Seven native and four introduced herbs namely $Thymus$$quinquecostatus$, $Chrysanthemum$$zawadskii$ var. $latilobum$, $Rosmarinus$$officinalis$, etc. were selected for analysis of the DPPH radical scavenging and anti-microbial activity of their extracts. These perennial herbs are classified as $Labiatae$ and $Compositae$ except for $Saururus$$chinensis$ and can be propagated through seedling and cuttage. These edible herbs are used as medicinal as well as ornamental plants. Their extract has electron donating ability which ranges from 69.7 to 78.7% for native herbs and 67.4 to 75.4% for introduced herbs. Native herbs have higher (3.54%) average DPPH radical scavenging than introduced herbs. In native herbs, maximum DPPH radical scavenging activity was observed in $Agastache$$rugosa$ (78.7%) followed by $Saururus$$chinensis$ while $Chamaemelum$$nobile$ showed highest activity among the introduced herbs. Many herbs viz. $Saururus$$chinensis$, $Chrysanthemum$$zawadskii$ var. $latilobum$ and $Solidago$$virga-aurea$ var. $gigantean$ showed excellent anti-microbial activity against gram positive $Enterococcus$$faecalis$, maximum (80.0%) by $Saururus$$chinensis$. Other herbs viz. $Solidago$$virga-aurea$ var. $gigantea$, $Chrysanthemum$$zawadskii$ var. $latilobum$, $Salvia$Salvia $plebeia$, $Chrysanthemum$$indicum$, $Rosmarinus$$officinalis$, $Chamaemelum$$nobile$ and $Lavandula$$stoechas$ showed anti-microbial activity against gram negative $Citrobacter$$freundii$. Especially, the inhibition of colony growth of $Citrobacter$$freundii$ was highest in the extract of $Chrysanthemum$$zawadskii$ var. $latilobum$, and $Chamaemelum$$nobile$.
Currently, urban greening projects and research are attracting attention as a way to mitigate urban heat island phenomenon. In this study, nitrogen fixative bacteria were isolated and their effects on plant growth were confirmed. First, enrichment was performed in a nitrogen-free medium to isolate the nitrogen-fixing bacteria, and the colony showing high growth in a medium with limited nitrogen source was isolated and purified. Separated bacterial isolates were reduced by more than 90% acetylene by ARA and indirectly confirmed the activity of nitrogenase by ethylene production. Cedecea sp. MK7 and Enterobacter sp. Y8 with confirmed reproducibility were selected as nitrogen fixative bacteria. Nitrogen fixing bacteria were applied to the growth of perennial rye grass, and it was found that the dry weight increased to 34.80 mg (186.60%) compared with the control with 18.65 mg dry weight. After plant growth, microbial community analysis of soil applied by bacteria showed similarity to the control group. Therefore, in this study, it is expected that the efficiency will be increased if plant growth is promoted by using nitrogen fixing bacteria in urban greenery system.
The alkaline phosphatase activity (APA) of two dam reservoirs and inflowing streams were measured monthly in 2000. During summer months in 2001, the vertical profiles of APA and related parameters were also examined in one of the reservoirs. The APA was relatively high during the summer season in the epilimnion while it was almost invariable in the hypolimnion. A small increase in APA was observed at just above the bottom. The APA fluctuation was independent of the concentration of soluble reactive phosphorus. It was assumed that APA is not indicative of the phosphorus availability status. An examination of size-fractionated samples suggested that APA in reservoirs was attached to particles larger than $0.4{\mu}m$, whereas in streams it existed in a dissolved form. There was a positive significant correlation between chlorophyll a concentration and APA in the photic zone. In the aphotic zone, APA correlated positively with the colony count of heterotrophic bacteria, but not with microscopic total bacterial counts.
Contamination levels of aerobic colony counts, coliforms and pathogenic bacteria were tested in fried rice dishes to monitor quality for risk management. The prevalence of Bacillus cereus in dishes from 8 Chinese-style restaurants and 2 institutional foodservices was 10%, and the bacteria's contamination levels was 3.47 log CFU/g. Echerichia coli, Staphylococcus aureus, and Salmonella were not detected in any of the 10 samples. However, for their aerobic colony counts and numbers of isolated coliforms, the samples were 30% and 70% over the microbial criteria, respectively, for ready-to-eat foods presented in the Korean Food Code. This suggests that fried rice dishes, although cooked with oil at high temperature, require special care. For the prediction of the growth curve of B. cereus spp. in the fried rice samples, an experiment design of 3 storage temperatures ($7^{\circ}C$, $35^{\circ}C$, $57^{\circ}C$) x 5 storage times (0 h, 2 h, 4 h, 6 h, 24 h) was applied. The sample exposed to $35^{\circ}C$ showed no B. cereus spp. at 0 h; however, there was a tendency of slow growth (1.0 log CFU/g) after 4 hours of storage and then faster growth at 6 h (3.7 log CFU/g) and 12 h (4.7 log CFU/g), showing a growth rate of 0.56 log CFU/g/hr. These results indicate that fried rice, despite being heat-treated, can become heavily contaminated with B. cereus spp. when held over 2 hours at room temperature. However, the samples stored at $7^{\circ}C$ and $57^{\circ}C$ over 24 hours were not contaminated with B. cereus. Based on these results, management guidelines for controlling B. cereus are suggested.

The main purpose of this study was to evaluate the acid resistance and antimicrobial effect of fluoride-laser combined application. Recently extracted third molars were used. $5{\times}3mm$ of the buccal and lingual specimens were exposed and incipient artificial carious lesions were formed by keeping them in the artificial cariogenic solution for 5 days. They were divided into five groups and treated with fluoride and laser according to the predetermined regimen. The acid resistance was compared between groups by chemical quantitative analysis of the calcium and phosphorous released into the test solution after single or combined application of fluoride and laser. The antimicrobial effect of each group was analyzed by counting the number of colony forming units after microbial incubation. The results from the present study can be summarized as follows; 1. Experimental groups showed lower values in calcium and phosphorous contents as well as in $CFU/m{\ell}$(colony forming units) than control group. Combined application groups showed lower values than single application groups. 2. Acid resistance and antimicrobial effect by fluoride and laser treatment were confirmed in this study. 3. Based upon the above-mentioned results of this study, it can be assumed that the use of laser-fluoride combined application may provide the child and adolescent patient population with antimicrobial effect as well as acid resistance. Further studies using various materials and experimental conditions are being encouraged.
This study was conducted to compare colony forming unit (CFU) of microorganisms in closed and open soilless culture systems for estimating the possibility for potential disease occurrence. Samples were collected at four different positions in four commercial greenhouses with closed or open soilless culture system using rock wool or coir as substrate, respectively. The distance between sampling positions was 3 cm starting from the substrate and the surface area of each position was $25cm^2$. The CFU of fungi was significantly higher in the open system, while that of bacteria was not significantly different but showed relatively lower in the closed system. Samples collected at the plastic surface of the substrates where little environmental effects occurred from drainage showed lower CFU than any other positions. The principal component analysis showed that samples collected on the drainage pathway highly affected the changes in microbial population in the greenhouse. These results indicated that greenhouses with closed soilless culture are expected to have more advantageous conditions for restraining the microbial growth, resulting in the lower potential of disease occurence in greenhouse ecosystem.
As Standards and Specifications of the Saengsik-classes has been established since 2005 by KFDA. The microbial Standards and Specifications of the Saengsik-classes is as follows; no detection in Escherichia coli, colony forming unit less then 1,000/g in Bacillus cereus, colony forming unit less then 100/g in Clostridium perfringens respectively. Contamination levels of Total aerobic bacteria, Escherichia coli, Bacillus cereus and Clostridium perfringens in Saengsik-classes were monitored. Total aerobic bacteria counts in Saengsik-classes was $1{\times}10^1{\sim}5.3{\times}10^7cfu/g$, for Bacillus cereus $1{\times}10^2{\sim}9{\times}10^2cfu/g$, for Clostridium perfringens $1{\times}10^1cfu/g$. Escherichia coli, was not isolated from all Saengsik-classes. Thess results will provide information for introduction of HACCP system to ensure microbial safety of Saengsik-classes.

The purpose of this study was to determine the level of toothbrush bacterial growth, whether the dryness of the toothbrush head differs depending on the number of holes in the head, and to use these results as a reference for future toothbrush design. Two-millimeter holes were created on the head of the toothbrushes in groups of three, one, or zero holes. We made the solution with Streptococcus mutans, and the toothbrushes were placed in the solution and agitated. The toothbrushes were shaken to remove moisture and allowed to air-dry. The toothbrush heads were swabbed with saline and then placed in two inoculation groups. The first group was inoculated with a 102 dilution of the S. mutans culture and the second was inoculated with the original culture. After incubation, bacterial colony numbers were measured. The number of holes on the toothbrush head correlated with a decrease in number of cultured bacterial colonies. Our model of a toothbrush head with holes indicated that these holes in the toothbrush head were effective in reducing the level of microbial contamination and that a greater number of holes creates an improved toothbrush sanitation effect. The average number of colonies on the head of toothbrush by number of holes was high, followed by the number of holes 0, 1 and 3, and the average number of colony among toothbrush heads was same. The use of a toothbrush with holes between the toothbrush head indicates that it is effective in reducing the level of microbial contamination between the toothbrush head and toothbrush and the higher the number of holes, the better the effect.

This study investigates characteristics of diesel degradation and variations of microbial community with the soil enrichment cultures. The cultures has yellow(YE-5) and transparent color's(WH-5) colony on solid plate medium. The bacillus type of YE-5 and WH-5 cultures showed diesel degradation at the rate of 99.07mg-Diesel/$L{\cdot}day$ and 57.82mg-Diesel/$L{\cdot}day$ in the presence of 1%(v/v) initial diesel concentration. Diesel degradation was 1.7 times faster than WH-5 culture. YE-5 or WH-5 culture could degrade a wide range of diesel compounds from $C_8$ to $C_24$. Microbial community analysis by PCR-DGGE technique shows that Psedomonas, Klebsiella, Escherichia and Stenotrophomonas as proteobacteria take role on the diesel degradation. uncultured Senotrophomonas sp. was only detected with YE-5 culture. It is concluded that proper combination of the microorganism should be present to stimulate the degradation of diesel and further studies are recommended for the effect of uncultured Senotrophomonas sp. or Escherichia hermannii on diesel degradation.
To establish basic techniques for protoplast fusion of Coriolus versicolor several factors affecting protoplast formation and regeneration were investigated. Protoplast isolation was at maximum with 2.5-day cultured mycelia of C. versicolor treated with the combination of two enzymes, Novozym 234 (10 mg/ml) and cellulase Onozuka R-10 (15 mg/ml), for 3-4.5 hours at $30^{\circ}C.$ As an osmotic stabilizer for stabilizing the protoplast, 0.6 M sucrose was the best for formation and regeneration of the protoplast from the mycelia of the fungus and the regeneration frequency was 3.48%. Protoplast fusion was made by a modified method of Peberdy using PEG (M.W. 4,000). The fusion frequency between two mutants of C. versicolor was 1.86% and the fusion products showed differences in growth rate and colony morphology.
