were more available than PAHs associated with coal-derived fraction. Therefore, although total PAH reductions were small, the removal of PAHs from the more easily available sediment fraction (clay/silt) may result in a significant environmental benefit owing to a reduction in total PAH bioavailability. By using naphthalene as a model PAH compound, biodegradation of naphthalene under denitrifying condition was assessed in microcosms containing MHS. Naphthalene spiked into MHS was degraded below detection limit within 20 days with the accompanying reduction of nitrate. With repeated addition of naphthalene and nitrate, naphthalene degradation under nitrate reducing conditions was stable over one month. Nitrite, one of the intermediates of denitrification was detected during the incubation. Also the denitrification activity of the enrichment culture from MHS slurries was verified by monitoring the production of nitrogen gas in solid fluorescence denitrification medium. Microorganisms capable of degrading naphthalene via denitrification were isolated from this enrichment culture.
미생물에 의한 금속이온의 환원은 탄소와 금속의 생지화학적 순환에 영향을 줄 뿐만 아니라 또한 금속, 방사성원소, 그리고 유기물로 오염된 지하수와 토양의 정화에 있어서 중요한 역할 가능성을 시사하고 있다. 지구의 극한 환경(예: 심해저 퇴적, 알칼리성 호수 등)에서 서식하는 철환원 박테리아를 분리하여 금속이온의 환원과 광물 형성 등의 실험에 이용하여 본 결과에 의하면, 이들 철환원 박테리아는 Fe(III), Mn(IV), Cr(VI), Co(III), and U(VI)이온 등을 환원시킬 뿐만 아니라, 자철석($Fe_3$$O_4$), 능철석($FeCO_3$), 방해석($CaCO_3$), 능망간석($MnCO_3$), 비비아나이트 [$Fe_3$($PO_4$)$_2$ .$8H_2$O], 우라니나이트(UO) 등의 광물을 형성한다. 철 환원 박테리아에 의한 광물 형성과 금속이온의 환원에 영향을 미치는 주요소는 대기의 조성, 화학 조성, 및 박테리아의 종이다. 호열성 철환원 박테리아는 철수화물과 금속이온(Co, Cr, Ni) 등을 동시에 환원시켜 금속 치환된 자철석을 합성하며, 또한 석탄회 등을 이용하여 탄산염 광물을 형성하여 대기 중의 이산화탄소를 고정하는 역할을 하기도 한다. 따라서 미생물에 의한 금속이온이 환원은 자연계에서 철과 탄소의 지화학적인 순환에 영향 미치며, 또한 미생물에 의한 자철석의 합성은 산업적으로 많은 이용가치가 있을 것으로 본다.
식물을 이용하여 토양이나 지하수 등의 매질로부터 오염물을 제거하는 기술인 phytoremediation은 지난 몇 년간 확용잠재성이 높은 환경복원기술로 많은 관심을 받아왔다. 본 연구에서는 현장조건을 모사한 그린하우스에서 이러한 phytoremediation에 의한 디젤로 오염된 토양의 정화효과를 조사하였다. 처리실험에 적합한 식물을 선정하기 위하여 인위적으로 디젤로 오염시킨 토양에서 증가에 따른 대상식물인 알팔파 옥수수, 피, 물피의 생장율 저하정도를 관측하여 내성이 강한 알팔파를 정화식물로 이용하였다. 디젤오염토양의 처리실험은 공기주입, 알팔파 파종, 공기주입과 알팔파 파종이 함께 적용된, PVC 파이프형태의 처리구에서 오염토양의 정화정도를 약 100일간 관측하였다. 칼럼 실험결과, 오염물의 제거속도에 기여하는 식물의 긍정적 효과가 관측되었으며 또한 지속적으로 토양내로 공기를 공급 하여줌으로써 Phytoremediation의 처리효율을 더욱 향상시킬 수가 있었다. 실험종료 후 각각의 처리구에서 토양 깊이별 미생물의 활성을 간접적으로 측정한 결과, 이러한 식물의 정화효과는 근권에 있는 토양미생물에게 유용한 뿌리 분비물을 지속적으로 제공함으로써 미생물의 활발한 번식 및 분해활동을 유지시켜 오염물의 분해를 더욱 촉진시키는 것으로 보인다.
본 연구는 국내에서 냉장보관창고 온도에 대한 조사를 수행하여, 온도분포를 추정하였고, 이를 미생물 위해평가의 입력변수로 활용할 수 있도록 적정 확률분포 모델을 제시하였다. 국내 냉장보관창고의 온도분포는 최저 $-3.2^{\circ}C$, 최대 $14.9^{\circ}C$, 평균 $2.55{\pm}3.55^{\circ}C$로 나타났고, $10^{\circ}C$이상 비율은 2.5%로 나타났으며, 대부분의 냉장창고 온도는 설정온도보다 높은 것으로 나타났다. 공간 위치별 온도분포는 상단(2.4~4 m) $0.8{\pm}1.69^{\circ}C$, 중단(1.5~2.4 m) $0.59{\pm}1.68^{\circ}C$, 하단(0.7~1.5 m) $0.65{\pm}1.46^{\circ}C$로 중단 온도가 가장 낮았으며, 위치별 온도차이는 최대 $1.11^{\circ}C$로 공간상에서 온도가 일정하게 유지되는 것이 아니라 어느 정도의 편차가 존재하는 것으로 나타났다. 이상의 수집된 온도자료는 @RISK 를 이용, 적합성 검정(GOF: K-S와 A-D test)을 수행하여, MRA에서 활용할 수 있는 국내 냉장창고 온도분포에 대한 가장 적합한 확률분포모델로 LogLogistic(-4.189, 5.9098, 3.2565)을 선정하였다.
산소가 고갈된 혐기성 환경의 유기물 분해 및 물질순환에서 철 환원반응의 생태/환경적 중요성에 대해 고찰하였다. 다양한 해양환경에서 유기물 분해 시 철 환원이 차지하는 중요성은 미약한 수준에서 거의 $100\%$에 이르기까지 그 범위가 극단적으로 다양하게 나타났다. 일반적으로 철 환원은 Fe(III)의 농도가 높은 곳에서 황산염 환원보다 중요한 유기물 분해 경로로 나타나, 유기물 분해에서 철 환원의 중요성은 철 환원세균이 이용 가능한 Fe(III)의 공급정도에 의해 결정되는 것으로 인식되었다. 산소공급이 미약한 연안혐기성 퇴적토 내에서 Fe(III)의 공급은: (1)조석에 의한 퇴적물 내 공극수의 교환(tidal flushing): (2)저서동물에 의한 생물교란: (3)식생의 유무에 따른 퇴적물의 산화/환원 상태의 변화 등에 의해 주로 영향을 받는 것으로 나타났다 철 환원세균에 의한 유기물 분해 및 다양한 금속원소의 전환기능을 이용한 특정 유기오염원과 금속오염원의 생물정화는 우리나라와 같이 부영양화된 연안생태환경의 개선 및 독성 유t무기 오염원의 생물정화 등 연안역의 환경친화적 관리가 절실히 요구되는 환경에서 생태/환경공학 분야의 유용한 해결수단으로 간주된다.
식물과 근권미생물을 이용해 토양 오염물질을 제거하는 rhizoremediation의 효율을 높이기 위해서는 오염물질을 제거함과 동시에 식물 생장을 촉진시키는 미생물 자원 개발이 필요하다. 본 연구에서는 중금속 및 유류 복합 오염 토양에서 서식하고 있는 옥수수와 톨페스큐의 근권으로부터 중금속(구리, 카드뮴 및 납) 내성을 가진 근권세균을 순수분리하였고, 식물 생장 촉진능, 중금속 내성능 및 디젤 분해능을 정성적으로 평가하였다. 그 결과 중금속 내성, 식물 생장 촉진 활성 및 디젤 분해능을 가진 6종의 균주를 분리하였다. 옥수수 근권에서 분리한 CuM5와 CdM2 균주는 Cupriavidus sp.로 동정되었다. 톨페스큐 근권에서 분리한 CuT6, CdT2, CdT5 및 PbT3는 각각 Fulvimonas soli, Cupriavidus sp., Novosphingonium sp. 및 Bacillus sp.로 동정되었다. Cupriavidus sp. CuM5와 CdM2는 중금속 내성과 디젤 분해능은 상대적으로 낮았으나, 식물 생장 촉진능이 상대적으로 우수하였다. 6종 중에서 디젤 분해능이 가장 우수한 균주는 Cupriavidus sp. CdT2와 Bacillus sp. PbT3이었다. 특히, Bacillus sp. PbT3는 3종의 중금속에 대해 상대적으로 우수한 내성을 가졌고 식물 생장 촉진능도 우수하였다. 본 연구에서 분리한 근권세균은 유류와 중금속 복합 오염 토양을 정화시키며 식물 생장을 촉진시키는 새로운 미생물 자원으로 활용 가능하다.
아셀렌산염(selenite, ${SeO_3}^{2-}$)을 환원시키는 세균 SE4-1을 복합 산업단지 내 위치한 오염된 하천 퇴적토로부터 분리하였고, 계통분석결과 이 균주는 Citrobacter freundii와 가장 유사하였다. 이 균주는 혐기적 환경에서 아셀렌산염을 원소상 셀레늄(elemental selenium, $Se^0$)으로 환원시켰고, 그 형상은 구형으로 확인되었다. 이러한 고체상의 침전은 셀레늄 산소음이온의 이동성을 줄이고 생물가용성을 줄여, 생태계 독성을 줄이는 잠재성을 제시해 줄 수 있을 것이다.
This study was to Investigate the consumer's understanding and purchasing foods, which were produced by organic agricultured method. The results of this study were as follows: 1. When shopping foods, the items which were deeply considered were taste of family and food safety. 2 Contaminants which were considered to be dangerous were, in decreasing order, agricultural chemical residue, food additives, environmental contaminant, and microbial hazard. Consumer's perception to agricultural chemical residue In foods was 'be much serious' and over 4.0 by Likert 5 scale. 3. Among of characteristics which was associated with organic farmming foods, taste and nutrition were considered the least important characteristic, whereas safety and good health were considered the most important characteristic. 4. The foods which were thought to be polluted by agricultural chemical residue were, in decreasing order fruit, vegetable, cereal, egg, meat, milk, and fish & shellfish. 5. The respondants' purchasing degree for organic farmming foods was 'often purchase' and 3.44 by Likert 5 scale. These result showed significant difference for age(p<0.05), for education level (p<0.05), for income(p<0.05) and for food expenditure(p<0.05), respectively 6. The type of organic farmming food which frequently purchased were, in decreasing order, vegetable. fruit, egg, cereal, milk, and meat. The reasons why consumer purchased organic farmming food were, in decreasing order, no pesticide/therbicides, no artifical fertilizer, no growth regulators, residue free, and good quality. 7. 78% of respondants Indicated that they bought organic farmming food in this year increasely or same as last year and 91.9% of respondants Indicated that they plan to buy organic farmming foods in next year.
This study was conducted to investigate the tolerance of some resistant fungal strains from soils contaminated with heavy metals. Various fungal strains were isolated from soil samples collected from studied sites which heavy metals and other pollutants have been emitted in effluents for several years. Fungi isolated belong to different genera; however, Penicillium spp. showed the most frequent species. The microbial number was remarkably higher in the control soil than contaminated soil samples collected from mining areas. $Pb^{2+}$ and $Zn^{2+}$ had the highest concentration in the polluted soils ranging from 89 - 3,521 ppm and 98 - 4,383 ppm, respectively. The minimum inhibition concentrations (MICs) of $Pb^{+2}$ and $Zn^{+2}$ showed the highest values against the fungal strains. $Ni^{+2}$ and $Co^{+2}$ were the lowest contaminants in the polluted soils with the concentration of 5 to 12.1 ppm and 1.8 to 4.8 ppm, respectively. The tested resistant strains showed the strongest inhibition for $Ni^{+2}$ and $Co^{+2}$ up to 200-400 ppm. Cadmium was the most highly toxic heavy metal for most of strains, however, 1 mM of $Cr^{3+}$, $Cu^{2+}$ and $Pb^{2+}$ accelerated the growth of Penicillium verrucosum KNU3. $Cu^{+2}$ and $Zn^{+2}$ at concentration of 1 mM did not affect the growth rate P. funiculosum KNU4. Tolerance of fungal species to heavy metals appears to be strain and origin dependent.
저칼로리 감미료로서 장내 미생물 균총을 개선한다고 알려진 대두 올리고당을 전처리 및 Steffen법을 이용하여 대두 침지액으로부터 분리, 회수하였다. 전처리로서 pH를 $3.5{\sim}4.0$으로 조절하거나, 염화칼슘을 당 대비 8%(w/w)를 첨가하여 대두 침지액 중에 함유된 단백질을 약 $25{\sim}30%$가량 제거할 수 있었다. 또한 대두 침지액에 염화칼슘과 산화칼슘 분말을 각각 당 대비 약 20%, $100{\sim}120%$씩 첨가한 후 $5^{\circ}C$에서 20분 동안 반응시키는 Steffen법을 이용하면 대두 침지액 중에 존재하는 대두 올리고당의 약 85%를 saccharate 형태로 분리, 회수할 수가 있었다. 이산화탄소로 당을 탈착시켜 회수한 결과 최종적으로 약 80%의 수율로 당을 회수할 수가 있었으며 단백질은 약 80% 정도 제거할 수가 있었다. 회수된 당액의 당조성은 대두 침지액과 유사하였다.
